The Anti-Wrinkle Mechanism of Ganoderma lucidum mycelial with Acorus gramineus callus in UVB Treated HaCaT Keratinocytes

  • Skin is continuously exposed to a variety of environmental stresses, including ultraviolet (UV) radiation. UVB is an inherent component of sunlight that crosses the epidermis and reaches the upper dermis, leading to increased oxidative stress, activation of inflammatory response and accumulation of DNA damage among other effects. In the present study, the anti-wrinkle mechanism of Acorus gramineus callus culture supernatant (GB-AGS-PSC) was elucidated in UVB treated HaCaT keratinocytes. GB-AGS-PSC prevented the matrix metalloprotease 1 (MMP-1), elastin, and pro-collagen product and cytotoxicity and SOD inhibition. Quantitative polymerase chain reaction showed that GB-AGS-PSC-treated cells displayed dose-dependent increase in messenger RNA expression levels of Aquaporin 3 (AQP3), Keratin 1(KRT1), fillagrin, and hyaluronan synthase-2 (HAS 2) and decreased expression levels of matrix metalloproteinase-3, -9, and -13 in UVB treated HaCaT keratinocytes. Additionally, GB-AGS-PSC suppressed TNF-α, IL-1β, and IL-8 product for inflammatory responses in UVB treated HaCaT keratinocytes. Therefore, GB-AGS-PSC may be useful as an anti-photoaging resource for the skin.

  • Background: The flowers of Inula japonica (Inulae Flos) have long been used in traditional medicine for the treatment of inflammatory diseases. In the present study, we investigated the anti-inflammatory properties of Inulae Flos Extract (IFE). Methods: The anti-inflammatory effects of IFE against nitric oxide (NO), $PGE_2$, TNF-${\alpha}$, and IL-6 release, as well as NF-${\kappa}B$ and MAP kinase activation were evaluated in RAW 264.7 cells. Results: IFE inhibited the production of NO and the expression of inducible nitric oxide synthase (iNOS) in LPS-stimulated RAW264.7 cells. In addition, IFE reduced the release of pro-inflammatory cytokines, such as TNF-${\alpha}$ and IL-6. Furthermore, IFE inhibited the NF-${\kappa}B$ activation induced by LPS, which was associated with the abrogation of $I{\kappa}B-{\alpha}$ degradation and subsequent decreases in nuclear p65 and p50 levels. Moreover, the phosphorylation of ERK, JNK, and p38 MAP kinases in LPS-stimulated RAW 264.7 cells was suppressed by IFE in a dose-dependent manner. Conclusion: These results suggest that the anti-inflammation activities of IFE might be attributed to the inhibition of NO, iNOS and cytokine expression through the down-regulation of NF-${\kappa}B$ activation via suppression of $I{\kappa}B{\alpha}$ and MAP kinase phosphorylation in macrophages.

Anti-inflammatory Effects of Gastrodia elata Extract in Lipopolysaccharide-stimulated BV-2 Microglia Cell

  • Gastrodia elata Blume is used for a variety of purposes including treatment of inflammation in the Korean medicine. The present study investigated whether the G. elata extracts have the anti-inflammatory effect on lipopolysaccharide(LPS)-stimulated BV-2 microglia cells. G. elata extracts showed an anti-inflammatory effects in the morphological and nitric oxide(NO) analysis, especially in hexane extract. So we investigated the hexane extract from G. elata in the following experiments. Hexane extract significantly inhibited the secretion of NO with protein level of inducible nitric oxide synthase in LPS-stimulated BV2 microglia cells. Hexane extract also inhibited LPS-stimulated inflammatory responses involving the degradation of cytosolic inhibitory(I)-${\kappa}B{\alpha}$ and the translocation of nuclear factor(NF)-${\kappa}Bp65$ to nucleus in LPS-stimulated BV-2 microglia cells by morphological analysis. Western blot analyse confirmed that I-${\kappa}B{\alpha}$ and NF-${\kappa}Bp65$ showed a similar pattern as morphological analysis. Our results suggest that G. elata extracts, especially hexane extract, may act as a therapeutic agent for inflammatory disease in the central nervous system through a selective regulation of NO production and NF-${\kappa}B$ activation.

