Protective Effect of Kaempferol on Cultured Neuroglial Cells Damaged by Induction of Ischemia-like Condition

  • This study was performed to evaluate the cytotoxicity induced by ischemia-like condition (ILC) in cultured neuroglial cells (C6 glioma cells). The protective effect of kaempferol (KAE), flavonoid against the cytotoxicity induced by ILC induction was assessed. In addition, antioxidative effects of KAE were done by colorimetric assays. Cell viability and the antioxidative effects such as DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lipid peroxidation (LP) were analyzed. ILC induction decreased cell viability in a dose-dependent manner, and the $XTT_{90}$ value (low cytotoxicity value) and $XTT_{50}$ value (high cytotoxicity value) were determined during ILC induction for 15 and 40 minutes, respectively. The butylated hydroxytoluene (BHT) antioxidant significantly increased cell viability damaged by the ILC-induced cytotoxicity. In the protective effect of KAE on ILC-induced cytotoxicity, KAE protected the ILC-induced cytotoxicity by the significant increase of cell viability, and also it showed DPPH-radical scavenging ability, SOD-like ability and inhibitory ability of LP. From these results, it is suggested that ILC induction showed cytotoxicity in these cultures and the oxidative stress is involved in the ILC-induced cytotoxicity. While, KAE prevented ILC-induced cytotoxicity by antioxidative effects. In conclusion, natural products like KAE may be a putative therapeutic agent for the treatment of disease associated with oxidative stress such as ischemia.

  • In the course of screening for antioxidant compounds by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, methanol extract of Cyperus difformis (Cyperaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of three phenolic compounds, rosmarinic acid (1), luteolin (2) and caffeic acid (3). Their structures were elucidated by spectroscopic studies. Three compounds showed the significant antioxidative effects on DPPH and nitric oxide radical scavenging effects, riboflavin and xanthin originated superoxide quenching activities. Among them, compound 3 showed the most significant antioxidative effects on those four antioxidant tests. Compound 3 showed better antioxidative activities than vitamin C.

Antimicrobial Effects of Flavone Analogues and Their Structure-Activity Relationships

  • It has been well known that the use of Saccharomyces cerevisiae can cause fungemia in critically ill patients and flavone shows an antimicrobial effect on S. cerevisiae. Therefore, we have investigated the activities of thirteen flavone analogues on S. cerevisiae in our studies. Because flavonoids including flavones have antioxidative effects, we try to carry out the activity studies of flavone analogues in vitro and in vivo. In addition, the relationships between the structures of flavone analogues and their biological activities, such as antimicrobial and antioxidative effects, were elucidated using Comparative Molecular Field Analysis calculations. Of the flavone analogues tested here, 3,2'-dihydroxyflavone showed both good antimicrobial and antioxidative activities.

Effect of Dried Powder and Juice of Aster Scaber on Lipid Metabolism and Antioxidative Capacity in Rats (참취의 건분 및 녹즙이 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder. Antioxidative Effects of Green Tea Powder Diet Against Ethanol-Induced Oxidative Damage in Rat Brain Regions (뇌 조직에서 알코올 투여에 대한 녹차 건분의 항산화 효과)

  • The present study investigated the protective effects of green tea against acute ethanol-induced lipid peroxidation and the change of antioxidative enzyme activities in various regions of rat brain : cortex, cerebellum, striatum and hippocampus. The following parameters were examined : malondialdehyde(MDA) levels and activities of superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px). Male Sprague-Dawley rats were given the experimental containing 1% green tea powder or control diet for 4 weeks, and at the end of feeding diet group received acute ethanol(5g/kg body weight) or equicaloric sucrose solution intragastrically. Green tea powder significantly decreased MDA levels in the striatum compared to control-non alcohol treated group to 1% green tea-non alcohol treated group without altering the antioxidative enzyme activities. Green tea resulted in a significant increase in GSH-Px activities in the hippocampus compared to either control-non alcohol treated group(0.043units/mg protein) or 1% green tea-non alcohol treated group(0.071units/mg protein). In conclusion, these results suggest that moderate consumption of green tea leaves can exert protective effects against ethanol-induced oxidative stress in brain regions, by reducing MDA concentrations in the striatum and enhancing GSH-Px activities in the hippocampus. Effects of acupuncture at Gansoo(B-18) and (Chungwan (CV-12) on Serum Lipid Composition and Antioxidative Capacity in Rats Fed Peroxidized Lipids (간수(B-18) 및 중완(CV-12) 자침이 과산화지질(過酸化脂質)을 급여(給與)한 흰쥐의 혈청지질(血淸脂質)강하 및 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objectives : To investigate the effects of acupuncture at Gansoo (B-18) and Chungwan (CV-12) on lipid metabolism in rats fed peroxidized lipid diet. Methods : Effects of acupuncture at Gansoo and Chungwan on plasma and liver lipid composition and antioxidative capacity were investigated in rats fed peroxidized lipids. Results : Although the level of plasma total cholesterol and triglyceride showed a tendency to decrease in the acupuncture group, the plasma HDL-cholesterol concentration showed no significant difference. The level of liver total cholesterol and triglyceride showed no significant difference in all treatment groups. Thiobarbituric acid( TBARS ) values in plasma and liver showed a tendency to decrease in the acupuncture groups. The plasma GOT and GPT activities showed low values in the acupuncture groups. The liver glutathione peroxidase, superoxide dismutase and catalase activity showed high values in the acupuncture groups. Conclusions : The results suggest that acupuncture at Gansoo (B-18) and Chungwan (CV-12) may have an influence on lipid metabolism via enhancing antioxidative capacity.

Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

Physical Characteristics and Antioxidative Capacity of Major Seaweeds

  • Seaweeds is a rich sources of dietary fibers exerting a number of physiological properties. However, the reported dietary fiber contents of seaweeds are not consistent and vary widely. Also. a limited number of studies on the biological effects of specific seaweeds have been reported. In this study, water-holding capacity, viscosity and antixidantive activity of major dietary seaweeds were measured to assess their physiological effects. Results showed that total dietary fiber contents ranged from 28 to 51% of dried weight, and large proportions of dietary fiber were insoluble fibers. Water-holding capacity was highest in sea mustard being 1310% , while laver, sea tangle, and green laver exhibited 943, 854 and 815%, respectively. The viscosity of seaweed samples was 20 to 40 cP in sea mustard and sea tangle, while laver and green laver possessed much lower values. All seaweed samples revealed a weak, albeit significant electron donating ability. Also, lipid peroxidation was reduced by 7 to 18%. However, there was no difference in antioxidative activity among seaweeds and sample concernations used. These results imply that most commonly used seaweeds possibly exert parts of their physiological effects through their water-holding, gel-forming , and/or antioxidative activities.

Antioxidative and Cytoprotective Effects of Isoflavones Isolated from Pueraria thunbergiana Flowers (갈화에서 분리한 이소플라본의 항산화 및 세포보호효과)

  • Antioxidative and cytoprotective effects of tectorigenin and glycitein isolated from the pueraria thunbergiana and its derivative, genistein, were determined. Among these three compounds, tectorigenin and glycitein bearing 6-methoxyl groups in both isoflavones showed significant free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase (XOD) generating superoxide anion radical. Tectorigenin only showed a slight inhibitory effect on XOD. We further studied the inhibitory effects of these isoflavones on the lipid peroxidation in rat liver microsomes induced by enzymatic and non-enzymatic methods. Each of them exhibited inhibitory effect on both ascorbic $acid/Fe^{2+}-{\;}and{\;}ADP/NADPH/Fe^{+3}-induced$ lipid peroxidation. Moreover, tectorigenin exhibited the highest protection of hydrogen peroxide damage on HepG2 and Vero cells among the three isoflavones, in the cytoprotective assay. It was suggested that the pattern of antioxidative and cytoprotective effect of isoflavones could be crucially by the aromatic substitution of oxygen-containing groups.

Antioxidative Effects of Volatile Oil and Oleoresin Extracted from Rosemary, Sage, Clove and Nutmeg (Rosemary, Sage, Clove 및 Nutmeg의 휘발성 및 비휘발성 성분의 항산화성)

  • This study was designed to investigate the antioxidative effects of volatile oil and oleoresin of rosemary mary (Rosmarinus officinalis Linne), sage (Salvia officinalis Linne), clove (Syzygium aromaticum Merrill) and nutmeg (Myristica fragrans Houttuyn) using the Rancimat and POV test. The antioxidative effects of steam volatile oils of rosemary, sage clove. and nutmeg were not statistically significant, at the levels of 0.01, 0.05, 0.1% added to lard. The antioxidative effects of oleoresin of above 4 spices were increased with the oleoresin concentration. The comparison of the results between Rancimat and POV test of above 4 spices was almost same.
