The antihypertensive effect of clonidine administered concurrently with labetalol or metoprolol were studied with spontaneously hypertensive rats (SHR). The changes of heart rate were also observed in the same rats. Every drug was orally administered single dose after prechecking the systolic blood pressure and heart rate of SHR. The blood pressure of SHR in concurrently administered group was more significantly decreased than in alone administered group. The effective and stable decrease of blood pressure was maintained at the group of clonidine with labetalol (0.05+50mg/kg) for 9 hours. The group of clonidine with metoprolol (0.05+100mg/kg) manifested more marked decrease of blood pressure than the group of metoprolol (100mg/kg) alone for 9 hours. The diminishing effect of heart rate was enhanced in group of administering clonidine with labetalol, decreasing the dose of labetalol from 50mg/kg via 25mg/kg to 12.5mg/kg. On the other hand, in the group that clonidine was administered concurrently with metoprolol, the diminishing effect of heart rate was decreased with decreasing doses of metoprolol from 100mg/kg via 50mg/kg to 25mg/kg.
Symplocarpus renifolius, a folk medicinal herb has been used for treatment of hypertension, rheumatis, tetanus, ringworm, cough and epilepsy in north and middle part of Korea. The extracts from the root of S. renifolius were evaluated for antibacterial, antihypertensive and analgesic activities. The crude extract of the root of S. renifolius showed antibacterial activity against Gram(+) bacteria and dose dependantly decrease the blood pressure of SHR. The chloroform extract of the roots of S. renifolius was also exibits analgesic action in mice.
Fish skin waste accounts for part of the solid waste generated from seafood processing. Utilization of fish skin by bioconversion into high-grade products would potentially reduce pollution and economic cost associated with treating fish processing waste. Fish skin is an abundant supply of gelatin and collagen which can be hydrolyzed to produce bioactive peptides of 2-20 amino acid sequences. Bioactivity of peptides purified from fish skin includes a range of activities such as antihypertensive, anti-oxidative, antimicrobial, neuroprotection, antihyperglycemic, and anti-aging. Fish skin acts as a physical barrier and chemical barrier through antimicrobial peptide innate immune action and other functional peptides. Small peptides have been demonstrated to possess biological activities which are based on their amino acid composition and sequence. Fish skin-derived peptides contain a high content of hydrophobic amino acids which contribute to the antioxidant and angiotensin-converting enzyme inhibitory activity. The peptide-specific composition and sequence discussed in this review can be potentially utilized in the development of pharmaceutical and nutraceutical products.
We collected wild mushroom, Sarcodon aspratus from Deogyu Mt. of Muju, Jeollabuk-do and physiological functionalities of its water extract were investigated. The water extracts from S. aspratus showed the highest antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of 74.3% and also showed high anti-gout xanthine oxidase (XOD) inhibitory activity (59.6%). However, tyrosinase inhibitory activity was very low (17.3%), and antioxidant activity and SOD-likely activity were not detected. The ACE inhibitory activity of S. aspratus fruiting body was the highest when powder of the fruiting body was shaked at $30^{\circ}C$ for 24 h by distilled water. Furthermore, the XOD inhibitory activity was also the highest by extraction at $50^{\circ}C$ for 24 h with water.
His-His-Leu (HHL), a tripeptide derived from a Korean soybean paste, is an angiotensin-I-converting enzyme (ACE) inhibitor. We report here a method of producing this tripeptide efficiently by expressing tandem multimers of the codons encoding the peptide in E. coli and purifying the HHL after hydrolysis of the peptide multimers. The HHL gene, tandemly multimerized to a 40-mer, was ligated with ubiquitin as a fusion gene (UH40). UH40 was inserted into vector pET29b; the UH40 fusion protein was then produced in E. coli BL21. The recombinant UH40 protein was purified by cation-exchange chromatography with a yield of 17.3mg/l and analyzed by matrixassisted laser desorption ionization (MALDI) time-of-flight (TOF) mass spectrometry and protein N-terminal sequencing. Leucine aminopeptidase was used to cleave a 405-Da HHL monomer from the UH40 fusion protein and the peptide was purified using reverse-phase high-performance liquid chromatography (HPLC) on a C18 HPLC column, with a final yield of 6.2mg/l. The resulting peptide was confirmed to be HHL with the aid of MALDI-TOF mass spectrometry, glutamine-TOF mass spectrometry, N-terminal sequencing, and measurement of ACE inhibiting activity. These results suggest that our production method is useful for obtaining a large quantity of recombinant HHL for functional antihypertensive peptide studies.
