Objective : The aim of this study is to report the effect of Hani-maehwa laser(fractional mode) treatment on acne scar. Methods : The patient with atrophic facial acne scars was treated with 5 sessions of laser therapy at 4-week intervals. The therapeutic response to treatment was assessed at 3 months after last laser session. The treatment effect was evaluated by standardized photography, anti aging, skin brightness and flatness. That was measured by micro fluorescence measurement camera(ECO SKIN, Cuvitz Inc.) in 16(left zygoma) and 26(right zygoma) spot. And side effects were also checked. Results : Atrophic facial acne scars have improved markedly. Anti aging, skin brightness and flatness were improved. Especially anti aging and flatness were noticeably improved. Adverse effects were not reported.Conclusions : Korean medicine cautery method applies to high level laser(CO2 Hani-maehwa laser). These are different designs but same principle. It can be considered that laser therapy is effective method for treating acne scars.
Tests of stability and toxicity, and clinical evaluation of anti-acne activity suggest that cryptotanshinone, a constituent of the roots of Salvia miltiorrhiza Bunge, is an effective active ingredient for acne vulgaris treatments. Acne vulgaris, called acne or pimples, is the most common disease of the pilosebaceous follicle unit of the skin. It affects nearly 80% of people between the ages of 11 and 30. Approximately 30% of teenagers have acne of sufficient severity to require medical treatment. Acne is a follicular disorder of the skin. It occurs in specialized pilosebaceous units on the face and body. Acne develops when these specialized follicles undergo pathologic alterations that result in the formation of non-inflammatory lesions (comedones) and inflammatory lesions (papules, pustules and nodules). An abnormality of keratinizing epithelium of these follicles, thought to be due to the action of sebum synthesized and secreted by the androgen-sensitive sebaceous glands, leads to inflammation induced by the follicular bacterium Propionibacterium acnes. Therapy involves treatments that modify these pathogenic factors and includes drugs with antikeratinizing, antibacterial and antiseborrheic actions. Acne vulgaris is a very frequent disease, seen primarily in adolescents, involving the sebaceous follicles. Acne vulgaris is characterized by a great variety of clinical inflammatory and non-inflammatory lesions: comedones, papules, pustules, nodules, cysts and scars. Acne vulgaris is a multi-factorial disease. Although its pathogenicity is unclear, extensive studies have shown that hyperseborrhea, superinfection by P. acnes and endocrinologic androgenic changes play a role in the development of acne vulgaris.
본 연구는 우리나라의 야산이나 길가, 들판 등에 흔히 자라는 여러해살이 풀이며, 오래 전부터 민간요법 및 한방에 유용 한 약재로 사용되어 지고 있는 선학초(Agrimonia pilosa Ledeb.)를 이용하여 여드름 유발균에 대한 항균활성을 평가하고, 이를 토대로 항여드름 관련 화장품 소재를 개발하는데 그 목적이 있다. 선학초를 methanol로 추출한 후, 이 추출물을 여드름 유발균주인 P. acne를 이용하여 minimum inhibitory concentration (MIC) 및 paper disk diffusion assay로 항균 효과를 측정하였으며, 추출물에 대한 온도 및 pH 변화에 따른 안정성을 평가하였다. 추출물을 이용하여 P. acne에 대한 MIC 값은 0.005 mg/mL로 측정되었으며, 이는 대조군으로 사용한 항생제인 triclosan의 MIC 값(0.004 mg/mL)과 비교했을 때 시료가 선학초 전초에 대한 crude extract임을 고려하면 매우 항균활성이 높은 것으로 평가되었다. 추출물에 대한 paper disk diffusion assay를 수행한 결과, 대조군으로 사용한 triclosan 처럼 높은 활성을 나타내는 것으로 측정되었으며, Staphylococcus aureus를 비롯한 여러 종의 호기성 균주에도 선학초추출물은 항균활성이 우수한 것으로 평가되었다. 선학초 추출물의 온도, pH 안정성을 평가한 결과, 70, 80, 90, 100, $121^{\circ}C$ 등 다양한 온도 조건과 $pH 2{\sim}pH 11$의 변화에 있어서도 그 효능이 변하지 않고 안정된 상태로 유지되는 것으로 관찰되었다.
