Early Growth Response of Broilers to Dietary Lysine at Fixed Ratio to Crude Protein and Essential Amino Acids

  • The effects of dietary lysine (Lys) at a fixed ratio to crude protein (CP) and essential amino acids (EAA) on early growth response of broilers were studied. Four diets were formulated to contain similar metabolizable energy (ME, 2,950 kcal/kg) but contained graded levels of incremental Lys (1.1, 1.2, 1.3 and 1.4%) while also increasing the dietary CP and EAA (methionine, methionine+ cystine, threonine and tryptophan) to maintain a constant ratio with Lys. Each diet was fed at random to 10 replicates of 6 chicks each throughout the experimental period (1-21 d). At the lowest concentration of Lys of 1.1% (19.04% CP), body weight gain (BWG) was lowest and feed conversion ratio (FCR) was poorest. The BWG increased and FCR decreased linearly as dietary Lys increased upto 1.3% (22.5% CP). Lowest feed consumption was observed in the dietary group that contained 1.1% Lys (19.04% CP) in the diet. Increasing the concentration of Lys to 1.2% (20.77% CP), significantly increased the feed consumption. The concentrations of protein, calcium, phosphorus and cholesterol in serum were not influenced by the variation in Lys contents in the diet. The humoral immune response as measured by antibody titre in response to SRBC inoculation was significantly lower in the diets containing 1.1% Lys compared to 1.4%. It is concluded that the Lys requirement of broilers is 1.3% (22.5% CP) during 0 to 21 days of age for eliciting optimum performance when a fixed ratio of Lys to CP (1:17.31) and essential AA is maintained (1:0.47 Met; 1:0.56 Thr; 1:0.17 Try).

Relative Performance and Immune Response in White Leghorn Layers Fed Liquid DL-methionine Hydroxy Analogue and DL-methionine

  • The relative performance and immune response was evaluated in White Leghorn layers fed liquid DL-methionine hydroxyl analogue-free acid (MHA-FA) relative to dry DL-methionine (DLM) in maize-soybean-sunflower based diets. Three graded levels of methionine (Met) from DLM or MHA-FA were added to the basal diet containing 0.27% Met on an equimolar basis to achieve 0.30, 0.36 and 0.42% Met in the diet. Each diet was fed ad libitum to 25 replicates of one bird (individual feeding) each, from 24 to 40 weeks of age. A regime of 16 h light was provided and all the layers were kept under uniform management throughout the experimental period. None of the parameters studied were influenced by the interaction between source and level of Met in diets. Similarly, the majority of parameters, except for daily feed consumption and immune response (influenced by level) and egg specific gravity and shell thickness (influenced by source), were not affected by either source or level of Met in the diets. Feed consumption was significantly lower in the birds fed a diet containing 0.42% Met compared to those fed lower levels of Met. The cutaneous basophilic hypersensitivity response to PHA-P and antibody titre (32 and 40 wk) to inoculation of sheep red blood cells increased significantly by increasing the concentration of Met in the diet from 0.30 to 0.36%. Thus, the Met requirement for immune competence was higher than for optimum production. The source of Met significantly influenced the egg specific gravity and shell thickness. The specific gravity and shell thickness of eggs increased significantly when MHA-FA was used as the source of Met in the diet compared to DLM. From the study it is concluded that Met requirement for immune competence (360 mg/b/d) is higher than for optimum production (300 mg/b/d). MHA-FA was comparable with DLM as a source of Met for production performance and immunity, when the bioavailability of MHA-FA was considered as 88% of DLM. Further, MHA-FA improved egg shell quality compared to DLM.

