Physicochemical Properties and Antioxidative Activities of Rapeseed Meal (유채박의 이화학적 특성 및 항산화 효과)

  • This research was to investigate physicochemical properties and antioxidative activities of rapeseed meal for the development of functional cosmetic material. Seventeen kinds of amino acid at rapeseed meal were found and glutamic acid concentration was significantly the highest (28.4 mg/g), followed by glycine, proline, arginine, isoleucine, and aspartic acid. Among various vitamins, cloline content was the highest (459.1 mg/kg), followed by niacin, tocopherol, and pantothenic acid. Among various fatty acids of rapeseed meal, oleic acid was the highest (36.7%), followed by linoleic acid and linolenic acid. DPPH radical scavenging activities of methanol and acetone extract of rapeseed meal at 2.0 mg/mL were 80.4 and 78.9%, respectively. The methanol and acetone extracts of rapeseed meal were a stable at the range of pH 3-9 on DPPH radical scavenging activity. The maximum reducing powers of methanol and acetone extract of rapeseed meal at 4.0 mg/mL were 0.7 and 0.68 OD 700 nm, respectively. The maximum superoxide inhibition activities of hot water, acetone, and methanol extract of rapeseed meal were 70.2, 75.2, and 81.4%, respectively. These results showed that the methanol and acetone extract of rapeseed meal can be used as a new source of functional cosmetic material.

Evaluation of the Antioxidant and Antiproliferative Properties of a Hot-water Extract from Gulfweed, Sargassum fulvellum

  • Sargassum fulvellum (gulfweed) is a widespread seaweed in the coastal areas of northeast Asia. In the present study, we identified the phenolic compounds present in aqueous and ethanolic extracts of S. fulvellum and evaluated their antioxidative properties and their abilities to block cell proliferation using in vitro assays: antioxidant activity was assessed by using a DPPH assay and superoxide anion scavenging activity, anti-tyrosinase activity, and anti-proliferative activity were assessed using MTT and lactate dehydrogenase [LDH] assays in vascular smooth muscle cells. The hot-water ($65^{\circ}C$) extract had a higher phenol content than the ethanolic extract. The hot-water extract showed a statistically significant increase in free radical scavenging activity and a greater ability to reduce proliferation of vascular smooth muscle cells stimulated with platelet-derived growth factor-BB. Taken together, hot-water extracts of S. fulvellum may be an important source of antioxidative and antiproliferative agents.

Antioxidative and Cytotoxic Effects of Ethanol Extracts from Elaeagnus multiflora (뜰보리수 에탄올 추출물의 산화적 스트레스 억제 효과와 암세포 증식 억제 효과)

  • Elaeagnus multiflora, generally referred to as the cherry silverberry, is a plant. Elaeagnus multiflora fruit, leaves, and roots have been traditionally utilized in China as a treatment for cough, diarrhea, itch, and foul sores, and even cancer. More importantly, it is being investigated as a food that is capable of reducing the incidence of cancer, and also as a means of halting or reversing the growth of cancers. Considering the dearth of information regarding the medicinal properties of Elaeagnus multiflora, we assessed the antioxidative and cytotoxic effects of Elaeagnus multiflora by examining its scavenging effects on the 1,1-diphenyl-2-picryl-hydrazyl(DPPH) radical, its inhibitory effects on lipid peroxidation, and its inhibitory effects on cancer cell proliferation in HeLa cells, MCF-7 cells, and SNU-638 cells via MTT assay. Ethanol extracts of Elaeagnus multiflora flesh and seed inhibited DPPH radical production($36.91{\pm}1.00$, $94.17{\pm}0.37$) and lipid peroxidation ($28.18{\pm}5.02$, $40.30{\pm}1.45$, respectively). The Elaeagnus multiflora seed is believed to exert a possible antioxidative effects against the DPPH radical. The ethanol extracts of Elaeagnus multiflora flesh and seed exerted the cytotoxic effects on Hela cells ($6.93{\pm}1.92$, $84.97{\pm}0.47$), MCF-7 cells($5.45{\pm}0.41$, $84.97{\pm}0.47$), and SNU-638 cells($19.39{\pm}0.43$, $76.84{\pm}0.63$) used in this study. This result suggests that Elaeagnus multiflora seeds in contrast to its flesh, is believed to exert a possible anticancer effect. Elaeagnus multiflora seeds are considered to be a the candidate for preventative and dietetic treatment as an anticancer functional food.

