Contrasting Correlation in the Inhibition Response of ADP-induced Platelet Aggregation and the Anti-coagulant Activities of Algal fucoidans Derived from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu)

  • Sulfated fucans are known to have both anti-thrombotic and anti-coagulant activities. In this study, the variation in platelet aggregation and anti-coagulant activities was investigated in vitro with regard to administered dose, molecular weight distribution, sulfate content, and sugar composition in two algal fucoidans from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu). The anti-coagulant activity largely correlated with sulfate content and with molecular weight distribution in a dose-dependent manner. However, both fucoidans demonstrated inhibitory responses to ADP-induced platelet aggregation in dose- and structure-dependent manners that contrasted with the anti-coagulant activity. Neither molecular weight distribution nor sulfate content greatly affected platelet-aggregation inhibition (PA-inhibition) by the fucoidan fractions, whereas anti-coagulant activity was sensitive to these structural factors. Interestingly, an E. bicyclis fucoidan fraction exhibited almost complete PA-inhibition at a treatment dose of 500 mg/mL while retaining weak anti-coagulant activity. In conclusion, these observations suggest that fucoidan may be a useful anti-thrombotic or anti-platelet agent in various arterial thrombotic disorders, including post-vascular intervention with controlled bleeding complications, due to its anti-coagulant modulating activity.

  • The purpose of this study is to investigate the anti-coagulant effects of Polygoni Mutiflori Radix extracts. The results were as follows : 1. This study show that Polygoni Mutiflori Radix extracts have Anti-coagulant significant effects on Prothrombin time in vitro test. 2. This study show that Polygoni Mutiflori Radix extracts have Anti-coagulant significant effects on Activated Partial Thromboplastin Time vitro test.

Synthesis and Biological Activity of Aspirin Derivatives

  • Aspirin has been widely used as analgesic and anti-inflammatory drug. Recently, it was elucidated that aspirin have anti-coaggregatory effect in low dose. This study was carried out to investigate the synthesis of aspirin derivatives from aspirin and aromatic compound of antioxidant and its biological activities. Synthesis of aspirin derivatives was prepared by esterification in the presence of 1, 1-carbonyldiimidazole. Biological activities was examined using effect of anti-coagulant on bleeding time, effect of antioxidant and effect of anti-platelet aggregation. As a result, SJ-101 showed strong antioxidative activity and anti-coagulant activity among four compounds. Anti-platelet aggregation of SJ-101 was examined by collagen, ADP, PAF method. SJ-101 exhibited more stronger activity to aspirin at collagen aggregation reaction. These finding demonstrates that SJ-101 is usefull as care drug of aging and old-disease because of its has antioxidant activity, anti-coagulant activity and anti-platelet activity.

  • Many plants and animal have long been known to have medicinal effects and therefore have been used as medicines. There are many substances that show various pharmacologic efficacy such as anti-tumor efficacy, anti-inflammatory efficacy, cholesterol-lowering efficacy, anti-coagulant of blood efficacy and anti-bacterial efficacy. I summarized the recent advances in research on physiologically functional foods. The pharmacological efficacy of dietary fiber, chitin & chitosan, DHA(docosahexaenoic acid), mushroom, alginic acid and herbs have selected as topices for discussion. I was examining the anti-coagulant activity of herbs, I discovered that Eugenia caryophyllata T. (clove) had a relatively high anti-coagulant activity.

  • Anti-coagulant activities of 25 species of medicinal plants which have been related to "blood" in ethnobotany, were evaluated by plasma recalcification time test. Among these, Artemisiae Herba showed the strongest activity. Its anti-clotting component was purified by using DEAE-cellulose, Sephadex G-75, and Sephadex LH-20 column chromatography. Its active compound has an average molecular weight of approximately $10^{4}$ and it was turned to be an acidic polysaccharide composed of galacturonic acid and rhamnose (7 : 2) by chemical analysis and spectral data.

  • Methanol extract of Loranthus yadoriki showed the prolongation effect of bleeding time in vivo using mice in dose dependent manner. From the MeOH extract of Loranthus yadoriki, compound-A was isolated by the activity guided isolation method using silicagel column chromatography. The anti-coagulant activity was evaluated by the bleeding time test in vivo and plasma recalcification time test in vitro. Compound A showed moderate anti -coagulant activity on plasma recalcification time in vitro.

  • Platelets play critical roles in both hemostasis and thrombosis. It was reported that platelet aggregation is associated with an increase in superoxide production and can be inhibited by hydroxyl radical scavengers. In the course of our search for the anti-platelet, anti-coagulant and/or anti-oxidative components from plants, the MeOH extract of Crassula cv. "Himaturi" (Crassulaceae) was observed to have both anti-aggregatory and anti-coagulant effects. A novel compound, 12-O-(4'-O-methyl-galloyl)-bergenin (1), was isolated as an active component from the EtOAC soluble fraction. (omitted)

  • Angelica genuflexa (Umbelliferae) is a perennial herbaceous plant which has also been variously reported as A. koreana and Ostericum koreanum. The MeOH extract was reported to have strong anti-thrombotic potential in the acute thrombosis model. In our preliminary testing, the MeOH extract and one of the solvent fractions (90% MeOH fr.) were observed to have both platelet anti-aggregating and anti-coagulant effects. Five coumarins, isoimperatorin (1), pabulenol (2), isooxypeucedanin (3), oxypeucedanin hydrate (4) and osthol (5) were isolated from the MeOH extract of Angelica genuflexa in the course of searching for anti-platelet and anti-coagulant components.   • Persimmon has been considered to have therapeutic values for various diseases in Korea. Dried persimmon has been applied to wounded parts for anti-inflammatory and analgesic activities. Anti-coagulant fraction from Persimmon stem was purified through gel filtration, phenyl Sepharose, DEAE-Sephadex and additional gel filtration column chromatographies. Its molecular weight was estimated to be 130,000 ∼ 180,000. By element analysis, its main components were C, H, and O. The anti -coagulant was heat- stable and completely inhibited after periodate oxidation, indicating that it was a complex carbohydrate.

  • Acyranthes bidentata has been used as anti-arrthritic, aphrodisiac, anti-viral, anti-spasmodic, anti-hypertensive, anti-coagulant and anti-tumor agent. In this study, we evaluated anti-inflammatory effect of polysaccharide extract from Acyranthes bidentata. Acyranthes bidentata polysaccaride decreased $IL-1{\alpha}$, IL-6, COX-2, and total NO synthesis. Our results point to the potential use of Acyranthes bidentata polysaccaride as a cosmeceutical agent for inflammatory cutaneous symptoms.