Lipopolysaccharide로 유도된 Raw 264.7 cell에서 물레나물(Hypericum asctron)의 Pro-inflammatory 억제 효과 (Inhibitory effect of Hypericum ascyron on pro-inflammatory responses in lipopolysaccharide-induced Raw 264.7 Cells)

  • 물레나물(Hypericum ascyron)은 예로부터 식 약용으로 사용되어져 왔으나, 물레나물의 항염증 효과와 mechanism에 대한 연구가 매우 부족하여 물레나물 추출물의 염증 생성 반응에 관여하는 기전을 규명하고자 하였다. 물레나물은 열수와 90% ethanol 추출물에서 각각 29.75, 31.82 mg/g으로 높은 phenolic 함량을 나타내었다. 물레나물 추출물의 hyaluronidase 저해 활성을 측정한 결과 $50-200{\mu}g\;phenolics/mL$ 농도에서 열수와 90% ethanol 추출물이 각각 0.00-14.81, 15.33-47.49%의 저해 활성을 나타내어 항염증 효과가 있는 것으로 판단되었다. 물레나물 추출물의 세포 독성을 측정한 결과, 열수와 90% ethanol 추출물에서 각각 $30-100{\mu}g/mL$ 농도에서 독성이 관찰되어 열수 추출물의 농도 구간을 $10-50{\mu}g/mL$, ethanol 추출물은 $5-20{\mu}g/mL$으로 선정하였다. LPS로 자극한 Raw 264.7 cell에서 iNOS, COX-2와 같은 염증성 매개체뿐만 아니라 pro-inflammatory cytokine의 생성과 발현은 물레나물의 열수와 90% ethanol 추출물에 의하여 농도 의존적으로 억제되는 것을 확인하였다. 물레나물 추출물의 염증 억제 효과는 iNOS와 COX-2를 억제함으로써 염증 반응에 관련된 물질인 NO, $PGE_2$, $TNF-{\alpha}$, IL-6, $IL-1{\beta}$ 생성을 억제하는 기작을 가지는 것으로 판단되었다. 따라서 물레나물 추출물은 다양한 염증성 질환에서 매개 물질들의 과발현에 의해 야기되는 질병을 치료하기 위한 치료제로 다양하게 활용할 수 있는 천연물 소재로 사용될 수 있을 것으로 판단되며, 물레나물의 기능성 식품 산업화를 위한 유용한 기초 자료가 될 수 있을 것으로 판단되었다.

당지질로 유도한 염증반응에서 Piceatannol의 항염증 기전 연구 (Mechanism Underlying the Anti-Inflammatory Action of Piceatannol Induced by Lipopolysaccharide)

  • 본 연구에서는 염증반응을 조절하는 다양한 신호전달체계를 중심으로 분자생물학적 방법을 통해 piceatannol의 항염증 기전을 규명하였다. LPS로 염증반응을 유도한 Raw 264.7 대식세포에서 piceatannol은 iNOS의 발현 억제를 통해 NO의 생성을 감소시키고 염증성 사이토카인(TNF-${\alpha}$, IL-6, IL-$1{\beta}$)의 생성을 감소시켰다. 염증반응을 조절하는 신호전달체계 중 piceatannol은 LPS에 의해 유도된 $I{\kappa}B$의 분해와 p65의 핵으로의 이동을 억제하고, LPS에 의해 유도된 SAPK/JNK의 인산화를 억제하였다. 또한 piceatannol은 LPS와 IL-6(LPS에 의해 증가됨)에 의한 STAT3의 활성화를 억제하였다. 뿐만 아니라 piceatannol은 Nrf2의 핵 내 축적을 야기하고 ARE의 transcriptional activity를 증가시켜 HO-1의 발현을 증가시켰다. 본 연구의 결과, piceatannol은 NF-${\kappa}B$와 AP-1, STAT3 신호전달의 억제를 통해, 그리고 HO-1의 발현 증가를 통해 항염증 효과를 나타내었다(Fig. 8).

