DMBA로 유발한 Mouse 피부암에 대한 Astaxanthin이 함유된 난황의 항암효과 (Inhibition of DMBA-Induced Mouse Epidermal Carcinogenesis by Astaxanthin-Containing Egg Yolks)

  • Anticarcinogenic activity of astaxanthin-containing egg yolks (designate AEY) was investigated for 7,12-dimethylbenz[a]anthracene (DMBA)-induced two stage mouse epidermal carcinogenesis. Female ICR mouse (6-7 weeks of age) were house in a humidity-and-temperature-controlled facility and subjected to feed and water ad libitum. AEY (10 mg/0.2 ml acetone) was painted on the back of mice 7 days, 3 days and 5 min before DMBA treatment (50 nmole/0.2 ml acetone). One week later after DMBA treatment, 6 ${\mu}g$ tetradecanoyl 12-phorbol 13-O-acetate (TPA) dissolved in 0.2 ml acetone was applied on the mouse twice weekly over a period of 22 weeks. No sample was given to control mice. Control egg yolk (CEY) and astaxanthin-containing oil (designate AO) from Phaffia rhodozyma were used as positive controls. Mouse treated with AEY exhibited 10 tumors per mouse whereas control mouse exhibited 15 tumors per mouse, the fact that 33% reduction of tumor per mouse by AEY treatment. Tumor incidence was also reduced to 15% by AEY treatment when compared to that of control group. Such effects were also seen in CEY and AO treatment groups, but leaser extent. AO gave reduction of food intake and body weights relative to those of AEY and CEY, indicating toxicity of AO. These results suggest that AEY exhibits anticarcinogenic activity for DMBA-induced mouse epidermal carcinogenesis.

사매의 KATOIII 위암세포(胃癌細胞)에 대(對)한 항암효능(抗癌效能) (Effects of Duchesnea indica on Human Stomach Cancer Cells(KATOIII))

  • Background : Death by cancer, malignant tumor, is on the increase around the world. In South Korea, cancer is the number one cause of death, and, in South Korea, stomach cancer is the most common form of cancer. This may be due to genetic and enviornmental factors. Because stomach cancer is so prevalent here, this study was undertaken. Objectives: This study was conducted to investigate the anti-cancer effects of Duchesnea indica(Andr.) Focke. in human stomach cancer cells(KATOIII). Materials & Methods: For human stomach cancer cells KATO III cells were obtained from Korea Cell Line Bank. The water decoction of Duchesnea indica(Andr.) Focke was added to cultures. First, morphological changes were examined under an inverted microscope. The destruction of stomach cancer cells was measured through Trypan blue exclusion testing, and suppression of viability of stomach cancer cells was measured via MTT assay. Anti-cancer mechanisms were assessed by analyzing the cell cycle. Results & Conclusions : In morphologic change, stomach cancer cells showed the withdrawn and floating appearance that is typical in cellular impairment. In each test group more stomach cancer cells were killed than in the control group to a statistically significant degree. Each test group showed more suppression of viability of stomach cancer cells than that of the control group to a statistically significant degree. Analysis of the cell cycle showed that Duchesnea indica(Andr.) Focke inhibited division of stomach cancer cells. This experiment yielded empirical evidence for Duchesnea indica(Andr.) Focke as a statistically significant anticarcinogen, specifically against stomach cancer.

도라지 추출물 첨가에 의한 돌나물의 항발암 상승효과 (Enhancement of Anticarcinogenic Effect by Combination of Sedum sarmentosum Bunge with Platycodon grandiflorum A. Extracts)

