Influence of Nitrogen, Phosphorous, Potassium and Sucrose on the Shoot and Bulb Growth of Allium victorialis var. platyphyllum (산마늘의 신초와 구근생장에 미치는 질소, 인, 칼륨 및 당의 영향)

  • Effects of nitrogen (N), phosphorous (P) and potassium (K) on the shoot and bulb growth of wild garlic ($Allium$ $victorialis$ var. $platyphyllum$) were studied by adopting $in$ $vitro$ culture. These macronutrients influenced the growth of both the shoot and bulb of garlic depending upon their application doses. A minimum of 3% potassium nitrate ($KNO_3$) as a source of nitrogen was found to be critical for shoot elongation while higher concentrations were inhibitory. Garlic bulb growth was profuse on the usual $KNO_3$ strength and sucrose (7%), followed by $KNO_3$ (9.4 mM) supplement. On providing 41.22 mM ammonium nitrate ($NH_4NO_3$) as nitrogen source highest shoot growth was observed while 82.45 mM $NH_4NO_3$ as a source of nitrogen supported high bulb growth. With regard to potassium a good shoot growth was observed in medium that contained 0.31 mM $KH_2PO_4$ and 3% sucrose, while bulb growth was high on 2.5 mM $KH_2PO_4$ and 7% sucrose. These experiments may thus direct the development of excellent growth conditions for the commercial production of edible wild garlic.

  • Water is an essential resource for humans, but fresh water becomes scarce due to population growth and contamination of limited resources. Membrane technology has been widely used for water treatment, and forward osmosis is a process which does not need high hydraulic pressure for the operation. However, there are needs for (1) development of novel draw solutes causing low internal concentration polarization and reverse salt flux for high water flux, and (2) development of economic recovery method of the draw solutes in the diluted draw solution. Previous researches on the draw solute include $NaHCO_3$ which can be regenerated by about $60^{\circ}C$ heating, sucrose which can make potable water without additional process, and magnetic nanoparticles which can be regenerated by external magnetic field. Using the principles of forward osmosis process, sea water desalination, wastewater treatment, refinement of proteins, energy generation using pressure retarded osmosis process, preparation of diluted fertilizer, and growing algae for biofuel can be conducted. This paper summarizes characteristics of ideal draw solutes, recovery method of the draw solutes, and various application examples.

Simultaneous Removal of Phenol and Hexavalent Chromium by Rhodococcus sp. CP01 (Rhodococcus sp. CP01에 의한 페놀과 6가 크롬이온의 동시 제거)

  • Simultaneous reduction of Cr(VI) and degradation of phenol was observed in batch and bench-scale continuous stirred tank reactors using Rhodococcus sp. CP01 isolated from leachate. The strain CP01, which was capable of utilizing phenol as a sole source of carbon and energy, completely reduced added hexavalent chromium (0.25 mM) to its trivalent form during 60 hr batch assay under optimal conditions (pH 7.0 and 1,000 mg/L of phenol concentration). The rates of Cr(VI) reduction and phenol degradation were estimated as 4.17 $\mu$M Cr(VI) and 38.4 mg phenol.$L^{-1}{\cdot}hr^{-1}$, respectively. The continuous culture experiment was conducted for 46 days using synthetic feed containing different levels of chromate (0.0625 to 0.25 mM) and phenol(1,000 to 4,000 mg/L). With a hydraulic retention time of 100 hr, Cr(VI) reduction efficiency was mostly 100% for influent Cr(VI) and phenol concentrations of 0.125 mM and 3,000 mg/L, respectively. During quasi-steady-state operation, specific rate of Cr(VI) reduction was calculated as 0.34 mg Cr(VI).g $protein^{-1}{\cdot}hr^{-1}$ which was comparable to reported values obtained by using glucose as growth substrate. The results suggest the potential application of biological treatment for detoxification of wastewater contaminated simultaneously with Cr(VI) and pheonol.

