Anti-oxidative and anti-inflammatory effects of Odukhwan and Sasinhwan in RAW264.7 cells (오덕환(五德丸)과 사신환(四神丸)의 항산화 및 항염증 효과)

  • Objectives : The aim of this study was verification of the anti-oxidative and anti-inflammatory effects of Odukhwan(ODH) and Sasinhwan(SSH) in mouse macrophage, RAW264.7 cells. Methods : To investigate the anti-oxidative effect and scavenging activities of DPPH radical, superoxide anions, nitric oxide and peroxynitrite were measured. Cytotoxic activity of extract of ODH and SSH on RAW264.7 cells was measured using MTS assay. To proof the reductive activity of intracellular oxidation, DCFH-DA assay was performed. The nitric oxide(NO) production was measured and pro-inflammatory cytokines and $PGE_2$ were measured by ELISA kit. The levels of inducible nitric oxide synthase(iNOS), cyclooxygenase-2(COX-2) and nuclear NF-${\kappa}B$ p65 expression were detected by western blot. Results : After those analyses, we bring to a conclusion as follows. Both herbal formulations scavenged DPPH radical and nitric oxide. But ODH had no scavenging activity of superoxide anions and SSH had low scavenging activity in peroxynitrite. And the results indicated that ODH and SSH inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of IL-$1{\beta}$ and IL-6 production in RAW264.7 cells. They also have suppression effects of LPS-induced NF-${\kappa}B$ activation. Conclusions : ODH and SSH have anti-oxidative and anti-inflammatory effects and they may be a part of database for development of new anti-oxidative and anti-inflammatory drugs.

Anti-oxidative Effect of Blueberry Duke Extract in Caenorhabditis elegans (블루베리 듀크 추출물의 예쁜꼬마선충 내의 항산화 효과)

  • We investigated the anti-oxidative effect of the blueberry duke (Vaccinium corymbosum L., Ericaceae) ethanol extract in Caenorhabditis elegans model. The ethanol extract of blueberry duke showed relatively significant DPPH radical scavenging and superoxide quenching activities. To prove antioxidant activity of the extract, we checked the activities of superoxide dismutase (SOD), catalase, intracellular ROS, and oxidative stress tolerance in C. elegans. In addition, to verify if the increased stress tolerance of C. elegans by treating with the extract was due to regulation of stress-response genes, we checked SOD-3 expression using a transgenic strain. As a consequence, the blueberry duke ethanol extract increased SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Besides, blueberry duke ethanol extract-treated CF1553 worms showed higher SOD-3::GFP intensity.

Anti-inflammatory Effects, Analgesic Effects, Antipyretic Effects, Anti-oxidative Effects, and Histamine Permeation Inhibition Effects of Modified Gamgil-tang per Extraction Method (추출 방법에 따른 감길탕가미방(甘桔湯加味方)의 항염증(抗炎症), 진통(鎭痛), 해열(解熱), 항산화(抗酸化) 및 Histamine 투과 억제효과)

  • Objectives : Modified Gamgil-tang is a prescription commonly used for respiratory diseases. This thesis was carried out to check the treatment effects and diversity of drug formulation by comparing extraction method of ethanol and water of modified Gamgil-tang. Methods : All experiments were carried out with water and 50% ethanol extraction for comparison. In vivo experiment, hyaluronidase inhibitory effects and trypsin inhibitory effects were tested to measure the anti-inflammatory effects activity. Scavenging effects of DPPH free radical, xanthine oxidase inhibitory effects and inhibition on TBA-RS formation were experimented to measure anti-oxidative effects. With the in vivo experiment, ICR group mice and SD group rats were used as experimental animals. An anti-inflammatory effects experiment were carried out to measure the action on carrageenin-induced hind paw edema: analgesic effects were measured using writhing syndrome induced by 0.7% acetic acid in mice: antipyretic effect was measured using endotoxin, and inhibitory effects of increase vascular permeability induced by 0.5% histamine were measured. Results : For extraction of glycyrrhizin contents, ethanol extract was extracted 2 times of that of water extract. Anti-inflammatory effects showed high in ethanol extract. Anti-oxidative effects measured high in ethanol extract. No significant result was found in inhibition on TBA-RS formation. Analgesic effects were found to be similar in water and ethanol extract. Antipyretic effects were found to be stronger in water extract. Inhibitory effects of increase vascular permeability induced by 0.5% histamine showed stronger in ethanol extract. Conclusion : By measuring anti-inflammatory effects, analgesic effects, antipyretic effects, anti-oxidative effects, and histamine permeation inhibition effects both in water extract and ethanol extract after adding agents such as Mentha Herba, Gardenias Fructus, and propolis to existing Gamgil-Tang, ethanol extract was found to be more effective in anti-inflammatory effects, analgesic effects, anti-oxidative effects, and histamine permeation inhibition effects. The converse was found for antipyretic effect.