  • 본 연구는 다른 산채류에 비해 칼슘이 칼슘이 특히 많은 우수한 식품으로 예부터 물김치나 겉절이 무침, 돌나무 김치로 많이 이용되어온 돌나물의 암세포 증식억제 및 암예방 효과를 검색하고 나아가 도라지 추출물과의 암세포 증식억제 상승효과를 보기 위하여 실시하였다. 돌나물의 암세포 증식억제 효과를 MTT assay로 실험한 결과, 3종의 암세포주(HepG2, HeLa, MCF-7) 모두 돌나물의 ethylether 분획층(SSMEE)에서 아주 높은 암세포 증식억제 효과를 보였으며, ethylacetate 분획층(SSMEA)에서도 유의적인 암세포 증식억제 효과를 나타내었다. 또한, 돌나물 분획물의 암예방 QR유도 활성을 HepG2 세포주를 이용하여 실험한 결과, 다른 분획층에 비해 비극성 용매층인 ethylether 분획층(SSMEE)과 hexane 분획층(SSMH)에서 유의적으로 QR유도 활성을 증가시키는 것으로 나타났다. 한편, 돌나물과 도라지와의 일정비 조합을 통해 상승효과를 본 결과, $IC_{50}$/이 되는 도라지의 butanol층 (PGMB)dfm 일정량 첨가했을 때 용매별 돌나물의 분획층 모두에서 암세포 증식억제를 상승시키는 효과를 나타내었고, 암예방 QR 유도활성 효과도 훨씬 상승하였으므로, 돌나물 조리시 유사한 부재료를 사용하는 도라지의 성분을 일정량 첨가하여 겉절이 무침이나 김치로 이용될 때 돌나물 성분의 생리활성 효과가 더욱 상승될 것으로 사료되며, 이후 돌나물의 생리활성 물질 구조 및 기전 규명에 유익한 자료가 될 것으로 사료된다.

CLA (Conjugated linoleic acid) 급이수준에 따른 역돔의 CLA 축적량 (Accumulation of the Conjugated Linoleic Aacid (CLA) in Tilapia ( Tilapia nilotica) Fed Diets on Various Levels of CLA)

  • 기능성물질인 CLA를 양어사료에 첨가하여 기존의 어류와는 다른 기능성 역돔 생산을 시도하였다. 사육 8주간 매 2주마다 어체내의 CLA축적도를 측정한 결과 8주 후 $1\%$ CLA 첨가구에서는 41.3 mg/g of fat였으며, $10\%$ CLA 첨가구에서는 180.9mg/g of fat으로 역돔의 정상적인 성장을 고려하지 않는다면 $10\%$의 사료내 첨가도 가능한 것으로 생각된다. 그러나 정상적인 성장을 고려하면 CLA의 적정 첨가량은 $1\%$로 나타났으며, 그 이상의 첨가시에는 성장 저하 현상을 나타내었다. 합성된 CLA는 총 6 종류의 이성체로 구성되어 있었고, 그 중 c9, t11 및 t10, c12 이성체가 $85\sim90\%$를 이루고 있었으며, 첨가된 CLA 함량에 따라 축적되는 양도 달라졌다. 사육 8주 후, 육 및 간 중의 지방산 조성은 n-9, 3 계열은 거의 화를 나타내지 않았으나, n-6계열은 첨가된 CLA의 함량에 따라 육 및 간에 축적되는 양은 $5\sim27\%$로, $1\%$ CLA 첨가구가 가장 높았고, $10\%$ CLA 첨가구가 가장 낮은 함량비를 나타내었다.

백화사설초(白花蛇舌草) 추출물(抽出物)의 항균실험(抗菌實驗) 및 SOD류사활성(類似活性), 전자공여능(電子供與能)에 관(關)한 연구(硏究) (Study on SOD like activity and Electric donor ability of Hedyotis diffusa $W_{ILLD}$)

  • In order to study on SOD like activity and Electric donor ability of Hedyotis diffusa $W_{ILLD}$, testing antibacterial effect on E. coli O157 which causes infectious inflammatory disease; measunng inhibitory effect on tyrosinase which stimulates melanin formation; and measuring the influence of it to SOD like activity, DPPH and TBARS which are related to ageing and carcinogenesis. The results of these experiments, are as follows. 1. In the antibacterial experiment with the extract of Hedyotis diffusa $W_{ILLD}$ on E. coli O157, there was no change in proliferation from the beginning of culture to two hours after, but proliferation inhibiting effect on E. coli O157 was detected from three hours after the beginning of culture. 2. Tyrosinase inhibitory effect was measured as $0.39{\pm}0.026%$. Compared with the control group, the effect was very slight. 3. The SOD like activity of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 21.33${\pm}$ 4.264%. Compared with the extracts of several other herbs, it was much more significant. 4. The DPPH of the extract of Hedyotis diffusa $W_{ILLD}$. was $53.3{\pm}0.91%$ when 0.02g was used and $83.5{\pm}1.82% $ when 0.05g was used. The result when 0.05g was used was more significant than 0.02g was used. 5. The TBARS of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 0.724 ${\pm}$0.O04MDAppm. Compared with the extracts of several other herbs, the result was more significant. From these results, we found that Hedyotis diffusa $W_{ILLD}$. can be used in therapy of dysentery with bloody stool and fever which is caused by infection with E. coli O157 and that it can be also used effectively as age resister or anticarcinogen.