본 연구는 배롱나무 꽃(Lagerstroemia indica flower)을 항여드름 화장품 소재로 제안하기 위한 것으로 배롱나무 꽃의 에탄올 추출물(Lagerstroemia indica flower extract, LIFE)을 이용해 피지조절과 항염, 항산화 효능을 확인하였다. 피지조절 효능을 평가한 결과 LIFE가 팔미트산으로 과도하게 유도된 피지생성을 65%까지 억제함을 확인하였다. 이어서 LIFE가 여드름 병변에서 주로 나타나는 Cutibacterium acnes (C. acnes)에 대한 항균작용과 lipase 활성억제능이 있으며, C. acnes로부터 분비되는 독성인자에 의한 염증반응도 감소시킬 수 있음을 확인하였다. 또한 LIFE가 대식세포에서 LPS로 유도된 염증성 매개인자인 일산화질소(NO)와 prostaglandin E2 (PGE2)의 분비를 각각 75%, 54%까지 억제함을 확인하였으며, 아스코르브산과 비슷한 정도의 높은 DPPH 라디칼 소거능 또한 있음을 확인하였다. 이 결과들은 천연 추출물인 LIFE가 피지 생성 조절과 함께, C. acnes에 대한 항균 및 lipase 활성 억제, 항염, 항산화능을 가짐으로써 복합성 질환인 여드름을 완화 및 치료하는 항여드름 소재로 활용될 수 있음을 제시한다.
Leukocytes are reportedly the first line of the innate immune defense and essential for the control of common bacterial infections. Therefore, in this work, the antibacterial activity of crocodile leukocyte extract against Propionibacterium acnes was evaluated, and we also characterized the related activity of skin infection. The leukocyte extract showed the minimum inhibitory concentration to be $100{\mu}g/ml$ to P. acnes. SEM imaging demonstrated that the leukocyte extract adversely affected P. acnes cell permeability in a concentration-dependent manner. Furthermore, the crocodile leukocyte extract could significantly reduce proinflammatory markers and decrease inflammatory signs in infected mouse ears. The crude leukocyte extract was further purified using FPLC and RP-HPLC. The resulting fraction F5 was indicated as the anti-acne peptide-containing fraction. The molecular mass of the peptide contained in F5 was calculated to be 4,790.5 Da. N-Terminal sequencing revealed the amino acid sequence as GPEPVPAIYQ, which displays similarities to immunoglobulin A and leucine-rich repeat neuronal protein. This is the first reported amino acid sequence of a crocodile leukocyte extract that possesses anti-acne activity. To attempt to use it in a prototype cosmetic, an anti-acne gel containing crude crocodile leukocyte extract was formulated, resulting in seven gel formulations (G1, G2, G3, G4, G5, G6, and G7). The formulations G5, G6, and G7 exhibited 2-fold higher anti-acne activity than G1-G4. Investigation of accelerating stability studies of anti-acne gel formulations G5, G6, and G7 demonstrated that a low storage temperature ($4^{\circ}C$) is suitable for maintaining the physical properties and biological activity of the anti-acne gel products.
