Tuber borchii Shapes the Ectomycorrhizosphere Microbial Communities of Corylus avellana

  • In this study, eight-month-old ectomycorrhizae of Tuber borchii with Corylus avellana were synthesized to explore the influence of T. borchii colonization on the soil properties and the microbial communities associated with C. avellana during the early symbiotic stage. The results showed that the bacterial richness and diversity in the ectomycorrhizae were significantly higher than those in the control roots, whereas the fungal diversity was not changed in response to T. borchii colonization. Tuber was the dominant taxon (82.97%) in ectomycorrhizae. Some pathogenic fungi, including Ilyonectria and Podospora, and other competitive mycorrhizal fungi, such as Hymenochaete, had significantly lower abundance in the T. borchii inoculation treatment. It was found that the ectomycorrhizae of C. avellana contained some more abundant bacterial genera (e.g., Rhizobium, Pedomicrobium, Ilumatobacter, Streptomyces, and Geobacillus) and fungal genera (e.g., Trechispora and Humicola) than the control roots. The properties of rhizosphere soils were also changed by T. borchii colonization, like available nitrogen, available phosphorus and exchangeable magnesium, which indicated a feedback effect of mycorrhizal synthesis on soil properties. Overall, this work highlighted the interactions between the symbionts and the microbes present in the host, which shed light on our understanding of the ecological functions of T. borchii and facilitate its commercial cultivation.

  • Experimental infection and histopathological study using a deleterious aquatic fungus (Saprolegnia diclina type I) were carried out in the eels (Anguilla japonica) to know what pathological changes the fungus would produce in the affected eels. The eels in group A and B which were treated with fish net were not susceptible to experimental infection. In group C which the cuticle was scraped by artificial treatment, the aquatic fungi were invading all the eel bodies including the desquamated lesions. Histopathologically, the affected eels were markedly reduced in number of goblet cells and showed moderate damage of the clavate cells. Nodular formation involving ecchymotic hemorrhage was seen in some affected areas. The eels survived to 25 days after experimental infection.

The Antimicrobial, Antioxidant, and Tyrosinase Inhibitory Activities of Solvent Extracts of Asterina pectinifera and Asterias amurensis (별 불가사리(Asterina pectinifera) 및 아므르 불가사리 (Asterias amurensis)추출물의 항균, 항산화 활성 및 미백 효과)

  • This study examined the antimicrobial, antioxidant, and tyrosinase inhibitory activities of bioactive compounds extracted from two starfish, Asterina pectinifera and Asterias amurensis, using solvent extraction after $Protamex^{TM}$ hydrolysis. Methanol and acetone fractions collected by stepwise extraction from specimens were subjected to silica gel column chromatography (SGCC) (200 mesh and 400 mesh), followed by high-performance liquid chromatography (HPLC). Two fractions (7:3 and 5:5 chloroform : methanol ratio, v/v) eluted using silica gel column chromatography from the two starfishes showed higher antimicrobial activity against Propionibacterium acnes and dermatophyte fungi (Epidermophyton floccosum, Microsporum audouinii, Trichophyton ferrugineum, Trichophyton mentagrophytes, and Trichophyton rubrum), antioxidant activity ($EDA_{50}$, mg/mL), and tyrosinase inhibitory activity compared to the other fractions. The final fractions obtained from Asterina pectinifera (RT 7.53, 8.93, and 10.48 min) and Asterias amurensis (RT 5.02 min) by SGCC (400 mesh) and HPLC from two SGCC fractions (200 mesh) showed 8.94 and 15.59 mg/mL antioxidant activity ($EDA_{50}$) and 46.89 and 40.19 % tyrosinase inhibitory activity, respectively. Extracts from starfishes are potential cosmetic basic material.

Inhibitory Effects of Seaweed Extracts on Growth of Malassezia furfur and Malassezia restricta

  • Fifty seven species of common seaweed from the coast of Korea were screened for antifungal activity against Malassezia species. Seaweeds as a source of bioactive compounds are able to produce a great variety of secondary metabolites with different activities. There are numerous reports on the biological activities of seaweeds against human pathogens, fungi, and yeasts, but only few contain data regarding inhibitory effects against Malassezia sp., a major cause of dandruff and seborrheic dermatitis. To help address this paucity of information, this work was carried out to examine the antifungal effects of seaweed extracts against M. furfur and M. restricta. Of the fifty seven species of marine algae screened for their potential antifungal activity, only 17 species (29.8%) exhibited inhibitory activity. In agar disc diffusion method, the ether extracts of Corallina pilulifera, Enteromorpha linza, Laminaria japonica, Symphyocladia latiuscula and Ulva sp. showed strong antifungal activity. To identify major constituents in seaweed extracts, four selected extracts were analyzed on' a GC-MS equipped with a flame ionization detector, and compared to spectral data from databases WILEY229.LIB and NIST107.LIB. Most constituents in seaweed extracts are fatty acid-related compounds. When we evaluated any acute toxicity, the ether extracts of the selected four species were not toxic in mice. According to these results, it can be suggested that these seaweed extracts are valuable for the development of therapeutic agents in treating dandruff and seborrheic dermatitis. Further investigations to determine its bioactive compound(s) are currently in progress.