Beneficial Effects of Dietary Antiacarcinogenic Conjugated Linoleic Acid(CLA) on the Performances of Laying Hens and Broilers

  • Effects of conjugated linoleic acid(CLA), known as an effective anticarcinogen in several aminal models, on the egg production and egg weight of laying hens, and the weight gains of broilers were investigated. CLA was synthesized from corn oil by the alkaline isomerization method and purified by the low-temperature precipitation method. Diets for laying hens and for broilers were synthesized to meet the specification of their NRC standard rationals. Two separated experiments(Experiment I and II) were conducted for laying hens. in experiment I, 45 hens(300 days of age) were divided into 15 hens per treatment group; each hen was housed in wired cage located in a temperature and humidity-controlled house and adopted to the control diet. One week later, each group was subjected to one the four treatment groups for 5 weeks : control, 1.0% CLA, 2.5% CLA and 5.0% CLA diets. Diet and water were ad libitum. The condition of experiment II was the same as that of experiment I except for the addition of 5% corn oil diet and the extension of feeding period to 7 weeks. Egg production, egg weight and feed intake were recorded every week. Forty-five broilers(10day of age) were adopted to the control diet for a week and then switched to the treatment diets for 5 weeks : control, 1.0% CLA, 2.5% CLA, 5% CLA and 5% corn oil, Body weight and feed intake of broilers were measured every week. Diets supplemented with various amounts of CLA enhanced the egg production and increased the egg weight regardless laying hen's age(150 days or 300 days) as compared to control diet. The most effective diet for the egg production and egg weight of young hens(150 days of age) was found to be 1.0% CLA diet, but relatively higher CLA diet(2.5% CLA) was required for old hens (300 day of age) to obtain similar results as seen in younger hens. All hens treated with CLA ate greater amount of feed than control hens. Broilers treated with various amount of CLA ate less feed as compared to control ones, but the body weight gain was greater than the control broilers. These results indicate that CLA enhanced the egg production and agg weight of laying hens, and increased the body weight gain of broilers with less diet consumption.

들깨의 볶음처리와 산가수분해에 의한 세포모델계 Quinone Reductase 활성유도능의 변화 (Modulation of Cellulalr Quinone Reductase Inducibility by Roasting Treatment and Acid Hydrolysis of Perilla)

  • 발암물질의 해독에 관여하는 2상 효소계의 지표효소인 QR을 활성화시키는 암예방성분의 존재여부를 탐색한 이전의 연구에서 들깨박의 메탄올 추출물이 hepa1c1c7 cell에서 높은 QR 유도활성을 나타내었다. 본 연구에서는 탈지공정에 앞서 실시되는 볶음과정에서 비활성상태로 존재하던 QR inducer가 활성상태로 유리되는 것으로 가정하고 산가수분해와 볶음처리를 한 후, 각각의 메탄올 추출물에 대한 QR 유도활성을 측정하였다. $100^{\circ}C$에서 30, 60, 120분간 산가수분해시킨 날들깨박의 메탄올 추출액은 가수분해시간이 증가할수록 세포의 QR 활성유도능이 증가하는것으로 나타났다. $180^{\circ}C$$200^{\circ}C$에서 5, 10, 20분간 볶은 들깨박의 경우, QR유도 활성은 날들깨박에 비해 높았으며, 묶음시간이 걸어질수록 증가하는 경향을 보였다. 볶은 들깨박의 메탄올 추출액은 농도가 증가함에 따라 비례적으로 QR 효소 활성을 증가시켰다. 탈지들깨박이 동물조직의 QR 효소활성에 미치는 영향을 생쥐를 이용하여 확인한 결과, 볶은 들깨박의 메탄올 추출물은 QR 효소활성을 간과 위에서 유의적으로 증가시켰고, 날들깨박은 간과 폐에서 유의적으로 효소활성을 증가시켰다. 이렇듯 들깨박은 동물의 여러기관에서 암예방의 지표효소인 QR을 유도하는 것으로 나타나, 발암 물질로부터 생체를 보호하는 물질을 함유하고 있을 가능성이 높은 것으로 추정된다.