  • Scopoletin은 Erycibe obtusifolia, Aster tataricus, Foeniculum vulgare 그리고 Brunfelsia grandiflora라는 식물들에서 발견된 성분이다. 이것은 신 혈관형성 억제와 항알러지 효과를 가지고 있다고 알려져 있다. 본 연구에서는 대식세포인 Raw264.7 cells 에서 scopoletin의 항염증 효과에 대하여 조사되었다. 먼저 Scopoletin이 LPS로 염증이 유도된 Raw264.7 cells에서 nitric oxide (NO) 생성과 식균작용(phagocytosis)에 대하여 어떠한 효과가 있는지 평가하였다. Scopoletin은 NO생성과 식균작용을 감소시키는 것으로 나타났다. 뿐만 아니라 NO와 prostaglandin E2 (PGE2)의 생성과 관련된 inducible nitric oxide synthase (iNOS)와 COX-2의 단백질 발현을 감소시키는 것으로 나타났다. 특히 염증성 cytokine인 interleukin-1β (IL-1β), interleukin-6 (IL-6) 그리고 TNF-α의 발현을 조사 하였는데, IL-1β and IL-6의 발현은 scopoletin의 처리에 의해 농도 의존적으로 감소하였다. tumor necrosis factor-α (TNF-α)는 scopoletin을 8 hr 처리하였을 때 감소하는 것으로 나타났다. 이러한 결과는 scopoletin이 iNOS, COX-2, IL-1β, IL-6, TNF-α 또한 phagocytosis 그리고 NO생성을 감소시켜 항염증 효능을 나타내어, 향후에 항염증제로서 활용의 잠재성이 있다고 판단된다.

RBL-2H3세포에서 생지황약침액의 FcεRI 신호전달을 통한 β-hexosaminidase분비와 Cytokine생성 억제 효과 (Inhibitory Effect of Rehmannia Glutinosa Pharmacopuncture Solution on β-hexosaminidase Release and Cytokine Production via FcεRI signaling in RBL-2H3 Cells)

  • Background: Type I allergy is involved in allergic asthma, allergic rhinitis, and atopic dermatitis which are accompanied by an acute and chronic allergic inflammatory responses. Rehmannia glutinosa is a traditional medicine in the East Asian region. This study examined whether a Rehmannia Glutinosa pharmacopuncture solution (RGPS) had anti-allergic or anti-inflammatory effects in antigen-stimulated-RBL-2H3 cells. Methods: We determined the effect of RGPS on cell viability using the 3-[4,5-dimethylthiazolyl]-2,5-diphenyltetrazolium bromide (MTT) assay. We also examined the effect of RGPS on the release of ${\beta}$-hexosaminidase and the secretion of IL-4 and TNF-${\alpha}$ using ELISA. In addition, we evaluated the effect of RGPS on the mRNA expression of various cytokines; IL-2, IL-3, IL-4, IL-5, IL-13 and TNF-${\alpha}$ using RT-PCR. Furthermore, we assessed the activation of mitogen-activated protein kinases (MAPKs) and nuclear factor (NF)-${\kappa}$B using Western blotting after RGPS treatment. Results: We found that RGPS ($10^{-4}$ to $10^{-1}$ dilution) did not cause any cytotoxicity. We observed significant inhibition of ${\beta}$-hexosaminidase release and suppression of the protein secretion of IL-4 and TNF-${\alpha}$ and mRNA expression of multiple cytokines in antigen-stimulated-RBL-2H3 cells after RGPS treatment. Additionally, RGPS suppressed not only the phosphorylation of MAPKs, but also the transcriptional activation of NF-${\kappa}$B in antigen-stimulated-RBL-2H3 cells. Conclusions: These results suggest that RGPS inhibits degranulation and expression of cytokines including IL-4 and TNF-${\alpha}$ via down-regulation of MAPKs and NF-${\kappa}$B activation in antigen-stimulated-RBL-2H3 cells. In conclusion, RGPS may have beneficial effects in the exerting anti-allergic or anti-inflammatory activities.

