Streptoalloteichus hindustanus ATCC 31219로부터 아미노글라이코사이드계 항생제에 내성을 지정하는 유전자의 클로닝 및 염기서열 결정 (Cloning and Sequencing of Resistance Determinants to Aminoglycoside Antibiotics from Sterptoalloteichus hindustanus ATCC 31219)

  • Streptoalloteichus hindustanus ATCC 31219, a nebramycin complex producer, is similar to Streptomyeces tenebrarius in a viewpoint of resistance to a wide range of aminoglycoside antibiotics. S. tenebrarius has resistance mechanisms of 16s rRNA methylation and aminogycoside modification. However, it is not known whether resistance mechanisms of Stall. hindustanus are the same as in S. tenebrarius. Therefore, we have tried to isolate resistance determinants from Stall. hindustanus. Two different types of aminoglycoside resistance determinants were isolated from Stall. hindustanus and expressed in Streptomyces lividans TK24. The apramycin resistance gene (amr) and the tobramycin resistance gene (tmr) isolated from Stall. hindustanus showed broad resistance spectrum against a dozen of aminoglycoside antibiotics. The complete nucleotide sequences of apramycin resistance gene (amr) were determined. The deduced amino acid sequence of the amr gene of Stall hindustanus ATCC 31219 showed extensive sequence homology to the 16s rRNA methylase gene (kamB) of S. tenebrarius.

  • Antibiotic resistance in human pathogens is a major public health issue. Some of the resistance problem can be attributed to the transfer of resistant bacteria from animals to people and the transfer of resistance genes from animal pathogens and commensal bacteria to human pathogens. Control measures include improvements in food hygiene to reduce the spread of zoonotic bacteria to people via the food chain. However, to specifically address the issue, the medical profession must control misuse and overuse of antibiotics in hospitals and community practice. In addition, the livestock industries and their advisors must reduce and refine the use of antibiotics in animal production and replace antibiotics with alternative disease control measures as much as possible.

  • Two hundred and twenty-seven strains of Escherichia coli isolated from 25 hens (12 hens received tetracycline neomycin and sulfadimethoxine, and 13 hens not received antibiotics) were studied for the drug resistance and distribution of R factors. About 74 per cent of E. coli strains isolated from hens of a herd received antibiotics were resistance to tetracycline (TC) streptomycin (SM), chloramphenicol (CM), kanamycin (KM), ampicillin (AP) and sulfisomidine (Su), alone or in combination thereof, but only a hen among a herd not received antibiotics excreted E. coli resistant to TC and SM. Among resistant strains, about 7% were found to be resistant to TC and SM, whereas 93% were resistant to three or more antibiotics. The most common pattern was the quadruple resistant to SM, TC, KM and Su (28.7%), and followed by triple ones to SM, TC and Su (25.3%), and SM, TC and KM (24.7%). About 84% of resistant strains carried R factors which were transferable to the recipient by conjugation.

  • Streptmyces fradiae NRRL B1195 and Streptomyces kanamyceticus IFO 13414 are highly resistant to the antibiotics they produce. The ribosomes of these organisms are found to be susceptible to the antibiotics, but the cell free extract of S fradiae is found to contain a phosphotransferase and an acetyltransferase which inactivate kanamycin and neomycin, and that of S. kanamyceticus an acetyltransferse which inactivates kanamycin and neomycin. The resistance of these organisms against streptomycin is found to be due to the resistant ribosomes; actually streptomycin activates their ribosomal systems for the synthesis of polyphenylalanine.

Characteristic of Antibiotic Resistance of Foodborne Pathogens Adapted to Garlic, Allium sativum L.