Objectives: This study was designed to investigate antihypertensive effect by single adminstration of Chunghyul-dan in stroke patients. Methods: This is a 31-patient case series from retrospective chart review of inpatients who diagnosed with stroke at a Kyung Hee University Korean Medicine Hospital (Seoul) between October 2016 to October 2017. We reviewed patient's charts with transient and sudden increases in blood pressure who were treated with Chunghyul-dan and analyzed blood pressure over time. In addition we classified patients by using the standardized predictive models for Korean medical diagnostic pattern-identification to find out if there are some differences in responder ratios. Results: Thirty one patients who took Chunghyul-dan were identified. 60 minutes, 120 minutes after 1200mg administration, systolic and diastolic blood pressure decreased significantly compared to baseline. We could not identify the difference between the groups of each pattern-identification because of small number of some groups. Conclusions: Although limited by its retrospective nature, this study suggests that Chunghyul-dan may be effective as short term antihypertensive method for stroke patient.
Objectives : Aim of this study was to investigate the antihypertensive effect of Gyeok-Pal Sang-Saeng-Yeok-Chim acupuncture treatment in hypertensive patients. Methods : The research subjects were a total of 26 hypertension patients. Eligible participants had systolic blood pressure ${\geq}140mmHg$ or diastolic blood pressure ${\geq}90mmHg$ with antihypertensive drug. Both the experimental and control group were divided into 13 participants respectively with homogeneous consideration. The experimental group had acupuncture treatment, Gyeok-Pal Sang-Saeng-Yeok-Chim, whereas the control group didn't have any treatment. Blood pressure was measured before and after acupuncture treatment for a total of 12 times. Results : After 4 weeks of treatment we could identify a decrease in both systolic and diastolic blood pressure in acupuncture, Gyeok-Pal Sang-Saeng-Yeok-Chim, treated group compared to that of control group. The given results by measuring time on blood pressure were as follows: Significant decrease in systolic blood pressure from the $1^{st}$ to $12^{th}$ day of visit(P<0.05) while diastolic blood pressure did not show significant decrease. Conclusions : The results suggest that Gyeok-Pal Sang-Saeng-Yeok-Chim acupuncture treatment is effective in decreasing blood pressure but with statistical restriction compared to that of control group.

The pharmacological profile of KR-31081, a newly synthesized AT1 receptor antagonist, was evaluated in pithed rats, conscious renal hypertensive rats (RHRs) and conscious furosemide-treated beagle dogs. In pithed rats, KR-31081 (i.v.) induced a non-parallel right shift in the dose-pressor response curve to angiotensin II (ID50: 0.05 mg/kg) with a dose-dependent reduction in the maximum responses; this antagonistic effect was about 40 times more potent than losartan (ID50: 1.74 mg/kg) which showed competitive antagonism. KR-31081 did not alter the responses induced by other agonists such as norepinephrine and vasopressin. In RHRs, orally given KR-31081 produced a dose-dependent and long-lasting (>24 h) antihypertensive effect with a higher potency to losartan (ED20: 0.30 and 3.36 mg/kg, respectively). In furosemide-treated dogs, orally given KR-31081 produced a dose-dependent and long-lasting (>8h) antihypertensive effect with a rapid onset of action (time to Emax: 1-1.5 h) and 20-fold greater potency than losartan (ED20: 0.41 and 8.13 mg/kg, respectively). These results suggest that KR-31081 is a potent, orally active AT1 receptor antagonist useful for the research and diagnostic tools as an added exploratory potential.
Forty eight strains of yeast were cultured in potato dextorse(PD) broth at $30^{\circ}C$ for 24 hr and centrifuged with 12,000 rpm for 20 min. After concentrated the cultures, antihypertensive angiotensin I-converting enzyme(ACE) inhibitory activities of its concentrates were investigated. Among them, the concentrates from Saccharomyces cerevisiae Y183-3 showed the highest ACE inhibitory activity of 71.8%. The ACE inhibitor from Saccharomyces cerevisiae Y183-3 was maximally produced when Saccharomyces cerevisiae Y183-3 cultured in PD broth at $30^{\circ}C$ for 36 hr.
