Antioxidant Activity of Persicaria perfoliata Extracts (며느리배꼽 추출물의 항산화 활성)

  • In this study, the antioxidative effects, inhibitory effects on tyrosinase, elastase of Persicaria perfoliata extracts were investigated. The deglycosylated fraction of extract ($12.38{\mu}g$/mL) showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of P. perfoliata extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extract ($0.35{\mu}g$/mL) showed the most prominent ROS scavenging activity. The protective effects of P. perfoliata extract/fractions on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. perfoliata extracts suppressed photohemolysis in a concentration dependent manner ($1{\sim}50{\mu}g$/mL) except the deglycosylated fraction of extract. The inhibitory effect of P. perfoliata extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects ($IC_{50}$) on tyrosinase were determined with ethyl acetate fraction of P. perfoliata extract ($136.00{\mu}g$/mL) and deglycosylated fraction of extract ($68.10{\mu}g$/mL). Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. Inhibitory effects ($IC_{50}$) on elastase were determined with ethyl acetate fraction of P. perfoliata extract ($67.20{\mu}g$/mL) and deglycosylated fraction of extract ($43.50{\mu}g$/mL). These results indicate that extract/fractions of P. perfoliata can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Extract/fractions of P. perfoliata can be applicable to new functional cosmetics for antioxidant, antiaging.

o-Dihydroxyisoflavone Derivatives from Highly Aged Korean Fermented Soybean Paste by Jang Yang Process and Its Biological Activity (장양(藏釀) 기술이 사용된 전통 고숙성 된장에서 유래한 o-Dihydroxyisoflavone 유도체의 생리활성)

  • Doenjang (Korean fermented soybean paste) is a unique fermented food in Korea. It has been traditionally manufactured from soybeans, by Jang Yang process. We focused on the newly formed compound in highly aged Doenjang and its biological activity. One new o-dihydroxyisoflavone, 7,3',4'-trihydroxyisoflavone and two known o-dihydroxyisoflavone derivatives were isolated from 5-year-old Doenjang and evaluated as potent antioxidant and whitening effect by comparing with other known isoflavone. 7,8,4'-Trihydroxyisoflavone (compound 1), 7,3',4'-trihydroxyisoflavone (compound 2) and 6,7,4'-trihydroxyisoflavone (compound 3) inhibited DPPH (diphenyl-1-picrylhydrazyl) formation by 50 % at a concentration of $21.5{\pm}0.2$, $28.7{\pm}0.4$ and $32.6{\pm}0.6$ ($IC_{50}$) respectively, whereas daidzein showed weak DPPH radical scavenging activity. In superoxide scavenging effect were measured in one assay. Compound 1 ($IC_{50}=18.10{\pm}0.2{\mu}M$) and 2 ($IC_{50}=10.54{\pm}0.4{\mu}M$) show significant inhibitory activity and greater effect than L-ascorbic acid. But compound 3 and daidzein showed lower inhibition activity. Also, o-dihydroxyisoflavone derivatives evaluated as potent inhibitors on tyrosinase activity and melanin formation in melan-a cells. Compound 1 ($IC_{50}=11.21{\pm}0.2{\mu}M$), compound 2 ($IC_{50}=5.23{\pm}0.6{\mu}M$) exhibited significant inhibitory effect on tyrosinase activity. Furthermore, those compounds are significantly suppressed the cellular melanin formation by 50 % at a concentration of $12.23{\pm}0.7{\mu}M$ (1) and $7.83{\pm}0.7{\mu}M$ (2). This result suggests that 7,3',4'-trihydroxyisoflavone from highly aged Doenjang could be used as an active ingredient for cosmetics.