Effect of Gossypol from Cottonseed Meal Diets on Some Clinico-biochemical Parameters and Humoral Immune Response of Crossbred Calves Fed Barley or Sorghum

  • Effects of feeding of 9.95 mg free gossypol/kg live weight through cottonseed meal (CSM) were studied in 20 intact male calves fed barley or sorghum as source of cereal during the experimental duration of 210 days. Serum concentration of total protein, albumin, globulin and their ratio did not vary because of protein (gossypol) or cereal sources. Serum level of cholesterol and urea were lower (p<0.05) in sorghum than barley fed calves. Feeding of gossypol through CSM enhanced (p<0.05) serum cholesterol. An interaction between protein and period was observed with respect to serum concentrations of urea, creatinine and alanine transferase. The levels of serum creatinine and alanine transferase increased (p<0.05) following 120 days of experimental feeding in calves fed CSM diets compared to the control animals fed groundnut meal diets. No effect of feeding gossypol was, however, evident on the serum enzymes viz. alanine and aspartate transferases and alkaline phosphatase. Moreover, the source of cereal and protein did not appear to influence the metabolic profile of the calves. Humoral immune response, measured through antibody titre against Brucella abortus S99 innoculation, revealed a delayed and depressed seroreactivity indicative of immunocompromisation because of the phytotoxin gossypol. In conclusion, the feeding of gossypol at the designated levels, although had no deleterious clinico-biochemical manifestations, affected the humoral immune response of the calves.

Study on the immunogenicity of synthetic polypeptide vaccine derived from Theileria sergenti merozoite (Theileria sergenti merozoite의 합성 polypeptide 백신의 면역원성에 관한 연구)

  • Western immunoblot analysis of antigen of T sergenti merozoite revealed that the immunodominant proteins of this organism were characterized as the 18KD, 29KD, 34KD, 45KD and 105KD in Korea. The 34KD and 45KD among those immunodominant proteins of the parasite were isolated and their amino acid sequences from the $NH_2$-terminus were determined and synthesized. They respective polypeptides were cationized to enhance their antigenicity, fortified with Freund's adjuvant and tested for immunogenicity in rabbits and cattle. The results obtained were as follows; 1. Theileria sergenti merozoite antigen was shown in 120KD, 100KD, 66KD, 45KD, 34KD and 30KD in western immunoblot using serum of rabbits immunized with 34KD synthetic polypeptide and 70KD, 58KD, 55KD and 45KD using bovine serum. In western immunoblot, 45KD, 34KD and 30KD were recognized by immunized rabbits, and 50KD and 45KD by cattle sera immunized with 45KD synthetic polypeptide, respectively. 2. The ELISA utilizing the synthetic polypeptides demonstrated significant antibody response to the respective peptides. After the 2nd booster injection, an OD of 0.760(preimmunization 0.132) in rabbits and an OD of 0.645(preimmunization 0.488) to 34KD synthetic polypeptide in cattle were observed. In animals immunized with 45KD synthetic polypeptide, after the 2nd booster injection, an OD of 0.640(preimmunization 0.144) in rabbit, and an OD of 0.776 (preimmunization 0.477) in cattle were measured. 3. After the 2nd booster the reciprocal IFA titer was 1:64 in rabbits and 1:512 in cattle immunized with the 34KD synthetic polypeptide. The IFA titre was observed as 1:512 in rabbit and 1:1,024 in cattle in immunized with the 45KD synthetic polypeptide.

Growth Performance, Humoral Immune Response and Carcass Characteristics of Broiler Chickens Fed Alkali Processed Karanj Cake Incorporated Diet Supplemented with Methionine

