Screening for Antitumor Efficacy fro the wild plants in Korea(II) (韓國産 抗腫瘍性 資源의 Screening에 대하여(II))

  • Many plants, which collected from Korea, were applied to antitumor and cytotoxic screeing tests against sarcom 180 a ascitec in mice, V-79 KB and P388 cultured cells. The results are summarixed as follows:1) The total packed cell volum method has been used for the antineoplastic screening for from natural higher plants in Korea. By this method, we have found out that the root, leaf and stem of Tripterygiu, regelii Spragne & Taketa having strong antineoplastic activity and also Rumex Japonicus Houtt. Eragrositis ferru-ginea Beauv. and Patrinia scabio-saefolia Fischer showed significant activity to anticancer tumor while cynanchum wilfordii Hemsley, and Rosa polyantha Sieb. et Zacc. showed slight activity to antitumor. 2) Among the 13 tested plants, the root and stem of Tripterygium regelii Spragne & Taketa and Amethystanthus excisus Nakai showed strong antitumor activity by the V79 cytotoxic cell screening test. 3) Twelve plants, which are glowing in mountainous area of Korea tested to anticancer activity. From the results, Eragrositis ferru-ginea Beauv., Angelica gigas Nakai, Geranium sibiricum L., Patrinia scabio-saefolia Fisher, Cynanchum wilfordii Hemsley, and Rubia akane Nakai have been proved to be anti-cancer plants by using P388 cell cultured method. 4) Tripterygiu, resgelii Spragne & Taketa, Eragrositis ferru-ginea Beauv., Patrinia scabio-saefoli Fisher, Cynanchum wilfordii Hemsley and Rasa polyantha Sieb. et Zacc., var. genuina Thunb. showed strong anti-tumor activity both total packed cell volume method and Cytotoxicity method.

Analysis of Research Subject Network in the Field of Oncogene (암유전자 연구주제 네트워크 분석)

  • Purpose: Health technology research & development is an important area to leading future. This study examined the current trends for 'oncogene' based on the research subject network to deduce a research front. Method: Papers were extracted from PubMed database using MeSH term for studies on 'oncogenes' and further categorized as papers published by Korean. Keywords were collected from all of articles. Research subject network was generated by keywords. Research subject network was analyzed by weighted degree centrality based social network analysis and transition of research subjects was analyzed by the time series. Results: On 'oncogenes', 'Genes, ras', 'Apoptosis', 'Signal Transduction' had a high degree centrality and currently 'Antineoplastic Agents', 'Prognosis', and 'Tumor Markers, Biological' were widely conducted. Conclusion: Consistency of research trend pattern was found by analyzing oncogene network with compromised to international vs. domestic trends. Analyzing keyword networks in various subject area, those will allow us to predict the research progress and propose evidence of research & developmental strategy.

The Longitudinal Study on the Calorie and Protein Intakes and Food Choices in Gastrectomy Patients who Receiving Adjuvant Chemotherapy (수술 후 Cisplatin을 투여받는 위암 환자의 구강영양섭취 실태 분석)

  • This study was conducted to investigate nutrient and food choices in gastric cancer patients receiving Cisplatin after surgery. Ten patients were followed from the fist day of the first cycle to the last date of the 6th the cycle of the chemotherapy. The subjects kept daily self record of dietary intake and the period of nausea/vomiting during 6 cycles. Using Computer Aided Nutritional Analysis Program, the degree of Calorie, carbohydrate, protein, fat and fluid intakes according the chemotherapy period. The reseacher developed food intake rating scale, and then three dietitians analysed the oral intakes according to the type of foods. As the results of this study, during the chemotherapy cancer patients are intakes much fewer calorie, protein and fluids than recommended dietary allowance. Oral intake was worsen as treatment proceed. During the chemotherapy periods most of the patients choose fruits, vegitables, steam rice, porridge, yogurt and the beam soup to overcome nausea and vomiting. In order to promote oral intake for chemotherapy patients, the researcher strongly suggest that indiviual food preform should be considered.

  • Alcohol extracts of six plant namely Coix agrestis L$_{OUREIRO}$, Chloranthus Japonicus (Siebi) N$_{AKAI}$, Ajuga Spectabilis N$_{AKAI}$, Juglans mandsuria Max., Cirsium Macki var Ussuriense $K_{ITA}$ and Sasamorpha Purpurascense (Hack) N$_{AKAI}$ were subjected to the screening tests for anti-tumor activities against SN-36 Leukemia, Sarcoma 180 and Ehrlich ascites carcinoma. Of the six extracts, Ajuga Spectabilis N$_{AKAI}$ and Coix agrestis L$_{OUREIRO}$ were found to be potential in survival effect of tumor transplanted mice.

