33종 생약 추훌물의 담배모자이크바이러스(TMV)와 오이모자이크바이러스(CMV)에 대한 감염 억제효과 (Inhibitory Effect of Extracts from 33 Medicinal Herbs Against TMV and CMV Infection)

  • 동북아시아 지역에서 전통적으로 진귀하게 사용되어온 생약자원 28과 33종의 추출물을 이용하여 주요 식물바이러스에 대한 감염억제활성을 조사하였다. 온실 생물검정 시험결과, 19과 21종의 메탄올 추출물이 담배모자이크바이러스(TMV)와 오이모자이크바이러스(CMV)에 대하여 항바이러스 활성을 보였다. 대부분의 공시한 사료들은 약한 항바이러스 활성은 보였으나, 목질진흙버섯의 메탄올 추출물은 TMV에 대하여 $98.7{\pm}1.3%$의 강한 항바이러스 활성을 보였다. 이는 CMV에 대하여도 $97.2{\pm}2.2%$의 높은 활성을 보였다. 또한, 오배자의 메탄올 추출물은 TMV 및 CMV에 대하여 $98.7{\pm}1.1%$이상의 억제효과를 보였다. 금후의 연구에서는 효과적이고 새로운 형태의 식물바이러스 방제제 개발에 있어서 매우 유용할 수 있는 이들 선발된 생약자원 유래의 항바이러스 활성물질의 구명이 필요하다.

수종 한약 처방 전탕액(煎湯液)의 허피즈바이러스에 대한 효능 검색 (Screening for Various Herb Medicines Extracts HSV 1-2)

  • Objective : In order to find antiviral compounds against Herpes simplex virus type I(HSV-1) and II(HSV-2) from herb medicines, a convenient virus-induced cytopathic effect(CPE) inhibition assay was introduced. Methods : Fourteen purchased herbal medicines, and their toxicity of infected cell and anti-viral activities were evaluated. Among them, the major part of herbal medicines showed cell stability compared with the contrast. Results : Cytotoxic concentration (CC) of the $H_2O$ extracts of Hyongbangpaedoksan against HSV-1 and HSV-2 was 181.12. This is high level cytotoxic concentration compared with the contrast. Therefore, we assumed that the high level cytotoxic concentration of herbal medicine play a major role in improvement of antiviral activity at the first infective cell. But antiviral effect was unable to figure out for selective index(Sl)=CC50/EC50. The other herbal medicines were unable to showed potent anti-HSV activity. Conclusions : The antiviral activation using herbs in this thesis have unlimited objects, to select research object will help to show the direction of antiviral drug development that have less side effect and more excellent efficiency.

항바이러스 효과에 응용되는 한약의 연구 동향 (A tendency of herbal medicines on anti-viral effect)

  • In order to find antiviral effect against Human immunodeficiency virus(HIV), Herpes simplex virus type I(HSV-1) and II(HSV-2) from herb medicines, publicated 29 paters on anti-viral effect of herbal medicines and a convenient virus-induced cytopathic effect (CEP) inhibition assay was introduced. The major virus on experiment are HIV, Hepatitis B virus and HSV-1,2. Those of other studies showed inhibition of infected virus DNA replication and screening test of herbal medicines. More than 15 extractions were prepared by pure water boiling from herbal medicines, and their toxicity of infected cell and anti-viral activities were evaluated. Among them, the major part of herbal medicines showed cell stability compared with the contrast. Cytotoxic concentration (CC) of the $H_2O$ extracts of Padoo against HIV was <4.0, Hyungbangpaedoksan against HIV was 9.3, Whangyonhaedoktang against HIV-1 and HSV-2 was 15.3. These are high level cytotoxic concentration compared with the contrast. But antiviral effect was unable to figure out for selective $index(SI)=CC_{50}/EC_{50}$. The other herbal medicines were unable to showed potent anti-HIV and anti-HSV activity. The antiviral activation using herbs in this thesis have unlimited objects, to select research object will help to show the direction of antiviral drug development that have less side effect and more excellent efficiency.

Feline calicivirus에서 항바이러스 활성을 가지는 천연식물자원 탐색 (Screening of Antiviral Activity from Natural Plants against Feline Calicivirus)

  • 노로바이러스는 인간에게 급성위장염을 일으키는 식중독 바이러스로, 최근 전세계적으로 노로바이러스 식중독 사고가 증가하고 있다. 따라서 본 연구에서는 다양한 생리활성 작용이 있다고 알려진 향신료, 차(茶), 식물성 한약재등 11종의 천연물질을 대상으로 노로바이러스의 대체모델인 feline calicivirus (FCV 에 대한 항바이러스 활성을 조사하였다. 각 추출물에 대한 항바이러스 활성은 end point dilution assay 방법인 50% tissue culture infectious dose ($TCID_{50}$)으로 바이러스 감염가를 측정하였다. 녹차(Camellia sinensis L.) 추출물의 경우 3.13 mg/ml의 농도에서 1 시간만에 FCV가 완전히 불활성화 되었으며, 홍화씨 (Carthamus tinctorius L.) 추출물은 6.25 mg/ml의 농도에서 5시간 만에 바이러스가 불활성화 되어 녹차추출물이 FCV에 대한 항바이러스 활성이 가장 뛰어난 것으로 조사되었으며, 농도 및 반응시간에 유의적으로 항바이러스 활성을 나타내는 것으로 관찰되었다. 녹차 및 홍화씨 추출물 등과 같은 천연식물 자원을 이용하여 FCV에 대한 항바이러스 활성을 조사한 결과는 궁극적으로는 노로바이러스에 의한 식중독 저감을 위한 기초자료에 이용될 수 있을 것으로 기대된다.

