감맥대조탕 복합추출물의 만성스트레스 유발 생쥐의 항우울 작용에 관한 실험연구 (Experimental Study on the Anti-Depressant Effects of Gammakdaejo-tang Complex Extracts in Rats Induced with Chronic Mild Stress)

  • Objectives: To experimentally assess the anti-depressant effects of Gammakdaejo-tang Complex Extracts (GMDJ-Tang) in rats. Methods: Twenty Wistar Hannover (160~170 g) stressed rats were treated with different concentrations of GMDJ-Tang extracts i.e., 0, 62.5 mg/kg, 125 mg/kg, and 250 mg/kg. Chronic mild stress was induced by food deprivation, empty bottles, forced swimming, flickered light, tilt cages, shaking cages, high density breeding, water deprivation, and by soaking the litter cover according to fixed schedule. Blood and brain tissue samples were collected for biochemical analysis. Tests included serotonin and ELISA assays. Results: GMDJ-tang increased the weight of treated rats as well as levels of serotonin, BDNF and TrkB; however, the differences were not significant. In contrast, the extracts significantly decreased blood glucose in stressed rats. GMDJ-tang extracts did not significantly impact Serum AST, ALT, leukocytes, erythrocytes and thrombocytes when comparing treatment groups to control rats. Likewise, hemoglobin, hematocrit and PLT increased in treatment groups following treatment with GMDJ-tang, but this change was without significance. Conclusions: These results suggest that GMDJ-tang can alleviate chronic mild stress in rats, possibly through anti-depressant activity.

섬백리향 정유의 진통, 항염증, 정신적인 작용 (Anti-nociceptive, Anti-inflammatory, Mental Effects of Essential Oil from Thymus magnus)

  • Thymus magnus is an endemic (Ulleung Island) species in Korea. This plant is used as diaphoretics and carminatives in traditional medicine. In the literature, few scientific assays were realized on this species, such as antibiotic (Streptococcus pneumoniae, Staphylococcus aureus, Salmonella enteritidis, and S. typhimurium) and antifungal activities. In order to clarify whether essential oil of T. magnus have pharmacological effects, anti-inflammatory, sedative, anti-depressant, analgesic, and sleep-prolonged effects were investigated using animal models. From this study, the following conclusions were attained; 1) Essential oil of T. magnus did not show any acute toxicity on mice when orally administered at the dose of 2-3 g/kg body weight. 2) Essential oil of T. magnus possessed strong anti-inflammatory activity, similar to that of a positive control prednisolone. 3) Essential oil of T. magnus had excellent analgesic activity, comparable to that of aspirin. 4) The essential oil of T. magnus possessed strong sleep-prolonged effect on pentobarbital induced-sleep test in mice model. 5) In the hot plate test, the essential oil of T. magnus had moderate effect. 6) And the essential oil of T. magnus had no significant effects in forced-swimming test and open-field test.

부자(附子)의 우울증 억제효과에 관한 연구 (Anti-depressant Effect of the Extracts of Aconitum carmichaeli)

  • Objectives : The water and methanol extracts of Aconitum carmichaeli(Aconiti Tuber Preparat) were investigated for their anti-depressant effects. Methods : In this study, reserpine-induced hypothermia test, tail suspension test and hot plate test. Additionally, the brain monoamine oxidase activity was determined in vivo. Results: In the reserpine-induced hypothermia test, both extracts suppressed the fall of body temperature compared to the control group in a dose-dependent manner, suggesting the inhibition on hypothermia. In the tail suspension test, the methanol extract dose-dependently reduced the duration of immobility by 28.4% at a dose of 1 g/kg compared to control group, which is more effective than the water extract. In the hot plate test, the water extract and methanol extract increased the jump latency time compared to the control group, showing the inhibition rate of 198% and 182%, respectively, at a dose of 1 g/kg. Methanol extracts potently inhibited the brain monoamine oxidase activity in an in vivo assay compared to the control group, showing 84.6% inhibition, but the water extract revealed very weak activity. Conclusions : Above results suggested that the extract of Aconitum carmichaeli can be useful for the prevention and treatment of depression.

권삼(拳蔘)이 지혈(止血).소염작용(消炎作用) 및 중추신경계(中樞神經系)에 미치는 영향(影響) (Effects of Bistortae Rhizoma on Hemostasia, Anti-inflammatory Action and Central Nervous System)

  • Objective : The purpose of these research was to investigate effects of water extract of Bistortae Rhizoma(BRE) on the hemostasia, anti-inflammatory action and central nervous system. Methods : we used mice and rats administered with the extract of the above herbs. Results : BRE decreased the permeability of evans blue into peritoneal cavity and cotton pellet granuloma formation. BRE did not decrease the acetic acid induced writhing syndrome and the histamine induced mouse paw edema. BRE inhibited the pentylenetetrazole and the strychnine induced convulsion. BRE shortened the bleeding time and plasma prtrombin time. BRE did not affect on the proliferation of Balb/c 3T3 cells. Conclusions : these results suggest that the effects of BRE are the hemostasia, anti-inflammatory action, and mild depressant activity of central nervous system.

