추출방법에 따른 소목 심재의 항산화 및 항당뇨 활성 평가 (Evaluation of Antioxidant and Anti-diabetic Effects of Sappan Lignum by Extraction Method)

  • Objectives : The heartwood of Sappan Lignum has been used since ancient times as an ingredient in folk medicines against anti-bacterial and anti-anemia purposes. Many bioactive constituents have been derived from this biomass such as chalcones and homoisoflavonoids. In the current investigation, the antioxidant and anti-diabetic properties using DPPH and $ABTS^+$ radicals scavenging, ${\alpha}-glucosidase$, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Sappan Lignum. Methods : In our continuing investigation for bioactive natural ingredients, the antioxidant and ${\alpha}-glucosidase$ inhibitory properties of Sappan Lignum extracts were prepared from different extraction methods and the biological efficacies were investigated in vitro. The antioxidant properties were evaluated employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the anti-diabetic effects of Sappan Lignum extracts were tested via ${\alpha}-glucosidase$ and AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested samples showed dose-dependent radical scavenging and ${\alpha}-glucosidase$ inhibitory activities. Among the tested extracts, the 80% methanolic extract of Sappan Lignum was showed the most potent activity with an $IC_{50}$ value of $82.3{\pm}1.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. While, $ABTS^+$ radical scavenging activity of 80% methanolic extract was higher than those of other extracts. Also, ${\alpha}-glucosidase$ inhibitory and AGEs formation effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Sappan Lignum can be considered as a new effective source of natural antioxidant and anti-diabetic materials.

인동등지골피탕(忍冬藤地骨皮湯)이 Streptozotocin으로 유발된 흰쥐에서의 항당뇨 효과에 대한 연구 (Anti-diabetic Effect of Indongdeungjikolpi-tang in Streptozotocin-induced Diabetic Rats)

  • Objectives: Indongdeungjikolpi-tang(IJT) is used as a traditional treatment of diabetes in oriental clinincs. This study aimed to evaluate the anti-diabetic effect of Indongdeungjikolpi-tang(IJT) in streptozotocin(STZ)-induced diabetic rats. Methods: Diabetes was induced by intraperitoneal injection of streptozotocin(STZ; 60 mg/kg BW) to Sprague-Dawley male rats. Experimental animals(six per group), were treated by oral administration of IJT(100 and 500 mg/kg BW) and glibendimide(3 mg/kg), a known antidiabetic drug for comparison, during 4 weeks. We measured the levels of glucose, insuline, triglyceride, creatinine and urea in sera of each group. An oral glucose tolerance test(OGTT) was also performed in all groups. Results: IJT(100 and 500 mg/kg) significantly reduced blood glucose levels and increased plasma insulin levels in STZ-induced diabetic rats. IJT also significantly reduced the plasma levels of tryglyceride, creatinine and urea in STZ-induced diabetic rats. The OGTT results showed a significant improvement in glucose tolerance in IJT-administrated rats. Conclusions: These data indicate that IJT may improve glocose homeostasis in STZ-induced diabetes, which could be associated with stimulation of insulin secretion.

Antioxidant Properties of Water Extract from Acorn

  • Antioxidant and anti-diabetic activities of acorn were evaluated by its potential for scavenging stable DPPH free radical, inhibition of lipid peroxidation, reducing power, and inhibiton of ${\alpha}-glucosidase\;and\;{\alpha}-amylase$. The water extract of acorn exhibited strong antioxidant and antidiabetic related activities in the tested model systems. Solvent fractionation of the water extract revealed that the water fraction and the EtOAc fraction had strong antioxidant activity, and inhibitory activity on ${\alpha}-glucosidase\;and\;{\alpha}-amylase$. The water fraction exhibited higher DPPH radical scavenging activity ($EC_{50}=7.19{\mu}g/mL$) than that of ${\alpha}-tocopherol\;(EC_{50}=7.59{\mu}g/mL)$. It is considered that water extract of acorn has the potential for natural antioxidant and anti-diabetic products.

