소양인(少陽人) 형방패독산(荊防敗毒散) 전탕액이 노화쥐의 신장과 비장 세포의 항산화능에 미치는 영향 (Anti-Oxidative Effect of Soyangin Hyeongbangpaedok-san Decoction in Kidney and Spleen Cells of Aged Rats)

  • 1. Objectives: The purpose of this study is to find out the Anti-Oxidative effects of Soyangin Hyeongbangpaedok-san(HBP) in kidney and spleen cells of aged rats. 2. Methods: Aged rats used in this experiment were 6, 52, 68 weeks old. Each age group was divided into three groups again. One group was given no treatment, another group was dosed normal saline and the other group was dosed HBP decoction. The antioxidant effects of HBP decoction were measured by the levels of SOD, GSH, MDA and NO. 3. Results: and Conclusions: 1) The activity of SOD was significantly increased in kidney cells of 68w-HBP group. Deterioration of the activity of SOD in kidney cells was significantly suppressed according to increasing in age of the week. 2) The level of NO was significantly decreased in kidney cells of 68w-HBP group. Deterioration of The level of NO in kidney cells was significantly suppressed according to increasing in age of the week. 3) The activity of SOD was increased in spleen cells of 52w and 68w-HBP group. 4) The level of GSH was significantly increased in spleen cells of 68w-HBP group.

Effect of Mixture Composed of Jeju' Scoria and Ecklonia cava on Anti-inflammation

  • The purpose of this study is to investigate the anti-inflammation effect of the mixture, consisting of a series of different ratio of Ecklonia cava extract and scoria. Also, to make more functional scoria powder into cosmetic material, studies on the toxicity by cell viability assay. Scoria is found in large amounts in Jeju Island, as an adsorbent of heavy metal ions ($Ni_2{^+}$, $Zn_2{^+}$, and $Cr_3{^+}$) in an aspect of its efficient utilization. Marine plants such as Ecklonia cava contain high amounts of polyphenolic antioxidants. The purpose of this study was to examine the anti-oxidative and anti-inflammation effects of combination of mixture of Eckloina cava extrat and scoria with optimal ratio. Therefore, this study suggested that combination of mixture of Eckloina cava extrat and scoria and its attenuated the oxidative and inflammatory reactions.

육미지황환(六味地黃丸)의 elastase 활성과 DPPH, NO 소거능에 미치는 영향 (Effects of Yukmigeehwang-hwan Extracts on the Elastase Activity and DPPH and NO Scavenging Activities Original Articles)

  • Objectives : Elastic fibers are found in the skin, lungs, arteries, veins and other structures. Elastases destroy the elastic fibers and cause the emphysema and pulmonary hypertension. Oxidative stress is needed for these pathologic changes. Accordingly, present study was designed to investigate the effect of Yukmigeehwang-hwan extracts (YHE) on elastase activity and anti-oxidative effects of YHE. Methods : The inhibitory effects on elastase and DPPH and NO free radical scavenging activities of YHE were measured. Results : The elastase activity was significantly inhibited by YHE. YHE significantly scavenged DPPH and NO free radicals as well. Conclusion : YHE showed the elastase-inhibiting effects and anti-oxidative activities in vitro. These results suggest that YHE may have potential roles in the treatment of pulmonary emphysema and pulmonary hypertension.

은갑방가감(銀甲方加減)이 혈전병태유관인자(血栓病態有關因子)와 산화적손상(酸化的損傷)에 미치는 영향(影響) (The Effect of Eungapbang-gagam on Thrombus Disease Related Factors and Oxidative Stress)