Human Skin Safety Test of Green Tea Cell Extracts in Condition of Allergic Contact Dermatitis

  • Various kinds of positive effects of green tea extracts had been studied for long time which included anti-inflammation, anti-aging, and cardiometabolic effects. Although topical steroid and non-steroidal calcineurin inhibitors may control clinical symptoms of allergic contact dermatitis, some of patients also present allergic reaction to these topical agents. Therefore, we have tried green tea extracts for managing this skin disorder with expectation of anti-inflammatory effect without potential side effects including skin irritation and toxic responses. The toxicity test of green tea extract also did not show any sign of irritation in the skin throughout the test period. Moderate severity of allergic contact dermatitis presented satisfactory clinical outcome at second week follow-up which was final visit of outpatient. This result mean that green tea extract has a positive effect for managing allergic contact dermatitis but its potency and efficacy seem to be so not strong enough to control moderate severity allergy skin lesion. In this pilot study, we were able to conclude that green tea cell extracts might be applied for potential anti-inflammatory soaking without skin toxicity.

Effects of Daegangwhal-Tang Hot Aqueous Extract on Anti-inflammation and Anti-oxidation in RAW 264.7 Macrophage

  • Background: The objective of this study was to determine the effects of Daegangwhal-Tang (DGHT) hot aqueous extract on production of inflammatory mediators and antioxidants in RAW 264.7 macrophage. Methods: DGHT was extracted with water, filtered, concentrated and freeze-dried to perform. Cytotoxicity of DGHT extract was performed by MTT assay. Activated macrophages were treated with varying concentrations of DGHT extract (10, 50, 100 and $200{\mu}g/mL$), and nitric oxide (NO) and prostaglandin E2 ($PGE_2$) concentrations were measured to detect anti-oxidative effects. Interleukin-6 (IL-6), interleukin-1 beta ($IL-1{\beta}$) and tumor necrosis factor-alpha($TNF-{\alpha}$) concentrations were also measured to detect inflammatory responses to DGHT Results: Cytotoxicity of DGHT extract at concentrations of 10, 50, 100 and $200{\mu}g/mL$ were not observed. NO production was significantly decreased in the DGHT hot aqueous extract $200{\mu}g/mL$ concentration group. $PGE_2$, IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ production was significantly decreased in the DGHT hot aqueous extract 100 and $200{\mu}g/mL$ concentration groups. DGHT hot aqueous extract appeared to have DPPH free radical scavenging capability at all of concentrations, but did not exceed 50%. Conclusion: These results suggest that DGHT hot aqueous extract has concentration-dependent anti-inflammatory and anti-oxidative effect.

Vitamin C Is an Essential Factor on the Anti-viral Immune Responses through the Production of Interferon-${\alpha}/{\beta}$ at the Initial Stage of Influenza A Virus (H3N2) Infection

  • L-ascorbic acid (vitamin C) is one of the well-known antiviral agents, especially to influenza virus. Since the in vivo antiviral effect is still controversial, we investigated whether vitamin C could regulate influenza virus infection in vivo by using Gulo (-/-) mice, which cannot synthesize vitamin C like humans. First, we found that vitamin C-insufficient Gulo (-/-) mice expired within 1 week after intranasal inoculation of influenza virus (H3N2/Hongkong). Viral titers in the lung of vitamin C-insufficient Gulo (-/-) mice were definitely increased but production of anti-viral cytokine, interferon (IFN)-${\alpha}/{\beta}$, was decreased. On the contrary, the infiltration of inflammatory cells into the lung and production of pro-inflammatory cytokines, tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-${\alpha}/{\beta}$, were increased in the lung. Taken together, vitamin C shows in vivo antiviral immune responses at the early time of infection, especially against influenza virus, through increased production of IFN-${\alpha}/{\beta}$.
