  • Various peptides derived from food are among the most potent physiologically active agents known, and include anticancer peptides, angiotensin converting enzyme(ACE) inhibitor exhibiting antihypertension action, opioid peptides, antithrombotic peptides, hypocholesterolemic peptides, immunomodulators, calcium absorption enhancers, and other peptides. Hydrophobic peptides extracted from a Cheddar-type cheese slurry were fractionated by gel chromatography and repeated HPLC. A peptide fraction from HPLC showed high cytotoxicity on the tumor cell lines such as a human colon carcinoma, and comprised of Tyr, Ser, Leu, Gly, and others. Hypocholesterolemic peptides were isolated from peptic hydrolyzates of casein and soy proteins. Macropeptides of 1,000${\sim}$5,000 dalton were effective on reducing the cholesterol level of mouse serum. Peptides showing high Krigbaum hydrophobicity and ANS surface hydrophobicity resulted in high hypocholesterolemic effect and fecal steroid concentrations. Caseinomacropeptides (CMP) were isolated from whey powder and treated with soluble and immobilized trypsin to obtain antithrombotic peptides. One fraction from the CMP hydrolyzed with immobilized trypsin for 24h exhibited high antithrombotic activity with 52.5% inhibition of platelet aggregation. These results suggested that peptides from various milk products could be utilized as a good bioactive agents for developing health functional foods.

Evaluation of the Effects of Acorus gramineus and Acorus tatarinowii Extracts on a Rat Model of Arterial Thrombosis induced by Ferric Chloride

  • We investigated the antithrombotic effect of Acorus gramineus Soland (A. gramineus) from Korea and Acorus tatarinowii Schott (A. tatarinowii) from China in a rat model of arterial thrombosis induced by ferric chloride ($FeCl_3$). Thirty minutes prior to a 35% $FeCl_3$ application, Sprague Dawley rats were intraperitoneally injected with saline, A. gramineus and A. tatarinowii (100 mg/kg), respectively. Occlusion time of rats injected with A. gramineus was delayed significantly compared to that of the vehicle and A. tatarinowii. Thrombus weight was meaningfully decreased in rats injected with A. gramineus compared to the vehicle. Additionally, A. gramineus inhibited collagen fiber damage in vessel compared to the vehicle, but A. tatarinowii did not show a significant effect. Our results show that A. gramineus and A. tatarinowii from the same genus have different antithrombotic effects, and especially A. gramineus has a better antithrombotic effect than A. tatarinowii.

Hypolipidemic and Antithrombotic Effects of Increasing Intake of Linolenic Acid Derived from Perilla Oil in Rats

  • This study investigated the hypolipidemic and antithrombotic effects of linolenic acid derived from Korean perilla oil. The experimental rats(male, Sprague-Dawley) were divided into 5 groups using a Randomized Complete Block Design and fed one of the five following diets : DO*/O#. D4/O, D4/4, D4/8, or D4/20(D*/# represents the ratio of linoleic to linoenic acid as the percentage of total dietary energy intake) for 4 or 8 months. Bleeding time and whole blood clotting time were determined and the composition of serum and platelet lipids analyzed. Comparisons from the DO/O to the D4/20 group showed that serum lipids (total lipid, triglyceride, total cholesterol, and HDL-cholesterol) gradually decreased with increasing linolenic acid intake - the hypolipidemic effect. The composition of platelet fatty acids[the ratio of eicosapentaenoic acid(EPA)/arachidonci aci(AA)] increased gradually with increasing linolenic acid intake. Higher linolenic acid intake increased bleeding time and whole blood clotting time, and decreased malondialdehyde(MDA) production in the platelets, though no significant differences. These results suggest that linolenic acid derived from perilla oil appears to suppress the conversion of linoleic acid to AA and the EPA transformed from linolenic acid appears to suppress the conversion of AA to TXA2. Since TXA2 is a platelet-aggregating and vasoconstricting agent, the redulction of TXA2 released by platelets with increasing intake of perilla oil containing a lot of linolenic acid confers an antithrombotic effect.

  • Twentyfive plant crude drugs with traditional reputation were selected for the antithrombotic screening. MeOH (80%) extract of each plant was given (500 mg/Kg, p.o.) to mice 1 hr prior to the thrombotic challenge (1.8 mg collagen plus 180 ug epinephrine/Kg, i.v.). Seven extracts (1, 2, 8, 10, 18, 22 and 23) showed either comparable or better recovery than aspirin (50 mg/Kg, p.o.) from paralysis caused by the pulmonary thrombosis. 1, 2, 8, 10, 18, 22 and 23 were fractionated to $CH_2Cl_2$ soluble (fr I) and $H_2O$ soluble (fr II) fractions and each fraction (100 mg/Kg, p.o.) was also tested. Mice treated with fr I of 2 (Angelica koreana), 10 (Corydalis spp.) or 23 (Schizonepeta tenuifolia) showed higher recovery and lower mortality rates than aspirin (50 mg/Kg, p.o.) treated mice.

The Effect of Aspalatone, a New Antithrombotic Agent, on the Specific Activity of Antioxidant Enzyme in the Rat Blood

  • The antioxidant efficacy of aspalatone, a new antithrombotic agent, has been recognized in the neurotoxic model and in the cardiotoxic model in proliminary studies. We examined the specific activity of antiosidnat enzyme in the rat blood following administrations of aspirin, maltol, aspirin together with maltol, salicylmaltol (major metabolite of aspalatone) and aspalatone, respectively. Our assessment showed that salicylmaltol, maltol, aspalatone enhanced antiperoxidative activity. In addition, neither aspirin nor combination of aspirin and maltol, showed any significant effect on the activity of antioxidant enzyme. Because $H_{2}$$O_{2}$ accumulation may stimulate the thrombogenesis in blood, the result suggests that the induction of blood antiperoxidative activity produced by aspalatone may have beneficial effects on the thrombogenesis.