Contents of Antioxidative Components from Pulpy and Seed in Wild Haw (Crataegus pinnatifida BUNGE) (자생 산사(Crataegus pinnatifida BUNGE)과육 및 씨의 항산화성분 함량)

  • This study was conducted to evaluate the contents of antioxidative components from pulpy and seed in wild haw (Crataegus pinnatifida $B_{UNGE}$). Pulpy and seed of haw were smashed, then measured for color properties, antioxidative components of ascorbic acid, phytic acid, proanthocyanidin, anthocyanin, total carotene, ${\beta}$-carotene, lycopene, chlorophyll a, b and tannin. The $a^*$, $b^*$ and $C^*$ values of seed were significantly lower than pulpy, but $L^*$ and $H^{\circ}$ values were higher than that of pulpy. Ascorbic acid contents of pulpy and seed were found to be $10.89{\pm}1.69mg/100g$ and $1.45{\pm}0.16mg/100g$, respectively. Phytic acid, proanthocyanidin, total carotene and tannin contents of pulpy and seed were $689.17{\pm}3.63mg/g$, $597.78{\pm}2.93mg/g$; $355.61{\pm}19.39mg/g$, $49.12{\pm}4.97mg/g$; $8.32{\pm}0.42mg%$, $0.80{\pm}0.01mg%$; $7.53{\pm}0.09mg/g$, $1.02{\pm}0.03mg/g$, respectively. Similarly, ${\beta}$-carotene, lycopene, chlorophyll a and chlorophyll b contents of pulpy also displayed higher values than that of seed. On the contrary, anthocyanin content of seed ($4.24{\pm}0.33mg/L$) was remarkably higher than pulpy ($0.99{\pm}0.62mg/L$). The results showed that pulpy could be severed as great natural antioxidant and biohealth functional food.

Separation and Characteristics of Antioxidative Substances in Fermented Soybean Sauce (양조간장으로부터 항산화성 물질의 분리 및 그 특성)

  • Fermented soybean sauce was fractionated by gel filteration chromatography and the physico-chemical properties of each fraction were investigated for the study on the separation and characteristics of antioxidative substances in the soybean sauce. The brown coloured melanoidin fractions(MF-I) collected by Sephadex G-10 column were found to contain nitrogeneous compounds and to have strong antioxidative activity and reducing power. The freeze dried MF-I was further fractionated with Sephadex G-50 and G-100 successively and then MF-II and MF-III fractions were observed to be the most effective antioxidant and reducing power among the fractions. The UV-VIS and IR absorption spectra of the each fraction were also determined and discussed.

ANTIOXIDATIVE ACTIVITIES OF SOME DIETARY FIBERS DETERMINED BY AN NIR EMISSION SPECTROSCOPY

  • Constituents of several .representative seaweeds, such as wakame Undaria pinnatifida; hijikia Hizikia fusifome; and kombu Laminaria japonica, were found to have fairly large reaction rates determined by quenching experiments of emission spectra in the near-infrared region (1max: 1270 nm) from singlet oxygen (102). Emission spectra of singlet oxygen generated from an aqueous solution of Rose Bengal under irradiation with a green laser (330 nm) were measured by a near-infrared (NIR) emission spectrometer constructed in our laboratory. The quenching experiments were as follows: Intensities of emission spectra were measured in the absence (I0) and in the presence of the seaweed constituents (I): Ratios of I0/I were plotted against every concentration of the quenchers (Stern-Volmer plots) which gives a straight line. The slope of each line gives a kqt value which gives a quenching constant kq value (an antioxidative constant against singlet oxygen) when the t value (half-life time of singlet oxygen in the solvent used) was given. The determined reaction rates are between 103-105 (g/l)-ls-1; the larger ones are as large as that of ascorbic acid, 8.4 ${\times}$ 104 (g/1)-ls-1. Most of these seaweed constituents also showed antioxidative activity against auto-oxidation and superoxide as well as their immunological enhancing activity. These results suggest a possibility that dietary fibers which are supposed to prevent the large-intestine cancer by their physical properties may prevent the cancer, at least in parts, by their chemical, antioxidative activity.