  • Purpose: In this study, we investigated the anti-thrombotic efficacy of "Eungapbang-gagam(EGB)" currently used in clinical treatment of PID Methods: We studied inhibitory effect of platelet cohesion, suppressive effect of GPIIb/IIIa activity, inhibitory effect of $TXB_2$ and $PGE_2$ biosynthesis, and oxidative damage suppression effects of "EGB" in vitro. Also, suppression of pulmonary embolism and changes of related factors in dextran coagulation condition model were studied in vivo. Results: In this study, EGB extract showed dose-dependent inhibitory effect on platelet coagulation induced by ADP, epinephrine, collagen, arachidonic acid. Also it showed dose-dependent inhibition effect on GPIIb/IIIa activities compared to the control group. EGB extract significantly suppressed the decrease of speed of bloodstream caused by blood coagulation in dextran coagulation condition model and increased the number of platelets and amount of fibrinogen, and decreased the APTT in dextran coagulation condition model compared to the control group. EGB extract showed dose-dependent decrease of oxidative damages caused by DPPH and superoxide anion radicals, whereas dose-dependent increase of superoxide dismutase like activity was observed compared to the control group. Conclusion: We confirmed the anti-thrombosis and anti-oxidative efficacy of "Eungapbang-gagam". Various clinical applications of "Eungapbang-gagam" as well as use of data for the construction of EBM is anticipated.

제주 자생 물사과 가지 유래 Flavonoid 화합물의 항산화 활성 (Anti-oxidative Activities for the Flavonoids of the Syzygium aqueum Burm.f. Alston Branches from Jeju Island)

  • 본 연구에서는 물사과 가지 추출물 및 용매 분획물의 항산화 활성과 세포보호효과를 검색하고 유효성분을 분리하여 화학구조를 규명하였다. 물사과 가지 추출물 및 용매 분획물의 DPPH 및 $ABTS^+$ 라디칼 소거 활성을 측정한 결과, 에틸아세테이트 및 부탄올 분획물에서 우수한 라디칼 소거 활성이 있음을 확인하였다. 또한 $H_2O_2$로 유도된 세포 손상에 대한 세포보호효과를 확인한 결과, $20{\mu}g/mL$의 농도에서 추출물 및 에틸아세테이트 분획물에서 보호효과가 나타남을 확인하였다. 항산화 활성이 좋은 에틸아세테이트 분획물에서 유효성분을 찾고자 medium pressure liquid chromatography (MPLC)를 실시하여 2개의 화합물을 분리하였으며 $^1H$$^{13}C$ NMR 데이터 분석 및 문헌 비교를 통하여 화학구조를 동정하였다; pinocembrin (1), desmethoxymatteucinol (2). 분리된 화합물에 대한 DPPH 및 $ABTS^+$ 라디칼 소거 활성을 측정한 결과, 화합물 1, 2 모두 농도의존적으로 우수한 라디칼 소거 활성이 나타났다. $100{\mu}M$의 농도에서 화합물 1, 2는 $H_2O_2$로 유도된 세포손상에 대하여 세포보호효과를 나타냈다. 이러한 연구 결과를 바탕으로 물사과 가지 추출물은 천연 항산화 소재로써 활용 가능할 것이라 사료된다.

산화동물 모델 흰쥐 간세포에 대한 지구자(枳椇子)와 지구지(枳椇枝)의 항산화 효과 연구 (Study on Antioxidant Effect of Hoveniae Semen cum Fructus and Hoveniae Ramulus on Liver Cells Isolated from Oxidatively Stressed Rat)

  • Objectives : The Objective of this study is to compare the anti-oxidative effect of the Hoveniae Semen cum Fructus (HScF) to that of Hoveniae Ramulus (HR) in vivo. Methods : The extracts of HScF and HR were tested in vivo for their anti-oxidative effects. SD Rats were injected with the AAPH (2.2'-azobis (2-aminodinopropane) hydrochloride) to induce the acute oxidization stress, and the oxidized rats were administered with the HScF and HR decoction orally. The RBC, WBC and PLT populations in plasma, and the blood chemistry levels of the rats were measured to observe the overall changes in the blood. The anti-oxidative effects of the HScF and HR decoctions were investigated on the liver tissues of the oxidated rats. Results : 1. Thrombocyte decreased significantly in the group medicated with HScF. 2. Blood glucose increased significantly in the group medicated with HR. 3. SOD activity increased significantly in both the group medicated with HScF and that with HR. 4. NO concentration decreased significantly in both the group medicated with HScF and that with HR. 5. The catalase content increased significantly in both the group medicated with HScF and that with HR. Conclusions : The HScF and HR extracts have an anti-oxidative effect.