  • To investigate anti-diabetic component from the node of lotus rhizome (Nelumbo nucifera; Nymphaeaceae), activity guided isolation was conducted. One amino acid was isolated from active fraction of the aqueous methanolic extract. The structure of this compound was identified as tryptophan (1) by the analysis of spectroscopic evidences and comparisons with the data of authentic samples. Tryptophan reduced the blood glucose level significantly in glucose-fed hyperglycemic mice compared with glucose-treated group and exhibited 44.3% of activity compared with tolbutamide,.

  • Flavonoids consist a wide range of naturally occurring compounds which are exclusively found in different fruits and vegetables. These medicinal herbs have a number of favourable biological and therapeutic activities such as antioxidant, neuroprotective, renoprotective, anti-inflammatory, anti-diabetic and anti-tumor. Troxerutin, also known as vitamin P4, is a naturally occurring flavonoid which is isolated from tea, coffee and cereal grains as well as vegetables. It has a variety of valuable pharmacological and therapeutic activities including antioxidant, anti-inflammatory, anti-diabetic and anti-tumor. These pharmacological impacts have been demonstrated in in vitro and in vivo studies. Also, clinical trials have revealed the efficacy of troxerutin for management of phlebocholosis and hemorrhoidal diseases. In the present review, we focus on the therapeutic effects and biological activities of troxerutin as well as its molecular signaling pathways.

죽염의 약리작용 평가 (Pharmacological Evaluation of Bamboo Salt)

  • Bamboo salt has been used for the purpose of precaution and treatment of certain diseases including cancer. Therefore, present study was carried out to ascertain the effects of bamboo salt upon anti-cancer, anti-hypertensive, and anti-diabetic activities as well. To examine the anti-cancer activity of bamboo salt, ICR mice implanted with 1$\times$l0$^{6}$ cells of sarcoma 180 intraperitoneally had been treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 60 days using adriamycin (2 mg/kg) as a positive control. Neither survival rate nor body weight had been significantly influenced by all the treatments indicating that bamboo salt A did not exert the anti-cancer effect on ICR mice. Anti-hypertensive activity was examined in spontaneously hypertensive rats (SHR) which had been administered with bamboo salt A, crude salt, and reagent-grade NaCl (0.1, 0.5, and 1.0% in drinking water) for 28 days using hydralazin (2 mg/kg) as a positive control. Blood pressure and heart rate were measured at 1, 3, and 4 weeks after the starting date. Significant anti-hypertensive activity was not observed in any treated group compared to the positive control group. In order to determine if bamboo salt had anti-diabetic activity, rats in which diabetes had been induced by streptozotocin (45 mg/kg, i.m.) were treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 28 days using insulin (50 U/kg, s.c..) as a positive control. Blood samples were taken and analyzed at 1,2, and 4 weeks after the starting date. Bamboo salt did not cause any decreasing effect on the blood glucose levels. These results clearly demonstrated that bamboo salt A did not exert anti-cancer, anti-hypertensive, or anti-diabetic activities in the present experimental animals.

A Vinegar-processed Ginseng Radix (Ginsam) Ameliorates Hyperglycemia and Dyslipidemia in C57BL/KsJ db/db Mice

  • Having idea to develop more effective anti-diabetic agent from ginseng root, we comprehensively assessed the anti-diabetic activity and mechanisms of ginsam in C57BL/KsJ db/db mice. The db/db mice were divided into 4 groups; diabetic control (DC), ginsam at a dose of 300 or 500 mg/kg (GS300 or GS500) and metformin at a dose of 300 mg/kg (MT300). Ginsam was orally administered for 8 weeks. GS500 reduced the blood glucose concentration and significantly decreased an insulin resistance index. In addition, GS500 reduced the plasma non-esterified fatty acid, triglyceride, and increased high density lipoprotein-cholesterol as well as decreased the hepatic cholesterol and triglyceride. More interestingly, ginsam increased the plasma adiponectin level by 17% compared to diabetic control group. Microarray, quantitative-polymerase chain reaction and enzyme activity results showed that gene and protein expressions associated with glycolysis, gluconeogenesis, and fatty acid oxidation were changed to the way of reducing hepatic glucose production, insulin resistance and enhancing fatty acid $\beta$-oxidation. Ginsam also increased the phosphorylation of AMP-activated protein kinase and glucose transporter expressions in the liver and skeletal muscle, respectively. These changes in gene expression were considered to be the mechanism by which the ginsam exerted the anti-diabetic and anti-dyslipidemic activities in C57BL/KsJ db/db mice.

