十六味流氣飮의 抗癌 및 抗癌劑 副作用에 미치는 영향 (Influences on the Anticancer and Inhibitive Effects of the Secondary Effects by Anticarcinogen of Shibyukmiyouki-Eum)

  • Shibyukmiyouki-Eum was a drug that treated carbuncle and cellulitis. So, the purpose of this study was to investigate effect of Shibyukmiyouki-Eum on the anticancer and inhibitive effects of the secondary effects by anticarcinogen We used Shibyukmiyouki-Eum extract(SYE) with freeze-dried, 8wks-old male mice(balb/c, ICR) and cancer cell lines(L1210. S-180) for this Study, The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). and measurement of WBC, RBC, hemoglobin and platelet was tested by a automated hematology analyzer. The results of this study were obtained as follow; SYE was showed significantly cytotoxicity on the L1210 and S-180 cell lines. increased proliferation of thymocytes decreased by anticarcinogen. In combined effects of SYE and vincristine(0.005㎎/kg), SYE was significantly inhibited proliferation of L1210 cells and significantly increased proliferation of thymocytes. Also SYE was significantly increased count of WBC. platelet and increased count of RBC, hemoglobin. These results suggest that SYE has not only anticancar action but inhibitive effects of on the secondary effects by anticarcinogen.

  • Sunjeonhwadok-Tang was a drug that treated carbuncle and cellulitis. So, the purpose of this study was to investigate effects of Sunjeonhwadok-Tang on the proliferation of cancer calls and immunocytes focusing around combined effects of anticarcinogen. We used Sunjeonhwadok-Tang extract(SHT) with freeze-dried, 8wks-old male balb/c mice and cancer cell lines(L1210, Sarcoma-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results : 1. SHT was significantly showed cytotoxicity on the L1210 cell lines. 2. SHT was significantly increased proliferation of thymocytes and splenocytes in vitro. 3. In combined effects of SHT and vincristine(0.005 mg/kg), SHT was significantly inhibited proliferation of L1210 cell lines, but was not inhibited proliferation of Sarcoma-180 cell lines compared with positive control group. 4. In combined effects of SHT and vincristine(0.005 mg/kg), SHT was significantly increased proliferation of thymocytes and splenocytes compared with positive control group. 5. In combined effects of SHT and vincristine, SHT was significantly increased proliferation of thymocytes and splenocytes in normal mice. 6. In combined effects of SHT and vincristine, SHT was significantly inhibited proliferation of L1210 cells in L1210 cells transplanted mice 7. In combined effects of SHT and vincristine, SHT was significantly increased proliferation of L1210 cell in L1210 cells transplanted mice. The present author thought that SHT had action of anti-cancer and immuno-activity, and in combined effects of vincristine, SHT had recoverable effects on damage by anticarcinogen.

  • Objective : The purpose of this study was to investigate effects of Taklihwajung-Tang on the proliferation of cancer cells land immunocytes focusing around combined effects of anticarcinogen. Materials and Method : We used Taklihwajung-Tang extract(THT) with freeze-dried, 8wks-old male balb/c mice and cancer cell lines(L1210, S-180) for this study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MIT assay). Results and Conclusion : The results of this study were obtained as follow ; 1. THT was significantly showed cytotoxicity on the L1210 cell lines and S-180 cell lines. 2. THT was significantly increased in the proliferation of thymocytes and splenocytes in vitro. 3. In combined effects of THT and vincristine(0.005mg/kg), THT was significantly inhibited proliferation of S-180 cell lines compared with positive control group. 4. In combined effects of THT and vincristine(0.005mg/kg), THT was significantly decreased in the weight of sarcoma compared with positive control group. 5. In combined effects of THT and vincristine, THT was significantly inhibited the hematological side reaction compared with positive control group. The present author thought that THT had action of anti-cancer and immune-activity, and in combined effects of vincristine, THT had recoverable effects on damage by anticarcinogen.