Polymerase Chain Reaction for the Rapid Detection of Listeria monocytogenes in Foods Using HlyA Gene Primers (HlyA유전자 Primer를 이용한 PCR에 의한 식품으로부터 Listeria monocytogenes의 신속 검출 방법)

  • The study was conducted to develop a rapid method for the detection of Listeria monocytogenes in foods via polymerase chain reaction (PCR) technique using hemolysin gene (hlyA) primers. Specificity and sensitivity of PCR, optimal conditions for PCR and application of hlyA gene primers for the detection of L. monocytogenes from milk and beef were investigeted. Each of the 20 L. monocytogenes strains gave a single 713 bp band, but other Listeria sup. and other bacteria did not show any bands. As few as 1 pg of L. monocytogenes DNA or 2.4$\times$10$^4$L. monocytogenes cells could be detected with hlyA gene primers. PCR product was most improved at 20~30 cycle in terms of removal of tailing and sensitivity. Also, the sensitivity was significantly improved by the further 10~15 cycle after 20 cycle PCR amplication. Milk (10 mL) and beef (10 g) samples were inoculated with L. monocytogenes at the concentrations ranging from 0 to 10$^{7}$ CFU/mL or g to determine the best sensitivity of PCR for the rapid detection of L. monocytogenes. PCR assay could detect 2 cells in milk with repeating PCR amplication and 2.6$\times$10$^2$cells in beef sample after 24 hr enrichment growth at 35$^{\circ}C$ in LEB.

  • Molten salt electrorefining process achieves uranium deposits at cathode using an electrochemical processing of spent nuclear fuel. In order to recover pure uranium from cathode deposit containing about 30wt% salt, the adhered salt should be removed by cathode process (CP). The CP has been regarded as one of the bottle-neck of the pyroprocess as the large amount of uranium is treated in this step and the operation parameters are crucial to determine the final purity of the product. Currently, related research activities are mainly based on experiments consequently it is hard to observe processing variables such as temperature, pressure and salt gas behavior during the operation of the cathode process. Hence, in this study operation procedure of cathode process is numerically described by using appropriate mathematical model. The key parameters of this research are the amount of evaporation at the distillation part, diffusion coefficient of gas phase salt in cathode processor and phase change rate at condensation part. Each of these conditions were composed by Hertz-Langmuir equation, Chapman-Enskog theory, and interphase mass flow application in ANSYS-CFX. And physical properties of salt were taken from the data base in HSC Chemistry. In this study, calculation results on the salt gas behavior and optimal operating condition are discussed. The numerical analysis results could be used to closely understand the physical phenomenon during CP and for further scale up to commercial level.

A Study on the Manufacturing Characteristics and Field Applicability of Engineering-scale Bentonite Buffer Block in a High-level Nuclear Waste Repository (고준위폐기물처분장 내 공학규모의 균질 완충재 블록 성형특성 및 현장적용성 분석)

  • The objective of this study is to propose a new methodology to fabricate a reliable engineering-scale buffer block, which shows homogeneous and uniform distribution in buffer block density, for in-situ experiments. In this study, for the first time in Korea, floating die press and CIP (Cold Isostatic Press) are applied for the manufacture of an engineering-scale bentonite buffer. The optimized condition and field applicability are also evaluated with respect to the method of manufacturing the buffer blocks. It is found that the standard deviation of the densities obtained decreases noticeably and that the average dry density increases slightly. In addition, buffer size is reduced by about 5% at the same time. Through the test production, it is indicated that the stress release phenomenon decreases after the application of the CIP method, which leads to a reduction in crack generation on the surface of the buffer blocks over time. Therefore, it is confirmed that the production of homogeneous buffer blocks on industrial scale is possible using the method suggested in this study, and that the produced blocks also meet the design conditions for dry density of buffer blocks in the AKRS (Advanced Korea Reference Disposal System of HLW).

Dissipation characteristics of mandipropamid and thiamethoxam for establishment of pre-harvest residue limits in lettuce (상추의 생산단계 잔류허용기준 설정을 위한 농약 Mandipropamid 및 Thiamethoxam의 잔류소실특성 연구)

  • The dissipation characteristics and kinetics of fungicide mandipropamid and insecticide thiamethoxam in lettuce under greenhouse conditions were investigated at three different lettuce-growing fields for estimating the pre-harvest residue limits (PHRLs). The analytical methods were fully validated for the quantitation of pesticide residues using High-Performance Liquid Chromatography-Photo Diode Array detector or Ultraviolet-Visible Detector and applied to real samples. The lettuces suitable for shipment were harvested during 10 days including pre-harvest interval after treatment at the recommended dose by safe-use guidelines. The initial mean residues in different fields were 6.68-17.87 and 4.96-8.31 mg/kg for mandipropamid and thiamethoxam, respectively, which decreased to 16-54 and 14-44% in 10 days. The clothianidin, a metabolite of thiamethoxam, was detected in <0.02 to 0.37 mg/kg. The dissipation of both pesticides followed first-order kinetics over a period of 10 days after application. Based on the residue data, the mean dissipation rate constant (λ) and biological half-lives (T1/2) were estimated to be -0.1060 and 6.5 days of mandipropamid and -0.1236 and 5.6 days of thiamethoxam. The PHRLs for lettuce on the 10th and 5th day before harvesting were calculated to be 63.24 and 43.56 mg/kg for mandipropamid, and 44.66 and 25.88 mg/kg for thiamethoxam, with -0.0746 and -0.1091 of the upper 95% confidence intervals of dissipation rate constant, respectively. This work would be useful as guidance for adjusting the shipment date and contribute to stabilizing the income of farmers in Korea.

