Synergistic Antimicrobial Action of Thymol and Sodium Bisulfate against Burkholderia cepacia and Xanthomonas maltophilia Isolated from the Space Shuttle Water System

  • A combination of thymol and sodium bisulfate was found to be an effective biocidal agent against strains of Burkholderia cepacia and of Xanthomonas maltophilia that were found in the space shuttle water system. Potassium iodide (KI), the biocidal agent used in the past, had a minimum inhibitory concentration (MIC) of 50,000 ppm against the two B. cepacia (541 STS-81 and 1119 STS-91) strains, whereas that of thymol and sodium bisulfate was 2,400 and 950 ppm, which was 21 and 53 times lower than that of KI for B. cepacia, respectively. The MIC value for the combination of thymol and sodium bisulfate was 4 times lower than that for thymol or sodium bisulfate alone against B. cepacia (541 STS-81, 1119 STS-91) or Pseudomonas cepacia (ATCC 31941). The fractional inhibitory concentration (FIC) of the combination of thymol and sodium bisulfate for all organisms tested was less than 0.5, indicating a strong synergistic effect.

  • The hydroxyapatite(HAp) as a carrier the for ion exchange agent of $Ag^+$ions was prepared in semiconductor fabrication, The Ca/P molar ratio of the HAp was 1.65. The HAp is molded in shape of the antimcrobial ball and then sintered at 100$0^{\circ}C$ Ag-containing HAp(HAp-Ag) was prepared by incorporating $Ag^+$/ions in HAp crystals through an ion-exchange reaction in solutions containing 0.01M $AgNO_3$. The antimicrobial effect of HAp-Ag for bacteria such as Escherichial coli and Staphylococcus aureus has been Investigated. The concentrations of silver in the antimicrobial ball was determined by inductively coupled plasma and the amount of $Ag^+$ions was 9.0$\mu\textrm{g}$/g. The HAp-Ag exhibited excellent antimicrobial effect for bacteria such as E. coli and S. aureus. The bactericidal activity was considered to be caused by direct contact of bacteria with $Ag^+$ions in the HAp crystals. The HAp would likely to be possible as a carrier of antimicrobial metal ions such as Ag, Cu, and Zn by recycling of waste sludge in the semiconductor fabrication process.

천연 항균제의 침지와 항균 포장필름이 풋고추의 저장성에 미치는 영향 (Effect of Natural Antimicrobial Agent Dipping and Antimicrobial Packaging Films on the Keeping Quality of Green Chilli Peppers)

  • 풋고추를 수확하여 선도를 유지할 수 있도록 하기 위하여 천연항균소재를 이용한 항균용제와 항균포장필름에 대한 각각의 효과와 두 가지를 병행한 상승효과를 확인하기 위하여 풋고추를 항균용제에 침지 처리후 1$0^{\circ}C$에 저장하면서 품질변화를 측정하였다. 그 결과 항균용제와 항균포장필름에 대한 각각의 효과뿐만 아니라 두 가지를 병행하여 저장하는 경우 상승효과가 더욱 두드러지게 나타나는 것을 확인 할 수 있었다. 따라서 풋고추의 선도를 유지하는 데에 천연항균소재가 효과적으로 사용될 수 있는 것으로 나타났고, 이러한 용제가 실용가능성을 가질 수 있도록 다각도로 연구가 진행되어야 할 것으로 생각된다.

  • The most proper antibiotic must be selected after antimicrobial susceptibility test. If difference in antimicrobial susceptibility was significant between oral sampling sites, it is rationale to use the most susceptible antibiotic agent respond to dental procedure and object of treatment. This study examined sampling site variation from saliva, supragingival plaque and subgingival plaque of 16 children's oral microbes. The cultured bacterial isolates, which were Streptococcus viridans and Neisseria, were examined for 10 antimicribial drugs with the Bauer-Kirby agar disk diffusion method. The used drugs were Penicillin, Ampicillin, Oxacillin, Cephalothin, Imipenem, Gentamicin, Erythromycin, Vancomycin, Ciprofloxacin, Clindamycin. There was no significant difference between three sampling sites for antimicrobial susceptibility test of S. viridans and Neisseria and the sequence of susceptibility was agreed among them. In conclusion, it was suggested that antimicrobial susceptibility test from saliva, supragingival plaque and subgingival plaque of children have no significant sampling site variation.

