본 연구는 COHEE의 항염증 효과를 확인하기 위한 실험으로써 pro-inflammatory cytokine의 분비량 및 iNOS, COX-2, NF-κB와 MAPKs의 발현량을 관찰하였다. 먼저 MTT assay를 통해 COHEE가 세포 생존율에 있어 독성을 나타내지 않음을 확인한 후 동일한 농도로 추후실험을 진행하였다. COHEE에 의해 NO 분비량이 농도 의존적으로 감소하였으며 IL-6, TNF-α 및 IL-1β의 분비량 또한 유의적으로 감소하였다. 특히 100 μg/ml의 농도에서 LPS 단독처리구인 대조구에 비하여 IL-6, TNF-α 및 IL-1β의 분비량을 각각 66%, 43% 및 93% 억제시켰다. 이러한 결과가 전염증성 매개인자의 전사인자인 NF-κB와 MAPKs 경로에 의한 것인지 확인하기 위하여 발현량을 관찰한 결과, COHEE가 LPS 처리에 의해 현저히 증가한 단백질의 발현을 농도 의존적으로 유의성 있게 억제하였다. 또한 COHEE는 croton oil로 부종을 유발한 마우스모델에서 귀부종 억제효과를 나타내었고 250 mg/kg 농도에서 조직의 경피 및 진피 두께의 발달을 prednisolone 10 mg/kg 처리구와 유사한 정도까지 현저히 억제시키고 염증성 세포인 mast cell의 침윤 억제 효과도 확인하였다. 이를 통해 COHEE는 염증 반응의 전사인자인 NF-κB의 발현을 조절함으로써 iNOS와 COX-2의 발현을 억제하고 그에 따라 전염증성 매개인자인 NO, IL-6, TNF-α 및 IL-1β의 분비를 억제하여 항염증 활성을 가지는 것을 확인하였으며, 이 결과를 종합해볼 때, COHEE가 염증 치료제로써의 소재로 이용될 가치가 충분할 것으로 사료된다.
본 연구는 헛개나무 잎, 줄기, 뿌리로부터 얻어진 메탄올 추출물의 항염증 효과를 구명하기 위해서 수행되었다. LPS로 염증이 유도된 RAW264.7 세포 내 부위별 추출물을 동시에 처리하여 염증 매개성 물질인 NO 생성량 분석 결과 $40{\mu}g/m{\ell}$농도에서 뿌리(94.7%)와 줄기(42.6%)에서는 효과가 있는 반면 잎에서는 효과가 없는 것으로 확인되었다. 이 중 탁월한 효과를 보인 뿌리 추출물(RE)의 항염증 효과 및 관련 분자적 기전을 확인하였다. 그 결과, 염증 반응의 주요 경로인 NF-kB 및 MAPK 신호전달경로에서 RE가 LPS로 유도된 NF-kB의 핵 이동을 억제하고, ERK, JNK, p38의 인산화를 억제함으로써 iNOS, COX-2의 발현이 감소되고, NO와 pro-inflammatory cytokine (IL-6, $IL-1{\beta}$, $TNF-{\alpha}$)의 생성이 억제됨을 확인하였다. 또한 RE는 대식세포에서 Nrf2를 활성화시켜 항염증성 단백질인 HO-1 발현을 유도하여 항염증 효과를 나타내었다. 또한, RE로부터 주요 성분을 분리한 후 NMR과 MS 기기를 이용하여 구조 동정된 27-O-protocatechuoylbetulinic acid 화합물에서도 높은 항염증 효과를 확인하였다. 이러한 연구결과는 헛개나무뿌리와 그 주요성분은 의약품 소재 및 기능성 식품 등의 기능성 소재로 활용될 수 있는 기초적인 정보를 제공할 것으로 생각된다.