Toxicological effects of pesticides on loach in rice paddy (벼 재배 논 서식 미꾸리에 대한 농약의 영향)

  • A toxicological study of the pesticides for rice paddy to loach was conducted with iprobenfos 17% GR, diazinon 3 % GR, and butachlor 5 % GR to establish the field test method and their toxicological effects on loach under the actual field conditions. The type C of cage was more effective in the sense of a little stress from the cage. The nets for shading and for preventing the birds were necessary to maintain the water temperature and to prevent the predators. The cumulative mortality of loach exposed to iprobenfos 17% GR, diazinon 3% GR and butachlor 5% GR were 10, 55 and 22%, respectively, during 7 days of exposure. The averaged concentration of the pesticides in paddy water 2 days after application were 1.67, 0.22 and 0.26 mg/L, and mortalities were 7, 50 and 17%, respectively, representing the similar results with acute toxicity $(48h-LC_{50})$.

Gibberellin Production by Newly Isolated Strain Leifsonia soli SE134 and Its Potential to Promote Plant Growth

  • Very few plant growth-promoting rhizobacteria (PGPR) are known to produce gibberellins (GAs). The current study aimed to isolate a phytohormone-producing PGP rhizobacterium from soil and assess its potential to enhance plant growth. The newly isolated bacterium was identified as Leifsonia soli sp. SE134 on the basis of partial 16S ribosomal RNA gene sequence. Application of L. soli culture filtrate significantly increased the biomass, hypocotyl, and root lengths of cucumber seeds as compared with non-inoculated sole medium and distilled water treated controls. Furthermore, the PGPR culture was applied to the GA-deficient mutant rice cultivar Waito-C. Treatment with L. soli SE134 significantly increased the growth of Waito-C rice seedlings as compared with controls. Upon chromatographic analysis of L. soli culture, we isolated, detected and quantified different GAs; namely, $GA_1$ ($0.61{\pm}0.15$), $GA_4$ ($1.58{\pm}0.26$), $GA_7$ ($0.54{\pm}0.18$), $GA_8$ ($0.98{\pm}0.15$), $GA_9$ ($0.45{\pm}0.17$), $GA_{12}$ ($0.64{\pm}0.21$), $GA_{19}$ ($0.18{\pm}0.09$), $GA_{20}$ ($0.78{\pm}0.15$), $GA_{24}$ ($0.38{\pm}0.09$), $GA_{34}$ ($0.35{\pm}0.10$), and $GA_{53}$ ($0.17{\pm}0.05$). Plant growth promotion in cucumber, tomato, and young radish plants further evidenced the potential of this strain as a PGP bacterium. The results suggest that GA secretion by L. soli SE134 might prove advantageous for its ameliorative role in crop growth. These findings can be extended for improving the productivity of different crops under diverse environmental conditions.

Study on the Characteristics of Odor Emitted from Swine Facilities (양돈시설에서 발생하는 악취특성에 관한 연구)

  • We studied the characteristics of odor emitted from 40 swine facilities across the country under various conditions like rearing densities, management style, ventilation system and swine manure handling characteristics, based on the olfactory and analytical evaluation. Odor concentrations (D/T) measured from swine facilities were respectively an average 4,055 D/T at liquid manure storage tanks on aeration, an average 913 D/T at slurry manure storage, an average 506 D/T at finishing swine facilities and an average 201 D/T at composting facilities. The higher rearing densities and slurry accumulation volume in finishing swine house, the more odor concentration (D/T) was increased. But The odor concentration (D/T) in finishing swine house did not show significant difference according to application of microbial additives. 9 odor compounds ($NH_3$, $H_2S$, MM, DMS, DMDS, PA, n-BA, n-VA, i-VA) were detected at swine facilities and the main odor compounds were volatile sulfur compounds such as $H_2S$, MM and volatile fatty acids compounds such as n-BA, n-VA, i-VA. 97.5% of swine farms surveyed in this study is located within 300 m of residential area and it is easy to bring odor complaints.
