A New Attempt to Establish the Extrinsic Aging Hair Model to Evaluate The Response to Aging in Physical Property (모발 노화에 따른 물성변화와 외인성 노화모델의 개발)

  • Human tissue undergoes aging by the oxidant damage via structural change and its physical properties. The skin aging process is well known and many evaluations have been conducted. However, studies on hair aging were relatively few and thus care for aging hair is difficult. This study aims to fabricate an aging hair and identify anti-aging effect with known ingredient in anti-aging. First of all, physical properties of aging hair of age 60s by physiologically intrinsic factors were compared to those of the hair made by various extrinsic factors such as several chemical reactions and iteration numbers of the treatments. The extrinsic aging hair of this study relates to the less amount of lipid and to the hair of perm treated once accordingly, wherein several physical properties, preferably comprise roughness and tensile strength, present a novel concept of the intrinsic aging hair. The penetration of peptide into the aging hair was leading the extrinsic hair towards more structurally directed a younger hair. In addition to the structural change, the penetration of the peptide enhanced texture and tensile strength of the aging hair. These patterns have been also found in addition of propolis. For the first time, these qualitative studies exhibit that indeed our extrinsic aging hair well describes the anti-aging efficacy as a receptor for a cross-linker and the ingredients of human hair.

CopA3 peptide from Copris tripartitus induces apoptosis in human leukemia cells via a caspase-independent pathway

  • Our previous study demonstrated that CopA3, a disulfide dimer of the coprisin peptide analogue (LLCIALRKK), has antibacterial activity. In this study, we assessed whether CopA3 caused cellular toxicity in various mammalian cell lines. CopA3 selectively caused a marked decrease in cell viability in Jurkat T, U937, and AML-2 cells (human leukemia cells), but was not cytotoxic to Caki or Hela cells. Fragmentation of DNA, a marker of apoptosis, was also confirmed in the leukemia cell lines, but not in the other cells. CopA3-induced apoptosis in leukemia cells was mediated by apoptosis inducing factor (AIF), indicating induction of a caspase-independent signaling pathway.

Interactions of Tricyclic Isoxazole Analogues with ${\alpha}_{2c}$-Adrenoceptor by Homology Modeling (상동성 모델링을 이용한 Tricyclic Isoxazole 유도체와 ${\alpha}_{2c}$-Adrenoceptor의 상호작용)

  • Adrenoceptor has been considered to be an important target in psychiatric disorders. Based on x-ray structures of bovine rhodopsin, we established homology model of ${\alpha}_{2c}$-adrenoceptor (ADA2C_rat) and then analyzed docking from binding model of receptor-ligand complex with high-active compound No.29 in tricyclic isoxazole analogues (1-30). We observed that the N (1.907 $\AA$) and O (1.712 $\AA$) atoms of isoxazole ring on the docked ligand (No.29) formed H-bonding interaction with O-H of Ser5.32 and carmeron phenyl ring centroid of tricyclic isoxazole formed $\pi-\pi$ interaction at 3.342 $\AA$ distance with phenyl ring centroid of Phe6.52. According to predictions of blood-brain distribution (logBB) through penetration of blood-brain barrie (BBB) and polar surface area (PSA) of the ligands, the high-active compound No.29 has values of logBB=-0.203, PSA=67.50, respectively. These results suggest that the high-active compound No.29 is a novel anti-depressant with the characteristics such as dopamine and serotonin.

The study on stabilization of Retinol-Nanoemulsion using Skin Lipid Matrix(SLM)

  • In cosmetic area, retinol is prominent ingredient for anti-wrinkle but unstable against light, heat, oxygen and so on. Therefore the stabilization of retinol is required. Here, we capsulated doubly retinol in the SLM(Skin Lipid Matrix) that makes three dimensional lamellar structure similar to skin, after formation of primary liposome (retinol-nanoemulsion). First, we make primary liposome from retinol / hydrogenated lecithin / polysorbate20 / caprylic & capric triglyceride / ethanol / and so on, and the mean diameter to 70 nm, using microfluidizer passed three times at 800 Bar, repeatedly. Then we produce DC-liposome (doubly capsulated-liposome) that was encapsulated primary liposome with SLM made of hydrogenated phosphatidyl choline / caprylic & capric triglyceride / 1, 3-butylene glycol / ceramide3 / cholesterol /etc. We measured for color stability against light and heat with chromameter. As a result of this experiment, we observed DC-liposome was more than from 1.5 to 3 times as stable as general liposome. Livability of retinol has improved from 2 to 6 times when we analyzed it by HPLC. Also, penetration effect of DC-liposome has improved.

  • Corrosion of reinforced concrete embedded in concrete is a deterioration phenomenon due to intrusion of embodied or Airborne chloride ions. Corrosion of a embedded steel increases the volume of the rebar and causes damage to the structure such as cracking and peeling of the concrete. This causes penetration of various corrosive factors and accelerates the corrosion of reinforcing bars, which has a serious effect on the durability of the structure. Researches on the corrosion phenomenon of these rebars by electrochemical methods have been carried out for a long time, but it is a lack of research in Korea. Therefore, in this study, one of electrochemical experimental methods, Tafel extrapolation method, was used to evaluate the performance of reinforcing bars according to the amount of NaCl and LiNO2 added to concrete.