본 연구에서는 천연식물자원인 참모자반 에탄올 추출물(SFEE)의 항염증 활성을 알아보기 위하여 in vitro 및 in vivo 실험을 진행하였다. 먼저 SFEE의 세포독성을 살펴보기 위하여 RAW 264.7 대식세포를 이용하여 세포 생존율을 살펴본 결과, SFEE $100{\mu}g/mL$의 농도까지 전혀 독성을 나타내지 않았다. 또한 항염증 효과를 알아보기 위하여 in vitro type으로 LPS로 염증을 유도한 RAW 264.7 세포에 대하여 NO, IL-6, $TNF-{\alpha}$ 및 $IL-1{\beta}$ 분비량과 iNOS, COX-2 및 $NF-{\kappa}B$와 같은 염증성 단백질의 발현량을 측정하였다. 대식세포 배양액의 $NO{_2}{^-}$ 농도를 측정한 결과 SFEE 첨가 농도에 따라 유의적으로 분비량이 감소하는 것을 확인하였으며, $100{\mu}g/mL$의 농도에서 90% 이상 억제됨을 확인하였다. 또한 $100{\mu}g/mL$의 농도로 첨가 시 염증성 사이토카인인 IL-6, $TNF-{\alpha}$ 및 $IL-1{\beta}$ 분비량을 각각 79, 73, 87% 억제함을 확인하였으며, NO 생성과 연관이 있는 iNOS 단백질과 COX-2 및 $NF-{\kappa}B$ 단백질의 발현이 현저히 억제됨을 확인하였다. 이러한 SFEE의 항염증 효과를 동물모델에서 확인하기 위해 in vivo 실험을 진행한 결과 croton oil로 유도한 마우스 귀 부종을 유의적으로 억제함을 확인하였고, 조직검사 결과 염증 반응에 의한 경피 및 진피 두께의 발달을 현저히 억제시키고 염증성 세포인 비만세포의 조직으로의 침윤을 억제하여 항염증 효과를 가짐을 입증하였다. 결론적으로 참모자반 에탄올 추출물은 염증 반응의 전사인자인 $NF-{\kappa}B$의 발현을 억제하여 iNOS와 COX-2의 발현을 억제시키고 그에 의해 NO, IL-6, $TNF-{\alpha}$ 및 $IL-1{\beta}$ 분비를 억제하여 항염증 활성을 가지는 것을 확인하였으며, 따라서 참모자반 에탄올 추출물이 염증성 질환의 예방 및 개선에 유용할 것으로 사료된다.

Background: Alcohol is one of the most commonly used psychoactive drugs. Due to its addictive characteristics, many people struggle with the side effects of alcohol. Korean Red Ginseng (KRG) is a traditional herbal medicine that is widely used to treat various health problems. However, the effects and mechanisms of KRG in alcohol-induced responses remain unclear. Therefore, the purpose of this study was to investigate the effects of KRG in alcohol-induced responses. Methods: We investigated two aspects: alcohol-induced addictive responses and spatial working memory impairments. To determine the effects of KRG in alcohol-induced addictive responses, we performed conditioned place preference tests and withdrawal symptom observations. To assess the effects of KRG in alcohol-induced spatial working memory impairment, Y-maze, Barnes maze, and novel object recognition tests were performed using mice after repeated alcohol and KRG exposure. To investigate the potential mechanism of KRG activity, gas chromatography-mass spectrometry and western blot analysis were performed. Results: KRG-treated mice showed dose-dependent restoration of impaired spatial working memory following repeated alcohol exposure. Furthermore, withdrawal symptoms to alcohol were reduced in mice treated with KRG and alcohol. The PKA-CREB signaling pathway was activated after alcohol administration, which was reduced by KRG. However, the levels of inflammatory cytokines were increased by alcohol and decreased by KRG. Conclusion: Taken together, KRG may alleviate alcohol-induced spatial working memory impairments and addictive responses through anti-neuroinflammatory activity rather than through the PKA-CREB signaling pathway.
Eosinophils play critical roles in the maintenance of homeostasis in innate and adaptive immunity. Although primarily known for their roles in parasitic infections and the development of Th2 cell responses, eosinophils also play complex roles in other immune responses ranging from anti-inflammation to defense against viral and bacterial infections. However, the contributions of pattern recognition receptors in general, and NOD-like receptors (NLRs) in particular, to eosinophil involvement in these immune responses remain relatively underappreciated. Our in vivo studies demonstrated that NLRC4 deficient mice had a decreased number of eosinophils and impaired Th2 responses after induction of an allergic airway disease model. Our in vitro data, utilizing human eosinophilic EoL-1 cells, suggested that TLR2 induction markedly induced pro-inflammatory responses and inflammasome forming NLRC4 and NLRP3. Moreover, activation by their specific ligands resulted in caspase-1 cleavage and mature IL-1β secretion. Interestingly, Th2 responses such as secretion of IL-5 and IL-13 decreased after transfection of EoL-1 cells with short interfering RNAs targeting human NLRC4. Specific induction of NLRC4 with PAM3CSK4 and flagellin upregulated the expression of IL-5 receptor and expression of Fc epsilon receptors (FcεR1α, FcεR2). Strikingly, activation of the NLRC4 inflammasome also promoted expression of the costimulatory receptor CD80 as well as expression of immunoregulatory receptors PD-L1 and Siglec-8. Concomitant with NLRC4 upregulation, we found an increase in expression and activation of matrix metalloproteinase (MMP)-9, but not MMP-2. Collectively, our results present new potential roles of NLRC4 in mediating a variety of eosinopilic functions.