Antioxidative Effect and Active Component Analysis of Gnaphalium affine D. DON. Extracts (떡쑥 추출물의 항산화 효과 및 활성 성분 분석)

  • In this work, the antioxidative effects and active component analysis of Gnaphalium affine D. DON. (G. affine) extracts were investigated. All experiments were performed with 70% ethanol extract, ethyl acetate fraction and aglycone fraction of the G. affine extract. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of ethyl acetate fraction ($6.15{\mu}g/mL$) of the G. affine was higher than that of (+)-${\alpha}$-tocopherol ($8.89{\mu}g/mL$), which is known as a reference control. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the 70% ethanol extract ($1.60{\mu}g/mL$), ethyl acetate fraction ($0.075{\mu}g/mL$) and aglycone fraction ($2.28{\mu}g/mL$) of extract of G. affine on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system using the luminol-dependent chemiluminescence assay were much higher than that of L-ascorbic acid ($6.88{\mu}g/mL$). The cellular protective effects of 70% ethanol extract (${\tau}_{50}$ = 52.0 min) and aglycone fraction of the extract (${\tau}_{50}$ = 60.6 min) on the $^1O_2$-induced cellular damage of human erythrocytes were exhibited the higher protective effect than (+)-${\alpha}$-tocopherol (${\tau}_{50}$ = 38.0 min), known as a lipophilic antioxidant. TLC and HPLC were used to analyse active components in the aglycone fraction of the extract. Results showed that luteolin and apigenin were main components. These results suggest that the G. affine extract can be applied to an effective antioxidant in scavenging ROS including radicals.

Melanogenesis Inhibitory and Antioxidant Activities of Phellinus baumii Methanol Extract (장수진흙버섯 메탄올 추출물의 멜라닌 생성 저해작용)

  • Phellinus baumii is a medicinal mushroom used in Asian countries for a long period of time. The purpose of this study was to investigate the skin whitening activities of methanol extracts from fruiting bodies of P. baumii. To evaluate the antioxidant activities of the extract, polyphenol and flavonoid contents, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and chelating activity on ferrous ions were studied. For assay of skin whitening activities, tyrosinase and DOPA inhibitory activities, and tyrosinase and melanin synthesis inhibitory activities of B16/F10 melanoma cells treated with the methanol extract were investigated. The total polyphenol content of P. baumii methanol extract was 4.19. DPPH scavenging ability of P. baumii methanol extract was 88.26% in $25{\mu}g/mL$ concentration. We tested tyrosinase inhibitory activity and melanin contents in melanoma cells. The tyrosinase activity was inhibited to 65.17% at the concentration of $125{\mu}g/mL$ and melanin synthesis was inhibited to 57.61% at the concentration of $25{\mu}g/mL$. Overall, the experimental results showed that P. baumii methanol extract had inhibitory activities of tyrosinase and melanin synthesis by dose dependent manner in B16/F10 melanoma cells. Strong ultra-violet absorption spectra in the range of 270~370 nm indicated that ethanol extract of P. baumii could protect the skin from UV. Therefore, P. baumii methanol extract might be used for development of skin whitening, anti-UV and skin care agents.

Antioxidative Effect and Component Analysis of Niaouli (Melaleuca quinquenervia) Leaf Extracts (니아울리 잎 추출물의 항산화 효과 및 성분 분석)

  • The antioxidative effects and component analysis of the Melaleuca quinquenervia leaf extracts were investigated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from dried M. quinquenervia leaves. The DPPH (1,1-phenyl-2-picrylhydrazyl) scavenging activity ($FSC_{50}$) of ethyl acetate fraction ($10.05{\mu}g/mL$) of M. quinquenervia leaf extracts was similar to (+)-${\alpha}$-tocopherol($8.89{\mu}g/mL$) known as a typical antioxidant. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethyl acetate fraction ($1.61{\mu}g/mL$) and aglycone fraction ($1.07{\mu}g/mL$) of leaf extracts of M. quinquenervia on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were similar to that of L-ascorbic acid ($1.50{\mu}g/mL$). The cellular protective effect of the extracts on the rose bengal sensitized photohemolysis of human erythrocytes was increased in a concentration dependant manner ($1{\sim}50{\mu}g/mL$). Especially, the cellular protective effects of Aglycone fraction (${\tau}_{50}=158.80min$) and 50% Ethanol extract (${\tau}_{50}=50.1{\pm}0.2min$) on the $^1O_2$-induced cellular damage of human cells were exhibited the higher than (+)-${\alpha}$-tocopherol (${\tau}_{50}=38.0min$). TLC and HPLC were used to analyse active components in the ethylacetate fraction of the extracts. Results showed that avicularin and quercetrin were active components of the extracts. These findings suggest that the M. quinquenervia leaf extracts can be applied to new cosmetics products as an effective antioxidant ingradient.

