The antioxidant activities of brownish natural dyeing agents, extracted from seven kinds of plants, were tested. Total polyphenol content, DPPH and ABTS radical scavenging activities, and singlet oxygen quenching effect were determined for hot water extracts and floral waters of plants. DPPH and ABTS radical scavenging activites increased with increasing amounts of the extracts from Uncaria gambir R. and Terminalia chebula R. displayed remarkable scavenging effects at concentrations below 0.1 mg/mL, in comparison with the positive control, ascorbic acid. However, antioxidant effects of the floral water, obtained from steam distillation of tested plants, were inefficient at concentration below 0.2 mg/mL. In particular, the natural dyeing agent effectively suppressed singlet oxygen induced by photosensitizer in in vitro assay systems. The concentrations ($IC_{50}$) required to exert 50% of singlet oxygen were 120 and $190{\mu}g/mL$ for hot water extracts from Uncaria gambir R. and Phellinus linteus, respectively. Among all the tested samples, the Uncaria gambir R. and Phellinus linteus extracts contained higher amount of total phenolic contents. The results suggest that naturally occurring dyeing agents are beneficial as natural antioxidants, encouraging further extensive studies.
Aralia continentalis Kitagawa (Araliaceae), known as "Dokwhal" in Korea, has been widely used in traditional Korean medicine for analgesia, neuralgia, sweating, and rheumatism. The biological activity was estimated with methanol extracts of from cultivated roots and adventitious roots of A. continentalis. DPPH and ABTS activities showed the highest activity in methanol extract of adventitious roots at 175.6 and $279.7{\mu}g/mL$ ($RC_{50}$), respectively. Antioxidant activity of methanol extract of the adventitious roots was higher than other samples. In the antibacterial activity assay (paper disc method), the methanol extract of adventitious roots showed activity against S. aureus, S. epidermidis, B. subtilis, and S. enterica. This study demonstrated that adventitious roots of A. continentalis, which was produced for the first time in the roots of A. continentalis, can be used as a natural antioxidant and antibacterial agents.

Melatonin, which is produced from tryptophan, exerts various biological functions, including the regulation of circadian rhythm, sedative agents, and antioxidant ability. Therefore, we conducted two experiments with early-weaned rats and pigs to investigate the antioxidant and sedative effects of melatonin. In the rat experiment, a total of 42 rats (21 days old) were used, and the antioxidant capacity was determined. Next, we used 120 early-weaned piglets (21 days old) to conduct a 5-week experiment to evaluate the reductive effect of melatonin on energy-wasting movement, including roaming and fight states. Dietary melatonin supplementation significantly improved growth in both rats and pigs compared to the control groups. Additionally, rats fed a melatonin-supplemented diet showed advanced antioxidant capacity with a decrease in hepatic malondialdehyde concentration compared to rats fed a basal diet. Moreover, dietary melatonin ingestion increased resting and feeding behaviors and reduced roaming and fight behaviors during Days 8-21 compared to the control diet group. Collectively, early weaned animals given dietary melatonin supplementation showed improved growth through upregulation of hepatic antioxidant capacity and minimization of energy-wasting behavior, including roaming and fight states, after pigs' social hierarchy establishment.

In this study, derivatives of trimethoxybenzene were investigated as inhibitors of melanogenesis. We examined the effects of methyl 3,4,5-trimethoxybenzoate (MTB), ethyl 3,4,5-trimethoxybenzoate (ETB), methyl 3,4,5-trimethoxycinnamate (MTC), and ethyl 3,4,5-trimethoxycinnamate (ETC). First, the inhibitory effects of these agents on melanin production were evaluated using α-melanocyte-stimulating hormone (α-MSH)-stimulated B16F10 melanoma cells. We found that all derivatives decreased α-MSH-induced melanin production in B16F10 melanoma cells; ETC showed a strong inhibitory effect at half of the concentration of the other derivatives. As tyrosinase is considered a key enzyme of melanogenesis, we also examined whether the derivatives inhibited tyrosinase activity. MTC and ETC reduced mushroom tyrosinase activity and expression levels of α-MSH-induced B16F10 cellular tyrosinase protein. Inhibitory effects of all derivatives on α-MSH-induced B16F10 cellular tyrosinase activity were shown in a dose-dependent manner. Additionally, the derivatives were exposed to diphenylpicrylhydrazyl free radical to examine their antioxidant characteristics. All derivatives showed considerable antioxidant activity, which was 2-fold higher than that of arbutin. In conclusion, the trimethoxybenzene derivatives, including MTB, ETB, MTC, and ETC exerted anti-melanogenic and antioxidant effects on α-MSH-stimulated melanogenesis, demonstrating their potential for use as novel hypopigmenting agents and antioxidants.
EGCG[(-)-epigallocatechin gallate], is a major component of green tea has been considered as a major antioxidant constituent. It has been considered as potential chemopreventive and chemotherapeutic agents. However, very little is known about the cellular actions by which EGCG mediates its therapeutic effects. Various aspects of antioxidant activity of EGCG were evaluated in this study. EGCG itself did not show significant cytotoxicity. Significant 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was observed in all ranges of concentration ($0.8-100{\mu}g/ml$) used in this study. Protective effect of EGCG against hydrogen peroxide induced cell death was observed. Relatively high lipid peroxidation inhibitory activity were detected ($IC_{50}$ was about $20{\mu}g/ml$). EGCG also dose-dependently enhanced the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) in V79-4 cells. In concentrations of $100{\mu}g/ml$ of EGCG, activities of SOD, CAT and GPX were measured as 36.9 U/mg of protein, 22.9 U/mg of protein and 17.8 U/mg of protein, respectively. When these values were compared with those of the control groups (24.9 U/mg of protein, 14.9 U/mg of protein and 11.7 U/mg of protein), the relative increases were calculated as 48, 54 and 52%, respectively. Taken together, our findings suggest that EGCG can act as an antioxidant by scavenging radicals and enhancing antioxidant enzyme activities.
The antibacterial and antioxidant potentials of extruded ginseng extract (EGE) with 60% ethanol and methanol were investigated. The inhibitory activity of the EGE in Gram-positive bacteria was significantly higher than in Gram-negative bacteria. Higher antibacterial activity was observed with methanol ginseng extract when moisture content and barrel temperature were 20% and $115^{\circ}C$, respectively, that diameter of inhibition zone at 1,500 mg/mL was $15.40{\pm}0.13\;mm$ for Bacillus subtilis and $9.31{\pm}0.05\;mm$ for Salmonella typhimurium. The amount of total phenolics was highest in extruded ginseng at 20% moisture content and $115^{\circ}C$ barrel temperature. Especially, a positive correlation was observed between the total phenolic content and antioxidant activity of the extracts. In the 2,2-diphenyl-1-picryhydrazyl radical (DPPH) system, all tested extract of extruded ginseng at 20% moisture content exhibited very strong antioxidant properties when compared to red ginseng with percent scavenging effect of 23-35% at 20mg/mL. In conclusion, it can be said that the extracts of extruded ginseng could be used as natural antimicrobial and antioxidant agents in the food preservation.

