Effects of Daegangwhal-Tang Hot Aqueous Extract on Anti-inflammation and Anti-oxidation in RAW 264.7 Macrophage

  • Background: The objective of this study was to determine the effects of Daegangwhal-Tang (DGHT) hot aqueous extract on production of inflammatory mediators and antioxidants in RAW 264.7 macrophage. Methods: DGHT was extracted with water, filtered, concentrated and freeze-dried to perform. Cytotoxicity of DGHT extract was performed by MTT assay. Activated macrophages were treated with varying concentrations of DGHT extract (10, 50, 100 and $200{\mu}g/mL$), and nitric oxide (NO) and prostaglandin E2 ($PGE_2$) concentrations were measured to detect anti-oxidative effects. Interleukin-6 (IL-6), interleukin-1 beta ($IL-1{\beta}$) and tumor necrosis factor-alpha($TNF-{\alpha}$) concentrations were also measured to detect inflammatory responses to DGHT Results: Cytotoxicity of DGHT extract at concentrations of 10, 50, 100 and $200{\mu}g/mL$ were not observed. NO production was significantly decreased in the DGHT hot aqueous extract $200{\mu}g/mL$ concentration group. $PGE_2$, IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ production was significantly decreased in the DGHT hot aqueous extract 100 and $200{\mu}g/mL$ concentration groups. DGHT hot aqueous extract appeared to have DPPH free radical scavenging capability at all of concentrations, but did not exceed 50%. Conclusion: These results suggest that DGHT hot aqueous extract has concentration-dependent anti-inflammatory and anti-oxidative effect.

Anti-inflammatory activity of Ganoderma lucidum by inhibition of NF-κB p65 phosphorylation

  • Ganoderma lucidum, an oriental polypore fungus and medicinal mushroom, has a long history of use for promoting health and longevity in Korea, China, and other Asian countries. This study was aimed at determining the anti-inflammatory activity and mechanism of action of Ganoderma lucidum in murine macrophage RAW 264.7 cells. Ganoderma lucidum was extracted with ethanol and freeze-dried. The anti-inflammatory effect (nitrite production) of Ganoderma lucidum extracts was tested using a nitric oxide (NO) colorimetric assay. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was performed to quantify the mRNA expression of cytokines including tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$), interleukin $(IL)-1{\beta}$, and IL-6. Western blotting was performed to measure the expression levels of inflammation-related proteins, such as inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), nuclear factor kappa B ($NF-{\kappa}B$) p65, and phosphorylated $NF-{\kappa}B$ p65. The NO colorimetric assay showed that NO production increased with the treatment of lipopolysaccharide in (LPS)-activated RAW 264.7 macrophages and decreased with the cotreatment of Ganoderma lucidum extracts and LPS. Ganoderma lucidum extracts repressed the mRNA expressions of cytokines, which were increased after the LPS treatment. In addition, Ganoderma lucidum extracts inhibited the LPS-induced expression of iNOS and COX-2 and the LPS-induced phosphorylation of $NF-{\kappa}B$ p65. These results suggest that the Ganoderma lucidum extracts exert an anti-inflammatory activity by inhibiting $NF-{\kappa}B$ related proteins and cytokines.

Experimental Effects of Taklidanggui-tang on the Anti-Cancer and Immuno-Action (탁리당귀탕이 항암 및 면역작용에 미치는 실험적 효과)

  • Taklidanggui-tang was a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effect of Taklidanggui-tang on the anti-cancer and proliferation of immunocytes, nitric oxide(NO) production of peritoneal macrophages. We used Taklidanggui-tang extract(TDT) with freeze-dried, 8wks-old male mice, and L1210 cell lines for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follow ; TDT was showed cytotoxicity on the L1210 cell lines, increased significantly proliferation of thymocytes and splenocytes in vitro. TDT inhibited significantly proliferation of L1210 cells, increased significantly proliferation of immunocytes in L 1210 cells transplanted mice. And TDT was extended significantly mean survival days in 5-180 cells transplanted mice, but TDT did not increased NO production from peritoneal microphages in L1210 cells transplanted mice. This results suggest that TDT has anti-cancer and immuno-action.