  • Sarcoidosis is an inflammatory disease involving multiple-organs with an unknown cause. The new onset of sarcoidosis associated with therapeutic agents has been observed in 3 clinical settings; tumor necrosis factor antagonists in autoimmune rheumatologic diseases, interferon alpha with or without ribavirin in patients with chronic hepatitis C or melanoma, and antineoplastic agent-associated sarcoidosis in patients with hematologic malignancies. Here, we report a female patient who developed sarcoidosis after capecitabine treatment as an adjuvant chemotherapy for sigmoid colon cancer. To our knowledge, this is the first report of a capecitabine-induced sarcoidosis.

Cytotoxic Activities of Herbal Drugs Against Human Cancer Cell Lines (Ⅱ) (인체암세포주에 대한 천연자원의 세포독성 검색 (Ⅱ))

  • In our continuing search for new antineoplastic agents from natural products, one hundred and thirty-five herbal drugs were extracted with petroleum ether/ether (1:1), ethyl acetate and methyl alcohol, successively and their cytotoxicities were evaluated against A549 (human lung carcinoma) and SK-OV-3(human ovary adenocarcinoma) cell lines. Among them, fifteen kinds of ether extracts, eighteen kinds of ethyl acetate extracts and seven kinds of methanol extracts showed significant cytotoxic activities (above 70% inhibition) against A549 cell lines at a concentration of $40\;{\mu}g/ml,$ while ten kinds of ether extracts, thirteen kinds of ethyl acetate extracts and six kinds of methanol extracts demonstrated significant cytotoxic activities against SK-OV-3 cell lines at the above same concentration.

  • For the search of new antineoplastic agents from natural resources, two hudred and one kinds of oriental medicinal drugs were extracted with petroleum ether/ether(1:1), ethyl acetate and methyl alcohol, successively and their cytotoxicities were evaluated against A549 (human lung carcinoma) and SK-OV-3 (human ovary adenocarcinoma) cell lines. Among them, thirty kinds of ether extracts, forty-one kinds of ethyl acetate extracts and nine kinds of methanol extracts showed significant cytotoxic activities (above 70% inhibition) against A549 cell lines at a concentration of $40\;{\mu}g/ml$. And also, twenty-four kinds of ether extracts, thirty-one kinds of ethyl acetate extracts and six kinds of methanol extracts showed significant cytotoxic activities against SK-OV-3 cell lines at the same concentration.

  • Atypical teratoid rhabdoid tumours (ATRTs) are the most common malignant central nervous system tumours in children ${\leq}1year$ of age and represent approximately 1-2% of all pediatric brain tumours. ATRT is a primarily monogenic disease characterized by the bi-allelic loss of the SMARCB1 gene, which encodes the hSNF5 subunit of the SWI/SNF chromatin remodeling complex. Though conventional dose chemotherapy is not effective in most ATRT patients, high dose chemotherapy with autologous stem cell transplant, radiotherapy and/or intrathecal chemotherapy all show significant potential to improve patient survival. Recent epigenetic and transcriptional studies highlight three subgroups of ATRT, each with distinct clinical and molecular characteristics with corresponding therapeutic sensitivities, including epigenetic targeting, and inhibition of tyrosine kinases or growth/lineage specific pathways.

  • Mollugin has been reported to have various biological activities including antineoplastic, antitumor, antiviral against the hepatitis B virus, anti-aging and antimutagenic activities. An effective and concise synthesis of mollugin in two steps including kinetic control from the cheap starting material 1,4-naphthoquinone has been introduced, and mollugin derivatives thus prepared are screened for their inhibition ability against the hepatitis C virus (HCV) and the dihydrobenzochromene structure might be an additional anti-HCV agent as a new leading compound.

  • A strongly cytotoxic fraction (Fc-2, ED50=0.0003$\mu\textrm{g}$/ml) against L1210 cell was obtained from the root, seed and fruit of Trichosanthes kirilowii. Administration of Fc-2 prolonged the life span of the BDF1 mice bearing L1210 and the ICR mice bearing S-180 by 135%, and 130%, respectively. The Fc-2 affected L1210 cells were enlarged in their diameters two or three times in comparison with the untreated ones. When 20mg/kg of Fc-2 was administered, intraperitoneally, all the mice were killed.