Anti-oxidative Effects of Dendrobii Herba on Toxic Agent Induced Kidney Cell Injury (석곡(石斛)의 항산화 효과)

  • Objectives : This study was carried out to determine if Dendrobii Herba have protective effect against cell injury induced by various toxic agents in rat kidney slices. Water(DWe) and methanol(DMe) extracts were prepared for this experiment. Methods : Cell injury was estimated by measuring lactate dehydrogenase(LDH). Lipid peroxidation was examined by measuring malondialdehyde, a product of lipid peroxidation. Results : DMe prevented the LDH release by $CCl_4$, menadione, tert-butyl hydroperoxide and mercury treatment in vitro in kidney slices, but DWe prevented the LDH release by $CCl_4$ and mercury. DMe also prevented reduction in GSH and lipid peroxidation induced by $CCl_4$ and mercury. Conclusion : Thus, DMe may have more powerful efficacy on anti-oxidative effects when compared with DWe. And further studies have to be followed concerned with extraction of Dendrobii Herba and its change of effects.

Anti-Oxidative Effects of Scutellariae Radix (황금(黃芩)의 항산화 효과)

  • Objectives : This study was carried out to determine if Scutellariae Radix have protective effect against cell injury induced by various toxic agents in rat kidney slices. Anti-Oxidative Effect of Jeokbaekhaogwanjoong-tang Distillate on Spleen Cells of Aged Rats (적백하오관중탕 증류액이 노화 흰쥐 비장세포의 항산화능에 미치는 영향)

  • Objective : The purpose of this study is to investigate the anti-oxidative effect of Jeokbaekhaogwanjoong-tang (JGT) distillate in spleen cells of aged rats.Methods : This experiment was performed using spleen cells of 10w, 52w, 72w old sprague dawley (SD) rats. The spleen cells of 10, 52, 72 weeks old rats were divided into three groups; DW group, Vit. C group, and JGT group. The cells of DW group were treated with distilled water. The cells of Vit.C group were treated with Vitamin C. And the cells of JGT group were treated with JGT distillate. After the treatment of DW, Vit.C, and JGT, the levels of superoxide dismutase (SOD), glutathione (GSH), nitric oxide (NO) and lipid peroxidation (MDA) were measured.Results : In the spleen cells of 72 weeks old rats, SOD activity was significantly increased in the JGT group compared to those in the DW group and the Vit.C group. In the spleen cells of 52 weeks old rats, GSH activity was significantly increased in the JGT group compared to the Vit.C group, and NO concentration was significantly decreased in the JGT group compared to the DW group and the Vit.C group. MDA concentration was not changed significantly in any cell groups by the treatment of JGT distillate.Conclusions : Jeokbaekhaogwanjoong-tang (JGT) distillate may have an anti-oxidative effects in the spleen cells of aged rats. Further investigation is needed for the anti-aging effect of JGT.

The Anti-oxidative and Anti-inflammatory Effect of Lonicera Japonica on Ulcerative Colitis Induced by Dextran Sulfate Sodium in Mice (금은화 추출물의 항산화, 항염증 효과가 Dextran Sulfate Sodium으로 유도된 생쥐의 궤양성 대장염에 미치는 영향)

  • Objectives : This study was to investigate the anti-oxidative and anti-inflammatory effect of Lonicera japonica water extracts (LE) on Ulcerative Colitis Induced by DSS (Dextran Sulfate Sodium) in Mice. Methods : Colitis was induced by DSS in Balb/c mice. The sample group was divided into three. The mice in control group were not inflammation-induced. The pathological group was composed of untreated colitis elicited mice. The experimental group was administered Lonicera japonica water extracts (LE) after colitis elicitation. The effects on ulcerative colitis were evaluated the anti-oxidant effect, inhibition of COX-2 mRNA expression, the morphological change of colonic mucosa, decrease effect of HSP 70 and COX-2 in mucosa. Results : The SOD ability of LE was dose-dependently increased and the LPS-induced COX-2 mRNA expression of LE was dose-dependently decreased. LE showed the protective effects on DSS-induced experimental colitis. LE inhibited shortening of colon length, the hemorrhagic erosion in colonic mucosa. LE also showed the decrease effect for HSP70 and COX-2 in mucosa. Conclusions : The current results demonstrate the clinical utility of LE in traditional medicine and indicate the possible treatments for ulcerative colitis from natural products. Further investigations for exact mechanisms will be needed.