Antioxidant capacity and hepatoprotective effect of ethyl acetate fraction from shoot of Aralia elata on alcohol-induced cytotoxicity (두릅 아세트산 에틸 분획물의 산화방지 효과 및 알코올에 대한 간세포 보호효과)

  • To evaluate physiological effect of Aralia elata, in vitro antioxidant activity and hepatic protective effects were investigated. Ethyl acetate fraction from Aralia elata (EFAE) had higher total phenolic content than other fractions (n-hexane, chloroform, and distilled water layers). EFAE also showed significantly greater radical scavenging activity against 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH), than other fractions. Moreover, EFAE showed dose-dependent inhibitory effect of malondialdehyde (MDA). Hepatoprotective effects of EFAE against ethanol- and $H_2O_2$-induced oxidative stress and cytotoxicity in H4IIE and HepG2 hepatic cells were examined using 2',7'-dichlorofluorescein diacetate (DCF-DA) and 3-[4,5-dimethythiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay. The results showed that EFAE reduced cellular oxidative stress, and increased hepatic cell viability. In addition, EFAE inhibited ethanol-induced lipid accumulation in HepG2 cells. Finally, physiological substances of EFAE were analyzed using high performance liquid chromatography (HPLC), and the major bioactive compounds identified were 3,5-dicaffeoylquinic acid and chlorogenic acid.

Antioxidative and Antimicrobial Activities of Methanol Extract from Rosmarinus officinalis L. and Their Fractions (로즈마리 추출물 및 분획물의 항균·항산화 활성)

  • This study was performed to evaluate the antioxidant and antimicrobial activity of methanol extract from Rosmarinus officinalis L. and its fractions. The ethyl acetate fraction of rosemary had a higher antioxidant activity in both DPPH ($3.22\;{\mu}g/ml$) and ABTS ($5.05\;{\mu}g/ml$) compared to other extracts and fractions. Based on the results of the FRAP assay, the ethyl acetate fraction of rosemary showed a value of $5.9{\pm}0.3\;{\mu}M/{\mu}g$, and buthanol fraction and rosmarinic acid exhibited values of $4.8{\pm}0.2\;{\mu}M/{\mu}g$ and $5.1{\pm}0.1\;{\mu}M/{\mu}M$, respectively. Measurements of the antimicrobial activities of the extracts, fraction against gram positive, negative bacteria revealed that the methanol extract, hexane, ethyl acetate, and chloroform fraction of rosemary caused Staphylococcus aureus and Escherichia coli to form clear zones greater than 12 mm. Furthermore, the methanol extract and chloroform fraction showed high antibacterial activity, with inhibition zone exceeding 13 mm. The methanol extract and chloroform fraction of rosemary had broad antimicrobial spectrums and low MIC values. Therefore, methanol extracts of rosemary could serve as potential antibacterial agents to inhibit pathogen growth in food and hand sanitizers.

Protective Effect of Isoflavone, Genistein from Soybean on Singlet Oxygen Induced Photohemolysis of Human Erythrocytes ($^1O_2$으로 유도된 사람 적혈구의 광용혈에 있어서 대두의 아이소플라본인 제니스테인의 보호작용)

  • Protective effects of natural components including genistein (4',5,7-trihydroxyisoflavone) from Glycine max MERRILL on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. Genistein $(10{\sim}100\;{\mu}m)$ suppressed photohemolysis in a concentration-dependent manner, and was more effective than the lipid peroxidation chain blocker, ${\alpha}$-tocopherol (Vit. E). Glycoside of genistein, genistin, the water-soluble antioxidant, L-ascorbate, and the iron chelator, myo-inositol hexaphosphoric acid dodecasodium salt (sodium phytate) did not exhibit protective effect against photohemolysis. L-Ascorbate and sodium phytate stimulated photohemolysis at high concentration $(500\;{\mu}m)$. ${\alpha}$-Carotene 3,3'-diol (lutein), a singlet oxygen $(^1O_2)$ quencher, exhibited pronounced protective effect, an indication that $^1O_2$ is important in photohemolysis sensitized by rose-bengal. Reactive oxygen scavenging activities $(OSC_{50})$ of natural antioxidants including genistein on reactive oxygen species (ROS) generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were in the order of sodium phytate > L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin. $OSC_{50}$ value of genistein, genistin, ${\alpha}$-tocopherol, L-ascorbate, and sodium phytate were 41.0, 109.0, 9.0, 5.2, and $0.56{\mu}m$ respectively. The order of free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity $(FSC_{50})$ was L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin. These results indicate that genistein can function as an antioxidant in biological systems, particularly skin exposed to solar UV radiation by scavenging $^1O_2$ and other ROS, and to protect cellular membranes against ROS.

