Anti-Oxidative and Anti-Inflammatory Effects of Cheongajihwang-Tang Extract on RAW264.7 Cells (청아지황탕(靑蛾地黃湯)의 RAW264.7 Cell에서 항산화 및 항염증 효능 연구)

  • Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects.

Effects of Anemarrhenae Rhizoma on anti-oxidative activities (서로 다른 품종 3종 지모(知母)의 항산화 효과)

  • Objective : This study was carried out to investigate diagnostic difference and anti-oxidative effect of 3 different Anemarrhenae Rhizoma (AR) extracts. Methods : Microscopic examination was used to distinguish histological differences. And eliminative ability of several kinds of free radicals was also measured. Results : In microscopic examination, we can distinguished three different lineage of AR with different structure of vascular bundles and secretory structures. The extract of AR show profitable abilities of elimination of DPPH free radical, ABTS free radical and hydrogen peroxide. Conclusion : So, it can be concluded that AR extract has an anti-oxidative effects. Especially indeciduous lineage was most effective to remove free radicals.

Anti-oxidative effects of Mori Folium, Mori Ramulus and Mori Cortex Radidus (상엽(桑葉), 상지(桑枝) 및 상백피(桑白皮)의 항산화 활성 연구)

  • Objective : This study was designed to investigate the anti-oxidative potentials to wide the usage of Morus alba L. derived from medicinal herbs on cosmoceutical fields. Methods : Anti-oxidative potentials were investigated by using several kinds of assays including electron donating ability, and the total contents of phenolic compounds were also measured. In some cases TLC (Thin Layer Chromatography) method were used to identify the patterns of phenolics in herbal extracts. Results : Aurones which are sub-types of flavonoids were observed in methanol extract of Mori Cortex Radidus, and total phenolic contents in Mori Cortex Radidus were more than in methanol extracts of Mori Folium and Mori Ramulus. Various kinds of phenolic compounds were observed in chloroform fraction of Mori Cortex Radidus. Conclusion : According to these results, the chloroform fraction of Mori Cortex Radidus could be the most possible resource that has potentials on anti-oxidant agents on cosmoceutical fields.

Optimal Steaming Condition of Gastrodia elate Blume (Chunma) using Response Surface Methodology (RSM) (반응표면분석법을 이용한 천마의 최적증자조건 설정)

  • The Chunma requires a simple pre-treatment, due to the inconveniences of preservation, toxicities, and pungencies in intact state. Various steaming conditions were examined to establish the anti-oxidative characteristics of Chunma using the Response Surface Methodology (RSM). Total phenolics and electron-donating ability were optimal at 94.43 and $101.67^{\circ}C$ for 42.38 and 41.09 min, respectively. Anti-oxidative (Al) and nitrite-scavenging ability were optimal at $100^{\circ}C$ for 40 min. Optimal steaming conditions were determined as 96{\sim}107{\circ}C$ for $28{\sim}46min$ min, almost identical to the predicted conditions of $100^{\circ}C$, 40 min determined using the superimposed contour map. Anti-oxidative activities (AI) under the optimal conditions were $95.9%{\sim}115.6%$ of those determined under predicted conditions, an indication of the high reliability of the calculated values.

Effects of Dietary Alpha-lipoic Acid on Anti-oxidative Ability and Meat Quality in Arbor Acres Broilers

