유근피(楡根皮)가 전신적(全身的) 및 국소적(局所的) 아나필락시스에 미치는 효과(效果) (Effect of Ulmi radicis Cortex Extract on Systemic and Local Anaphaylaxis in Rats)

  • Ulmi radicis cortex is a herb medicine which has been used for the treatment of such allergic disease as urticaria, allergic rhinitis and athma. To assess the contribution of an aqueous extract of Ulmi radicis cortex(URC) in systemic anaphylaxis, we used compound 48/80 as a fatal anaphylaxis inducer in rats. URC inhibited anaphylactic shock 100% with a dose of 1.0 mg/g body weight (BW) 1 hr before injection of compound 48/80. URC significantly inhibited serum histamine levels induced by compound 48/80. URC (1.0 mg/g BW) also inhibited to 79.1% passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. URC dose-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. Moreover, URC had a significant inhibitory effect on anti-DNP IgE-induced histamine release or tumor necrosis $factor-{\alpha}$ production from RPMC. The level of cAMP in RPMC, when URC was added, significantly increased compared with that of normal control. These results indicate that URC may possess strong antianaphylactic effect.

알레르기 반응에 미치는 건중탕의 효과 (Effects of Gunjung-tang on Allergic Reaction)

  • Gunjung-tang(GJT) has been reported at Shanghanlun(傷寒論), which has been used for the treatment of general weakness, digestive organ disease and the treatment of allergic disorders in clinical medicine. However, its effect in experimental models remains unknown. In a rat and mouse model, the role of GJT was examined in mast cell-dependent allergic reactions and secretion of inflammatory cytokines. GJT inhibited anti-dinitrophenyl IgE antibody-induced passive cutaneous anaphylaxis reaction by Intraperitoneal pretreatment. GJT inhibited both IL-1β and TNF-α secretion on egg albumin induced allergic tissues and compound 48/80 induced thymus. Furthermore, GJT enhanced Band T lymphocytes proliferation. Our findings provide evidence that GJT inhibits the IgE-dependent allergic reactions and inflammatory cytokines secretion, and enhanced immune response.

Propolis 투여가 Ovalbumin으로 유발된 마우스의 알러지성 천식에 미치는 영향 (Effects of Propolis on the Ovalbumin-Induced Allergic Asthma in Mice)

  • Objective : Propolis (PP) has been used in oriental medicine. PP has various biological activities. However, its immunoregulatory and anti-inflammatory activities have not been well studied. In this study, I investigated these activities of PP by using ovalbumin-induced allergic asthma in mice. Methods : To examine the effect of PP on allergic asthma, mice were sensitized with $100{\mu}g$ of OVA and 1mg of aluminum potasssium sulfate (Alum; Sigma) intraperitoneally on day 0 and 7. On day 14, mice were challenged on consecutive 3 days with 5% OVA and AHR was assessed 24 hours after the last challenge. To examine severity of AHR, I examined the population of eosinophiles and T cells in spleen and lung and cytokine production in T cells. Futhermore, I examined histological changes during the OVA-induced allergic asthma. Results and Conclusion : PP reduced the population of eosinophil and CD4+ T cells on the OVA-induced AHR mice model. PP also inhibited IL-4 production but increased INF-g production in T cells. These results suggest that PP may be beneficial material for allergic asthma.

홍삼유래 ginsenosides Rh1의 단핵구 U937 세포 유착조절 효과 (Regulatory Effect of Ginsenosides Rh1 on Monocytic U937 Cell Adhesion)

  • G-Rh1에 의한 단핵구 세포주인 U937 세포의 유착조절 능을 조사하여 다음과 같은 결론을 얻었다. 1. G-Rh1은 CD29 항체 (MEM101A) 처리에 의해 유도된 세포-세포간 유착현상을 유의적으로 억제하였다. 2. G-Rh1은 fibronectin처리에 의해 유도된 U937 세포-fibronectin간 유착현상을 유의적으로 억제하였다. 3. G-Rh1은 CD29의 세포표면 발현 수준을 유의적으로 감소시켰다. 최근 활발히 G-Rh1의 약리작용 연구들이 진행되고 있다. 특별히, 본 화합물은 항알러지, 피부염증질환 치료효과, 항암효과 및 여성호르몬 유사기능 등이 있는 것으로 보고된 바있다. CD29-매개성 세포유착과정이 다양한 염증과정, 알러지 반응 및 암세포의 이동 및 전이성과 관련이 있다는 관점에서 볼 때 본 연구결과는 이들 약물이 갖는 치료기전의 하나가 CD29 기능저해에서 비롯될 수 있음을 제시한다고 하겠다. 이후 G-Rh1의 억제 기능에 관한 기전 이해를 위해 세포유착 유도 신호전달 단백질의 활성을 포함한 다양한 분자적 수준에서의 추가적인 실험들을 진행하고 한다.