Antiviral Potential of the Genus Panax: An updated review on their effects and underlying mechanism of action

  • Viral infections are known as one of the major factors causing death. Ginseng is a medicinal plant that demonstrated a wide range of antiviral potential, and saponins are the major bioactive ingredients in the genus Panax with vast therapeutic potential. Studies focusing on the antiviral activity of the genus Panax plant-derived agents (extracts and saponins) and their mechanisms were identified and summarized, including contributions mainly from January 2016 until January 2022. P. ginseng, P. notoginseng, and P. quinquefolius were included in the review as valuable medicinal herbs against infections with 14 types of viruses. Reports from 9 extracts and 12 bioactive saponins were included, with 6 types of protopanaxadiol (PPD) ginsenosides and 6 types of protopanaxatriol (PPT) ginsenosides. The mechanisms mainly involved the inhibition of viral attachment and replication, the modulation of immune response by regulating signaling pathways, including the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway, cystathionine γ-lyase (CSE)/hydrogen sulfide (H2S) pathway, phosphoinositide-dependent kinase-1 (PDK1)/ protein kinase B (Akt) signaling pathway, c-Jun N-terminal kinase (JNK)/activator protein-1 (AP-1) pathway, and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) pathway. This review includes detailed information about the mentioned antiviral effects of the genus Panax extracts and saponins in vitro and in vivo, and in human clinical trials, which provides a scientific basis for ginseng as an adjunctive therapeutic drug or nutraceutical.

한약재 식물 에탄올추출물을 이용한 인플루엔자 불활화백신 제작 (Preparation of an Inactivated Influenza Vaccine Using the Ethanol Extracts of Medical Herbs)

  • 코로나-19 펜데믹에서 볼 수 있듯이, 새로운 바이러스 감염병의 출현은 전 세계적으로 공중보건에 심각한 우려를 발생시킨다. 특히, 항바이러스제 및 백신의 부재는 감염병의 피해를 더욱 증가시킨다. 식물 유래 천연물은 안전하고 효과적인 항바이러스제 개발의 주요 공급원이다. 본 연구는 한약재 식물의 에탄올추출물의 항바이러스 활성을 분석함으로써 안전성과 효능을 갖는 새로운 항바이러스제 후보물질을 발굴하는 것을 목표로 하였다. 10종의 한약재 에탄올추출물의 항산화활성과 세포독성을 분석한 후 로타바이러스, A형간염바이러스, 독감바이러스에 대한 광범위한 바이러스 사멸활성을 분석하였다. 특히, 마가목과 감초의 추출물은 독감바이러스에 대한 강력한 사멸활성을 나타내었다. 또한, 마가목과 감초의 추출물로 사멸된 독감바이러스의 백신효능과 방어효능을 마우스 모델에서 검증하였다. 추출물로 사멸된 바이러스는 높은 수준의 중화항체를 유도하였으며 야생형 바이러스 공격접종을 효과적으로 방어하는 우수한 백신효능을 나타내었다. 본 연구의 결과는 한약재 유래 천연물을 기반으로 하는 항바이러스제와 사백신 제조를 위한 바이러스 불활화제 개발에 활용될 수 있는 가능성을 제시한다.

한약 단미제 분획의 Influenza virus type A 활성에 관한연구 (Study on the Influenza Type A Activity of Fractions of Korean Medicinal Herbs)

  • In order to search for anti-influenza virus type A agents from Korean medicinal herbs, we selected 100 medicinal herbs, based on a review of the Korean traditional medicine books. Four of 100 Korean medicinal herbs, MM-40, MM-55, MM-63, MM-110, exhibited very strong anti-influenza virus activity. The fractions of four medicinal herbs, which had very strong anti-influenza virus activity, were tested for antiviral activity by means of Haemagglutination inhibition test(HTT), 40% MeOH fraction of MM-40, $H_2O$ fraction of MM-55, 20% fraction of MM-63 3nd $H_2O$ fraction of MM-110 had strong anti-influenza virus activity at the range of $78{\mu}g/ml$ to $156{\mu}g/ml$, 1.56mg/ml to 100mg/ml, 6.25mg/ml to 50mg/ml and $48.7{\mu}g/ml$ to $780{\mu}g/ml$, respectively. These results of HIT indicated that fractions of Korean medicinal herbs might inhibit either attachment of virus to cell surface receptor or penetration of virus into cell during the initial stage of infection. In the cytotoxicity of fractions against red blood cells, 40% MeOH fraction of MM-40, 20% fraction of MM-63 and $H_2O$ fraction of MM-110 showed cytotoxicity at the range of $78{\mu}g/ml$ to 10mg/ml, 50mg/ml to 100mg/ml and $195{\mu}g/ml$ to 100mg/ml, respectively, whereas $H_2O$ fraction of MM-55 did not show cytotoxicity. In order to establish influenza virus adapted animal model, influenza virus type A were passaged 3 and 4 times successively in Balb/c mouse. As a result, we had 4 HA unit titers on the 5 days of 3rd passages and 7 days of 4th passages after infection, respectively.