  • Samul-tang (SMT), which was firstly described in (Hwajegukbang) Song dynasty, is well known remedy for blood diseases in Oriental medicine. SMT is traditional herbal-remedy composed of Rehmanniae Radix Preparat, Angelicae Gigantis Radix, Cnidii Rhizoma and Paeoniae Radix. Recently, SMT has known to have anti-oxidative action. However, the reports on anti-oxidantic action in neuroglial cells are rare. In addition, the exact mechanisms are unclear. For these reasons, we investigated the protective effects of SMT on cell death induced by oxidative stress using C6 glioma cells. In our results, SMT accelerated proliferation rates of C6 cells in vitro. In addition, levels of LDH release induced by oxidative stress were lowered by treatment with SMT. Finally, protective effects on cell death induced by chemicals such as paraquat and rotenone were observed. In conclusion, these results suggest the possibility to protect brain cell or neuronal cell from damage induced by oxidative stress.

Anti-Apoptotic Effects of Catalpol on Preimplantaion Porcine Embryos

  • Catalpol, an iridoid glucoside, isolated from the root of Rehmannia glutinosa Libosch. It possesses a broad range of biological and pharmacological activity including anti-tumor, anti-inflammation and anti-oxidant by acting as a free radical scavenger. Therefore, in this study, the effects of catalpol on blastocyst development, expression levels of reactive oxygen species (ROS) and apoptotic index were investigated in porcine embryos. After in vitro maturation and fertilization, porcine embryos were cultured for 6 days in porcine zygote medium 3 (PZM-3) supplemented with catalpol (0, 100, 200 and $400{\mu}M$, respectively). Blastocyst development not significantly improved in the catalpol treated group when compared with control group. Otherwise, the intracelluar levels of ROS were decreased and the numbers of apoptotic nuclei were reduced in the catalpol ($100{\mu}M$) treated porcine blastocysts (P<0.05). On the other hand, blastocyst development was significantly improved in the catalpol ($100{\mu}M$) treated group when compared with the untreated catalpol group under $H_2O_2$ ($200{\mu}M$) induced oxidative stress (P<0.05). Otherwise, the intracellular levels of ROS in catalpol ($100{\mu}M$) treated group were significantly decreased in the untreated catalpol group under $H_2O_2$ ($200{\mu}M$) induced oxidative stress (P<0.05). Furthermore, the total cell numbers of blastocysts were significantly increased (P<0.05) in the catalpol ($100{\mu}M$) treated group under $H_2O_2$ ($200{\mu}M$) induced oxidative stress, whereas numbers of apoptoic nuclei were significantly reduced (P<0.05). In conclusion, our results indicate that treatment of catalpol may have important implications for improving developmental competence and preimplantation quality of porcine embryos through its anti-oxidant and anti-apoptotic effect.

Anti-inflammatory and anti-oxidative effects of 3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride on β-amyloid-induced microglial activation

  • We aimed to assess the anti-inflammatory and antioxidative properties of KHG26792, a novel azetidine derivative, in amyloid ${\beta}$ ($A{\beta}$)-treated primary microglial cells. KHG26792 attenuated the $A{\beta}-induced$ production of inflammatory mediators such as IL-6, $IL-1{\beta}$, $TNF-{\alpha}$, and nitric oxide. The levels of protein oxidation, lipid peroxidation, ROS, and NADHP oxidase enhanced by $A{\beta}$ were also downregulated by KHG26792 treatment. The effects of KHG26792 against the $A{\beta}-induced$ increases in inflammatory cytokine levels and oxidative stress were achieved by increasing the phosphorylation of $Akt/GSK-3{\beta}$ signaling and by decreasing the $A{\beta}-induced$ translocation of $NF-{\kappa}B$. Our results provide novel insights into the use of KHG26792 as a potential agent against $A{\beta}$ toxicity, including its role in the reduction of inflammation and oxidative stress. Nevertheless, further investigations of cellular signaling are required to clarify the in vivo effects of KHG26792 against $A{\beta}-induced$ toxicity.