In this study, the antioxidant and anti-inflammatory effects of methanol extract (ME) and hot water extracts (HE) from the fruiting bodies of Ganoderma applanatum were investigated. The 1,1-diphenyl-2-picryl-hydrazy (DPPH) radical scavenging activity of 2.0 mg/mL ME (94.83%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME and HE were similar to that of BHT at 2.0 mg/mL, whereas lipid peroxidation activity of the ME and HE were significantly lower than that of BHT. High-performance liquid chromatography analysis showed that the G. applanatum fruiting bodies contained nine phenolic compounds, which might contribute to antioxidant and anti-inflammatory activities. The survival rate of RAW 264.7 macrophages treated with 2.0 mg/mL ME and HE were 65.23 to 68.12% at 2.0 mg/mL, thereby indicating that the extracts were slightly cytotoxic at the concentration tested. The extracts also inhibited the nitric oxide (NO)-mediated expression of inducible nitric oxide synthase (iNOS) protein in lipopolysaccharide-induced RAW 264.7 macrophages and carrageenan-induced paw edema in rats. The study results demonstrated that the fruiting bodies of G. applanatum possessed good antioxidant and anti-inflammatory activities, which might be used to develop novel anti-inflammatory agents.
This study investigated quality changes in fresh-cut burdocks treated with hot water and anti-browning agents. The combined treatment using both heat treatment and anti-browning agents delayed the browning of burdocks, especially for those dipped in hot water and a solution of green tea extract plus NaCl. This treatment reduced the respiration rate and inhibited the growth of microorganisms more than heat treatment alone. The organoleptic quality of burdocks treated with the combined method proved to be the best according to sensory evaluation. Hence, this combined treatment using heat and anti-browning agents can enhance overall quality of processed fresh-cut root vegetables by browning inhibition and shelf-life extension.
In order to discover new cancer chemopreventive agents from natural or synthetic products, a structurally diverse class of chemopreventive agents was evaluated using in vitro biomarker of inhibition of 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced ornithine decarboxylase (ODC) activity in cultured mouse epidermal 308 (ME)308 cells. As a results, apigenin, benzylisothiocyanate, curcumin, diallyl disulfide, N-(4-hydroxyphenyl)retinamide (4-HPR), menadione, miconazole, nordihydroguaiaretic acid (NDGA) and phenethyl isothiocyanate showed potent inhibitory effects in this process. A chemically diverse group of compounds was included in the evaluation, such as flavonoids, retinoids, isothiocyanates, sulfur-containing compounds and phenolic antioxidant compounds. These data are suggestive to understand the cancer chemopreventive potential mediated by these substances.
The interest of oligomeric proanthocyanidins(OPCs) as therapeutic agents against diseases involving radical damage is growing. Proanthocyanidins are a class of polyphenolic compounds in several plant species and are oligomers of flavan-3-ol monomer units. Polyphenols in green and black tea, grape seeds, grapes and wine have raised much attention but mountain grape seed has not been investigated intensively up to now This study investigated the total OPCs contents and the total antioxidant activity of mountain grape seeds. Total antioxidant activity using DPPH method was employed and OPCs contents were determined by means of the UV-VIS spectrophotometer. The total OPCs yield of mountain grape seeds was about 1.45 % and total antioxidant activity was 15.8 ${\mu}g/m{\ell}$.
