Antimutagenic Effects of Traditional Herbal Drugs on the Aflatoxin $B_1$ (민간 생약재의 아플라톡신 $B_1$에 대한 항돌연변이 효과)

  • The antimutagenic effects of 46 kinds of medicinal plants that have been used as traditional folk antitumor agents in Korea were studied by using Ames mutagenicity test. Most of the methanolic extracts from the plants which were used in this experiment showed strong antimutagenic activity toward aflatoxin B1(AFB1) in Salmonella typhimrium TA100 and TA98. However, N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) induced mutagenicity was not blocked by adding the methanolic extracts of the plants except persimmon leaves (Diospyros kaki Thunberg)and Elaeagnus umbellata.

Antimutagenic Effects on Methanol Extracts of Doenjang Made with Various Kinds of Water or Salt (물 및 소금 종류를 달리한 된장의 메탄올 추출물에서의 항돌연변이 효과)

  • The study was carried out to evaluate the antimutagenic effects in methanol extracts of Korean soybean paste (doenjang) added with various kinds of water (germanium water, painted maple sap) or salt (sun-dried salt, roasted salt, one time bamboo roasted salt, nine times bamboo roasted salt). Methanol extracts of germanium water doenjang (Ge-D) and painted maple sap (Acer mono Max) doenjang (PM-D) exhibited significant inhibitory activity ($56{\sim}62%$) against aflatoxin $B_1$ ($AFB_1$) by adding of 1 mg/plate in Ames test. Also, methanol extracts of Ge-D and PM-D showed stronger antimutagenic activity toward N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in SOS chromotest than traditional doenjang (TD). Methanol extracts of doenjang made with four kinds of salt revealed antimutagenic activity toward MNNG; especially, doenjang extracts using one-time bamboo roasted salt (B1-D) showed 94% inhibition at the concentration of 5 mg/plate. Methanol extracts of B1-D also had the strongest inhibitory effect against MNNG of doenjang made with different salts in SOS chromotest. As the results indicate, the various kinds of water and salt have had separate effects on the antimutagenic activity of doenjang; therefore, further research on various physiological functions of water or salt added traditional doenjang is needed.

  • The antimutagenic effects of buchu kimchi and Chinese cabbage kimchi and theri cytotoxic effects against human cancer cell line were investigated in the Salmonella typhimurium system and MTT assay, respectively. Leek and Chinese cabbage were aslo evaluated in the same system. Buchu kimchi was fermented at 15 $^{\circ}C$ for 4 days . Buchu kimchi samples showed somewhat higher antimutagenic effects against aflatoxin B1(AFB1) than CHinese cabbage kimchi in Salmonella typhimurium TA100 strain. There was no difference onthe antimutagenic activity according to the length of fermentation . Leek exerted stronger antimutagenicity against AFB1 than Chinese cabbage in the Ames assay. In MTT assay, 6-day fermented buchu kimchin revealed the highest cytotoxicity against AGS human gastric adenocarcinoma cells in which 62% and 82% of the inhibition were observed wiht the addition of 100ug, 400ug/well, respectively. Buchu kimchi samples caused 60~70% inhibition on the proliferation of HT-29 at 400ug/well. Leek exhibited higher antiproliferative effect against both AGS cells and HT-29 cells than Chinese cabbage in MTT assay. From these results, it is considered that buchu kimchi has stronger antimutagenic and in vitro anticancer effects than Chinese cabbage kimchi and the high inhibition rate of buchu kimchi probably results from leek, the major ingredient of buchu kimchi .

  • The antimutagenic effects of juices from green pepper(GP), red pepper leaf(RPL), red pepper(RP) and sweet pepper(SP) were examined by the Ames method using Salmonella typhimurium TA100. The juice supernatants of GP, RPL and RP showed antimutagenic activities against afltoxin B1(AFB1) in Salmonella typhimurium TA 100. The juice supermatants of GP and RPL also exhibited the inhibitory effects(p<0.05) to the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitro-quinoline-1-oxide(4-NQO). The juice of RP showed antimutagenic activities against indirect mutagen of AFB1, however, the activity was reduced at higher concentration(5.0%), furthermore, as the adding concentration of sample increased to 5.0%, it exhibited slight comutagenicith on direct mutagen of MNNG. Theantimutagenic activities of GP and RPL juices were reduced significantly after heating at 100℃ for 20min, supposing that the antimutagenic compound(s) in the juices were heat labile.

  • Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at $26^{\circ}C$ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TAlOO.