상백피에 의한 MC/9 비만세포의 활성 억제 조절 연구 (Suppressive effects of Morus alba Linne Root Bark (MRAL) on activation of MC/9 mast cells)

  • Objective : Morus alba Linne Root Bark (MRAL) is a medicinal herb in Korean Medicine, known for its anti-inflammatory and anti-allergic properties. However, its mechanisms of action and the cellular targets have not yet been found and the study was developed to investigate the allergic suppressive effect of MRAL. The purpose of this study is to investigate the allergic suppressive effects of MRAL on activation of MC/9 mast cells. Methods : Cytotoxic activity of MRAL (50, 100, 200, 400 ${\mu}g/mL$) on MC/9 mast cells measured using EZ-Cytox cell viability assay kit (WST reagent). The levels of interleukin-5 (IL-5), IL-13 and IL-4, IL-5, IL-6, IL-13 mRNA expression were measured by enzyme-linked immunosorbent assay (ELISA) and real-time PCR respectively. The expression of transcription factors such as GATA-1, GATA-2, NFAT, AP-1 and NF-${\kappa}B$ p65 DNA binding activity were measured by western blot and electrophoresis mobility shift assay (EMSA). Results : Our results indicated that MRAL (50 ${\mu}g/mL$, 100 ${\mu}g/mL$) significantly inhibited PMA/Ionomycin-induced production of IL-5 and IL-13 and the expression of IL-4, IL-5, IL-6 and IL-13 mRNA in MC/9 mast cells. Moreover, MRAL (50 ${\mu}g/mL$, 100 ${\mu}g/mL$) inhibited PMA/Ionomycin-induced GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos protein expression and NF-${\kappa}B$ p65 DNA binding activity in MC/9 mast cells. Conclusions : In conclusion, we suspect the anti-allergenic activities of MRAL, may be related to the regulation of transcription factors GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos and NF-${\kappa}B$ p65 DNA binding assay causing inhibition of Th2 cytokines IL-5 and IL-13 in mast cells.

연교적패독산(連翹敗毒散) 물 추출물(抽出物)의 마우스 Th1/Th2 사이토카인 조절(調節)에 의한 항알레르기 효과 (Effect of Yeongyupaedog-san on Cytokine Levels of Mouse Th1/Th2 Cells and Anti-allergic Activity in Ovalbumin-sensitized Allergic Inflammation Model)

  • This study was to evaluate the effect of Yeongyupaedog-san (YGPDS) on mouse Thl and Th2 cells' differentiation and ovalbumin (OVA)-induced allergic inflammation. The proliferation of mouse CD4 T cells and the secretion of Th1/Th2 cytokines under the influence of YGPDS extract were measured as well as the amount of ${\beta}-hexosaminidase$ in RBL-2H3 cells and the levels of $TNF-{\alpha}$ and 1L-6 secretion in Raw264.7 cells. BALB/c mice were orally administered with YGPDS extract and simultaneously inoculated with OVA to induce allergic reaction and measure the level of total IgE, OVA-specific IgE and the production of IFN- g, IL-4, IL-5 by the spleen cells. When mouse CD4 T cell were stimulated with anti-CD3 and anti-CD28 for 48 hours in various concentrations of YGPDS extract, it increased proliferation of CD4 cells by 11% in $100\;{\mu}g/^{ml}$ concentration but it showed an inhibition by 37% at $200\;{\mu}g/^{ml}$ CD4 T cells under Th1/Th2 polarizing conditions for 3 days with YGPDS resulted in mild decrease of IFN- g in Thl cells and significant decrease of IL-4 in Th2 cells at $500\;{\mu}g/^{ml}\;and\;100\;{\mu}g/^{ml}$ by 18% and 21%, respectively. YGPDS extract had a dose-dependent inhibitory effect on antigen-induced release of ${\beta}-hexosaminidase$ in RBL-2H3 cells. Treatment of YGPDS extract on LPS stimulated Raw 264.7 cells showed dose-dependent decrease in TNF-n production. Oral administration of YGPDS extract on OVA-induced allergic mice showed an inhibitory effect on the levels of total serum IgE and OVA-specific IgE by 25% and 34% , respectively. Culture of spleen cells with OVA resulted in significant increase of IFN- g by 44% and significant decrease of IL-4 and IL-5 by 56%, and 24%, respectively. The results show that YGPDS does not strongly induce mouse T cells to transform into Thl or Th2 but it has an anti-allergic effect in vitro, and that it also corrects the unbalance between the reactions of Th cells in allergic diseases.

