CD29 및 CD98 활성 매개에 의한 Jurkat T 세포의 유착과 그 활용 (Cell-cell Adhesion of Jurkat T Cells Induced by CD29 and CD98 Activation and its Application)

  • Cell-cell adhesion managed by various adhesion molecules plays an important role in regulating functional activation of cells. This event mediates attachment of inflammatory cells to endothelial cells, interaction of antigen-presenting cells with T cells and metastatic adherence of cancer cells to epithelial tissue cells. Therefore, this cellular response is considered as one of therapeutic target to treat various cancers and inflammatory diseases. To develop proper model for evaluation of functional activation of adhesion molecules, the ability of U937 and Jurkat T cells responsive to various adhesion inducers such as phorbal-12-myristate-13-acetate (PMA), staurosporin and monoclonal antibodies to CD29, CD43 and CD98 was investigated using quantitative cell-cell adhesion assay. U937 cells made more cell-cell clusters by the treatment of antibodies to CD29 and CD43 than Jurkat T cells, while Jurkat T cells exhibited increased cell-cell adhesion ability in CD98 antibody treatment. In agreement, the surface levels of CD29 and CD98 were highly observed in U937 and Jurkat T cells, respectively. Therefore, our data suggest that Jurkat T and U937 cells can be used for model system to evaluate functional activation of adhesion molecules such as CD29 and CD98.

고유감각과 전정감각 입력이 외상성 뇌손상 쥐의 BDNF 발현에 미치는 영향 (The Effect of Proprioceptive and Vestibular Sensory Input on Expression of BDNF after Traumatic Brain Injury in the Rat)

  • Purpose : The purposes of this study were to test the effect of proprioceptive and vestibular sensory input on expression of BDNF after traumatic brain injury in the rat. Subject : The control group was sacrificed at 24 hours after traumatic brain injury. The experimental group I was housed in standard cage for 7 days. The experimental group II was housed in standard cage after intervention to proprioceptive and vestibular sensory(balance training) for 7 days. Method : Traumatic brain injury was induced by weight drop model and after operation they were housed in individual standard cages for 24 hours. After 7th day, rats were sacrificed and cryostat coronal sections were processed individual1y in goat polyclonal anti-BDNF antibody. The morphologic characteristics and the BDNF expression were investigated in injured hemisphere section and contralateral brain section from immunohistochemistry using light microscope. Result : The results of this experiment were as follows: 1. In control group, cell bodies in lateral nucleus of cerebellum, superior vestibular nucleus, purkinje cell layer of cerebellum and pontine nucleus changed morphologically. 2. The expression of BDNF in contralateral hemisphere of group II were revealed. 3. On 7th day after operation, immunohistochemical response of BDNF in lateral nucleus, superior vestibular nucleus, purkinje cell layer and pontine nucleus appeared in group II. Conclusion : The present results revealed that intervention to proprioceptive and vestibular sensory input is enhance expression of BDNF and it is useful in neuronal reorganization improvement after traumatic brain injury.

Subject : Twelve Sprague-Dawley rats were randomly assigned into group I and group II. After traumatic brain injury, group I was housed in standard cage for 7 days. Group II was housed in therapeutic cage after balance training for 7 days. Method : Traumatic brain injury was induced by weight drop model and after operation they were housed in individual standard cages for 24 hours. After 7th day, the rats were sacrificed and cryostat coronal sections were processed individually in goat polyclonal anti-BDNF antibody. The morphologic characteristics and the BDNF expression were investigated in injured hemisphere section from immunohistochemistry using light microscope. Result : Immunohistochemical response of BDNF in lateral nucleus, purkinje cell layer, superior vestibular nucleus and pontine nucleus appeared very higher in group II than in group I Conclusion : The present result revealed that simultaneously application of balance training for proprioceptive and vestibular sensory stimulation input and therapeutic environment in traumatic brain injured rats is enhance expression of BDNF and it is facilitates neural plasticity.

A Temperature-Controllable Microelectrode and Its Application to Protein Immobilization

  • This letter presents a smart integrated microfluidic device which can be applied to actively immobilize proteins on demand. The active component in the device is a temperature-controllable microelectrode array with a smart polymer film, poly(N-isopropylacrylamide) (PNIPAAm) which can be thermally switched between hydrophilic and hydrophobic states. It is integrated into a micro hot diaphragm having an integrated micro heater and temperature sensors on a 2-micrometer-thick silicon oxide/silicon nitride/silicon oxide (O/N/O) template. Only 36 mW is required to heat the large template area of 2 mm${\times}$16 mm to $40^{\circ}C$ within 1 second. To relay the stimulus-response activity to the microelectrode surface, the interface is modified with a smart polymer. For a model biomolecular affinity test, an anti-6-(2, 4-dinitrophenyl) aminohexanoic acid (DNP) antibody protein immobilization on the microelectrodes is demonstrated by fluorescence patterns.

Investigations into the immunomodulatory activity of Ulmus davidiana Planch extracts

  • Objective: Although the effect of Ulmus davidiana Planch (UD) extracts on collagen-induced-arthritis (CIA) and bone metabolism has been studies, research on its effect on human immunomodulatory activity is further a due. The objective of the present study was to investigate the immunomodulatory activity of UD on cellular and humoral immunity. Methods : Oral administration of the ethanolic and water extracts of UD, at doses of 20, 100 and 200 mg/kg in mice, dose dependently potentiated the delayed type hypersensitivity reaction induced both by sheep red blood cells (SRBC) and oxazolone. Results : It significantly enhanced the production of circulating antibody titre in response to SRBC in mice. Extracts of UD failed to show any effect on macrophage phagocytosis. Chronic administration of UD extracts significantly ameliorated the total white blood cell count and also restored the myelosuppressive effects induced by cyclophosphamide. Conclusion : The present investigation reveals that UD extracts possesses immunomodulatory activity.