Blueberry, blackberry, and blackcurrant differentially affect plasma lipids and pro-inflammatory markers in diet-induced obesity mice

  • BACKGROUND/OBJECTIVES: Evidence indicates that berry anthocyanins are anti-atherogenic, antioxidant, and anti-inflammatory. However, berries differ vastly in their anthocyanin composition and thus potentially in their biological and metabolic effects. The present study compared hypolipidemic, antioxidant, and anti-inflammatory properties of blueberry (BB), blackberry (BK), and blackcurrant (BC) in a diet-induced obesity (DIO) mouse model. MATERIALS/METHODS: Male C57BL/6J mice were fed a high fat (HF; 35% fat, w/w) control diet or a HF diet supplemented with freeze-dried 5% BB, 6.3% BK or 5.7% BC for 12 weeks (10 mice/group) to achieve the same total anthocyanin content in each diet. Plasma lipids, antioxidant status and pro-inflammatory cytokines were measured. The expression of genes involved in antioxidant defense, inflammation, and lipid metabolism was determined in the liver, epididymal adipose tissue, proximal intestine, and skeletal muscle. Histological analysis was performed to identify crown-like structure (CLS) in epididymal fat pads to determine macrophage infiltration. RESULTS: No differences were noted between the control and any berry-fed groups in plasma levels of liver enzymes, insulin, glucose, ferric reducing antioxidant power, superoxide dismutase, and tumor necrosis factor ${\alpha}$. However, BK significantly lowered plasma triglyceride compared with the HF control and other berries, whereas BC significantly reduced F4/80 mRNA and the number of CLS in the epididymal fat pad, indicative of less macrophage infiltration. CONCLUSIONS: The present study provides evidence that BB, BK and BC with varying anthocyanin composition differentially affect plasma lipids and adipose macrophage infiltration in DIO mice, but with no differences in their antioxidant capacity and anti-inflammatory potential.

  • This study was performed to develop anti-obesity Phellineus linteus powder for application as food additives. P. linteus powders which contained anti-obesity lipase inhibitor were prepared by freeze drying and spray drying. Effects of various additives on the quality of their powders were investigated. 10% over addition of dextrin and gum-arabia was good in the rheological properties of their powders and optimal spray temperature was near $130^{\circ}C$. When their powders were sealed and storaged at room temperature for 6 months, its characteristics were not changed. Especially, changes of its colors was repressed by addition of 0.3 M oxalic acid, citric acid and ascorbic acid than those by non-treatment.

A comparative study on the phytochemical and anti-oxidant activity differences in HongJam prepared with various silkworm varieties

  • The aim of this study was to elucidate the difference in the amount of phytochemicals and anti-oxidant activities of the freeze-dried 5th instar 3rd day silkworm powder (FDSP) and 7 HongJams (HJs). Three primary colors of light of 8 silkworm powders revealed why each sample had unique color. The visual and absorption spectrum analyses of 80% methanol (MeOH) extracts of 8 silkworm powders revealed that white jade (WJ)-FDSP extracts showing strong forest-green colors had different absorption spectrum from 7 HJs. Among 7 HJs, the absorption spectrum of light green silkworm (LGS)-HJ extract was different from those of other HJ extracts. The amounts of total flavonoids and phenolic compounds of WJ-FDSP were 423.0±9.21% and 252.3±12.86%, respectively, more than those of WJ-HJ. Among 7 HJs, the amounts of total flavonoids and phenolic compounds in LGS-and red silkorm (RS)-HJ extracts were significantly more than those of WJ-HJ extract. Consistent with the amounts of phytochemicals, radical scavenging activity and reducing ability of WJ-FDSP extract were significantly higher that other HJ extracts. Among 7 silkworm powder extracts, LGS-HJ extract showed significantly higher radical scavenging activity and reducing ability than those of WJ-HJ extract. But, RS-HJ extract only showed significantly higher radical scavenging activity than that of WJ-HJ extract. Taken together, the amounts of phytochemicals in 8 silkworm powders contributed on their colors and anti-oxidant activities.