불환금정기산(不換金正氣散)이 마우스 Th1/Th2 분화(分化) 및 알레르기 염증 반응 조절에 미치는 효과 (Effect of Bulhwangeumjeonggi-san on Cytokine Levels of Mouse Th1/Th2 Cells and Anti-allergic Activity in Ovalbumin-sensitized Allergic Inflammation Model)

  • This study was to evaluate the effect of Bulhwangeumjeonggi-san (BS) on mouse Th1 and Th2 cells' differentiation and ovalbumin (OVA)-induced allergic inflammation. The proliferation of mouse CD4 T cells and the secretion of Th1/Th2 cytokines under the influence of BS extract were measured as well as the amount ${\beta}$-hexosaminidase in RBL-wH3 cells and the levels of TNF-${\alpha}$ and IL-6 secretion in Raw264.7 cells. When mouse CD4- T cell were stimulated with anti-CD3 and anti-CD28 for 48 hours in various concentrations of BS extract, it increased proliferation of CD4 cells by 14% in 50 ${\mu}g/m{\ell}$ concentration but it showed an inhibition in higher concentrations. CD4 T cells under Th1/Th2 polarizing conditions for 3 days with BS resulted in mild decrease of IFN- g in Th1 cells and mild increase of IL-4 in Th2 cell at 50 ${\mu}g/m{\ell}$ but the level of IL-4 decreased by 18% at 100 ${\mu}g/m{\ell}$. BS extract had a dose-dependent inhibitory effect on antigen-induced release of ${\beta}$-hexosaminidase in RBL-2H3 cells. Treatment of BS extract on LPS stimulated Raw 264.7 cells showed dose-dependent decrease in TNF-${\alpha}$ production. Oral administration of BS extract on OVA-induced allergic mice showed an inhibitory effect on the levels of total serum lgE and OVA-specific lgE by 50% and 55%, respectively. Culture of spleen cells with OVA resulted in significant increase of IFN- g by 25% and significant decrease of IL-4 and IL-5 by 53%, and 38%, respectively. The results show that BS does not strongly induce mouse T cells to transform into Th1 or Th2 but it has an anti-allergic effect in vitro, and that it also corrects the unbalance between the reactions of Th cells in allergic diseases.

유기농 콩 추출물의 항염증 및 항알레르기 효과 (Anti-inflammatory and Anti-allergic Effect of Extractsfrom Organic Soybean)

  • 유기농재배 콩을 다양한 용매에서 추출하여 그 추출물에 대한 항산화 효과, 항염증 효과, 대식세포의 세포사멸 보호 효과, 그리고 알레르기 저해효과를 비교하였다. DPPH 법과 SOD 효소의 활성 측정으로 항산화 능력을 평가하였으며, 항염증 효과는 LPS로 유도시킨 Raw 264.7 세포주에서 염증 매개 단백질인 Cox-2와 iNOS의 발현량을 Western blotting으로 조사하여 분석하였다. 또한 대식세포 보호 효과를 세포사멸 조절 단백질인 Bcl-xl의 발현량조사로 평가하였고, 1mM $H_2O_2$ 처리로 유도되는 세포사멸의 보호효과를 세포 수 측정법으로 조사하였다. 항알레르기 효과는 흰쥐의 비만세포인 RBL-2H3 세포주를 사용하여 IgE로 매개되는 비만세포의 탈과립화를 OPT assay로 측정하여 분석하였다. 그 결과, 유기 재배 콩의 다양한 추출물에서 항산화, 세포사멸 보호, 항염증 효과를 나타내었다. 특히 유기농 콩의 열수 추출물은 LPS로 유도시킨 염증반응을 효과적으로 억제하는 것으로 나타났다. 그러나 항알레르기 효과는 모든 추출물에서 히스타민 유리량이 약간 감소하는 것으로 나타나 유기농 콩의 특별한 효과를 확인할 수 없었다. 콩에는 많은 유용한 생리활성물질 뿐 만 아니라 알레르기 유발 물질도 존재하는 것으로 알려져 있지만 조추출물 상태에서는 특이적인 알레르기 유발보다 오히려 항알레르기 효과가 나타났다. 결론적으로, 만성 염증 및 천식, 그리고 아토피피부염과 같은 염증관련 질환에는 유기 재배된 콩이 좋은 억제 효과를 가지고 있다고 판단된다.

