  • One hundred and sixty one Escherichia coli strains isolated from 24 swine (11 swine fed with feedstuffs containing 7.5mg/kg of tetracycline and 13 swine not received antibiotic) were studied for the drug resistance and distribution of R factors. About 42 per cent of E. coli strains isolated from pigs of a herd fed with tetracycline (TC)-containing feeds were resistant to TC, streptomycin(SM), sulfisomidine(SA), ampicillin (AP) and kanamycin (KM), alone or in combination thereof, but none of the swine not receiving antibiotic containing feedstuffs excreted E. coli resistant to these drugs, Among resistant strains, 18.2% were found to be singly resistant to TC, whereas 81.8% were resistant to two or more antibiotics. The most common pattern was the triple resistant to TC, SM and SA(30.3%), and follolwed by double ones to TC and SM(24.2%). About one half of resistant strains carried R factors which were tranferable to the recipients by conjugation. In spite of feeding with feedstuffs containing only TC, high incidences of multiple resistance and R factors were observed in the E. coli isolated from these swine.

Interaction of Antibiotic with PAN and Cationic-Dyeable PET Fibers in Development of Infection Resistant Biomedical Materials

  • Interaction of a representative antibiotic, doxycycline(Doxy), with commercial poly(acrylonitrile) (PAN) and cationic-dyeable poly(ethylene terephthalate)(PET) fiber was studied in development of infection resistant biomedical materials. Regular PET was also employed for a comparison purpose. Their interactions were investigated at different treatment temperatures, times, and pHs. Fibers were also hydrolyzed by 1% NaOH for 1 or 2 hours at $85^{\circ}C\;and\;100{\circ}C$ to study effect of hydrolysis on antibiotic sorption. Infection-resistant characteristics of the substrates were evaluated by zone of inhibition (ZOI) test. Results revealed that a significant chemical change occurred in PAN and cationic-dyeable PET due to hydrolysis. Additional functional groups obtained by hydrolysis not only enhanced sorption of the antibiotics but also provided greater ZOI values, indicating substantial improvement in sustained infection resistance properties.

Prevalence and Antibiotic Susceptibility of Vancomycin-Resistant Enterococci in Chicken Intestines and Fecal Samples from Healthy Young Children and Intensive Care Unit Patients

  • The prevalence resistance genotype and antibiotic susceptibility of vancomycin-resistant enterococci (VRE) were determined. Prevalence of VRE in chickens, healthy children and intensive care unit (ICU) patients was 43.0%, 12.7% and 24.1%, respectively. Forty out of 56 isolates from chicken intestines were identified as Enterococcus faecium, and 12 were E. faecalis. All the isolates contained the vanA gene. Nine out of 13 VRE isolates from patients and two out of 21 from healthy young children were identified as E. faecium. The resistance types of E. faecium, E. gallinarium and E. casseliflavus were VanA, VanCl, and VanC2, respectively. The minimum inhibitory concentrations (MICs) of E. faecium, E. gallinarium, and E. casseliflavus to vancomycin were 512,8 and 4 g/ml, respectively. Specifically, E. faecium isolates were resistant to most of antibiotics except ampicillin and gentamicin. This is the first report of high VanA type VRE prevalence in nonhospitalized young children in Korea.

Rosmarinus officinalis 정유의 수종 항생제 감수성 및 내성 균주에 대한 억제효과 (In Vitro Inhibitory Activities of Essential Oils from Rosmarinus officinalis L. Against Antibiotic-Susceptible and Resistant Strains of Some Pathogenic Bacteria)

  • The in visto inhibitory activities of essential oils of the Rosmarinus officinalis as well as its main constituents were evaluated against antibiotic-susceptible and -resistant strains of Staphylococcus aureus, streptococcus pneumoniae, Salmonella enteritidis and S. typhimurium. The essential oil fraction of R. officinalis and its main components, 1,8-cineole and camphor, exhibited significant inhibitory activities against most of the tested strains in this study, with MICs(minimum inhibitory concentrations) racing from 0.5mg/ml to 16mg/ml. The total oil fraction showed higher activity than its main components, 1,8-cineole and camphor against S. aureus strains. No remarkable differences were evident between MICs of the susceptible and resistant strains of S. aureus. Among the tested strains, S. pneumoniae CCARM 3523, the resistant strain to norfloxacin, oxacillin and erythromycin exhibited significantly lower sensitivity to the tested oils than antibiotic-susceptible strain. The oils revealed mostly higher inhibitory activity against S. typhimurium than against S. enteritidis.