Occurrence of Fungal Species and Mycotoxins from Decayed Sugarcane (Saccharrum officinarum) in Egypt

  • Seventy-three fungal species belonging to forty-three genera were isolated from 40 samples of Saccharrum officinarum (collected from Naage-Hamadi canal in Qena Governorate, Egypt). Aspergillus, Trichoderma, Mucor and Pythium were the most common genera on the two isolation media. The dominant species of Aspergillus were A. niger, A. flavus, A. ustus, A. terreus and A. wentii. Some species were dominant on 40 g/l sucrose such as Aspergillus niger, A. flavus, Emericella nidulans, Trichoderma viride, Torula herbarum and Mamaria echinoeotryoides, while the dominant species on 10 g/l glucose were Mucor circinelloides, Aspergillus niger, Torula herbarum and Trichoderma viride. Mycotoxins including aflatoxins $B_1,\;B_2,\;G_1\;and\;G_2$, zearalenone and diacetoxyscirpenol were detected in the examined samples of Saccharrum officinarum. The mycelial growth of A. flavus, A. niger, Fusarium moniliforme and Torula herbarum decreased with the increase in Dimethoate concentrations, although 25 ppm was less effective than the higher levels of the insecticide ($75{\sim}200\;ppm$). Dimethoate stimulated the activity of Go-Tin A. niger, F. moniliforme and T. harbarum, while the Go-T activity was inhibited in A. flavus with the Dimethoate treatments.

Toxicity of agricultural chemicals on Lymnaea viridis the intermediate host of Fasciola hepatica (간질(肝蛭)의 중간숙주(中間宿主)인 애기물달팽이에 대한 몇가지 농약(農藥)의 독성시험(毒性試驗))

  • In the present study, the effects of 4 agricultural chemicals commonly used in this conuntry were experimentally assessed on Lymnaea viridis the intermediate host of Fasciola hepatica, which in non-target organism of these chemicals. The major habitat of the snail is rice paddies in Korea and many agricultural chemicals are used for weed, fungi or insect control in rice paddies and there is a general concern that certain levels of these chemicals could reach the aquatic ecosystem and possible alter the snail life. Agricultural chemicals used in this study included two herbicides, an insecticide and a fungicide. The tenth generation of laboratory reared snails were selected and exposed to the varying concentrations(0-100 ppm) of these chemicals. As concentrations and time of exposure increase, the per cent mortality increases(p<0.01). $LC_{50}$(lethal concentration for 50% mortality) values of these chemicals on snail after 96-hour exposure were variable; iprobenfos showed the highest acute toxicity(12.6 ppm), while carbofuran showed the lowest acute toxicity(74.5ppm). Sublethal concentrations of chemicals after 96-hour exposure were also variable ; bentazone showed the highest chronic toxicity(0.81ppm), while carbofuran showed the lowest chronic toxicity(5.04 ppm).

  • Formalin, 37% formaldehyde, has been employed as a chemical for controlling ectoparasites and aquatic fungi responsible for infectious fish of diseases in hatcheries and culture facilities (Roberts, 1978; Schnick, 1991; Rach et al., 1997). Regarding the commercial usage of formaldehyde in the aquaculture industry, the U.S. Food and Drug Administration (FDA) approved three commercial products as parasiticides in a species-specific manner: Paracide-F and Formalin-F for bluegill, catfish, largemouth bass, salmon, and trout and Parasite-S for all finfish (FDA, 1998). Withdrawal time for these products was legally zero when used as permitted under the regulations. With the increased production of cultured fish in Korea, such as olive Hounder Paralichthys olivaceus and black rockfish Sebastes schlegeli, application of formalin to diseased fish has become more frequent. Moreover, there is still some concern about environmental exposures caused by effluents from fish culture facilities. The purpose of the present study was to evaluate residues in fish resulting from therapeutic usage of formalin in the aquaculture industry and to document the rate of disappearance of formaldehyde in seawater treated with formalin. Evaluation of Fish Acute Toxicity and Preliminary Risk Assessment of Plant Extracts, Sophora, Neem and Derris (유기농업자재 고삼, 님 및 데리스 추출물의 어류에 대한 독성과 위해성 평가)

