  • Inordinate stress causes disorders of various systems in humans and activates defense mechanisms to maintain homeostasis in the body. Sleep is a vital, highly organized process regulated by complex systems of neuronal networks and neurotransmitters. Sleep is an essential biological process whose underlying regulating involves numerous anatomical structures and biochemical substances that can be compromised by stress and by the immune system. Gamma-amino butyric acid (GABA) is the main inhibitory neurotransmitter of the central nervous system, and activation of GABAA receptors is known to favor sleep. This study was conducted to evaluate the possible application of Lactobacillus brevis BJ20 fermentation to improve the functional qualities of sea tangle Saccharina japonica and oyster Crassostrea gigas. Antioxidant activity was determined by assaying levels of radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide. L. brevis BJ20 fermentation of sea tangle and oyster enhanced both antioxidant and antiinflammatory activities. These results suggested that L. brevis BJ20 fermented sea tangle and oyster could be used for alleviation of stress and to promote sleep.

Neuroprotective Effects of N-Acetyldopamine Dimers from Cicadidae Periostracum

  • The chemical investigation of the 90% EtOH extract from Cicadidae Periostracum led to the isolation and identification of seven known N-acetyldopamine dimers (1-7). These compounds were identified by comparing mass spectrometry data and NMR spectroscopic data with those previously reported. In this study, complete interpretation of 1D and 2D NMR data of 1 and 2 were reported for the first time. In addition, compounds 3 and 4 were isolated from this material for the first time. All isolates were obtained as racemic mixtures, as confirmed by chiral HPLC. Furthermore, we evaluated the neuroprotective activities of compounds 1-7 and found that compounds 1, 5, and 6 significantly attenuated rotenone-induced death of SH-SY5Y neuroblastoma cells at a concentration of 100 μM. Parallel to this result, compounds 3 and 6 displayed antioxidant effects in the cytoplasm, as determined by CM-H2DCFDA fluorescence intensity, while compounds 1 and 5 showed antioxidant effects in the mitochondria, as assessed by MitoSox fluorescence intensity. Overall, these results suggest that some of these compounds protect neuroblastoma cells by ameliorating the release of reactive oxygen species. Further studies are warranted to elucidate the underlying mechanisms by which these compounds exhibit antioxidant and neuroprotective actions.

고지방식이로 유도한 신장의 산화적 스트레스에 대한 자화지정(紫花地丁)의 항산화 효과 (Antioxidant Effect of Viola mandshurica W. Becker on the High Fat Diet-Induced Renal Oxidative Stress)

  • The prevalence of renal disease is increased with the overweight and obesity. High fat diet-associated oxidative stress increases production of reactive oxygen species (ROS) and induces apoptosis. There are two types of antioxidant defense mechanisms for oxidative stress. One is the enzyme defense mechanism by antioxidant enzymes such as superoxide dismutase (SOD), and catalase (CAT). The other is non-enzyme defense mechanism by signaling molecules such as nuclear factor-like 2 (Nrf-2), HO-1. In this study, we induced obesity in mice with high fat diet for six weeks and thereafter administered orally Viola mandshurica for 4 weeks. V. mandshurica is known to clear heat, detoxify and cool blood, and subside a swelling effect. In the V. mandshurica administered group, the immunoreactive signal of the Tunel staining was weaker than that of obesity group. Proapoptotic Bax, caspase 3 immunoreactives of the V. mandshurica administered group was lower than those of obesity group, whereas anti-apoptotic Bcl-2 immunoreactity was higher in the V. mandshurica administered group. Antioxidant enzyme mechanism such as superoxide dismutase 2 (SOD2), catalase (CAT) immunoreactives of the V. mandshurica administered group and Antioxidant non-enzyme mechanism such as Nuclear factor-like 2 (Nrf2), Heme Oxygenase 1 (HO-1) immunoreactives of the V. mandshurica administered group was higher than those of obesity group. These results demonstrate that V. mandshurica had the antioxidant and anti-apoptosis effects on obese mice.

Protective Effect Against Hydroxyl Radical-induced DNA Damage and Antioxidant Mechanism of [6]-gingerol: A Chemical Study