Propionibacterium acnes infection in skin tissue often causes acne vulgaris, commonly characterized by inflammatory papules, pustules, and nodules. Chitosan and its derivatives possess strong anti-inflammatory effects. In this study, the anti-inflammatory activity of chitosan-phytochemical conjugates on P. acnes-infected human skin keratinocytes (HaCaT) was evaluated. We designed a model of P. acnes-induced inflammation in viable HaCaT cells. Nitric oxide (NO), an inflammatory marker, was successfully elevated by P. acnes infection in HaCaT cells in a dose-dependent manner. Furthermore, the levels of NO were reduced by treatment with chitosan-phytochemical conjugates (chitosan-caffeic acid, -ferulic acid and -sinapic acid) in a dose-dependent manner. Among these conjugates, chitosan-caffeic acid exhibited the strongest NO suppression in HaCaT cells infected with P. acnes. The results obtained in this study suggest that chitosan-phytochemical conjugates could be used as a potential therapeutic agent against acne vulgaris.
피부 여드름 치료, 예방을 위한 천연물의 활성평가를 시도하였다. 335종의 약용 및 야생식물의 다양한 부위로부터 500종의 추출물을 조제하여, 여드름 원인세균인 Propionibacterium acnes에 대한 생육저지 활성을 평가한 결과, 21종 식물체에서 조제한 25종 추출물에서 활성을 확인하였다. 500종 추출물 중, 홀아비꽃대(지상부) 추출물에서 가장 강력한 활성이 나타났으며, 방기, 고삼, 오수유, 은행(씨), 상백피, 독활 및 호장근 추출물에서도 우수한 활성을 확인하였다. 특히 고삼, 상백피, 독활 등의 항여드름 활성은 기존 보고 및 민간요법과도 잘 일치하며, 홀아비꽃대 및 방기 추출물의 항여드름 균 활성은 본 연구에서 처음으로 확인하였다. 고삼을 대상으로 한 활성물질 분리결과, kuraridin과 kurarinone의 주요 활성물질임을 확인하였다. 본 연구결과는, 안정성이 확보된 식물추출물로부터 새로운 피부여드름 치료제 개발이 가능함을 제시하고 있으며, 현재 고삼 추출물 및 활성물질의 세포 독성 및 작용기전에 대한 연구가 진행중이다.
Acne is a relatively common disorder, especially in the adolescent or 20-30th young people. The condition is characterized by comedones, papules and pustules. Acne has social and psychological influence on the acne patients. But there is no perfect treatment of acne. It is known that Herba Houttuyniae has anti-inflammatory action and anti-bacterial action. Therefore, we studied the effect of 5% Herba Houttuyniae extract solution on the acne. The study was designed to compare the efficacy of 5% Herba Houttuyniae solution on the acne. The study was carried out on two groups-a experimental and a control group- for 4 weeks. Efficacy was determined by investigating counts of non-inflammatory open and closed comedones, and inflammatory papules and pustules once a week. After a experiment self-assessment was checked. As the result of the study, we found that a significant difference was observed in the mean counts and self-assessment of the experimental group. So we concluded that Herba Houttuyniae has a clinical effect on the acne.
Herbal mixture water extract of (Chrysanthemum morifolium, Portulaca oleracea, Sanguisorba officinalis, Sophora flavescens, Phellodendron amurense, Cnidium monnieri) which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, and Propionibacterium acne. The growth of Stapylococcus epidermis, and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW 264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 28-60%, and 48-72% , respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW 264.7 cells. The LPS-induced cytokine gene expression including IL-$1{\beta}$, TNF-$\alpha$, and IL-6 were dose-dependently suppressed by the mixture extract. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.
본 실험에서는 여드름 치료에 사용되던 국소 레티노이드제와 항생제를 대용 할 수 있고 화장품에 사용되는 원료로 추출하여 항여드름 치료제 또는 화장품 원료를 개발하기 위해 실험한 결과 broccoli proplyeneglycol 그룹에서 DPPH, superoxide radical, nitric oxide assay 에서 모두 vitamin C에 가까운 활성을 나타내었고 paper disk diffusion test에서는 broccoli ethanol과 broccoli hexane 그룹에서 P. acne의 저해 활성을 나타낸 것으로 보아 항여드름 치료제 또는 화장품 원료로써의 유용 가치가 있다고 사료 되어진다.