  • A study was conducted to see the effect of dietary incorporation of alkali (1.5% NaOH, w/w) processed solvent extracted karanj cake (SKC) supplemented with methionine on growth performance, humoral immune response and carcass characteristics of broiler chickens from 0 to 8 weeks of age. One hundred and twenty, day- old broiler chicks were wing banded, vaccinated against Marek' disease and distributed in a completely randomized design (CRD) into 3 groups of 40 chicks each, which was further replicated to 4 and fed on diet containing soybean meal and those of test groups were fed diets containing alkali (1.5% NaOH) treated SKC partially replacing soybean meal nitrogen of reference diet (12.5%) without or with supplementation of methionine (0.2%). Individual body weight of chicks and replicate-wise feed intakes were recorded at weekly intervals throughout the experimental period. Feed consumption from 1 to 14, 28, 42 and 56 d of age was recorded for each replicate and feed conversion efficiency (weight gain/feed intake) for the respective period was calculated. Mortality was monitored on daily basis. On 28$^{th}$ day of experimental feeding, two birds of each replicate in each dietary group (8 birds/diet) were inoculated with 0.1 ml of a 1.0% suspension of sheep red blood cells (SRBC) and the antibody titre (log 2) was measured after 5 days by the microtitre haemmagglutination procedure. After 42 days of experimental feeding, a retention study of 4 days (43-47 d) duration was conducted on all birds to determine the retention of various nutrients such as DM, N, Ca, P and GE. On 43$^{rd}$ day of experimental feeding, one representative bird from each replicate of a dietary treatment (4/dietary group) was sacrificed, after fasting for two hours with free access to water, through cervical dislocation to observe the weight of dressed carcass, primal cuts (breast, thigh, drumstick, back, neck and wing), giblet (liver, heart and gizzard), abdominal fat and digestive organs. The body weight gain of chicks fed reference diet and those fed diet incorporated with NaOH treated SKC (12.5% replacement) with or without methionine supplementation was comparable during 0 to 4 weeks of age. However, dietary incorporation of alkali processed SKC replacing 12.5% nitrogen moiety of soybean meal resulted in growth retardation, subsequently as evidenced by significantly (p<0.05) lowered body weight gain during 0 to 6 weeks of age in birds fed diet incorporated with alkali processed SKC at 6.43% without methionine as compared to those supplemented with methionine or reference diet. Dietary incorporation of alkali (1.5% NaOH) processed SKC replacing 12.5% of soybean meal nitrogen in the diet of broiler chickens had no adverse effect on feed conversion ratio during all the weeks of experimental feeding. The humoral immune response (HIR) as measured by the antibody titre in response to SRBC inoculation was comparable among all the dietary groups. No significant difference in the intake and retention of DM, N, Ca, P or GE was noted among the chicks fed reference and alkali processed SKC incorporated diets with or without methionine supplementation. None of the carcass traits varied significantly due to dietary variations, except the percent weight of liver and giblet. The percent liver weight was significantly (p<0.05) higher in the birds fed diet incorporated with alkali processed SKC as compared to that in other two groups. Thus solvent extracted karanj cake could be incorporated after alkali (1.5% NaOH, w/w) processing at an enhanced level of 6.43%, replacing 12.5% of soybean meal nitrogen, in the broiler diets up to 4 weeks of age, beyond which the observed growth depression on this diet could be alleviated by 0.2% methionine supplementation.

Immune response of eel against fish pathogen, Edwardsiella tarda (어류 병원성 세균 Edwardsiella 에 대한 뱀장어의 면역 반응)

  • To study the immune responses of the japanese eel. Anguilla japonica, fish were injected intraperitoneally with several types of Edwardsiella tarda antigen, i. e., FKC(formalin killed cells), HKC(heat killed cells) or LPS(lipopolysaccharide), and the changes of immunocytes numbers, phagocytosis and agglutination titre in the peripheral blood of the fish were investigated. The number of lymphocytes in the peripheral blood of eels were decreased until 6 hours after injection, and then were turn to normal levels after 24 hours of injection. However, the level were slightly increased and were remained after 24 hours. The number of neutrophils of FKC, HKC or LPS injected fish were the highest at 12 hours after injection and were decreased slowly after that. Three weeks after the injections, the agglutination of antibody titre of all immunized groups were reached at 128 and were remained this level thereafter. However 6 weeks after the injections, that in HKC injected fish were dropped the level up to 4. Fish were injected with LPS and the blood from the fish were bled after 12 hours. Then the blood were incubated with E. tarda. Six hours after incubation, the phagocytic index was reached the highest level, 28.3. One week after the LPS injection, the blood were again bled and incubated with E. tarda. The phagocytic index at this time was 3.9. The phagocytic indexes of the fish injected with FKC and HKC, treted as same LPS injected fish as above, were 18.8 and 10.7, respectively. The phagocytic index of the control fish was 1.2. The antibacterial activities of normal antiserum against E. tarda were shown for both FKC and LPS injected fish, but not for HKC injected fish. The RPS(relative percentage of survival) of HKC, FKC and LPS injected fish in the challenge test were 10%, 20% and 30%, respectively. These results suggest that the effect of protection of the eel which were injected with antigen were varied with the method of preparation of the antigen.