Effect of long term treatment of aqueous extract of Enicostemma littorale in Type 2 diabetic patients

  • We have evaluated the effect of long term treatment of Enicostemma littorale (E. littorale) in type 2 diabetic patients taking pills of aqueous extract of E. littorale regularly as a complimentary medicine for at least 9 months. The effects of E. littorale on glycemic control, lipid profile, cardiac function and DNA damage in these patients were compared with those who had not been regular in taking E. littorale but regular in taking other conventional anti-diabetics. Our data suggest that, E. littorale can maintain normal blood glucose, serum insulin, serum triglycerides levels of type 2 diabetic patients if taken regularly. E. littorale also improves insulin sensitivity, and normalize disturbed lipogram and elevated creatinine levels, thereby produces beneficial effect in preventing cardiovascular complications and may preserve the kidney function. The finding that E. littorale also prevents DNA damage suggest a long term effect in diabetic patients. E. littorale thus can be considered as safe supplementary therapy for a long term and effective management of type 2 diabetic patients.

  • Anti-diabetic effect of the exopolysaccharides (EPS) produced from submerged mycelial culture of Cordyceps sinensis (Cs) was studiedin a type II diabetic animal model (C57BL/6J ob/ob). This study was designed to determine whether Cs-EPS improves clinical symptoms of type II diabetes in ob/ob mice. After Cs-EPS treatment at doses of 200 mg/kg body weight, the fasting blood glucose levels decreased by 47% after 7 weeks compared with those of the control mice. According to the oral glucose tolerance test, the glucose levels recovered its baseline after 120 min in Cs-EPS-treated mice, although the blood glucose levels increased significantly after 30 min. On the other hand, the control group (not-treated) did not recovered its initial level of glucose after 120 min. Furthermore, food intake, body weight, total plasma cholesterol and triglyceride concentrations in ob/ob mice treated with Cs-EPS were significantly decreased, compared with those in control ob/ob mice. Cs-EPS treatment increased significantly the plasma insulin level and the expression of leptin mRNA in adipose tissue of Cs-EPS-treated ob/ob mice. From these results, it is demonstrated that Cs-EPS could be effective for regulating normal blood glucose levels by increasing the amounts of plasma insulin and leptin expression in ob/ob mice, indicating that this compound could be a candidate material as a dietary supplement to control hyperglycemia in patients suffering from type II diabetes.

Oxidative and Anti-oxidative Status in Blood of Streptozotocin-induced Diabetic Piglets

  • Eight LW${\times}$D crossbred, castrated weanling piglets were used to examine the effect of hyperglycemia by streptozotocin (STZ)-injection on oxidative and anti-oxidative status in circulating fluid. Every two of the eight piglets were intravenously administrated STZ at a dose of 0 (control), 100, 125 or 150 mg/kg BW, respectively, and on 15th day after the STZ-injection, some markers of the oxidative stress in circulating fluid were measured to evaluate oxidative and anti-oxidative status in the piglets. First, piglets with hyperglycemia were selected from the STZ-injected piglets as measured by the levels of fasting plasma glucose (FPG) during 2 weeks after the STZ-injection. Additionally, data obtained from the intravenous glucose tolerance test (IVGTT) on 14th day were analyzed. Secondly, the data obtained in this experiment were divided into the control group and the hyperglycemic (STZ) group, and compared. The FPG level or area under curve (AUC) for plasma glucose during the IVGTT in the STZ-induced diabetic piglets was slightly significantly (FPG, p = 0.070; AUC, p = 0.072) higher compared with the control. On the other hand, the plasma level of lipid peroxidation in the STZ-induced diabetic piglets was significantly (p<0.05) higher compared with the control. These results raise the possibility that STZ-induced diabetic piglets produced in this study can be used as a diabetic animal model to research the pathogenic mechanisms or therapy of complications in diabetic mellitus.
