Objectives: Polygonati Rhizoma (PR) has containing the bioactive compounds such as poly sccharide A,B,C, oligosaccharide, amino acid, it has reported to anti-diabetes and hypertension, atherosclerosis. In this study, we were evaluates antioxidant and anti-physical fatigue effects of PR and steamed PR.Methods : The sample was divided into 5 groups-PR0 (PR without steaming process), PR1 (PR with once steaming process), PR3 (PR with third steaming process), PR6 (PR with sixth steaming process), PR9 (PR with ninth steaming process). We measured anti-oxidant activity through contents of polyphenol, flavonoid and DPPH, ABTS free radical scavenging capacity. And, anti-physical fatigue effect was evaluated using the swimming test, and the AMPK protein expressions in soleus muscle.Results : As a result, polyphenol, flavonoid, DPPH, ABTS free radical scavenging capacity of PR were increased as steaming times. Anti-physical fatigue effects by swimming test, PR0 have significantly increased, but steamed PR groups were decreased. The AMPK protein expressions of PR0 and PR1 groups were increased comparing with PR3, PR6 and PR9. All groups had effects on decreasing TG, creatine in blood serum, but had no effects on TC in blood serum.Conclusions : In conclusion, PR with 9 steaming process was more excellent than not-processed PR in anti-oxidant effect such as DPPH, ABTS radical scavenging activity and contents of polyphenol, flavonoid, but, not-processed PR increased swimming times than processed PR. These results suggest that processed PR has anti-oxidant effect as steaming times, and not-processed PR may be a novel potential anti-physical fatigue agents than processed PR.
Objectives : Jakyakgamcho-tang has been used as analgesic and spasmolytic for muscle pain. It has reported to anti-diabetes, anti-inflammation, and neuro-protective effects. A clinical study of muscle fatigue and pain improvement effect of Jakyakgamcho-tang has been reported with increasing frequency of clinical use. However, the anti-fatigue effect of Jakyakgamcho-tang in animal model has not been studied. In this study, we compared anti-physical fatigue effects of water and 30% ethanol extract of Jakyakgamcho-tang. Methods : The amounts of components contained in water and 30% ethanol extract of Jakyakgamcho-tang were compared by ultra-performance liquid chromatography. Anti-physical fatigue effects were evaluated using weight-loaded forced swimming test. We also investigated the effects of Jakyakgamcho-tang on the change of fatigue parameters by blood biochemical analysis. Results : The relative amount of components of Jakyakgamcho-tang were about 19-53% higher in the 30% ethanol extract than in the water extract. In the swimming test, 30% ethanol extract showed a significantly greater anti-fatigue effects than conventional water extract of Jakyakgamcho-tang. The 30% ethanol extract of Jakyakgamcho-tang improved the exhausted swimming time (54%), and serum lactate dehydrogenase (48%) and lactic acid (60%) levels compared with water extract of Jakyakgamcho-tang. Conclusions : These results showed that differences in the amounts of components by different extraction were associated with differences of anti-fatigue effect of Jakyakgamcho-tang. Thus, the 30% ethanol extraction method could be applied to dose-reducing formulations of traditional herbal medicines.

Objective: Fatigue can decrease both quality of life and work efficiency. Ginseng is one of the most popular herbal treatments for improving personal health, with applications in treating fatigue. However, the exact mechanisms of anti-fatigue effects are still unclear. Thus, we investigated the effect of red ginseng powder (RGP) on exercise capacity and peripheral fatigue using both behavioral and molecular experiments in mice. Design: Four-groups behavioral and molecular experiment. Methods: Male 6-weeks-old ICR mice were treated with distilled water, 100, and 200 mg/kg RGP for 5 days via oral administration. The exercise capacity of each animal group was measured by locomotor activity, rota-rod, hanging wire, and cold swimming tests. Additionally, after performing the treadmill to induce fatigue, lactate expression and molecular experiments were investigated using mice gastrocnemius. Results: Mice treated with RGP exhibited increased exercise capacity in the behavioral tests. Additionally, RGP induced a dose-dependent decrease in lactate levels after high-intensity exercise, and Monocarboxylate transporter (MCT) 4 expression increased in groups treated with RGP. However, there was no significant change in MCT1. Conclusions: These results suggest that RGP exerts several anti-fatigue properties by lower lactate and improved exercise capacity. Increased MCT4 expression may also affect lactate transport. Thus, this study suggests that the anti-fatigue properties of RGP might be associated with MCT4 activity.
Objective: Chicken essence and branched chain amino acid (BCAA) supplementation has been recognized to significantly relieve fatigue. To obtain chicken essence with high amounts of BCAA, spent hens herein was used to prepare dripped chicken essence (SCE) and compared with commercial dripped chicken essence (CCE) for in vivo anti-fatigue effect. Methods: To determine the effect on anti-fatigue by dripped chicken essence, the exhaustive swimming was performed. Thirty-two 7-week ICR mice were divided into four groups, which included the control group (CG), CCE, SCE-1X and SCE-2X. The mice were given daily oral administration (0.012 mL/g body weight/d). The fatigue index analysis was conducted weekly. Results: The results showed that SCE had a higher BCAA level as expected, and mice treated with dripped chicken essence (CCE and SCE) could significantly improve exercise performance. The lower blood lactate level, blood urea nitrogen level and creatine phosphokinase activity were found in the supplement of SCE group compared with the CCE group, which suggested that the SCE possessed strong anti-fatigue ability. This could possibly be due to the higher content of BCAA. Conclusion: In this study, SCE promoted recovery from physical fatigue in mice and elevated endurance ability. Among them, the double dose (SCE-2X) showed the strongest anti-fatigue ability. Taken together, spent chickens could be a good source of chicken essence to improve the effect of anti-fatigue.