Physiological Activities of Commercial Instant Curry Powders and Individual Spices (시판 instant curry 및 curry 사용원료의 생리활성)

  • Physiological activities of hot water extracts of 10 commercial instant curry powders and 6 spices, were investigated. All spice extracts except ginger showed significant antioxidant activities on the autoxidation of linoleic curry acid (p<0.01). Antioxidant activities of clove and fennel were significantly higher than ${\alpha}-tocopherol$, instant curry powders, and other spices, Red pepper $(52.8{\pm}2.13%)$, clove, and coriander showed significant inhibitory activities against angiotensin-I-converting enzyme (p<0.001). Cytotoxic effects of instant curry powder and spices against human cancer cell lines were examined through MTT assay. Black pepper $(29.31{\pm}2.21%\;cytotoxic\;rate)$ and cardamon $(19.41{\pm}3.92%)$ were effective against MCF-7 (p<0.01), Clove $(42.92{\pm}5.57%)$ against HeLa (p<0.01). Ginger $(34.21{\pm}1.11%)$, cardamon, and black pepper against A172 (p<0.001), garlic $(82.88{\pm}0.53%)$ against SN12C (p<0.001), garlic $(71.63{\pm}0.38%)$, red pepper, ginger, fenugreek, SPC, cumin, and MPC against SNU-638 (p<0.001), and cassia $(82.84{\pm}16.92%)$ against A549 (p<0.001).

Antioxidants in Serum and Induced Sputum of COPD Patients (만성폐쇄성폐질환 환자의 혈청 및 유도객담에서의 항산화제)

  • Background : Although an oxidants and antioxidants imbalance has been considered in the pathogenesis of chronic obstructive pulmonary disease (COPD), there is a paucity of reports focussing on the smoking-induced changes of oxidants and antioxidants in COPD. Method : The concentration of antioxidants (ascorbic acid, uric acid, retinol, and $\alpha$- & $\gamma$-tocopherol) was measured in the serum and induced sputum of 30 healthy controls and 34 stable COPD patients using high performance liquid chromatography (HPLC). The inhibition of lipid peroxidation as an index of antioxidant capacity was measured in the serum by a TBA assay. Results : The serum concentration of ascorbic acid, $\alpha$-tocopherol, and retinol were significantly lower in the patients with COPD than in healthy controls ($484.8{\pm}473.3$ vs $1497.8{\pm}819.2\;pmol/L$, $48.38{\pm}17.34$ vs $73.96{\pm}26.29\;pmol/L$, p<0.001, and $9.51{\pm}8.33$ vs $15.01{\pm}5.88\;pmol/L$, p<0.05, respectively, mean$\pm$SD). However, there were little differences in the ascorbic acid and uric acid concentrations in the induced sputum between the COPD patients and the controls. The induced sputum to serum ratio of ascorbic acid was significantly higher in COPD patients compared with healthy control (0.375 vs 0.085, p<0.05). In the normal controls, the serum ascorbic acid concentration was lower in smokers than in nonsmokers ($1073{\pm}536$ vs $1757{\pm}845\;pmol/L$, p<0.05), but the level was still higher than that of the COPD patients (p<0.05). The serum retinol levels were correlated with $FEV_1$ in COPD patients (r=0.58, p<0.05). The products of lipid peroxidation were increased in normal smokers and COPD compared with norma1 nonsmokers ($115.56{\pm}19.93$ vs $120.02{\pm}24.56$ vs $91.87{\pm}20.71\;{\mu}mol/{\mu}mol$ Pi of liposome, p<0.05). Conclusion : Cigarette smoking may induce the dep1etion of serum antioxidants and this depletion of antioxidants is suggested to play a role in the pathogenesis of COPD.