  • Objectives:Spring water is widely known to relax muscles by promoting blood circulation. This study was conducted to analyze theantithrombotic effect of naturally carbonated hot spring water (NCHW) to assess its influence on blood circulation. Methods:Atopic dermatitis was induced in the skin of the mice used in this experiment. NCHW was applied, and the antithrombotic effect was assessed and compared with that of other interventions. The positive control group was treated orally with aspirin. Results:After 3 weeks of exposure to NCHW, the experimental groupshowed a significant antithrombotic effect. NCHW also produced inhibitory responses to both collagen- and ADP-induced platelet aggregation, whereas the group given aspirin reacted only to collagen-induced platelet aggregation. Conclusions:The experiment demonstrated the intrinsic antithrombotic effects of NCHW compared with those of artificially carbonated water, tap water, and aspirin. This result suggests the possibility that NCHW can be used as a supportive and alternative treatment for vascular diseases.

  • The freeze-dried powder of Lumbricus rubellus earthworm was administered orally to rats and its fibrinolytic and antithrombotic effects were investigated. The fibrinolytic activity of plasma was determined by measuring the plasmin activity of the euglobulin fraction and was increased to two-folds of the control at a dose of 0.5 g/kg/day and five times with 1 g/kg/day after 4-day administration. The antithrombotic effect was studied in an arterio-venous shunt model of rats. The thrombus weight decreased significantly from 43.2 mg to 32.4 mg at a dose of 0.5 g/kg/day after 8-day treatment. The level of fibrinogen/fibrin degradation product (FDP) in serum was elevated in a dose-dependent manner during the treatment period. On the 8th day after administration, the FDP value was increased to 7.7 $\mu\textrm{g}$g/ml compared with the control value of 3.3 $\mu\textrm{g}$g/ml. These results support that earthworm powder is valuable for the prevention and/or treatment of thrombotic conditions.

현호색산의 항혈전작용에 대한 연구 (The Experimental Studies on Antithrombotic Effects of Hyunhosaiksan)

  • The purpose of the present study was to investigate the effects of Hyunhosaiksan (HHS) on antithrombotic actions which include blood activation, thrombus removal, warming of circulating blood, and the control of pain on abdomen and lower and upper burning spaces. HHS significantly inhibited platelet aggregation induced by ADP and epinephrine in a HHS dose-dependent manner when analyzed by the Sigmoid Emax model in WinNonlin. EC50 values of HHS were 1.71 ㎍/ml and 0.004 ㎍/ml for ADP and epinephrine respectively. In the vivo study, HHS inhibited pulmonary embolism induced by collagen and epinephrine, which was however statistically insignificant. HHS increased number of platelets, APTT and volume of fibrinogen significantly as compared with the control group in dextran-induced thrombus model. Furthermore, HHS stimulated levels of blood flow in vivo though its effect was not observed in vitro. These results suggest that Hyunhosaiksan (HHS) can be used for treating numerous diseases related with blood aggregation and circulation problems. Further systematic investigations on the synergic effects among drugs used in the oriental medicine as well as in the western medicine in relation to thrombosis therapy would provide an important insight into the potential therapeutic applications.

  • This study was performed to investigate the antithrombotic activity and protective effect of hexane fraction of Kamihyulbuchukeotang (KHCTH) on brain injury by KCN and MCA occlusion a prescription of HCT added with Lumbricus and Notoginseng Radix. Experiemental parameters are brain ischemia by MCA occlusion assay, KCN-induced brain injury, pulmonary thrombosis and platelet aggregation assay. The results were summarized as follows; 1. KHCTH extracts significantly inhibited the duration of KCN-induced coma (67%) and mortality (80%). 2. KHCTH extracts significantly suppressed brain ischemic area and edema following MCA occlusion and protected neuron cells as compared with control data. 3. KHCTH extracts inhibited pulmonary thrombosis induced by collagen and epinephrine. 4. KHCTH extracts inhibited platelet aggregation induced by collagen, ADP as agonist up to 76.9% and 32.3% respectivey at 1 mg/ml more effective than water extract of KHCT These data suggested that KHCTH could be applied as the protector of brain ischemia and injury and antithrombotic agent.

Antithrombotic and Antiallergic Activities of Rhaponticin from Rhei Rhizoma Are Activated by Human Intestinal Bacteria

  • To evaluate the antithrombotic and antiallergic properties of rhaponticin extracted from Rhei Rhizoma, the in vitro and ex vivo inhibitory activities of rhaponticin and its metabolite, rhapontigenin, were measured. These compounds inhibited in vitro ADP- and collagen-induced platelet aggregation. Rhapontigenin was more potent, with $IC_{50}$ values of 4 and $70{\;}{\mu}g/ml$, respectively. In ex vivo ADP- and collagen-induced rat platelet aggregation, these compounds also exhibited a potent inhibitory effect. The antiplatelet aggregation effects of rhaponticin and rhapontigenin were more potent than those of aspirin. Rhapontigenin showed significant protection from death due to pulmonary thrombosis in mice. Rhapontigenin also showed the strongest inhibitory activity against $\beta-hexosaminidase$ release induced by DNP-BSA. These compounds inhibited PCA reaction in mice. Rhapontigenin intraperitoneally administered showed the strongest inhibitory activity and significantly inhibited PCA at doses of 25 and 50 mg/kg, with inhibitory activities of 48 and 85%, respectively. The inhibitory activity of orally administered rhaponticin was stronger than that of intraperitoneally administered rhaponticin. These results suggest that rhaponticin, in the rhizome of Rhei Rhizoma, is a prodrug that has extensive antiallergic and antithrombotic properties.