Fatigue is a common complaint and affects the quality of life in modern people. Physical stress may induce activation of certain immune cells. Fermented porcine placenta (FPP) has been used to alleviate fatigue. Inflammatory cytokines are produced by physical stress and results in symptoms of fatigue. However, the role of FPP on fatigue-associated inflammatory cytokine production has not been elucidated yet. Thus, we estimated the anti-fatigue effect of FPP and its active components, leucine (Leu) and lysine (Lys) in activated RAW264.7 macrophages and forced swimming test (FST) fatigue animal model. Pretreatment with FPP, Leu, or Lys significantly inhibited the lipopolysaccharide (LPS)-induced tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 production without inducing cytotoxicity on LPS-stimulated RAW264.7 macrophages. FPP, Leu, or Lys inhibited the production of nitric oxide and downregulated the expression of inducible nitric oxide synthase on LPS-stimulated RAW264.7 macrophages. Furthermore, caspase-1 activities increased by LPS were significantly reduced by FPP, Leu, or Lys. In the FST, inflammatory cytokine levels of the mice administrated with FPP, Lys, and Leu were significantly reduced compared with the control group at 21 days. Collectively, these results show that anti-fatigue effect of FPP and its active components, Leu and Lys might be derived from the down-regulating of inflammatory mediators.
Seahorse, a syngnathidae fish, is one of the important organisms used in Chinese traditional medicine. Hippocampus abdominalis, a seahorse species successfully cultured in Korea, was validated for use in food by the Ministry of Food and Drug Safety in February 2016; however. the validation was restricted to 50% of the entire composition. Therefore, to use H. abdominalis as a food ingredient, H. abdominalis has to be prepared as a mixture by adding other materials. In this study, the effect of H. abdominalis on muscles was investigated to scientifically verify its potential bioactivity. In addition, the anti-fatigue activity of a mixture comprising H. abdominalis and red ginseng (RG) was evaluated to commercially utilize H. abdominalis in food industry. H. abdominalis was hydrolyzed using Alcalase, a protease, and the effect of H. abdominalis hydrolysate (HH) on the muscles was assessed in C2C12 myoblasts by measuring cell proliferation and glycogen content. In addition, the mixtures comprising HH and RG were prepared at different percentages of RG to HH (20, 30, 40, 50, 60, 70, and 80% RG), and the anti-fatigue activity of these mixtures against oxidative stress was assessed in C2C12 myoblasts. In C2C12 myoblasts, $H_2O_2$-induced oxidative stress caused a decrease in viability and physical fatigue-related biomarkers such as glycogen and ATP contents. However, treatment with RG and HH mixtures increased cell viability and the content of fatigue-related biomarkers. In particular, the 80% RG mixture showed an optimum effect on cell viability and ATP synthesis activity. In this study, all results indicated that HH had anti-fatigue activity at concentrations approved for use in food by the law in Korea. Especially, an 80% RG to HH mixture can be used in food for ameliorating fatigue.

