Zoosporic Fungi Isolated From Four Egyptian Lakes and the Uptake of Radioactive Waste

  • Aquatic fungi from four brackish water lakes; Edku, Burullus and Manzala lakes which are located at the northern region of Egypt and Qarun lake that located in El-Fayoum city are reported in this manuscript. Twenty-nine fungal species which belong to 19 genera of aquatic fungi were recovered from water samples collected from the studied lakes. The most frequently isolated fungal species were Chytridium conferrop, Allomyces throughout and Rhizoclosmatium globosum. Thraustochytrium amoeboidum and Leptolegniella exoosporus have a moderately occurrence frequency. The maximum fungal count of recovered aquatic fungi was recorded in Burrullus lake followed by EdKu, Manzala and Qarun lakes. This study was extended to test the ability of six selected aquatic fungi(Brevilegniella keratinophila, Blastocladiella cystogena, Chytridium conferrop, Entophlyctis variabilis, Schizochytrium mangrovei and Thraustochytrium rosin), to uptake the radionuclide from their culture medium as a step to biologically treat the waste water or solution with radio-cesium and radio-cobalt. Fifty seven of Cs-137 and 35% of Co-60 could be removed from liquid waste by the selected aquatic fungi.

New Records of Three Aquatic Fungi Isolated from Freshwater in Samcheok and Yeongju, Korea (강원 삼척 및 경북 영주의 담수지역에서 발굴된 수생균류 3종의 국내 최초 보고)

  • Three aquatic fungi were isolated from samples of freshwater-deposited plant litter and foam collected in Samcheok, Gangwon-do, and Yeongju, Gyeongsangbuk-do, Korea. Based on their morphological characteristics and a phylogenetic analysis of the internal transcribed spacer (ITS) rDNA region, the three isolates NNIBRFG329, NNIBRFG339, and NNIBRFG19 were confirmed as aquatic fungi: Articulospora tetracladia, Margaritispora aquatica, and Aquanectria penicillioides. These species were known as aquatic fungi but neither species has been previously reported in Korea.

Diversity of Endophytic Fungal Strains from Jeju Aquatic Plants (제주 수생식물에서 분리한 내생균류의 다양성)

  • Endophytic fungi are present in host plants and contribute to resistance to biotic and abiotic stress. Aquatic plants are living in aquatic environment such as saltwater or freshwater and exposed more water stress than other land plants. In this study, we investigated 4 wetlands in Jeju and collected 11 aquatic plants. Exogenous microbes were removed by preprocessing of plants and endophytic fungal strains were isolated from the plants. We isolated 126 fungal strains from Namsaengi-pond, 22 fungal strains from Sujangdong-marsh, 44 fungal strains from Yongsu-reservoir and 32 fungal strains from Gangjeongcheon. The fungal strains were identified using internal transcribed spacer (ITS) region and analyzed the phylogeny and diversity. Endophytic fungi isolated from plants of Namsaengi-pond were classified to 30 genera, 19 families, 12 orders, 7 classes and 4 phyla. Endophytic fungi of Sujangdong-marsh were classified to 11 genera, 11 families, 6 orders, 5 classes and 4 phyla. Endophytic fungi of Yongsu-reservoir were classified to 13 genera, 12 families, 7 orders, 5 classes and 4 phyla. Endophytic fungi isolated from Gangjeongcheon were classified to 9 genera, 7 families, 5 orders, 2 classes and 1 phyla. Overall, they were divided 40 genera and Alternaria, Colletotrichum and Fusarium were isolated from 4 sites in common. By investigating the endophytic fungi in aquatic plants, it is for baseline data that determination of diversity and the ecological distribution of endophytic fungi.

Evaluation of Extracellular Enzyme Activity of Fungi from Freshwater Environment in South Korea (담수환경에서 분리한 곰팡이의 세포외분해효소 활성 탐색)

  • This study aimed to isolate and characterize fungi from freshwater environments in South Korea and evaluate their extracellular enzyme activities. Fungal strains were collected from various freshwater sources and identified using phylogenetic analysis. The isolated fungi included known aquatic hyphomycetes and previously unreported species. Extracellular enzyme, including those of protease, amylase, lipase, cellulase, laccase, and chitinase, activities were evaluated. Among the isolated strains, several showed high enzyme activity, suggesting their potential role in organic matter decomposition in freshwater ecosystems. This research expands our knowledge of the diversity and enzyme activities of the fungi in freshwater environments, contributing to our understanding of their ecological roles.

Mycota of Well Waters in Assiut, Egypt

  • The distribution and occurrence of aquatic zoosporic and terrestrial fungi were investigated in 21 well waters in Assiut governorate, Egypt. Using a zoospore capture technique, 923 colonies of aquatic freshwater fungi were recovered from well waters, out of which 811 colonies reached sexual maturity. These colonies were assigned to 23 species which belong to 11 genera. The most common genera were Achlya, Saprolegnia and Dictyuchus. Using two types of media, 35 species in addition to 2 varieties of terrestrial fungi which belong to 18 genera were also recovered. The most frequent glucophilic genera (recovered on glucose-Czapek's agar at $28^{\circ}C$) were Aspergillus, Penicillium and Fusarium. The results obtained on cellulose-Czapek's agar at $2^{\circ}C$ were basically similar to those on glucose agar and the most frequent genera were Aspergillus, Penicillium and Fusarium followed by Chaetomium and Cephalosporium.