알러지성 비염에 대한 청연산 추출물의 항염증효과 (The Anti-inflammatory Effect of Cheongyeonsan for Allergic Rhinitis)

  • Objectives This study investigated the effects of Cheongyeonsan for allergic rhinitis. Methods First, the GS/MS was used to analyze the effects of Cheongyeonsan by measuring inflammatory markers. Second, 3 groups of 10 6-week-old BALB/c mice were divided into Ctrl (no treatment), ARE (allergic rhinitis-induced without treatment), and CRT (allergic rhinitis-induced after Cheongyeonsan treatment) groups. Ovalbumin (OVA) was used as an antigen to induce allergic rhinitis and sensitization was performed by intraperitoneal injection of 0.1% OVA solution 21, 14, and 7 days before the onset of allergic rhinitis. Allergic rhinitis was induced by dropping OVA solution on the nasal cavity of each mouse for 5 days after the last sensitization. Seven days after the first induction, second induction was introduced by the same method. After making the section, MMP-9, substance P, $TNF-{\alpha}$, $NF-{\kappa}B$ p65, COX-2, iNOS and Nrf, apoptotic cells were observed by Masson trichrome staining, immunohistochemical staining, TUNEL of nasal mucosal tissues of each group. Results GC/MS results showed undecanoic acid. Masson trichrome results showed that the CRT group had less respiratory epithelial damage. Immunohistochemical staining showed CRT group had 58% decrease in MMP-9, 61% decrease in substance P, 55% decrease in $TNF-{\alpha}$, 38% decrease in $NF-{\kappa}B$ p65, 53% decrease in COX-2, 54% decrease in iNOS, 87% increase in Nrf compared to those of the ARE group. TUNEL showed a positive reaction of 84% increase in apoptotic cells greater than that of the ARE group. Conclusions Cheongyeonsan alleviates nasal mucosal damage and reduces inflammatory mediators from allergic rhinitis-induced mice.

甘草麻黃湯 추출물의 배합 비율에 따른 투여가 Ovalbumin으로 유발된 천식 생쥐에 미치는 영향 (The Effects of Gamchomahwang-tang extract According to the ratio of 2 compounds on the Ovalbumin-Induced Allergic Asthma in Mice)

  • Background and Objective : Asthma is a chronic inflammatory disease at the mucosa and is associated with excess production of Th2 cytokine and eosinophil accumulation in lung.Gamchomahwang-tangextract(GME) is one of the well known prescription used in oriental medicine for treating asthma. This study was designed to compare the anti-asthmatic effect of GME according to the ratio of 2 compounds.Methods : To examine the effects of GME on asthma, mice were sensitized with 100 ㎍ of OVA and 1 ㎎ of aluminum potassium sulfate(Alum; Sigma) intraperitoneally on day 1 and 15. From day 22, mice were challenged on 3 consecutive days with 5% OVA. The anti-asthmatic effects of GME were evaluated by enhanced pause(Penh), bronchoalveolar lavage fluids (BALF), inflammatory cytokine production and genes expression, serum IgE production. and histological change in lung tissue. GMEⅠ consists of ES and GU in the proportion 2:1(300 ㎎/㎏ group), GMEⅡ consist of ES and GU in the proprtion 4:1(300 ㎎/㎏ group).Results : GMEⅠ,Ⅱ generally inhibited lung inflammation, inflammatory cells infiltration and cytokine production and gene expression such as IL-4, IL-5 and IL-13 in BALF and serum IgE level. GMEⅡ significantly reduced the cytokine production and gene expression such as IL-4, IL-5 and IL-13 in BALF and GMEⅠ decreased cytokine production of IL-4, IL-13 in BALF and gene expression of IL-4, IL-5 in Lung. GMEⅡ potently inhibited the development of Penh and also reduced the number of eosinophil during OVA-induced AHR(airway hyper-reactivity). Overall the results show that GMEⅡ has more effect on inhibiting production, gene expression of cytokine, serum IgE level and development of Penh than GMEⅠ. Consequently, GMEⅡ might be more effective than GMEⅠ at inhibiting allergic asthma on the OVA-induced mice model.Conclusion : These results indicate that GME has a deep inhibitory effects on airway inflammation and hyperresponsiveness in mice model of asthma and that suppression of IL-4, IL-5, IL-13 expression and decrease of IL-4, IL-5, IL-13 production in BALF might contribute this effect. Hence, the results indicate that GME might be useful herbal medicine of allergic asthma. As a result, GMEⅡ mght be superior to GMEⅠ in the aspect of anti-asthmatic effect on the OVA-induced mice model.