Salt-water Processing-dependent Change in Anti-oxidative and Anti-inflammatory Effects of Cortex Eucommiae (염수초 포제법에 따른 두충의 항산화 및 항염증 활성 변화 비교연구)

  • Objectives The present study aimed to investigate the change in marker compounds, anti-oxidative and anti-inflammatory effects of salt-water processed Cortex Eucommiae. Methods To evaluate the influence of processing on anti-oxidant effect of Cortex Eucommiae, changes in total phenol, total flavonoid, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between processed and raw Cortex Eucommiae were assessed. In addition, nitrite assay was conducted to determine the influence of processing on anti-inflammatory effect of Cortex Eucommiae. Cell viability was also examined as to elucidate whether processing affects cytotoxicity of Cortex Eucommiae. Finally, high-performance liquid chromatography (HPLC) analysis was conducted to monitor changes in pinoresinol diglucoside amount of processed and raw Cortex Eucommiae. Results Salt-water processed Cortex Eucommiae showed higher total phenol and flavonoid amount, compared to raw Cortex Eucommiae. Furthermore, anti-oxidative activity of processed Cortex Eucommiae was improved as discovered in DPPH, ABTS, and FRAP assays. Anti-inflammatory effect of Cortex Eucommiae was also enhanced following salt-water processing, as evidenced in nitrite assay. HPLC analysis found that the amount of pinoresinol diglucoside, widely known as the marker compound of Cortex Eucommiae, increases through salt-water processing. All experiments were performed with non-toxic concentration of Cortex Eucommiae; processing did not affect the cytotoxicity of Cortex Eucommiae up to the currently adopted concentration. Conclusions The present results support that salt-water processing of Cortex Eucommiae is beneficial in terms of marker compound amount, anti-oxidative, and anti-inflammatory activities. Additional investigations are needed to standardize the processing method of Cortex Eucommiae.

Anti-aging and Anti-Oxidative Effect of SYG (SipYiMiGwanJungTang) in aged Rats (십이미관중탕(十二味寬中湯)의 노화(老化) 억제 효과에 대한 실험적(實驗的) 연구(硏究))

  • The purpose of this study is to investigate the anti-aging and anti-oxidative effects of oral administration with SYG (SipYiMiGwanJungTang) decoction in early aged rats. The results were as follows : 1. At the 52 week of the SYG group, the body weight gain was decreased. 2. At the 52 week of the SYG group, the activity of the SOD in the liver was significantly increased than the 10, 22 and 40 week. 3. At the 52 week of the SYG group, the level of the glutathione in the liver was significantly increased than the normal group. 4. At the 68 week of the SYG group, the activity of the catalase in the liver was significantly increased than the normal and control group. 5. At the 52 week of the normal group, the level of the NO in the liver was significantly increased than the 22 and 40 weeks. But, the level of the NO of the SYG group was not changed by the aging. 6. At the 52 and 68 weeks of the SYG group, the level of the MDA in the liver was significantly decreased than the normal and control groups. These results suggest that oral administration of SYG (SipYiMiGwanJungTang) decoction has anti-oxidative and anti-aging effects in early aged rats.

The Anti-oxidative and Immune-regulatory Effect of Chungsimyeonja-tang in Aged Rat (태음인(太陰人) 청심연자탕(淸心蓮子湯)이 SD rat 의 항산화(抗酸化) 및 면역조절효과(免疫調節效果)에 대한 실험적 연구)

  • 1. Objectives The purpose of this study is to investigate the anti-oxidative and immune regulatory effect of oral administration of Chungsimyeonja-tang decoction in aged rats. 2. Methods The rats in the normal group were 14 weeks old, and those in the control and Chungsimyeonja-tang groups were 40 weeks old. The rats in Chungsimyeonja-tang group were orally treated with chungsimyeonjatang decoction daily for 2 months. To observe the anti-oxidative effect and immune regulatory effect of Chungsimyeonja-tang, we performed blood chemistry analysis, histological analysis. 3. Results and Conclusions 1) The number of WBC in blood was significantly increased and the number of PLT in blood was significantly decreased by oral administration of Chungsimyeonja-tang decoction. 2) The levels of albumin, LDL cholesterol, ALP and GOT in serum were significantly decreased, and the level of glucose in serum was significantly increased by oral administration of Chungsimyeonja-tang decoction. 3) The concentrations of glutathione and catalase were significantly increased and the concentrations of NO and MDA were significantly decreased in the liver of early aged rats by oral administration of Chungsimyeonja-tang decoction. 4) The concentrations of SOD and catalase were significantly increased and the concentration of MDA was significantly decreased in the lung of aged rats by oral administration of Chungsimyeonja-tang decoction. 5) The levels of IL-4 and IL-6 were significantly decreased while the levels of IFN-${\gamma}$ were significantly increased in the spleen of early aged rats by oral administration of Chungsimyeonja-tang decoction.