Quality Characteristics and Antioxidative Effects of Dasik added with Lycii Fructus Extract (구기자 추출액을 첨가한 다식의 품질특성 및 항산화 효과)

  • The purpose of this study was to investigate the quality characteristics and antioxidative effect of Dasik prepared with Lycii fructus extract(LD). LD were added in ratios (w/w) of 0(C), 5(LD1), 10(LD2), and 15%(LD3), and then proximate compositions, physicochemical properties, sensory evaluations and antioxidative effect of the Dasik were measured. LD1~LD3 samples showed higher contents of moisture, crude lipid, crude protein and crude ash as well as pH and sugar content compared to control (p<0.001). The total phenol content of LD samples (LD1, 31.5% ; LD2, 66.1% ; and LD3, 69.7%) was higher than that of control (4.71%). DPPH radical scavenging activity of LD3 showed the highest activity of 65.3%, whereas that of control (27.0%) was the lowest among samples. Additionally, ABTS radical scavenging activity of LD samples was higher than that of control (C, 38.9% ; LD1, 53.6%; LD2, 80.5% ; and LD3, 83.3%). The results of texture analysis on all samples showed that control had the highest but LD3 was the lowest (p<0.001). In sensory evaluation, the scores of appearance, flavor, taste and overall preference for LD2 was significantly higher than the samples(C, LD1, and LD2). Form the findings, this study suggests that 10% addition of LD was effective for preparation of Dasik in the aspects of the consumer acceptability and functionality.

Antioxidative Activity of Sea Buckthorn and Quality Characteristics of Brown Rice Sulgidduk (비타민나무 잎 첨가 현미설기떡 제조 및 항산화 활성)

  • This study was performed to investigate the antioxidative properties of sea buckthorn (Hippophae rhamnoides L.; leaf, fruit and stem) and the quality characteristics of brown rice sulgidduk prepared using a powder of sea buckthorn leaves. First, the antioxidative activities of sea buckthorn were measured to choose the most effective part of this plant. By analyzing the measured values, we concluded that the effective part of sea buckthorn was its leaves. The $IC_{50}$ value of the DPPH radical scavenging activity and the ABTS radical scavenging activity in sea buckthorn leaves were $7.78{\mu}g/mL$ and $264.04{\mu}g/mL$, respectively. The total polyphenol and total flavonoid contents of sea buckthorn leaves were 3.80 mg/mL and .19 mg/mL, respectively. Therefore, the brown rice sulgidduk was prepared using a powder of sea buckthorn leaves in the weight ratio of 0%, .5%, 1.0%, 1.5%, 2.0% and 2.5%. For analyzing the quality characteristics of the prepared sulgidduk, proximate compositions, color and texture profiles were measured and a sensory evaluation was conducted. With an increase in the added content of the sea buckthorn leaf powder (SBLP), the L-value significantly decreased while the a-value and the b-value increased. In the case of texture profiles, the control group (control A) had a higher score for hardness than the case groups in which the SBLP was added. However, springiness, chewiness and adhesiveness were not significantly different among the groups. In the sensory evaluation, the sample containing 1.5% SBLP yielded the best results. Therefore, we suggest that the powder of sea buckthorn leaves is a good ingredient for increasing the consumer acceptability and functionality of sulgidduk.

Effect of Amino Acids Addition on Stability and Antioxidative Property of Anthocyanins (아미노산의 첨가가 anthocyanins 색소의 안정성과 항산화능력에 미치는 영향)

  • Effects of amino acids (Arg, Lys, Gly, Ile, Glu, Asp, and Met) on the color intensity, stability and antioxidative properties of anthocyanins extracted from grape skins were investigated. Intensity of anthocyanins was significantly increased by the addition of Asp. Except for basic amino acids such as Arg and Lys, stabilities of anthocyanins were significantly improved by the addition of other amino acids including neutral, acidic and sulfur containing amino acids during the storage at $30^{\circ}C$ at pH 3.5. In case of control anthocyanins was remained unchanged the intensity of red color decreased significantly during the storage whereas their antioxidative activity were unchanged. Although effects of amino acids addition on electron donating abilities of anthocyanins were not differentiated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method, the addition of Asp or Met resulted in increased ferric reducing ability which measured by FRAP (ferric reducing ability of plasma) assay.

Antioxidative Properties of Phenolic Compounds Extracted from Black Rice (흑미 색소물질에 함유된 페놀화합물의 항산화 특성)

  • The composition and antioxidative effects of phenolic compounds in black rice were studied. The contents of free and bound phenolic compounds extracted from black rice were 845.4 and 401.6 mg respectively per 100g sample weight. Free phenolic compounds had higher antioxidation ability than those of bound phenolic compounds. Solvent fractionation of free phenolic compounds revealed that butanol fraction had the highest phenolic compounds contents and antioxidative activity among other solvent fractions. Although butanol fraction showed lower lipid peroxidation inhibition (LPI) ability than that of $\alpha$-tocopherol and BHT, free radical scavenging ability was much higher than that of $\alpha$-tocopherol and BHT, as evidenced by electron donating ability (EDA) and benzoic acid hydroxylation inhibition (BAHI) assays.