Diversity of Endophytic Fungi Associated with the Roots of Four Aquatic Plants Inhabiting Two Wetlands in Korea

  • A total of 4 aquatic plants, Eleocharis kuroguwai Ohwi, Hydrocharis dubia Backer, Salvinia natans All., and Zizania latifolia Turcz., were sampled from representative two wetlands of South Korea. A total of 38 endophytic fungal strains were isolated from aquatic plants native to the Daepyeong wetland, and 27 strains were isolated from the Jilnal wetland. The internal transcribed spacer regions of fungal isolates were sequenced and a phylogenetic analysis was performed. In addition, endophytic fungal diversity from each wetland and host plant species was deduced. A total of 25 fungal genera were purely isolated, and 16 fungal genera were isolated from each of the two wetlands. Commonly isolated genera from both wetlands were Aspergillus, Cladosporium, Clonostachys, Fusarium, Leptosphaeria, Penicillium, and Talaromyces. This study revealed that fungal diversity varied with environmental conditions and by host plant in representative two wetlands.

Gibberellins Production and Identification of Endophytic Fungi Isolated from Aquatic Plant in Fresh Water (담수에 자생하는 수생식물에서 분리된 내생균류의 지베렐린 생산과 동정)

  • Aquatic plant Hydrocharis dubia (Blume) Backer was collected from the Dalsung wetland in Daegu. Sixteen endophytic fungi with different colony morphologies were isolated from the roots of aquatic plants. Waito-c rice (WR) seedlings were treated with fungal culture filtrates (FCF) for screening plant growth-promoting activity. In the results, HD1008 strain isolated from aquatic plant showed highest plant growth-promoting activity. The FCF of HD1008 strain was analyzed using gas chromatography mass spectrometry (GC/MS) with selected ion monitoring (SIM). Analysis of the FCF of HD1008 strain found that it contained gibberellins (GA) ($GA_1$, 1.2 ng/100 mL; $GA_4$, 5 ng/100 mL). Phylogenetic tree of HD1008 strain was constructed by partial internal transcribed spacer (ITS) region and partial beta-tubulin gene sequences. Therefore, we describe HD1008 strain as a new gibberellin-producing Penicillium trzebinskii based on morphological and molecular characteristics.

New Record of three Penicillium species isolated from the Freshwater Environment in Korea

  • In this study, we aimed to observe the diversity of aquatic fungi by collecting deposits of soil, plants, and plant litter from streams and rivers. Three Penicillium strains were isolated from soil in streams. Based on the morphological characteristics and phylogenetic analysis using DNA sequences of the internal transcribed spacer, β-tubulin and calmodulin genes, the isolates were identified as Penicillium guanacastense, P. saturniforme, and P. scabrosum. These three fungi have not yet been reported in Korea.

  • Three endophytic fungal strains were isolated from aquatic plants collected in Jeju. The fungi were identified by sequencing and phylogenetically analyzing their internal transcribed spacer (ITS) regions. Consequently, the three strains NNIBRFG2982, NNIBRFG2984 and NNIBRFG 3040 were confirmed as the fungal species Acremonium tubakii, Colletotrichum cliviae and Cylindrocarpon pauciseptatum, respectively, all of which are new to Korea.

Microbiological Investigation of the Frozen-Raw Sliced Fishes for Sushi Manufacturing (생선초밥용 냉동횟감의 미생물학적 조사)

  • This study assessed the contamination levels of total aerobic bacteria, fungi, coliforms, Escherichia coli, and Staphylococcus aureus, and qualitative analysis of Bacillus cereus, Salmonella spp., Listeria spp., and Vibrio spp. in four frozen-raw sliced fishes (cuttlefish, flatfish, salmon, and shrimp) for sushi production. The total aerobic bacteria, fungi, and coliforms were 2.95-3.38, 1.96-2.88, and 0.92-1.29 log CFU/g, respectively. In particular, shrimp was highly contaminated with total aerobic bacteria (3.38 log CFU/g) and fungi (2.88 log CFU/g). Over 3 log CFU/g of total aerobic bacteria was also detected in cuttlefish, flatfish, and salmon. Less than 1-2 log CFU/g of E. coli was detected in all frozen samples. S. aureus was detected at 2.25-3.13 log CFU/g in most samples. B. cereus was qualitatively detected at 25% in most samples, except for salmon (0%). Salmonella spp., L. monocytogenes, and Vibrio spp. were qualitatively detected at 25-50% of all four samples. The microbial contamination levels determined in the current study may be potentially used as basic data to perform microbial risk assessments of frozen-raw sliced fishes.