활성화된 단핵구의 동형성 세포 부착(동형성 응집)은 염증반응, 분화, 이동과 같은 생리학적, 병리학적 과정에서 중요한 역할을 한다. 매실 추출물은 항바이러스, 항균, 항암작용과 같은 효과를 보인다고 알려 져있다. 따라서, 매실 추출물은 단핵구의 동형성 응집 억제를 통해 염증반응을 조절할 가능성을 가진다. 본 연구에서는, 염증성 질환에서 매실 추출물의 치료효능을 뒷받침할 수 있는 분자적 기전을 조사하였다. 매실 추출물이 지질다당질(LPS)로 활성화된 단핵구의 동형성 응집을 억제함을 확인하였다. 이러한 효과는 LPS로 활성화된 THP-1 세포의 iNOS 단백질 발현 억제를 통해, 산화질소(NO) 생산의 감소로 조절되는 것을 발견하였다. 또한 NO 생성물질인 SNAP 처리 실험을 통해 단핵구 동형성 응집을 억제하는데 매실에 의한 NO 억제가 필수적인 기작임을 확인하였다. 게다가, 매실 추출물은 LPS로 유도된 IκB-α 의 인산화와 NF-κB의 핵내로의 이동을 현저하게 감소시키는 것을 확인하였다. 매실 추출물은 NO생성과 NF-κB 활성 억제를 통해 LPS로 활성화된 단핵구의 동형성 응집을 저해하고 이를 통해 항염증 효과를 유도할 수 있다는 결론으로부터 만성 염증성 질환의 치료와 예방에 매실 추출물의 효능을 제시하고자 한다.
본 연구에서는 참치유의 항염증 효과를 알아보기 위하여 in vitro 및 in vivo type의 실험을 진행하였다. 먼저 참치유(TO)가 세포독성을 가지는지에 대해 알아보기 위해 MTT assay를 진행한 결과, 모든 첨가 농도에서 세포독성을 가지지 않는 것을 확인하였다. 세포독성을 가지지 않은 농도에서의 항염증 효과를 알아보기 위하여 염증성 매개 인자인 NO, IL-6, TNF-α 및 IL-1β에 대한 항염증 효과를 살펴본 결과, TO의 첨가에 따라 각각 45%, 93%, 97%, 83%의 억제효과를 보임을 확인하였다. 또한 그 up-stream의 전사인자인 NF-κB 및 MAPKs와 전염증성 효소인 iNOS, COX-2의 발현을 효과적으로 억제하는 것을 확인하였다. In vivo type 실험의 일환으로 croton oil 유도 마우스 귀 부종에 대한 억제 효과를 살펴본 결과, TO의 처리에 의해 경피 및 진피 두께의 발달과, 염증 부위로의 mast cell 침윤이 억제됨을 확인하였다. TO의 효과적인 이용을 위해 그 안전성에 대한 평가로 급성 경구독성 평가를 진행한 결과, 경구독성을 유발하지 않음을 확인하였다. 본 연구 결과로 TO의 적용이 in vitro 및 in vivo type의 염증 모델에서 우수한 효과를 보임을 확인하여, TO 가 효과적인 염증 예방 및 치료제로의 활용 가능성이 충분함을 입증하였다.
이상의 실험 결과들을 요약하면, 포르말린을 측두하악관절 내로 주입하여 발생한 염증성 통증 행위반응은 QX-314의 투여로 감소할 수 있었다. 저농도의 QX-314의 진통작용은 TRPV1 통로를 이용하여 세포막 내로 이동하여 작용이 나타났으며 고농도의 QX-314는 TRPV1 통로와 무관하게 진통작용을 나타내었다. 이와 같은 결과는 측두하악관절 장애로 인해 발생되는 염증성 통증에 QX-314가 효과적인 치료제로 사용할 수 있다는 것을 말해주며, 특히 고농도의 QX-314가 세포막을 이동하는 경로에 대한 연구가 더 진행된다면 임상에서 QX-314가 진통제로서 사용할 수 있는 계기가 될 것으로 판단된다.