효모 추출물 SCE 및 그 분획 SCE-40의 항 우울 및 항 불안 효과 (Anti-depressant and anti-anxiety effects of Saccharomyces cerevisiae extract and its hydrolyzed fraction)

  • 효모 (Saccharomyces cerevisiae)는 인체에 무해한 Generally Recognized As Safe(GRAS)급으로 인정되며, 최근 효모 추출물이 정신적 긴장, 과민, 두통 같은 월경 전 증후군을 완화시키는 효과가 있음이 보고되어 있어, 효모추출물(Saccharomyces cerevisiae extract: SCE)과 그 분획(SCE-40)이 우울증과 불안증에서도 효과가 있는지 확인하고자 다음 실험을 실시하였다. 행동학적 검정으로 SCE의 항우울 효과를 확인하기 위해 SCE를 0, 1, 10, 100mg/kg의 농도로 웅성 ICR 생쥐에게 2주간 경구 투여한 후 강제수영검사(forced swimming test; FST)에서의 부동시간과, 또 다른 항우울 효과 검정 실험인 미현수검사(Tail Suspension Test: TST)에서의 부동시간을 측정하였다. 또한, SCE의 항불안 효과 검정을 위해서 SCE를 0, 1, 10, 100mg/kg의 농도로 웅성 ICR 생쥐에게 1회(1시간 전), 혹은 2주간 경구 투여한 후 고위십자미로검사(Elevated-plus-maze test : EPM)에서의 open arms에 머문 시간을 측정하였다 시험관내 실험으로는 웅성 백서의 대뇌피질에서의 SCE 및 SCE-40의 serotonin transporter (SERT), norepinephrine transporter(NET), 그리고 GABA의 ligand의 결합 억제능 측정과, serotonin (5-hydroxytryptamine: 5-HT), norepinephrine uptake 측정이 수행되었다. 행동적 실험 결과, FST에서 1회나 2주간 SCE 10mg/kg과 100mg/kg을 투여 받은 생쥐에서 생리 식염수만 투여 받은 생쥐보다 부동시간이 유의하게 감소됨을 보여 SCE에 의해 항우울 효과가 유발됨을 보였고, TST 실험에서도 유사한 결과가 나타났다. 또한, 항불안을 측정하는 EPM 실험에서도 1회나 2주간 SCE 10mg/kg과 100mg/kg을 투여 받은 생쥐에서 생리 식염수만 투여 받은 생쥐보다 open arms에서 머무는 시간이 유의하게 증가되어 SCE가 단기 또는 장기간의 항불안에 효과가 있음을 나타냈다. 시험관내 실험 결과에서는, SCE와 SCE-40이 SERT, NET, 그리고 GABA ligand의 결합 억제능이 있음이 확인되었고, SCE와 SCE-40은 serotonin(5-hydroxytryptamine: 5-HT)과, norepinephrine의 uptake를 억제하는 것으로 나타났다. 그러므로 본 연구는 효모 추출물(SCE)과 그 분획(SCE-40)이 행동적, 신경학적으로 항우울, 항불안에 효과가 있음을 확인하였으며, 효모 추출물(SCE)과 그 분획(SCE-40)이 안전한 천연식품으로서 우울증, 불안증 등의 관련 질환의 예방, 치료용 의약품 개발과 기능성 식품에 효과적으로 이용될 수 있음을 시사한다.

적작약(赤芍藥)이 강제수영부하시험에서 CRF, c-Fos 와 TH에 미치는 영향 (Effects of Paeoniae Radix Rubra on CRF, c-Fos and TH in the Forced Swimming Test)

  • Objectives : For the purpose of verifying the anti-depressant effect of Paeoniae Radix Rubra(PR), the expression of corticotropin-releasing factor(CRF), c-Fos and tyrosine hydroxylase(TH) was evaluated after performing the Forced Swimming Test(FST). Methods : Spraque-Dawley rats were ingested PR extract(100mg/kg, 400mg/kg, p.o.) for 3 times prior to FST. And the expression of corticotropin-releasing factor(CRF), c-Fos in the paraventricular nucleus(PVN) and tyrosine hydroxylase(TH) in the locus coelureus(LC) and ventral tegmental area(VTA) was measured immunohistochemically after FST. Results : The duration of immobility was significantly decreased in PR 100mg/kg Group and PR 400mg/kg Group, in comparison with the control group (p<0.001). The expression of CRF in the PVN was significantly decreased in PR 400mg/kg Group in comparison of the control group (p<0.05). The expression of c-Fos in the PVN was rather significantly increased in PR 100mg/kg Group in comparison with the control group, while almost no change was demonstrated in PR 400mg/kg Group. The expression of TH was significantly decreased in VTA in comparison with the control group (p<0.05), but the number of expression cells in LC was slightly decreased in case of PR 100mg/kg group while it was increased in case of PR 400mg/kg Group. Conclusion : Judging from the result of the aforementioned tests, Paeoniae Radix Rubra has decreased immobility. In addition, it has also decreased the expression of CRF and the expression of TH in VTA, while the expression of c-Fos and of TH in LC has no significance. Therefore, it is believed that Paeoniae Radix Rubra has an anti-depressant effect by decreased immobility through the reduced expression of CRF and TH in VTA.