  • [6]-Gingerol is known as the major bioactive constituent of ginger. In the study, it was observed to effectively protect against ${\bullet}OH$-induced DNA damage ($IC_{50}$ $328.60{\pm}24.41{\mu}M$). Antioxidant assays indicated that [6]-gingerol could efficiently scavenge various free radicals, including ${\bullet}OH$ radical ($IC_{50}$ $70.39{\pm}1.23{\mu}M$), ${\bullet}O_2{^-}$ radical ($IC_{50}$ $228.40{\pm}9.20{\mu}M$), $DPPH{\bullet}$radical ($IC_{50}$ $27.35{\pm}1.44{\mu}M$), and $ABTS{^+}{\bullet}$radical ($IC_{50}$ $2.53{\pm}0.070{\mu}M$), and reduce $Cu^{2+}$ ion ($IC_{50}$ $11.97{\pm}0.68{\mu}M$). In order to investigate the possible mechanism, the reaction product of [6]-gingerol and $DPPH{\bullet}$ radical was further measured using HPLC combined mass spectrometry. The product showed a molecular ion peak at m/z 316 $[M+Na]^+$, and diagnostic fragment loss (m/z 28) for quinone. On this basis, it can be concluded that: (i) [6]-gingerol can effectively protect against ${\bullet}OH$-induced DNA damage; (ii) a possible mechanism for [6]-gingerol to protect against oxidative damage is ${\bullet}OH$ radical scavenging; (iii) [6]-gingerol scavenges ${\bullet}OH$ radical through hydrogen atom ($H{\bullet}$) transfer (HAT) and sequential electron (e) proton transfer (SEPT) mechanisms; and (iv) both mechanisms make [6]-gingerol be oxidized to semi-quinone or quinone forms.

Profiling of Gene Expression in Human Keratinocyte Cell Line Exposed to Quantum Dot Nanoparticles

  • Quantum Dot (QD) nanoparticles are used in various industrial applications, such as diagnostic, drug delivery, and imaging agents of biomedicine. Although QDs are extensively used in many medical science, several studies have been demonstrated the potential toxicity of nanoparticles. The first objective of this study was to investigate the nanotoxicity of QDs in the HaCaT human keratinocyte cell line by focusing on gene expression pattern. In order to evaluate the effect of QDs on gene expression profile in HaCaT cells, we analyzed the differential genes which related to oxidative stress and antioxidant defense mechanisms by using human cDNA microarray and PCR array. A human cDNA microarray was clone set, which was sorted for a list of genes correlated with cell mechanisms. We tried to confirm results of cDNA microarray by using PCR array, which is pathway-focused gene expression profiling technology using Real-Time PCR. Although we could not find the exactly same genes in both methods, we have screened the effects of QDs on global gene expression profiles in human skin cells. In addition, our results show that QD treatment somehow regulates cellular pathways of oxidative stress and antioxidant defense mechanisms. Therefore, we suggest that this study can enlarge our knowledge of the transcriptional profile and identify new candidate biomarker genes to evaluate the toxicity of nanotoxicology.

Dysregulation of NRF2 in Cancer: from Molecular Mechanisms to Therapeutic Opportunities

  • Nuclear factor E2-related factor 2 (NRF2) plays an important role in redox metabolism and antioxidant defense. Under normal conditions, NRF2 proteins are maintained at very low levels because of their ubiquitination and proteasomal degradation via binding to the kelch-like ECH associated protein 1 (KEAP1)-E3 ubiquitin ligase complex. However, oxidative and/or electrophilic stresses disrupt the KEAP1-NRF2 interaction, which leads to the accumulation and transactivation of NRF2. During recent decades, a growing body of evidence suggests that NRF2 is frequently activated in many types of cancer by multiple mechanisms, including the genetic mutations in the KEAP1-NRF2 pathway. This suggested that NRF2 inhibition is a promising strategy for cancer therapy. Recently, several NRF2 inhibitors have been reported with anti-tumor efficacy. Here, we review the mechanisms whereby NRF2 is dysregulated in cancer and its contribution to the tumor development and radiochemoresistance. In addition, among the NRF2 inhibitors reported so far, we summarize and discuss repurposed NRF2 inhibitors with their potential mechanisms and provide new insights to develop selective NRF2 inhibitors.

Mechanisms involved in adenosine pharmacological preconditioning-induced cardioprotection

  • Adenosine is a naturally occurring breakdown product of adenosine triphosphate and plays an important role in different physiological and pathological conditions. Adenosine also serves as an important trigger in ischemic and remote preconditioning and its release may impart cardioprotection. Exogenous administration of adenosine in the form of adenosine preconditioning may also protect heart from ischemia-reperfusion injury. Endogenous release of adenosine during ischemic/remote preconditioning or exogenous adenosine during pharmacological preconditioning activates adenosine receptors to activate plethora of mechanisms, which either independently or in association with one another may confer cardioprotection during ischemia-reperfusion injury. These mechanisms include activation of $K_{ATP}$ channels, an increase in the levels of antioxidant enzymes, functional interaction with opioid receptors; increase in nitric oxide production; decrease in inflammation; activation of transient receptor potential vanilloid (TRPV) channels; activation of kinases such as protein kinase B (Akt), protein kinase C, tyrosine kinase, mitogen activated protein (MAP) kinases such as ERK 1/2, p38 MAP kinases and MAP kinase kinase (MEK 1) MMP. The present review discusses the role and mechanisms involved in adenosine preconditioning-induced cardioprotection.