  • Promoting the organic farming, much of the plant extracts used for controlling pests and fungi have been imported from China, India and Myanmar. But, it is so worrisome that aquatic animals such as muddy loach inhabiting in paddy field and common carps in river exposed to the pests and fungi likely be harmed. This study was conducted in order to evaluate the risks of aquatic animals influenced by the three plant extracts, i.e. Sophora flavescens, Azadirachta indica and Derris elliptica. The toxicities of common carp (Cyprinus Carpio), muddy loach (Misgurnus anguillicaudatus) and PEC (Predicted environmental concentration) exposed to the three plant extracts were estimated by the typical spray volume method. Risks were determined by the toxicity value as 48-hr $LC_{50}$ (Lethal concentration, median) or NOEC (No observed effect concentration) into PEC. 48-hr $LC_{50}$ of Common carp and NOEC by Sophora flavescens extracts was 7.9 and 6.2 mg/L, 26.8 and 21.8 mg/L by Azadirachta indica extracts and 47.0 and < 24.0 mg/L by Derris elliptica extracts, respectively. 48-hr $LC_{50}$ of Muddy loach and NOEC by Sophora flavescens extracts was 16.9 and 10.0 mg/L, 35.6 and 30.0 mg/L by Azadirachta indica extracts, and 73.9 and < 40 mg/L by Derris elliptica extracts, respectively. Therefore, acute toxicities of the three plant extracts for aquatic animals were proved to be very low level. PEC of Sophora flavescens extracts in paddy, drainage and river water was 68.0~3.0, 11.33~0.50 and 3.0~0.0018 mg/L, respectively. TER of Sophora flavescens extracts in the three water was 0.2~5.6, 1.5~33.8 and 2.6~4388.9, respectively. PEC of Azadirachta indica extracts in paddy, drainage and river water was 90.9~1.2, 15.2~0.2 and 4.8~0.00075 mg/L, respectively. TER of Azadirachta indica extracts in the three water was 0.4~29.7, 2.3~178.0 and 4.5~35733.3, respectively. PEC of Derris elliptica extracts in river water was 0.0063 mg/L. TER of Derris elliptica extracts in river water was 5222~15667.

Structures and some Properties of the Antimicrobial Compounds in the Red Alga, Symphyocladia latiuscula (참보라색우무에서 추출한 항균물질의 구조 및 특성)

  • Three antimicrobial compounds (SL-l, SL-2 and SL-3) were isolated and identified from the marine red alga, Symphyocladia latiuscula. In addition, their biological functionalities such as cytotoxicity and desmutagenic activity were investigated. From the cryophyllized S. JatiuscuJa, SL-l, SL-2 and SL-3 were purified by solvent extractions and HPLC.SL-2 was crystallized in benzene-diethyl ether solvent. On the EI-MS spectra, it was found that they had three bromines in their structure which showed typical signal strength ratios at $M^+, [M+2]^+, [M+4]^+, [M+6]^+ (13: 38: 37: 12)$. $SL-l$ was identified as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol ($C_8H_7Br_3O_3, MW=374$) by NMR and MS spectra. SL-2 was assigned as 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether ($C_8H_7Br_3O_3, MW=388$) and confirmed by X-ray crystallographic analysis. SL-3 was presumed as an isomer of SL-2. Methanol extract of the S. latiuscula showed antimicrobial activities against all strains tested (bacteria, 15 strains; yeasts, 17 strains; fungi, 4 strains), much or less. The strongest inhibition activity of the methanol extract was to the Vibrio mimicus ($50 {\mu}g/ml$) and V. vulnificus ($50 {\mu}g/ml$). The mice injected intraperitoneally with 3 mg of SL-l and 5 mg of 5L-2 showed no acute toxicity response. SL-2 showed higher desmutagenic activity than SL-l against PhIP and MeIQx.

Effect of Plant Extracted Essential Oil on Antimicrobial Activity for Fish Phathogens (식물 추출 정유가 어류의 병인체에 미치는 항미생물 효과)

  • Antimicrobial activity of the essential oil extracted from plants Artemisia princeps var. orientalis, Thuja orientalis and Chamaecyparis obtusa were tested against selected pathogenic bacteria and fungi of fish. At the concentrations above 500ppm, ingibitory effect of the oil of A. princeps var. orientalis was at its peak against Aeromonas hydrophila ATCC 14715, A. hydrophila CF-2, A. salmonicida ATCC 14174 and A. salmonicida EL-1 but the bacteria Edwardsiella tarda KBF-1, Vibrio anguillarum ATCC19264, V. ordalii ATCC33509 and Streptococcus sp. were insensitive. Likewise, the oil extract of T. orientalis showed the highest inhibitory activity against V. ordalii ATCC33509, E. tarda ECK-1, and E. tarda KBF-1 at 300ppm; however the activity was highest at 500ppm or A. hydrophila ATCC14715, A. hydrophila CF-2, A. salomonicida ATCC14174, A. salmonicida EL-1 and Streptococcus sp. SF-1. With increasing dose of C. obtusa oil, the inhibitory activity became more and more effective against A. hydrophila CF-2, A. salomonicida ATCC14174, E. tarda ECK-1 and Streptococcus sp. SF-1, but A. hydrophila ATCC14174, A. salmonicida EL-1, E. tarda KBF-1, V. anguillarum ATCC19264, V. ardalii ATCC33509 and gram positive bacteria (Streptococcus sp.) were somewhat resistant. A. princeps var. orientalis, T. orientalis and C. obtusa were also tested against Saprolegnia sp. at the oil concentrations of 10, 100, 500, 1,000, 1,500 and 2,000ppm. The inhibitory effect of the oil on the inhibit the mycelial growth of Saprolegnia sp. at 10ppm and completely inhibited at over 500ppm.