  • An experiment was conducted to evaluate the effects of dietary alpha-lipoic acid (LA) on growth performance, carcass characteristics and meat quality in Arbor Acres broilers. A total of 240 1-d-old male Arbor Acres broilers were randomly allocated to 4 dietary treatments (0, 300 ppm, 600 ppm, and 900 ppm dietary LA supplementation, respectively). Birds were slaughtered at 42 days old. Live body weight (BW), average daily gain (ADG), average feed intake (AFI), feed conversion ratio (FCR), dressing percentage, breast muscle percentage, thigh muscle percentage, abdominal fat percentage, muscle color (L*, a*, b*), pH values at 24 h postmortem, meat shear force value (SFV) and anti-oxidative ability were measured. Results showed that addition of 600 ppm or 900 ppm LA decreased BW (p<0.01), ADG (p<0.01) and AFI (p<0.05) compared with other diets. FCR was not affected by dietary LA content. LA had no marked effect on dressing percentage, breast muscle percentage or thigh muscle percentage. Abdominal fat percentage was lower (p<0.05) in the 900 ppm LA supplementation group than the control group. Dietary 900 ppm LA increased (p<0.05) breast and thigh muscle pH value at 24 h postmortem compared with the control treatment. Dietary LA increased thigh muscle a* value, though no significant difference was found in thigh muscle a* value among the treatments. Dietary LA significantly decreased breast muscle L* value (p<0.05), breast muscle b* value (p<0.01) and thigh muscle b* value (p<0.05). Broilers fed LA had higher breast muscle a* value (p<0.05) and thigh muscle L* value (p<0.05). All test groups had lower (p<0.05) breast muscle SFV than the control group. Dietary 600 ppm or 900 ppm LA both decreased (p<0.01) thigh muscle SFV compared with the control treatment. Dietary 900 ppm LA significantly increased (p<0.05) TAOC, SOD and GSHPx compared with no LA treatment. Broilers fed LA had lower (p<0.01) MDA compared with the control treatment. These results suggested that dietary LA enhanced the anti-oxidative ability and oxidative stability, and contributed to the improvement of meat quality in broilers.

Comparison of Anti-Oxidative and Cox-2 Promoter Activities of Lepidoptera Extracts (Lepidoptera 추출물에 의한 항산화 및 Cox-2 프로모터 활성 비교)

  • Lepidoptera (butterflies) extracts, traditionally employed as medicines, have various biological activities. Five species of Lepidoptera (Papilio maackii, Papilio xuthus, Pieris rapae, Eurema hecabe, and Sasakia charonda) were extracted with distilled water (DW), dimethyl sulfoxide (DMSO), ethanol (EtOH), and methanol (MeOH). Each extract was analyzed for anti-oxidant and anti-inflammatory activities using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay method, the ferric reducing ability of plasma (FRAP) test, and a cyclooxygenase-2 (COX-2) promoter assay. The results suggest that Lepidoptera extracts have valuable anti-oxidant and anti-inflammatory properties, supporting the idea that the extracts may serve as a food biomaterial(s) preventing oxidative processes and inflammatory damage.

  • Four isoflavonoids and three flavonoids, and a gallotannin were isolated from the fruits of Sophora japonica (Leguminosae). Their structures were identified as genistein (1), sophoricoside (2), genistein-4'-O--L-rhamnopyranoside (3), $genistein-4'-O-{\alpha}-L-rhamnopyranosly-(1{\rightarrow}2)-{\beta}-D-glucopyranoside$ (4), $kaempferol-3-O-{\alpha}-D-sophoroside$ (5), $kaempferol-3-O-{\beta}-D-glucopyranosyl-(1{\rightarrow}2)-{\alpha}-L-rhamnopyranosly-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (6), rutin (7) and gallic acid $4-O-{\beta}-D-(6'-O-galloyl)-glucopyranoside$ (8) by chemical and spectroscopic analysis and comparisons with previously reported spectral data. Compounds 3 and 8 were isolated for the first time from this plant. Anti-oxidative activity was evaluated for the isolated compounds. 8 exhibited potent anti-oxidative activity against the radical scavenging ability of DPPH with the $IC_{50}$ value of $17.1\;{\mu}g/ml$.