Immunological characteristics of Edwardsiella tarda grown under iron-restricted condition (철 결핍 조건에서 배양된 Edwardsiella tarda의 면역학적 특성)

  • The immunogenicity of Edwardsiella tarda was surveyed under two different culture conditions. In SDS-PAGE patterns of the outer membrane proteins (OMPs) extracts of E. tarda, grown under Trypic soy broth (TSB) and TSB supplemented iron chelate 2,2‘-dipyridyl iron-restricted condition, were examined. The results showed that the iron-regulated outer membrane protein (IROMPs) with molecular masses of 68 and 73 kDa were expressed by bacteria grown in iron-chelate TSB.The pathogenicity was examined by intraperitoneal injection with live E. tarda grown under TSB, iron-chelate TSB and iron-supplemented TSB. The result of pathogenicity test showed significantly high mortality in the group of live E. tarda grown under iron-chelate TSB.The effect of formalin killed cell (FKC) of TSB cultured bacteria and 2,2'-dipyridyl FKC (DP-FKC) of cultured bacteria on the iron-chelate TSB on the development of protective immunity in olive flounder was studied. The level of immune response was evaluated with immunized fish at 1, 2, 3 and 4 weeks after immunization. The numbers of specific antibody secreting cells (SASCs) showed significantly increased level at 2 week after immunization in each group. The agglutination titre of immunized fish was significantly high level at 3 weeks after immunization.The level of protection in olive flounder at 1, 2, 3 and 4 weeks after vaccination was examined by intraperitoneal challenge test with live E. tarda.

Pathological studies on the morphological and functional changes of lymphomyeloid tissues overpopulated with melano- macrophage centers in tilapia, Oreochromis niloticus (나일틸라피아 임파조직내 Melanomacropage center의 형성과다(形成過多)에 따른 형태(形態) 및 기능변화(機能變化)에 관한 병리학적(病理學的) 연구(硏究))

  • To induce an overpopulated melanomacrophage centers(MMCs) within spleen and kidney in tilapias, two methods were applied through the intraperitoneal inoculation of V. anguillarum FKC with a repeated dose of saline(the 1st induction group), and of colloidal carbon (the 2nd induction group). In the 1st group, both number and size of MMCs were slightly increased in spleen and head kidney. However in the 2nd group the two hemopoietic tissues were nearly occupied with quite a large number of MMCs. Regardless of induction groups, many of MMCs were confined within the walls of blood vessels in the spleen. Especially in the 2nd group, the MMCs without fibrous capsules often had concentrically or eccentrically located, thin-walled lumens of vessels, which strongly suggests to be ellipsoids. In head kidney, nearly all MMCs were located within or just around the lymphocytic areas but the precise relationship to blood vessel wall was not obvious. Despite of such overpopulated MMCs, no histopathological degenerative findings in hemopoetic parenchymas of both organs were recognized. To evaluate the effect on defensive function, tilapias of the 2nd group were challenged with E. tarda. Susceptabilities to E. tarda were never increased but rather significantly decreased compared to control. Weekly antibody titres in sera were determined for all induction groups, in which the titres in the 1st and 2nd groups were 4 or 8 times higher than in the control, and then remained high until the 4th week. Also with the hemopoietic function, cellular compositions of peripheral blood were analyzed at weekly intervals but no significant changes resulted. From those results, it is suggested that overcrowding of MMCs would not lead to any morphological as well as functional deteriorations of spleen and head kidney but have an intimate association with enhancement of protective immune system in tilapias.