커큐민이 건강에 미치는 효과 - 재평가 (Effects of curcumin on human health - A reassessment)

  • 커큐민은 산화방지능, 항염증, 항우울, 항균, 항종양 등의 다양하고 유익한 생리학적 활성이 있는 것으로 알려져 있다. 또한, 커큐민은 면역조절, 대사조절, 신경보호 효과도 가지고 있다. 지난 10년 동안 13,000편 이상의 연구 논문에서 커큐민의 건강상 이점에 대한 연구가 발표되었음에도 불구하고, 아직까지 전 세계 어느 나라에서도 커큐민을 의약품으로 시장에 출시하지 못하고 있다. 긍정적인 결과가 많음에도, 건강상의 이점을 내세우는 대부분의 연구는 커큐민의 다양한 화학적 거동을 고려하지 않은 시험관실험 및 동물실험에 기반으로 하고 있다. 커큐민은 PAINS이자 IMPs 중의 하나로 알려져 있다. 따라서 엄격한 연구 설계에도 불구하고, 인체적용시험 및 임상시험은 특별한 이점을 입증할 수 없었다. 이 논문에서는 커큐민의 다면적 특성을 검토하여 커큐민의 부정적 특성이 이로운 효과와 균형을 이룰 수 있도록 하고 건강상의 이점을 더 잘 이해할 수 있는 잠재적 연구 방법을 제안하고 있다.

타우린의 일반적 특성에 관한 선행연구 고찰 (General Characteristics of Taurine: A Review)

  • Taurine is one of the most abundant free ${\beta}$-amino acids in the human body that accounts for 0.1% of the human body weight. It has a sulfonic acid group in place of the more common carboxylic acid group. Mollusks and meat are the major dietary source of taurine, and mother's milks also include high levels of this amino acid. The leukocytes, heart, muscle, retina, kidney, bone, and brain contain more taurine than other organs. Furthermore, taurine can be synthesized in the brain and liver from cysteine. There are no side effects of excessive taurine intake in humans; however, in case of taurine deficiency, retinal abnormalities, reduced plasma taurine concentration, and other abnormalities may occur. Taurine enters the cell via a cell membrane receptor. It is excreted in the urine (approximately 95%) and feces (approximately 5%). Taurine has a number of features and functions, including conjugation with bile acid, reduction of blood cholesterol and triglyceride levels, promotion of neuron cell differentiation and growth, antioxidant effects, maintenance of cell membrane stability, retinal development, energy generation, depressant effects, regulation of calcium level, muscle contraction and relaxation, bone formation, anti-inflammatory effects, anti-cancer and anti-atherogenic effects, and osmotic pressure control. However, the properties, functions, and effects of taurine require further studies in future.

서울, 경기지역의 황기에 대한 인지도 및 이용실태 조사 (An Investigative Analysis of Recognition and Uses for Astragalus Membranaceus in Seoul and Kyunggido Area)

  • This study was conducted to investigate the recognition and uses of Astragalus membranaceus. The survey methodology employed was a questionnaire, which interviewed 272(37.8%) males and 448(62.2%) females from the Seoul and Kyunggido area. The main results are as follows: 40-50 years old like and eat Astragalus membranaceus more frequently than 10-30 years old. The most popular reason provided for the preference of Astragalus membranaceus was good health benefits. People indicating a dislike for Astragalus membranaceus had generally fewer previous experiences of eating this plant. The main purchasing place appears to be traditional markets and supermarkets. Astragalus membranaceus was acknowledged as having the following characteristics ‘excellent nutrition’(3.82), ‘natural food’(3.76), ‘possessing anti-cancer and anti-oxidization characteristics’(3.70) and ‘good for liver function, a depressant and good coelenteron effects’(3.62). However, it was not associated with a ‘cheap price’(3.02) and was regarded as ‘difficult for buy’(3.10). Recognition and eating experience was low for Astragalus membranaceus kimchi and rice bread, bread, sauce and Astragalus membranaceus added health drinks. However opinion of its taste was regarded highly.