Inflammatory mediators has been recognized as an important role in the pathogenesis of rheumatoid arthritis (RA). IL-17 is increasingly recognized as an important regulator of immune and inflammatory responses, including induction of proinflammatory cytokines and osteoclastic bone resorption. Evidence of the expression and proinflammatory activity of IL-17 has been demonstrated in RA synovium and in animal models of RA. However, the signaling pathways that regulate IL-17 production remain unknown. In the present study, we investigated the role of the phosphatidylinositol 3 kinase (PI3K)-Akt pathway in the regulation of IL-17 production in RA. PBMC were separated from RA (n=24) patients, and stimulated with various agents (anti CD3, anti CD28, PHA, ConA, IL-15). IL-17 levels were determined by sandwich ELISA and RT-PCR. The production of IL-17 was significantly increased in cells treated with anti-CD3 antibody, PHA, IL-15 or MCP-1 (P<0.05). ConA also strongly induced IL-17 production (P<0.001), whereas TNF-alpha, IL-1beta, IL-18 or TGF-beta did not. IL-17 was detected in the PBMC of patients with osteoarthritis (OA) but their expression levels were much lower than those of RA PBMC. Anti-CD3 antibody activated the PI3K-Akt pathway and activation of the PI3K-Akt pathway resulted in a pronounced augmentation of nuclear factor kappaB ($NF-{\kappa}B$). IL-17 production by activated PBMC in RA is completely or partially blocked in the presence of $NF-{\kappa}B$ inhibitor PDTC and PI3K-Akt inhibitor, wortmannin and LY294002, respectively. Whereas the inhibition of AP-1 and extracellular signal-regulated kinase (ERK)1/2 did not affect IL-17 production. These results provide new insight into that PI3K/Akt and $NF-{\kappa}B$ dependent signal transduction pathway could be involved in the overproduction of key inflammatory cytokine, IL-17 in rheumatoid arthritis.
Objective: This experimental study was performed to examine the in vitro and in viva anti-inflammatory and anti-allergic effects of Mori Cortex. Methods: Water extract of Mori Cortex was studied to its ability to stimulate or inhibit macrophage 264.7 cells to produce inflammatory and allergic mediators. Cytokines such as $IL-1{\beta}$, IL-6, IL-10 and $TNF-{\alpha}$ were measured by immunochemical assay. In vitro, the macrophages 264.7 were classified into four groups. One group was a normal group. The other group was a (-) control group stimulated with LPS. And the third group was a (+) control group pretreated for 1 hour with hydrocortisone. And the fourth group was a sample group pretreated for 1 hour with Mori Cortex. After pretreatment, macrophage were incubated with lipopolysaccharide(LPS) $100\;ng/m{\ell}$ for 12 hour and media collected and $IL-1{\beta}$, IL-6, IL-10 and $TNF-{\alpha}$ concentrations in supernatants were measured each by Enzyme linked immuno-soubent assay. Mori Cortex were used $50\;{\mu}g/m{\ell},\;100\;{\mu}g/m{\ell},\;250\;{\mu}g/m{\ell},\;500\;{\mu}g/m{\ell},\;and\;1,000\;{\mu}g/m{\ell}$. Hydrocortisones were used $10^{-8}M,\;10^{-7}M,\;10^{-6}M,\;10^{-5}M\;and\;10^{-4}M$. In vivo, the SD rats were classified into three groups. One group was a normal group injected with normal saline into the abdominal cavity. The other was a control group prescribed to compound 48/80 after normal saline injection. And the third was a sample group prescribed to compound 40/80 after Mori Cortex injection. Then, the release of histamine, IL-6 and $TNF-{\alpha}$ were measured. Results : In vitro, Man Cortex significantly increased the release of $IL-1{\beta}\;and\;TNF-{\alpha}$ by LPS-stimulated macrophage 264.7 cells. And it significantly decreased the release of IL-10. In IL-6, Mori Cortex of low concentration significantly decreased the release of IL-6, but that of high concentration acted in reverse. In vivo, Man Cortex didn't show significant inhibitory effects on the release of histamine and IL-6 in comparison with that of the control group. But it significantly increased the release of $TNF-{\alpha}$ in comparison with that of the control group.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