Phytotherapy in periodontics as an effective and sustainable supplemental treatment: a narrative review

  • Purpose: Periodontal disease is a chronic condition caused by microbial infection and mediated by the host's immune response. Phytotherapy is a therapeutic approach that utilizes a renewable resource capable of supplying less expensive medicines for the world's growing population. This review aimed to present clinical evidence on the use of complementary medicinal herbs in the treatment of periodontal diseases. Methods: Different databases were searched using the terms "herbal" and "periodontitis." All included studies were examined with a focus on herbal indications, type, and prescription length. Dentists' therapeutic and prophylactic herbal prescribing habits were also assessed. Results: Various herbs such as turmeric, neem, aloe-vera, pomegranate, catechu, tulsi, cloves, lemon grass, green tea, tea tree oil, peppermint, garlic, pineapple, oak bark, babul, bakul, sage, coriander, moringa, amla, guava, and grape seed extract have been used in the treatment of periodontitis. These herbs have been reported to exhibit a range of therapeutic effects, including anti-inflammatory, antiplaque, antihalitosis, antiresorptive, antioxidant, antibacterial, antifungal, antiviral, and antimicrobial properties. These components can be utilized in various forms such as mouth rinse, gel, oil, toothpaste, aqueous extract, mouthwash, or tooth powder. Conclusions: Several readily available herbal formulations are now available on the market and have been shown to be effective as supplemental periodontal phytotherapy. However, these should be used under the supervision of a dental professional to ensure optimal benefits and effectiveness. Therefore, it is necessary to improve the understanding of suggested herbal prescription practices among dental professionals.

  • Plants of the genus Hypericum (Family - Hypericaceae) are herbs, shrubs or small trees and are distributed chiefly in the temperate regions of the world. About 400 different species of Hypericum are available throughout the globe and 20 species occur in India, including a few cultivated in gardens. Almost all plants of the genus Hypericum are widely used in folk medicine. Several potent phytoconstituents from different Hypericum species have led to the isolation of antibacterial, antifungal and cytotoxic compounds. With the development of resistance and cross resistance with different microorganisms and the evolution of so many deadly diseases the screening and evaluation of the phytoconstituents so much so the development of varied phytoconstituents for the drug development for these deadly diseases is utmost essential in every aspects. The present review on the antimicrobial use of different Hypericum reports the findings from and extensive literature search on the Hypericum species around the globe that have been assessed for antimicrobial and antiviral activity. An attempt has been made through this review to summarize the information in this aspect in order to highlight the promising species of this genus which are worthy for further investigation as leads for drug development. Over 31 different Hypericum species have been reported to possess such activities with their varied number of phytoconstituents. Sixteen different constituents of six different classes of phytoconstituents have been reported to be present in different varieties of Hypericum, which may be considered responsible for this activity.

Traditional Unani Medicine in Flu-like Epidemics and COVID-19 during Pregnancy: A Literary Research

  • Background: The pandemic COVID-19 caused by a novel coronavirus SARS-COV-2 has spread like a forest fire. This disease may have serious consequences for pregnant women. Presently, no specific drugs or vaccines exist to battle this disease and researches are underway. Unani medicine has a unique role in prevention and management during epidemics. Here, we reviewed the overview of COVID-19 infection and pregnancy, concept and practices in Unani medicine for flu-like epidemics in general and pregnancy, and safety of Unani drugs for the prevention and treatment of mild symptomatic cases of COVID-19 during pregnancy. Methodology: Unani classical texts and pharmacopoeia were meticulously explored for concepts and practices for flu-like epidemic diseases. Further, we browsed scientific databases such as PubMed, Scopus and others for an overview, epidemics and Unani medicine, effectiveness and safety of Unani drugs in COVID-19 and pregnancy. Results: Unani medicine includes prevention and management of flu-like epidemic include quarantine and isolation, aromatic herbal drugs fumigation and spraying for environmental disinfection, ilaj bid tadbir for health promotion and use of health-protecting drugs and symptom-specific drugs in general and related to pregnancy. Lahsun, asalussus, behidana, banafsha, zanjabeel, unnab, etc are in use since antiquity for the prevention and treatment of asymptomatic and mildly symptomatic pregnant women during infectious and epidemic diseases. Conclusion: Currently, the aforementioned plants are proven for antiviral, antioxidant, immunomodulatory and anti-inflammatory activities, probably useful in the COVID-19 pandemic. Additionally, scientific studies have provided new insight into the mechanism underlying the therapeutic effect of Unani medicines that are safe in pregnancy.