  • Selenium is an essential nutritional element for several animal species and human. It has been also seen, that low levels of selenium in the diet can cause many diseases. This metalloid was defined like a "double face" element because it possesses antioxidant, antimutagen, anticarcinogen but also mutagenic and carcinogenic effects. Biological Effects of Black Colored Soybean

  • It has been believed that supplementation of human diets with soybean products markedly reduces human cancer mortality rates. Specifically the black colored soybean has been considered as having a medicinal effect since ancient times in Korea. Six genotypes of black colored soybean and a normal variety were investigated for their biological effect. While the isoflavone content of black colored soybean was higher than the normal soybean, saponin content of the seeds was not different among varieties. In the case of antioxidant activity, black colored soybean showed over 3 times increase in the active effect than the control variety. When feeding soybean extract, a general change of alcohol resolving effect, such as alcohol dehydrogenase activity, was observed in the blood and liver of rats.

  • Garlic (Alium sativum L.), originated in the Kirghiz region of Central Asia, is oneof the oldest cultivated plants. It has reached Europe via Egypt in the days of Pharaohs. Since then, the legendary medicinal properties attributed to garlic have attracted human interests for thousands of years. However, the research on the medicinal effect of garlic, mostly on this anticarcinogenic actions has been systematically performed only for the last tow or three decades. Many researches have proven that garlic inhibits neoplasia elicited by chemical carcinogens using in vitro or in vivo experimental systems. Attention has recently been focused on assessing the therapeutic or chemopreventive measures of garlic compounds against carcinogenesis in animals. The active principles of he garlic extract, mainly the sulfur compounds and their derivatives have been deeply scrutinized into their chemical, functional and medicinal properties.

  • Sensitising cancer cells and at the same time desensitizing normal cells is a double task in cancer management. Agents which can combat the debilitating side effects of cancer therapeutics and simultaneously synergize with anticancer agents in specifically targeting cancer cells are needed. Selenium, a proven anticarcinogen, gains due importance in terms of its efficacy to combat the side effects of cancer therapy. This study is a comparative analysis of the chemoprotective effects of selenium compounds, methyl selenol (generated from organic selenomethionine (5mmol/L ; METase 40U/L)) and sodium selenite (inorganic form)($30{\mu}M$) in peripheral blood human lymphocytes exposed to cisplatin and mitomycin. Biochemical alterations occurring in many cells during apoptosis include loss of plasma membrane phospholipid asymmetry, DNA fragmentation, and activation of caspase-3. The present study demonstrated that the selenium metabolite and selenite are efficient in protecting lymphocytes undergoing DNA damage and exerted their activity by reducing caspase 3 expression. Interestingly organic methylselenol (MeSe) was found to offer more protective effects compared to inorganic selenite (SeL), by reducing the induction of apoptosis by the cytotoxic agents. This suggests that MeSe and to a lesser extent selenite might have potential for assessment in clinical trials and could be considered as strong candidates in pharmacogenomics or in the nutriprotective arena.

Effects of Indole Oligomers Induced from Indole-3-carbinol on the Growth of MCF-7 Breast Cancer Cells

  • Inhibitory effect of indole oligomers induced from indole-3-carbinol(I3C) on the growth of breast cancer cells was studied. We gernerated the reaction mixtures (RXM) at ambient temperature by treating a stirred aqueous solution of I3C (typeically 0.25ml) at a concentration of 12 $\mu$mol/ml) with hydrochloric acid (typically 28$\mu$l of a 1 mmol/ml solution). RXM was fractionated by the column chromatography. The fractions with similar UV-pattern were further fractionated by HPLC and 3.3'-diindoylmethane (DIM) and other indole oligomers were identified. I3C, RXM, and it derived indole compounds were added to MCF-7 cells and cultured in the presence of 10-7M estradiol for 7 days. the growth-inhibitory effect of I3C and DIM on the growth of MCF-7 cell was very strong. The synthetic DIM also revealed antiproliferative effect on MCF-7 cel. The fractions containing high DIM content (77%), were most effective in inhibiting MCF-7 cell growth induced by estradiol. With these results, we suggest that I3C and DIM might have anticarcinogenic effect on the breast cancer.