  • Microorganisms causing bovine mastitis, swine diarrhea and swine pneumonia were isolated from farms in Chungbuk and Kyunggi Province and the isolates were identified using microbial identification system(MIS). The most common isolates from bovine mastitis were Staphylococcus sp.,Streptococcus sp., and Corynebacterium sp., those from swine diarrhea were Escherichia coli, Salmonella sp., and Clostridium perfringens, those from swine pneumonia were Bordetella bronchiseptica and Pasteurella multocida. Examination of 16 antibiotics against these pathogens revealed that the incidence of antibiotic-resistant microoganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. These results suggest that the use of antibiotics in a farm should be controlled in order to decrease the number of antibiotic-resistant microorganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. Detection of $\beta$-lactam Antibiotic-resistant Genes in Eschericia coli using DNA Chip from Porcine Fecal Samples

  • There prevalence of $\beta$-lactamases bacteria in animals has been increased since 1990s [1]. The resistance in E coli which is mediated by $\beta$-lactamases hydrolyze the $\beta$-lactam ring eventually inactivate the antibiotics [2]. Generally, $\beta$-lactamases can be classified into four main groups and eight subgroups according to their functional and structural characteristics [3]. The detection of $\beta$-lactam antibiotic-resistant bacteria by DNA chip has been described [4]. The chip has a specific probe DNAs that contained the $\beta$-lactam antibiotic-resistant genes which was labeled by multiplex PCR reaction with a mixture of primer sets that were designed to amplify specific gene. Here we report the susceptibility of enteropathogenic E. coli isolated from pigs in Korea using the DNA chip in detecting $\beta$-lactam antibiotic-resistant genes. (omitted)

천연 추출물을 이용한 염색포의 견뢰도와 항생제 내성균주에 대한 항균효능 비교 (Comparison of the Fastness of Dyed Fabric using Natural Extracts and its Antibacterial Efficacy against Antibiotic-resistant Strains)

  • This paper aims to compare and verify the antibacterial effects of cotton fabrics naturally dyed with extracts of Ulmus davidiana var. japonica Nakai, Caesalpinia sappan, Saururus chinensis, and Artemisia princeps against antibiotic-resistant strains of bacteria. After natural dyeing of the test fabric, Al, Cu, Fe mordants were used. The color fastness against washing, rubbing, and perspiration of the cotton fabrics dyed with the 4 types of extracts were mostly excellent. However, the color fastness against light showed poor results for all four types. As for the antibacterial test method, MRSA (ATCC 33591) was applied to the cotton fabrics dyed with the four kinds of extracts and cultured for 24 hours. After that, the bacteria that proliferated on the fabrics were collected and spread on a solid medium. The bacteria were measured to find out the bacteriostatic reduction rate for the antibiotic-resistant strains. As a result of the analysis, all four extracts showed a high bacteriostatic reduction rate of more than 99% when the copper mordant was used. Even with the lack of a mordant, the bacteriostatic reduction rate was high, at 99.9% for Caesalpinia sappan and 94.6% for Saururus chinensis.