  • Antibiotic resistance of foodborne pathogens adapted to garlic (Allium sativum Linn.) was determined in order to understand the relationship between antibiotic resistance and garlic. The Gram (-) strains of Escherichia coli and Salmonella typhimurium and the Gram (+) strains of Bacillus cereus and Staphylococcus aureus were subcultured consecutively in a garlic broth, and the surviving colonies on the agar were selected as the adapted strains. Minimal inhibitory concentrations (MIC) for 15 antibiotics on the adapted strains were determined on Muller-Hinton Infusion agar. Adaptation to 1.3%(v/v) garlic juice increased MIC for vancomycin, aminoglycoside, and erythromycin on B. cereus, and for ampicillin and erythromycin on E. coli O157:H7. MIC of aminoglycosides, chloramphenicol, and vancomycin on the adapted S. aureus increased. The adapted S. typhimurium was more resistant to penicillin and vancomycin than the non-treated strain. The adapted S. typhimurium and S. aureus lost their antibiotic resistance in non-garlic stress conditions. However, the adapted B. cereus was still resistant to erythromycin and vancomycin, and the adapted E. coli was also resistant to erythromycin. Antibacterial garlic might increase the antibiotic resistance of E. coli, B. cereus, S. aureus, and S. typhimurium and this resistance can continue even without the stress of garlic. Therefore, garlic as a food seasoning could influence the resistance of such pathogens to these antibiotics temporarily or permanently.

Effect of NaCl on Thermal Resistance, Antibiotic Resistance, and Human Epithelial Cell Invasion of Listeria monocytogenes

  • This study evaluated the effects of NaCl on heat resistance and Caco-2 cell invasion of Listeria monocytogenes in broth media and sausage. A 10-strain mixture of L. monocytogenes was inoculated in tryptic soy broth containing 0.6% yeast extract (TSBYE), and sausage formulated with 0, 2, 4, and 6% NaCl. The medium was stored at 7, 15, 20, and $25^{\circ}C$ for 3-16 d, and medium samples were withdrawn at the appropriate time and challenged to 55, 60, and $63^{\circ}C$ to evaluate the thermal resistance of the pathogen. Sausage samples were stored at 7 and $25^{\circ}C$, and they were exposed to $63^{\circ}C$ to evaluate thermal resistance. NaCl-habituated L. monocytogenes strains NCCP10811 and NCCP10943 were examined for 12 antibiotics and Caco-2 cell invasion assay (only L. monocytogenes NCCP10943). Bacterial populations of L. monocytogenes generally increased (p<0.05) during the heat challenge as NaCl concentrations increased in both TSBYE and sausage samples. The antibiotic resistance of L. monocytogenes was not observed ($p{\geq}0.05$) when it was exposed to a single concentration of NaCl in TSBYE, but the pathogen obtained resistance to some antibiotics when exposed to a sequential increase of NaCl concentration. Invasion efficiency of L. monocytogenes NCCP10943 was not increased ($p{\geq}0.05$) with NaCl concentration increase. These results indicate that NaCl may increase the resistance of L. monocytogenes to heat and to some antibiotics, but may not increase Caco-2 cell invasion of L. monocytogenes.

H2O2 Generating Ability and Multi-Drug Resistance of Lactic Acid Bacteria Required for Long-Term Inpatient Treatment with Antibiotic Resistance

  • Purpose: In our study, in order to find lactic acid bacteria (LAB) with multi-drug resistance to antibiotics, we isolated 140 strains from 15 types of kimchi commercially available in Korea and 20 types of Kimchi made at home from January to December in 2016, and investigated their H2O2 generating ability and multi-drug resistance to antibiotics. Methods: In order to observe the H2O2 generation ability of LAB, we performed the experiment with methods such as Rabe, Hillier, and Kang. To test the antibacterial susceptibility of LAB, we used the disc agar diffusion method using MRS agar (Difco, USA) according to the CLSI and WHO test methods. There are 18 types of antibiotic discs used. Results: Out of the total numbers of 140 strains, 6 strains of Ent. Faecium, 25 strains of L. plantarum, 1 strain of L. rhamnosus, 3 strains of L. sakei, 1 strain of L. acidophilus, 1 strains St. thermophilus, and 7 of unidentified strains generated H2O2. The antibiotic susceptibility of Ent. Faecium indicated SXT, OX, NA, and E; and the antibiotic susceptibility of L. plantarum indicated NA; and the antibiotic susceptibility of St. thermophilus indicated NA, CC, RA, CTT, CM, and P ; and the antibiotic susceptibility of L. rhamnosus indicated SXT, VA, NA and CTT; and the antibiotic susceptibility of 6 strains of L. sakei indicated SXT, OX, NOR, NA, CTT and CIP, all indicating antibiotic resistance. In the case of multi-drug resistance to antibiotics for 53 strains of L. antarum, 8-drug resistance was the most common with 25 strains, followed by 7-drug-resistant strains with 18 strains, 9-drug-resistant strains with 4 strains, 6-drug-resistant strains with 3 strains, 5-drug-resistant strains with 2 strains, and 17-drug-resistant strains with 1 strain. In the case of multi-drug resistance to antibiotics for Ent. Faecium 27 strains, 9-drug resistance was most commonly identified as 9 strains, 8-drug resistance was identified as 6 strains, 7- and 11 drug resistances were identified as 4 strains each, and 4- and 6-drug resistances were identified as 1 strain each. Conclusion: Ent. Faecium, L. plantarum, L. rhamnosus, L. sakei, and St. thermophilus, shown to have anantibacterial activity in previous studies on LAB and shown to have and H2O2 generating ability, antibiotic resistance and multi-drug resistance in this study, are expected to be able to play an excellent role for long-term inpatients to use as an alternative to antibiotics and to cope with emerging antibiotic resistance.