Antimutagenic and Antibacterial Activities of Korean and American Propolis (한국산과 미국산 프로폴리스의 항돌연변이 및 항균효과)

  • The antimutagenic activities of ethanol extracts of Korean and American propolis were tested using Salmonella Typhimurium TA98 with two indirect mutagens of 3-amino-1,4-dimethyl-5H-pyrido [4,3-b]indole (Trp-P-1) and 2-aminoanthracene (2-AA) with S9 mix. Additionally, their antimicrobial activities against acne-related pathogenic strains of Propionibacterium acnes, Staphylococcus Epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa were evaluated using both paper disk method and agar dilution method. Ethanol extracts of Korean and American propolis showed strong inhibitory effects, in a dose dependant manner, against the mutagenicities induced by Trp-P-1 and 2-AA. The antimutagenic effect of ethanol extracts of Korean propolis showed significantly higher protective activity than that of American propolis against the Trp-P-1 induced mutagenicity of S. Typhimurium TA98 at the lower concentration ($1-10\;{\mu}g$), but significantly lower protective activity at the higher concentration ($50-200\;{\mu}g$). The antimutagenic effect of ethanol extract of Korean propolis showed significantly higher protective activity than that of American propolis against the 2-AA induced mutagenicity at the concentration of $1\;{\mu}g$, but significantly lower protective activity than that of the American at the higher concentration ($50-200\;{\mu}g$). Both extracts showed strong antimicrobial activities against all the acne-related pathogens tested, with minimal inhibitory concentration (MIC) values in the range $1,500-5,000\;{\mu}g/mL$.

Study on Antimicrobial and Antimutagenic Activity of Horseradish ( Wasahia japonica) Root Extracts (고추냉이 뿌리의 항균활성 및 항변이원활성에 관한 연구)

  • The antimicrobial activities of horseradish ( Wasahia japonica) root extract against 4 kinds of food poisoning bacteria and 3 kinds of molfs were examined. The antimutagenic activity of horseradish ( Wasahia japonica) root extracts was also examined by Ames test with Salmonella tyhimurium TA 98 The antimicrobial activities of distilled water extracts from horseradish root were stronger than those of ethanol extracts, and stronger against molds than bacteria. Of the kinds of bacteria, Vibrio parahaemolyticus was best inhibited by the distilled water extracts from horseradish root. The antimicrobial activity of distilled water extracts from horseradish root were stronger against 3-Amino-1, 4-dimethyl-5H-pyrido{4,3-b}indole than 2-Amino-3,-8-dimethylimidazo-[4,5-f]quinoxaline.

Antioxidative and Antimutagenic Activity of Ethanol Extracts from Cuscutae Semen (토사자(Cuscutae Semen) 에탄올 추출물의 항산화 효과 및 항돌연변이 활성)

  • In this study, the antioxidant and antimutagenic activities of Cuscutae semen ethanol extract were evaluated. Antioxidant activity was measured by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and the Ames test was employed to determine the inhibition effect on mutagenicity in Salmonella typhimurium TA100. The extract showed significant free radical-scavenging activity towards the DPPH radical, and at a concentration of 400 ppm, its free radical-scavenging activity was similar to that of BHT. The $IC_{50}$ value of the extract was 89 ppm, indicating good antioxidant capacity. Moreover, at 5 mg/mL, the extract presented inhibitions of approximately 98.0% and 49.2% on mutagenicity induced by 4-nitroquinoline 1-oxide and sodium azide, respectively. The total polyphenols and flavonoid contents of the extract were 20.1 mg/g and 1.9 mg/g, respectively. Therefore, this study indicates that the ethanolic extract of Cuscutae semen has excellent antioxidative and antimutagenic potential.

Antimutagenic Effect of Doenjang(Korean Fermented Soy Paste) toward Aflatoxin (된장의 Aflatoxin $B_1$에 대한 항돌연변이 효과)

  • Antimutagenic effect of doenjag (Korean fermented soy paste) on mutagenesis induced by aflatoxin B1 (AFB1) in Salmonella typhimurium strains TA98 and TA100 was studied. AFB1 revealed maximum mutagenicity at dose level of 1 $\mu$g/plate with metabolic activation system in both strains. Strong antiutagenic activity toward AFB1 was completely inhibited at the level of 50% of the doenjang extract. At the same concentration 64-66% and 39-53% of the AFB1 induced mutageneses were blocked when the methanol extracts of raw and cooked soybeans were added in the system respectively Raw soybeans showed higher ihhibition rate to the mutagenicity than cooked soybeans but the fermented soybeans(doenjang) was the most effective (p<0.05) Other soybean fermented foods such as commercial doenjang natto and miso were also exhibited some antimutagenic activities however the traditional doenjang was the most effective and then commercial doenjang. Natto and miso were less effective.