The aim of this study was to evaluate the effect of hot water extract of peduncle obtained from Hovenia dulcis Thunb (HD) which is commercially developed for the protective effect on the alcoholic hepatotoxicity, on the endurance capacity for weight loaded forced swimming mice. The swimming times to exhaustion in mice fed 100 and 200 mg/kg HD for 2 weeks were prolonged 3.6 and 3.7 fold, and for 4 weeks 1.9 and 2.7 fold compared with each vehicle control ($42.8{\pm}20.5$ min and $67.7{\pm}47.8$ min, for 2 and 4 weeks), respectively. Blood biochemical parameters for ALT, AST, creatinine and BUN were not significantly different between from HD fed or control mice. Although HD fed mice swam over 2 fold longer time than vehicle control mice at 4 weeks, blood biomarkers of physical fatigue such as glucose, triglyceride and free fatty acid, lactate were not significant different and even tended to ameliorate. Hepatic lipid peroxidation and SOD activity did not significantly change in HD fed- and vehicle control exhausted swimming mice at 2 or 4 weeks. However, catalase activity in HD-fed mice was significantly increased in a dose-dependent manner compared with vehicle control mice. The present study indicates that HD improved physical fatigue and exercise performance in mice. Therefore, it has a potential for the pharmacological effect of anti-fatigue.

Objectives: We evaluated the anti-fatigue effects of Rh2-enriched Korean ginseng (Ginseng Rh2+) using a forced exercise-induced chronic fatigue mouse model. Methods: ICR male mice were subjected to running wheel for 1 h, 5 days/week during 4 weeks, and running velocity was gradually increased. Each running session was followed by oral administration of distilled water, Ginseng Rh2+ (150 or 300 mg/kg), or N-acetyl-L-cysteine (NAC, 100 mg/kg) 1 h later. The exercise tolerance and forced swimming test were performed to evaluate the fatigue condition. Results: Chronic forced exercise reduced the physical activity, as evidenced by the behavioral tests, which were notably ameliorated by Ginseng Rh2+ treatment. Ginseng Rh2+ treatment also attenuated the alterations of energy metabolism and oxidative stress in skeletal muscle tissues and/or sera, including malondialdehyde (MDA), lactate concentration and its related factors (lactate dehydrogenase, blood urea nitrogen, and glucose levels). Conclusion: These findings strongly suggest that Ginseng Rh2+ exerts a potent anti-fatigue effect through modulation of energy metabolism and oxidative response.
Background: In physical activity or labor, the human body is in a state of high intensity stress, and all parts or physiological functions of the body respond positively to maintain or balance the need for movement. The human body has many physiological changes in the process of movement, and fatigue is the external manifestation of various complex changes inside the human body. Fatigue is also a physiological mechanism of self-protection after the body reaches a certain level of activity, which can prevent the occurrence of life-threatening excessive functional failure. The generation of fatigue is a very complex process, and its mechanism has not been concluded yet. Therefore, it is an important work to search and screen the effective components of natural plants that have anti-fatigue effect and to explore their mechanism. Methods: This was a 8-week, randomized, double-blind, placebo-controlled clinical trial. A total of 110 subjects who passed physical examination were included according to the scheme design, and randomly divided into a test group which was given KRG and a placebo control group. The calculation is carried out according to the standard of sub-high-intensity exercise test. Results: There was no adverse effect on safety index of subjects after taking red ginseng capsule. After KRG treatment, subjective strength grade is significant lower than placebo treatment. Blood lactic acid content is significantly get lower after trial in KRG group, and significant lower than placebo group. Creatine phosphokinase(CK) content is significantly get lower after trial in KRG group, and significant lower than placebo group. Conclusion: According to the criterion in the test scheme, the result shows that KRG is helpful on relieving physical fatigue.

We investigated the effects of prolonged exposure in hot environmental condition and ingestion of fluid on various physiological variables including plasma glucose, lactate, the rating of perceived exertion (RPE), and heart rate as well as golf putting performance. Six male professional golfers were voluntarily participated in three different putting trials which were separated by seven days of time interval period. Three different putting trials were conducted at either 20℃ or 32℃, or 32℃ + Fluid ingestion. Performing 32℃ + Fluid ingestion trial, all subject ingested sport drink as much as their body mass was decreased. For each experiment, all subjects were undertaken total 48 putting, which separated by four x 12 putting in four different time points (i.e., Rest, 1 hr, 2 hr, and 3 hr). Plasma glucose concentration was significantly decreased with hot ambient condition but it was almost fully recovered by fluid ingestion. Plasma lactate concentration was significantly higher when subjects were exposed in hot environmental condition, and it did not change with fluid ingestion. There was a no different in putting performance and psychological fatigue level (performed by GRID test) at any environmental conditions. The RPE, commonly used for evaluating of physical fatigue level, was significantly dropped by fluid ingestion which indicates lower physical fatigue level. In addition to this, heart rate (HR) was also significantly decreased after fluid ingestion. Based on these results, it was concluded that the ingestion of fluid during prolonged exposure in hot ambient condition decrease the degree of physical fatigue levels and heart rate, which will possibly improve the golf performance when exposed in extreme weather condition in summer.