Milk Quality and Antimicrobial Resistance against Mastitis Pathogens after Changing from a Conventional to an Experimentally Organic Dairy Farm

  • The present study was to investigate the effect of the transition from conventional to organic dairy farming on the antimicrobial resistant pattern of pathogens in milk. A farm with tie-stall management, with an average herd size of 20 milking cows, was selected based on the owner' willingness to accept, for at least 6 months, the highly restricted protocol developed in this study. Comparisons of bacterial isolates and antimicrobial susceptibilities before changing to an organic farm system (BEFORE) and for 6 months after (AFTER) operating the experimental organic farm system were performed by Fisher's Exact Chi-square tests. Significant levels were defined at p<0.05. During the AFTER period, average frequency of antibiotic treatment was decreased from more than 3 cases/month to less than 1 case/month during which the antibiotic use was authorized only by the veterinarian. In total, 92 and 70 quarter milk samples from 24 and 18 cows during BEFORE and AFTER, respectively, were included in the study. Overall, isolates ranged from a non-resistant level for cephazolin to a very high resistant level to streptomycin (64.71% to 95.45%). Percentages of antimicrobial resistant isolates during BEFORE were significantly higher than during AFTER for ampicillin (43.48% and 5.88%, respectively) and streptomycin (95.45% and 64.71%, respectively). In conclusion, percentages of antimicrobial resistant isolates were decreased after 6 months of operating as an organic farm system.

Comparison of Antibiotic Resistance of Blood Culture Strains and Saprophytic Isolates in the Presence of Biofilms, Formed by the Intercellular Adhesion (ica) Gene Cluster in Staphylococcus epidermidis

  • To elucidate the question of whether biofilm formed by the intercellular adhesion (ica) gene cluster has influences on antibiotic resistance in Staphylococcus epidermidis, we compared 124 skin strains with strains isolated from 50 blood cultures that cause septicemic diseases. The results revealed that the blood culture isolates were more resistant to the antibiotics tested than the saprophytic isolates. Moreover, antibiotic multiresistance was more prevalent in the clinical isolates. In the blood culture isolates, $46\%$ of the strains were resistant to three or more antibiotics, whereas only $12\%$ of the saprophytic isolates were resistant to three or more antibiotics. Interestingly, these characteristics were highly correlated with the biofilm formed by the ica gene cluster. In biofilm-producing strains, $84\%$ of the blood culture isolates and $44\%$ of the saprophytic isolates were antibiotic multiresistant, whereas only $22\%=;and\;9\%$, respectively, were antibiotic multiresistant in biofilm-nonproducing strains. Additionally, in the biofilm-producing ica-positive strains, $89\%$ of the blood culture isolates and $57\%$ of the saprophytic isolates were antibiotic multiresistant. However, the rate of the antibiotic multiresistance in the ica-negative strains was very low, thus indicating that the biofim formed by the lea gene cluster in S. epidermidis is an important pathogenic factor in association with the antibiotic multiresistance.

Characterization of Extended-Spectrum-$\beta$-Lactamase Genotype TEM, SHV and CTX-M from Clinical Isolates of Klebsiella pneumoniae and Comparison with Antibiotic Susceptibility Test

  • Resent studies have reported increased isolation of extended-spectrum $\beta-lactamase$ (ESBL) producing strains at several hospital in Korea. We studied to investigate the isolation rates of ESBL strains from clinical isolates of Klebsiella pneumoniae and to characterize differences in types using analyses of genotyping and antibiotic susceptibility test. Antibiotic susceptibility test with confirmation of ESBL by double disk synergy test was performed on the 54 ESBL strains of Klebsiella pneumoniae from a hospital in Busan. Transfer of resistant gene in ESBL strains resistant to 3rd generated antibiotics was confirmed by transconjugation test using E. coli $RG176^{nal(r)}$. blaTEM, blaSHV, blaCTX-M genes were detected by PCR. ESBL producing strains had 100% of resistant rate to ampicillin, azteronam, cefazolin, cefepime and ceftriaxone ($\beta-lactam$ antibiotics). Forty strains of bla TEM$(74\%)$, 41 strains of bla SHV $(76\%)$, 23 strains of bla CTX-M $(43\%)$ were found, respectively. The strains had one or more genes. They had high resistant rates to $\beta-lactam$ antibiotics including cephalosporin. The resistant rates of strains with multiple resistant genes were higher than those of strains with single resistant gene.