하천에서 분리한 비소 내성세균의 유전적 특성 (Genetic Characteristics of Arsenic Compounds-Resistant Bacteria Isolated from Stream Water)

  • Several arsenic compound-resistant bacteria were isolated from Jung-Rang stream. The isolates, D-3, D-12, and D-14 were characterized phenotypically and genetically, and identified as Serratia liquefaciens, Klebsiella oxytoca, and Klebsiella pneumoniae, respectively. A plasmid of 67kb was found in Klebsiella oxytoca D-12 and designated as pMH12. Transfer of this plasmid from D-12 to E. coli HB101 was occurred, and the resulting transconjugant strains expressed the same level of heavy metal resistance as the donor strain. The physical presence of this plasmid in transconjugant was detected with agarose gel electrophoresis. Arsenite-sensitive derivatives of isolate D-12 were obtained with Mitomycin C treatment which cured pMH12. Antibiotics and heavy metal resistances were also examined to be used as a proper marker for the isolates in gene cloning. Isolate D-12 has resistance to several heavy metal ions such as $Cd^{2+}$ , $Zn^{2+}$ and $Hg^{ 2+}$ Also, all the other arsenite resistant isolates showed resistance to several heavy metal ions and antibiotics.

  • This report is to observe the rate of multiple resistance to various antibiotics among staphylococcus aureus and E. coli which were isolated with high frequency at our department during 1969. 1. One hundred fifty two strains(20.6%) resistant to four antibiotics multiple resistant was the highest incidence among 739 strains of Staphylococcus aureus. 2. Regarding the multiple antibiotic resistant of Staphylococcus aureus, 75strains(10.1%) to SA. PC. TC the three antibiotics resistant, have been observed, with the highest incidence, and next to this, 69 strains(9.3%) to SA. PC. SM. TC the four autibiotics resistant, and 68 strains(9.2%) to SA. PC. SM. CM. TC. SM. the six antibiotics resistant, have been observed in descending order. 3. The case of one hundred twenty seven strains(28.0%) of the six antibiotic multiple resistant among 454 strains of Escherchia coli was the highest incidence. 4. Concerning the multiple antibiotic resistant of Escherichia coli, 70 strains(l5.4%) to SA. SM. CM. TC. EM. FD. the six antibiotics resistant, have been observed, with the highest incidence, and next to this, 59 strains(l3.0%) to SA. SM. CM. TC. EM. FD. AC the seven antibiotics resistant, and 42 strains(9.3%) to SA. SM. CM. TC. EM. FD. KM. AC the eight antibiotics resistant have been observed in descending order.

  • Streptococcus sp. TR-1 which has inducible resistance to MLS antibiotics was isolated from soil samples in Korea. Streptococcus sp. TR-1 was cultured in Lysis broth, then a plasmid was isolated by modified Elliker method. Bacillus subtilis UOTO277 was transformed with that plasmid. This result showed that the plasmid has the gene relating with inducible resistance to MLS antibiotics. It was named pMB4 and its size was determined about 2.4 Kb by results of digestion with various restriction enzymes. Restriction endonuclease cleavage site map of pMB4 plasmid was made by double digestion of the plasmid. pMB4 plasmid has different restriction endonuclease site map from the other plasmids that have been discovered in Streptococcus sp. so far. And it could be identified that pMB4 plasmid does not have homology with ermK of Bacillus licheniformis EMR but has homology with ermC of Staphylococcus aureus from the results of Southern hybridization.