The Anti-oxidative and Anti-inflammatory Effect of Lonicera Japonica on Ulcerative Colitis Induced by Dextran Sulfate Sodium in Mice (금은화 추출물의 항산화, 항염증 효과가 Dextran Sulfate Sodium으로 유도된 생쥐의 궤양성 대장염에 미치는 영향)

  • Objectives : This study was to investigate the anti-oxidative and anti-inflammatory effect of Lonicera japonica water extracts (LE) on Ulcerative Colitis Induced by DSS (Dextran Sulfate Sodium) in Mice. Methods : Colitis was induced by DSS in Balb/c mice. The sample group was divided into three. The mice in control group were not inflammation-induced. The pathological group was composed of untreated colitis elicited mice. The experimental group was administered Lonicera japonica water extracts (LE) after colitis elicitation. The effects on ulcerative colitis were evaluated the anti-oxidant effect, inhibition of COX-2 mRNA expression, the morphological change of colonic mucosa, decrease effect of HSP 70 and COX-2 in mucosa. Results : The SOD ability of LE was dose-dependently increased and the LPS-induced COX-2 mRNA expression of LE was dose-dependently decreased. LE showed the protective effects on DSS-induced experimental colitis. LE inhibited shortening of colon length, the hemorrhagic erosion in colonic mucosa. LE also showed the decrease effect for HSP70 and COX-2 in mucosa. Conclusions : The current results demonstrate the clinical utility of LE in traditional medicine and indicate the possible treatments for ulcerative colitis from natural products. Further investigations for exact mechanisms will be needed.

Quality characteristics and preparation of Dasik using roasted mung bean (로스팅 녹두를 이용한 다식 제조 및 품질특성)

  • In this study, we manufactured mung bean Dasik (pattern pressed candy) after selecting the optimum roasting conditions through physiochemical analysis and sensory evaluation. Then anti-oxidative abilities of roasted mung bean were measured in order to develop beauty food (Dasik) using roasted mung bean. In the content of vitexin and isovitexin of roasted mung bean, about 10 times the vitexin in the seed(60.85 mg/g) is found in the skin of raw mung bean, and about 9 times the isovitexin in the seed(71.42 mg/g) is also found in the peel. As a result of analyzing the seed and peel of mung bean after roasting it for 10, 20 and 30 minutes respectively, the optimum roasting condition is thought to be $110^{\circ}C$ for 30 minutes as the contents of vitexin and isovitexin showed the highest values of 104.94 mg/g and 122.02 mg/g respectively when the mung bean was roasted at $110^{\circ}C$ for 30 minutes. In the anti-oxidative activity evaluation of the optimum mung bean Dasik, the total content of phenol was shown to be 0.15 mg/mL, and the total content of flavonoid was shown to be 0.026 mg/mL. The DPPH radical scavenging ability showed a high vitality of 58.19%, and the ABTS radical scavenging ability was shown to be 13.26%.

Anti-oxidative Effect of Chungpyesagan-tang in LPS Induced RAW 264.7 Cells (LPS로 유도된 RAW 264.7 세포에서 청폐사간탕(淸肺瀉肝湯)의 항산화 효과)

  • Objectives : This study was conducted to confirm the anti-oxidative effect of Chungpyesagan-tang(CPSGT) extract. Methods : In this study, MTT assay was performed to confirm cell viability, and DPPH and ABTS were performed to confirm radical scavenging ability. The ROS scavenging ability and the protective effect against DNA damage were confirmed by 2,7-dichlorofluorescin diacetate(DCF-DA) and 4',6-diamidino-2-phenylindole(DAPI) staining and comet assay. mRNA expression of Heme oxygenase-1(HO-1) was measured by real-time PCR, and expression of HO-1 and Kelch-like ECH-associated protein 1(Keap1) proteins was measured by western blot. Results : CPSGT was not cytotoxic at 50-400㎍/㎖. The radical scavenging activity was increased, and the ROS scavenging activity and the protective effect against DNA damage were increased compared to the LPS-treated group. The mRNA expression and protein expression of HO-1 were increased in a concentration-dependent manner. The protein expression level of Keap1 was decreased in a concentration-dependent manner. Conclusion : This suggests that CPSGT has an antioxidant effect and can be used as a potential material for skin diseases.