  • Ultra-wideband(UWB) radar is widely used in many penetration radar applications, such as ground-penetrating radar and foliage-penetrating radar, because it has many advantages in detecting concealed objects. One type of UWB radar system is random noise radar, which many be robust to jamming environment. However conventional random noise radar requires high-speed analog-to-digital convertor(ADC) for matched filtering. In this thesis, a pseudo-random noise radar system that maintains anti-jamming characteristics but does not require high-speed ADC is researched. and The UWB system is implemented in a low frequency system, and its performance has been demonstrated by experiment, which proves the concept of the proposed pseudo-random noise radar system.

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental facts play critical roles in cellular damage and aging. The extract of tinged autumnal leaves of maple tree(Acer palmatum) has proven to be a powerful antioxidant. The Acer palmatum extract is very effective on the stabilization of biological membranes( containing unsaturated fatty acid). We studied photoprotective effect of the extract against UVB-induced cytotoxicity in human keratinocytes. The extract improved cell viability comparing to control after UVB irradiation. In the determination test of pro inflammatory cytokines the extract decreased expression of interleukin 1 a and 6, which play an important role in inflammation and skin erythema caused by UV. We also studied property of varying cosmetic formulations on the percutaneous absorption of the extract. After 24 hour in vitro penetration study, the content of the extract was more highly detected in skin residue part. This result showed the extract had relatively high compatibility of skin in our emulsion system. On human skin, after appling the product containing the extract we obtained a good result of antiwrinkle effect by skin visiometer. In conclusion, the Acer palmatum extract is a photoprotective and very effective in stressed and aged skin care. And we can predict the extract mainly affects on the skin cell and tissue in our emulsion system.

  • The prodrug and antedrug concepts, which were developed to overcome the physical and pharmacological shortcomings of various therapeutic classes of agents, employ diametrically different metabolic transformations. The prodrug undergoes a predictable metabolic activation prior to exhibiting its pharmacological effects in a target tissue while the antedrug undergoes metabolic deactivation in the systemic circulation upon leaving a target tissue. An increased therapeutic index is the aspiration for both approaches in designing as well as evaluation criteria. The recent research endeavors of prodrugs include the gene-directed and antibody-directed enzymatic activation of a molecule in a targeted tissue, organ specific delivery, improved bioavailabilities and cellular penetration of nucleotides. As for antedrugs, emphasis in research has been based upon the design and synthesis of systemically inactive molecule by incorporating a metabolically labile functional group into an active molecule.

Research Trend of Antiviral Natural Products for Companion Animal (천연물 유래의 반려동물 항바이러스활성물질 연구 동향)

  • Recently, companion animal culture has grown rapidly and mature, raising interest in preventing and treating animal diseases. In particular, viral infection was a serious threat to companion animal health because there was no proper antiviral drugs. Synthetic antiviral drugs have limitations such as low efficiency, toxicity, and occurrence of resistant viruses. Therefore, attempts to find new anti-viral drugs from natural sources have continued. This review focused on the natural products and active substances that exhibit antiviral activity against three viruses: canine distemper virus (CDV), canine parvovirus (CPV), and feline calicivirus (FCV) that cause fatal diseases in dogs and cats. Natural plant extracts, flavonoids, polysaccharides, alkaloids and saponins showed antiviral activity with various mechanisms and differences in activity depending on the structure. Especially, quercetin and epigallocatechin-3-gallate (EGCG) showed antiviral activity through a multi-mechanism that interferes with the attachment and penetration stages of the virus and inhibits the viral polymerase within the cell. Some natural plant extracts showed a virucidal activity and showed the potential effect as a preventative agent to prevent the viral infection. This review is expected to provide research trend on the development of antiviral natural products for companion animals.

Effect of the Extract of Stachys Affinis on DNFB-induced Allergic Contact Dermatitis in Mice (DNFB에 의해 알레르기성 접촉성 피부염이 유도된 마우스에서 초석잠 추출물의 효과)

  • This study aims to evaluate the anti-inflammatory effect of 80% ethanol extracts of Stachys affinis (MQ) on 1-fluoro-2,4-Dinitrofluorobenzene (DNFB) induced atopic dermatitis (AD) in Balb/c mice. AD-like allergic contact dermatitis was induced by challenge of DNFB on the ear after DNFB sensitization on the back sides of mice. MQ alleivated clinical severity in AD-like skin lesions. In addition, ear thickness of epidermis and penetration of inflammatory cells in AD-like skin lesions were decreased by topical application of MQ. The the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-4 (IL-4) were measured in AD mice using ELISA kits. Levels of TNF-α and IL-4 in serum were significantly decreased by topical application of MQ. Therefore, this study could give a clinical basis that MQ could be a agent to prevent AD.