위식도 역류질환에 대한 한의학 연구 경향 분석: 국내 논문을 중심으로 (The Review of Domestic Research on Traditional Korean Medicine for Gastroesophageal Reflux Disease)

  • This study was conducted to examine the current status of traditional korean medicine studies on gastroesophageal reflux disease in Korea, identify deficiencies, and suggest the direction of future medicine research methods to lay the foundation for traditional korean medicine treatment. All domestic papers on the korean traditional treatment of gastroesophageal reflux disease were selected among the literature published until August 2022 in six domestic databases. A total of 52 selected research data were classified into experimental research papers, clinical research papers, and review papers. In experimental papers, to evaluate the effectiveness of treatment, improvement of esophageal mucosal lesions, anti-inflammatory mechanisms, antioxidant mechanisms, esophageal mucosal protection mechanisms, gastric peristalsis control, and gastric acid secretion inhibition mechanisms were used as evaluation measures. In the clinical research paper, the basis for diagnosis of cases was clinical symptoms through medical history listening and diagnosis through visits to hospitals in the past. The average treatment period was 40.7 days, and the duration of treatment was not significantly affected by the duration of the disease. The most widely used Korean medicine treatment intervention was herbal medicine. There were 3 literature review studies, 3 systematic literature review and meta-analysis studies, 1 comparative review study for clinical trial guideline development, all using Chinese papers. This study included all domestic papers on gastroesophageal reflux disease to identify the research trend of the Korean oriental medicine community, and based on this, it is meaningful to confirm areas that need to be supplemented in future research plans.

SH-SY5Y 세포주에서 하고초, 금은화, 황금 에탄올 추출물의 6-OHDA로 유도된 산화적 손상에 미치는 영향 (The Effect of Ethanol Extracts of Herba Prunellae, Flos Lonicerae and Radix Scutellaria on 6-OHDA Induced Oxidative Damage in the SH-SY5Y Cell Line)

  • Objectives : Categorized as 'cheongyeol' herbs, Herba Prunellae, Flos Lonicerae and Radix Scutellaria have been proven to have effect on degenerative brain disease, cerebrovascular disease and brain tumor because of their anti inflammation, antioxidant, or anticancer effects. In this study, we studied activity against reactive oxygen species and anti inflammation effect of these three 'Cheongyeol' herbs. Methods : We measured each herb's yield of ethanol extracts, phenolic contents and activities against DPPH, hydroxyl radical and superoxide anion. Also through 6-hydroxydopamine (6-OHDA) induced oxidative damage in SH-SY5Y human neuroblastoma cell line, we measured antioxidant effect and NO activity of the three herbs. From the three herbs, we chose Prunella Herba, which showed the highest antioxidant effect, and studied its cell survival rate and anti inflammation effect through COX-2 and iNOS. Results : All three herbs showed significant results, and especially Prunella Herba showed significant effect on phenol contents, antioxidant effect on various active oxygen and antioxidant, and anti inflammation effect through cell line. Conclusions : Further study of the origin concept of 'cheongyeol' and research into specific mechanisms and role in treatment of cranial nerve disease, seems warranted.

FDY003의 항산화활성 및 표준화 연구 (Study on Antioxidant Activity and Standardization of FDY003)

  • Objectives: FDY003 is a raw material for medicine consisting of a natural product that is expected to have the advantages of low side effects and high efficacy. In this study, we predict the efficacy and the standardization of the drug by method validation of anticipated index compounds and the measurement of antioxidant activity. Methods: FDY003 is prepared by extracting and purifying 70% of ethyl alcohol (EtOH). The method validation of cordycepin and chlorogenic acid was determined by high-performance liquid chromatography-photo diode array (HPLC-PDA) and the content of FDY003 was calculated. In order to monitor the biological activity of FDY003, antioxidant activity was measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric-reducing antioxidant power (FRAP). The equivalent values of antioxidants such as trolox, ascorbic acid, gallic acid, and caffeic acid were measured by ABTS and FRAP. Results: Chlorogenic acid and cordycepin were both found suitable for method validation in HPLC and FDY003 containing 9.92±0.50 and 17.97±0.27 ㎍/g, respectively. In DPPH, the electron donating ability (EDA) value of FDY003 was increased in a concentration dependent manner. FDY003 confirmed antioxidant activity by ABTS and FRAP. Conclusions: FDY003 contains certain components including cordycepin and chlorogenic acid and has antioxidant ability by various mechanisms. Therefore, it is expected that FDY003 is capable of various physiological activities including anti-cancer activity.