Effects of Gamisibgi-San on the Immunocytes and Cancer cell (면역세포(免疫細胞) 및 종양세포(腫瘍細胞)에 미치는 가미십기산(加味十奇散)의 효과(效果))

  • Objective : Gamisibgi-San was a drug that treated carbuncle and cellulitis. Materials and Method : We used Gamisibgi-San extract(GMSGS) with freeze-dried, 8wks-old male mice and cancer cell lines(L1210, S-180) for this Study. The cytotoxicity and proliferation of cells wat tested using a colorimetric tetrazoliun assay(MIT assay). Results and Conclusion : The results of this Study were obtained as follow ; 1. GMSGS was significantly showed cytotoxicity on the L1210 cell lines and S-180 cell lines. 2. GMSGS was significantly increased in the proliferation of thymocytes and splenocytes in vitro. 3. GMSGS was significantly decreased in the proliferation of L1210 cells in L1210 cells transplanted mice. 4. GMSGS was significantly decreased in the Weight of Sarcoma in S-180 cells transplanted mice. 5. GMSGS was significantly increased in the Period of Survive in S-180 cells transplanted mice. The present author thought that GMSGS had action of anti-cancer by becoming immunocytes activity(proliferation of thymocytes and splenocytes).

Isolation and Identification of Phenol Compounds from Acer tegmentosum and their Anti-inflammatory Activity (산겨릅나무로부터 페놀화합물의 분리 및 항염증 활성의 측정)

  • The Acer tegmentosum (3 kg) were extracted with boiled water and the freeze dried extract powder was partitioned with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc and n-BuOH fraction, six phenolic compounds were isolated through the silica gel, octadecyl silica gel and sephadex LH-20 column chromatography. On the basis of spectroscopic methods, such as $^1H$-NMR and $^{13}C$-NMR, and LC/MS, the chemical structures of the compounds as feniculin (1), avicularin (2), (+)-catechin (3), (-)-epicatechin (4), salidroside (5) and 6'-O-galloylsalidroside (6). In this study, compounds 1 and 2 have been first isolated from the A. tegmentosum. To provide insight into the effects of six compounds isolated from A. tegmentosum on inflammation, we investigated its effect on nitric oxide (NO) production in RAW 264.7 cells using lipopolysaccharide (LPS) stimulation. Compounds 1 and 6 slightly repressed NO production. Also, compounds 3 and 4 inhibited NO secretion with statistical significance. However, compounds 2 and 5 did not show any inhibitory effect on NO production.

Ramie Leaf Extracts Suppresses Adipogenic Differentiation in 3T3-L1 Cells and Pig Preadipocytes

  • The present study was carried out to evaluate the anti-obesity effect of different concentrations of extracts of hot air-dried ramie leaf (HR) and freeze-dried ramie leaf (FR) in 3T3-L1 cells and pig preadipocytes. To analyze the effect on cell proliferation, cells were treated with $25{\mu}g/mL$ or $100{\mu}g/mL$ HR or FR extract for 2 days. Cell differentiation was evaluated by measuring glycerol-3-phosphate dehydrogenase and lipoprotein lipase (LPL) activities and intracellular triglyceride content. Treatment with either HR or FR extracts inhibited the proliferation of 3T3-L1 cells and pig preadipocytes in a dose-dependent manner. HR extract treatment inhibited the differentiation of both cell types more effectively than FR treatment. The extent of triglyceride accumulation decreased significantly in both cells following either HR or FR treatment. Furthermore, LPL activity significantly decreased after treatment with HR or FR extract. These results indicated that HR and FR extracts may inhibit proliferation and differentiation of 3T3-L1 cells and pig preadipocytes. Further studies are needed to explore the anti-obesity effect of HR and FR extracts.

Effects of BojungIkkiTang-Gamybang on Protective of Cell Death and Anti-Oxidative in C6 Glioma Cell (보중익기탕가미방(補中益氣湯加味方)이 신경교세포의 세포사멸보호 및 항산화에 미치는 영향)

  • This study was designed to investigate the effects of BojungIkkiTang-Gamybang freeze dried powder (BITG) on proliferauion, protective of cell death induced by chemicals such as paraquat, hydrogen peroxide etc and anti-oxidative effects in C6 glioma cells. In our results, BITC accelerated proliferation rates of C6 cells in vitro. In addition, protective effects on cell death induced by paraquat and hydrogen peroxide. And, BITC did not have effects on SOD and total glutathione activities, but decresed malone dialdehyde activity. In conclusion, these results suggest the possibility of BojungIkkiTang-Gamybang to protect brain cell or neuronal cell from damage induced by oxidative stress. And also suggest that related mechanisms are involved in malone dialdehyde activity.