Antimicrobial Effect of Coptidis rhizome Extract against Mutans Streptococci and Periodontopathogens

  • The purpose of the study was to investigate the antimicrobial activity of the methanol extract of Coptidis rhizome against the type strains of cariogenic bacteria, Streptococcus mutans and Streptococcus sobrinus, and the periodontopathogens, Porphyromonas gingivalis, Prevotella intermedia, Treponema denticola and Aggregatibacter actinomycetemcomitans. The antimicrobial activities of the crude extract and the methanol extract fractions of Coptidis rhizome separated by silica gel chromatography were evaluated by determining the minimal bactericidal concentration (MBC) values, using the microdilution method. The cell viability test of the extracts of Coptidis rhizome on the KB cells was also studied by methyl thiazolyl tetrazolium (MTT) assay. Our results showed that the 11th fraction (F11) of the methanol extract had the greatest antimicrobial activity against the tested bacteria, with no associated cytotoxicity on the KB cells, upto a concentration of $50{\mu}g/ml$. These results suggest that the silica gel chromatography fraction F11 of the methanol extract of Coptidis rhizome, could be useful in the development of oral hygiene products as an antimicrobial agent for the prevention of dental caries and periodontal diseases.

Characteristics of Sophorolipid as an Antimicrobial Agent

  • Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm, respectively. Also, 100 ppm of sophorolipid inhibited $50\%$ of cell growth of plant pathogenic fungus, Botrytis cineria. However, sophorolipid showed no effect on Escherichia coli, indicating that its selective antimicrobial activity depended on the cell wall structure. Treatment of B. subtilis with sophorolipid increased leakage of intracellular enzyme, malate dehydrogenase, indicating a possible interaction of sophorolipid with a cellular membrane. Comparing lactone-type and acid-type sophorolipids, the former showed a higher antimicrobial activity. Supplementing other surfactants showed no significant effects on the antimicrobial activity. Animal study showed that 5 g of sophorolipid per kg body weight by oral administration caused no toxicity, and sophorolipid induced no irritation on the skin. These results show potential use of sophorolipid as an active ingredient in healthcare products.

  • Perilla frutescens var. crispa (Pfc), a herb belonging to the mint family (Lamiaceae), has been used for medicinal and aromatic purposes. In the present study, we analyzed the variation in the chemical composition of essential oils (EOs) obtained from five different genotypes of Pfc collected from different regions. Based on principal component analysis (PCA) and hierarchical cluster analysis (HCA), we identified three groups: PA type containing perillaldehyde, PP type containing dillapiole, and 2-acetylfuran type. To assess the correlation between EO components and antimicrobial activities, we compared classification results generated by PCA and HCA based on antimicrobial activity values. The findings suggested that the major compounds obtained from EOs of Pfc are responsible for their antimicrobial activities. Chemotypes of Pfc plants are essentially qualitative traits that are important for breeders. The present findings provide potential information for breeding Pfc as an antimicrobial agent.

Glycyl Norfloxacin 유도체의 합성과 항균작용 (Synthesis and Antimicrobial Activity of Glycyl Norfloxacin Derivatives)

  • The synthesis and antimicrobial activity of N-substituted glycyl derivatives of Norfloxacin were described. Norfloxacin was treated with chloroacetyl chloride to yield chloroacetyl norfloxacin (1). This compounds was treated with alkylamines to obtain quinolone carboxylic acids (2-6), which were reacted with pivaloyloxymethyl chloride to get pivaloyloxymethyl quinolone carboxylates (7-11). Free carboxylic quinolones (2-6) showed little stronger activities to their pivaloyloxymethyl esters (7-11). In quinolone analogues, longer alkyl chain compounds showed stronger activities than shorter one.