재첩가공품의 생리학적 특성과 이용 -재첩추출물의 항암효과와 면역활성증강 효과- (Physiological Effects and Utilization of Corbicula elatior Products -Effect of Cockle Extracts on Carcinogen-induced Cytotoxicity and Immune REsponse RElated to Its Antitumor Activity-)

  • Inhibitory effects of cockle extracts on carcinogen-induced cytotoxicity in C3H/10T1/2 cells were studied. Soup (62$\mu\textrm{g}$/mL), solubility (28$\mu\textrm{g}$/mL) and liposolubility (9 $\mu\textrm{g}$/mL) of the cockle inhibited 3-methyl-cholanthrene(MCA)-induced cytotoxicity in C3H/10T1/2 cells by 53 and 94%, respectively. These results suggest that the extracts cockle might have anticarcinogen-induced cytotoxicity of C3H/10T1/2 cells. The effects of cockle extracts on the immune response related to its antitumor activity in vitro and in vivo were investigated. The cockle extracts showed a direct cytotoxic effect on sarcoma-180 cells, tumor cells in vitro. Soup (0.49 mg/mL), solubility (0.11 mg/mL) and liposolubiliy (0.05 mg/mL) of the cockle markedly decreased the total numbers of sarcoma-180 cells, but not their viability. The phagocytic acitivity of peritoneal macrophage of mice was significantly augmented by these extracts of the cockle compared with that of control in vivo. These extracts also raised the phagocuytic index, indicating that the number of phagocytize dmicrobes per macrophage increased. Thus, cockle extracts might show a antitumor activity by enhancing the phagocytic cell activities.

콩 Saponin의 생리활성 기능과 함량변이 (Biological Activities of Soyasaponins and Their Genetic and Environmental Variations in Soybean)

  • There is much evidence suggesting that compounds present in soybean can prevent cancer in many different organ systems. Especially, soybean is one of the most important source of dietary saponins, which have been considered as possible anticarcinogens to inhibit tumor development and major active components contributing to the cholesterol-towering effect. Also they were reported to inhibit of the infectivity of the AIDS virus (HIV) and the Epstein-Barr virus. The biological activity of saponins depend on their specific chemical structures. Various types of triterpenoid saponins are present in soy-bean seeds. Among them, group B soyasaponis were found as the primary soyasaponins present in soybean, and th e 2, 3-dihydro-2, 5-dihydroxy-6- methyl-4H-pyran-4-one(DDMP)-conjugated soyasaponin $\alpha\textrm{g}$, $\beta\textrm{g}$, and $\beta$ a were the genuine group B saponins, which have health benefits. On the other hand, group A saponins are responsible for the undesirable bitter and astringent taste in soybean. The variation of saponin composition in soybean seeds is explained by different combinations of 9 alleles of 4 gene loci that control the utilization of soyasapogenol glycosides as substrates. The mode of inheritance of saponin types is explained by a combination of co-dominant, dominant and recessive acting genes. The funtion of theses genes is variety-specific and organ specific. Therefore distribution of various saponins types was different according to seed tissues. Soyasaponin $\beta\textrm{g}$ was detected in both parts whereas $\alpha\textrm{g}$ and $\beta$ a was detected only in hypocotyls and cotyledons, respectively. Soyasaponins ${\gamma}$g and $\gamma\textrm{g}$ were minor saponin constituents in soybean. In case group A saponins were mostly detected in hypocotyls. Also, the total soyasaponin contents varied among different soy-bean varieties and concentrations in the cultivated soy-beans were 2-fold lower than in the wild soybeans. But the contents of soyasaponin were not so influenced by environmental effects. The composition and concentration of soyasaponins were different among the soy products (soybean flour, soycurd, tempeh, soymilk, etc.) depending on the processing conditions.