안면신경마비의 치험례 (Case Report of Facial Nerve Paralysis)

  • Facial nerve paralysis(or Bell's palsy) which commonly occurs unilaterally, gives rise to paralysis of facial expression muscle. This condition is classified into symptomatic facial nerve paralysis due to intracranial tumor, post operative trauma, etc. and idiopathic facial nerve paralysis. To explain the etiology of idiopathic facial nerve paralysis, many hypothesis including ischemic theory, viral infection, exposure to cold, immune theory etc. were suggested, but there is no agreement at this point. The method to evaluate the facial nerve paralysis, when it occurs, consists of three stage scale method, image thechnics like CT and MRI, laboratory test to examine the antibody titers of viral infection, neurophysiologic test to evaluate the degree and prognosis of paralysis. Treatment includes medication, stellate ganglion block(SGB), surgery, physical therapy and other home care therapy. In medication, systemic steroids, vitamins, vasodilating-drug and ATP drugs were used. SGB was also used repeatedly to attempt the improvement of circulation and to stimulate the recovery of nerve function. Physical therapy including electric acupuncture stimulation therapy(EAST) and hot pack was used to prevent the muscle atrophy. When No response was showed to this conservative therapies, surgery was considered. After treating two patients complaining of Bell's palsy with medication(systemic steroids) and EAST, favorable result was obtained. so author report the case of facial nerve paralysis.

면역글로불린제제 효능평가를 위한 5종 B형간염 표면항원항체검출법의 비교 (Comparison of 5 Assays for Quantification of Antibody to Hepatitis B Virus Surface Antigen with Immunoglobulin G Preparations)

  • Five assays for anti-HBs were compared to improve potency test of Human lgG preparations. The three commercial EIA kits were optimized including dose response curve ranges and compared by conducting a co-laboratory study. After selecting the most reproducible EIA kit, methods comparison was performed with 22 samples in 5 different days. As a result, EIA (7.7 ${\pm}$ 5.3%) and MEIA (AxSYM: 3.7 ${\pm}$ 1.9%, IMx: 1.6 ${\pm}$ 0.8%) showed precision and accuracy (100.1 ${\pm}$ 12.6%). Therefore, the validated EIA assay was established and it is believed to be comparable to current MEIA.

나노 양자점 결합을 이용한 살모넬라 식중독균 검출 (Detection of Pathogenic Salmonella with a Composite Quantum Dot)

  • It is required to develop rapid methods to identify pathogenic Salmonella in food products for protecting and maintaining safety of the public health from Salmonellosis. The objective of the present study was to explore feasibility of the nanotechnology to detect pathogenic Salmonella rapidly in various samples. Sensitivity of the a composite quantum dot to detect Salmonella typhimurium in samples were evaluated. For selective detection of Salmonella, anti-Salmonella polycolonal antibody was utilized to capture and stain Salmonella. Quantum dots were attached onto Salmonella in the samples and produced fluorescent light. Fluorescence response of the composite quantum dot was measured with a commercial fluorescence meter. The fluorescence signal starts to increase with the samples in which higher concentration of the cells were contained. The sensitivity of the sensor was $10^6\;CFU/mL$ Salmonella spiked in PBS.

  • 단순포진 바이러스 감염을 유발하는Herpes simplex2형 바이러스의 감염에 관여하는 항원들과 중화항체 생산을 유발하는 주요 항원들의 위치를 확인하였다. Vero cell에 감염하였을 때 48시간 동안 31, 43, 59, 69 kDa 바이러스 항원들이 지속적으로 발현되었으며, 감염된 쥐에서 생산한 항체와의 반응에서는 51 kDa 항원이 가장 강한 반응을 보였다. 면역전자현미경으로 위치를 확인한 결과 colloidal gold가 바이러스 표면에 발견되는 것으로 보아 이 항원이 바이러스 표면에 존재하고 있는 것으로 확인되었다. 형광현미경 분석은 이 항원들이 감염된 세포 내에서 전반적으로 발견되었고 특히 세포 표면에서 많이 발현되고 있었다.

  • Eukaryotic cells continuously integrate intrinsic and extrinsic signals to adapt to the environment. When exposed to stressful conditions, cells activate compartment-specific adaptive responses. If these are insufficient, apoptosis ensues as an organismal defense line. The mechanisms that sense stress and set the transition from adaptive to maladaptive responses, activating apoptotic programs, are the subject of intense studies, also for their potential impact in cancer and degenerative disorders. In the former case, one would aim at lowering the threshold, in the latter instead to increase it. Protein synthesis, consuming energy for anabolic processes as well as for byproducts disposal, can be a significant source of stress, particularly when difficult-to-fold proteins are produced. Recent work from our and other laboratories on the differentiation of antibody secreting cells, revealed a regulatory circuit that integrates protein synthesis, secretion and degradation (proteostasis), into cell lifespan determination. The apoptotic elimination - after an industrious, yet short lifetime - of terminal immune effectors is crucial to maintain immune homeostasis. Linking proteostasis to cell death, this paradigm might prove useful for biotechnological purposes, and the design of novel anti-cancer therapies.
