홍마늘 추출물과 녹차, 식이섬유 및 녹차 식이섬유를 혼합한 3종의 복합물(R+T, R+F 및 R+TF)을 이용하여 고지방-콜레스테롤을 급이한 흰쥐에 대해 항비만 효과를 $in$$vitro$ 및 $in$$vivo$에서 측정하였다. 녹차 복합물(R+T) 및 녹차 식이섬유 복합물(R+TF)은 홍마늘 추출물(RG)보다 총 페놀 함량이 1.9~2.0배 높았으며, 콜레스테롤 흡착활성은 9.5~11.5배 높은 활성이었다. 실험군은 6군(정상군, 고지방-콜레스테롤 식이 급이군(HFC), R+T 급이군(HR+T), R+F 급이군(HR+F) 및 R+TF 급이군(HR+TF))으로 나누어 각 추출물을 식이에 1% 수준으로 4주간 급이 하였다. 실험군에서 최종 체중은 대조군에 비해 유의적인 감소를 보였으나, 식이효율은 대조군과 유의차가 없었다. 간장의 중량은 정상군에 비해 대조군이 2.0배 정도 증가되었으며, HR+T군과 HR+TF군에서 유의적으로 감소하였다. 내장지방과 부고환 주변지방 함량은 복합물 첨가군에서 대조군에 비해 유의적으로 감소하였다. 비만도 지수는 대조군에 비해 HR+TF군에서만 유의적으로 감소하였다. 총 지질, 총 콜레스테롤, 중성지질, LDL- 및 VLDL-콜레스테롤 등의 혈중 지질성분과 동맥경화 지수, 심혈관 질환 위험지수는 대조군에 비해 홍마늘 추출물 및 복합물 첨가군에서 유의적으로 감소하였으나, HR+T군, HR+F군 및 HR+TF군간에는 비슷한 수준이었다. GPT 활성은 복합물 첨가군간에 유의차가 없었으나, 홍마늘 추출물 첨가군보다는 유의적으로 낮은 활성이었다. 지질과 산화물 함량은 대조군에 비해 홍마늘 추출물과 복합물 첨가군에서 유의적으로 감소되었다. 항산화 활성은 HR+T군에서 가장 높았다. 따라서 홍마늘 복합물의 체내 지질 개선 및 항비만 효과는 시료 중의 녹차나 식이섬유의 총 페놀 함량 및 콜레스테롤 흡착활성에 기인된다고 생각된다.
Carnitine, hydroxycitric acid, and soy peptide have been known to be anti-obesity agents. The purpose of this study was to evaluate the combined effects of carnitine, hydroxycitric acid, and soy peptide mixture as a potential anti-obesity supplement in overweight women. Overweight premenopausal women (n=33; PIBW>110; 20 to 39 years) were randomized into two groups: the placebo group and the functional beverage group (the test group). Functional beverage was composed of 2000 mg soy peptide, 20 mg L-carnitine and 300 mg garcinia(40% hydroxycitric acid). Body weight and 3 day food dimes, biochemical measurements and computerized tomography were measured at baseline and 8-week. After 8-week consumption of functional beverage with usual diet and exercise, body weight fell an average of 1.4 kg (2.1%). Visceral fat area reduced an average of 7.8% at L1($69.6{\pm}8.7\;vs\;64.2{\pm}7.5\;\textrm{cm}^2$) and 5.1% ($60.7{\pm}4.9\;vs\;57.6{\pm}4.8\;\textrm{cm}^2$, p<0.05) at L4level after weight loss in the test group. Calf fat area in the test group showed about 10% reduction ($31.0{\pm}2.7\;vs=\;27.7{\pm}1.7\;\textrm{cm}^2$, p<0.05) after weight loss. These reductions in fat areas were not shown in the placebo group. There were tendencies of increase in serum levels of $\beta-hydroxybutyrate$, acetoacetate, and total ketones in the test group. There were 7% and 17% insignificant increase in fasting free fatty acid (FFA) and response area of FFA during oral glucose tolerance test(OGTT), respectively, in this group. ill addition, little weight loss in the test group showed 8% but not significant reduction in insulin response area during OGTT. In conclusion, this study shows that taking a mixture of carnitine, hydroxycitric acid, and soy peptide as a potential anti-obesity supplement for 8-week produced advantageous changes in the weight and visceral fat accumulation of overweight women.
본 연구는 대두 부산물인 순물과 침지수가 고지방 식이에 의해 비만이 유도된 마우스의 지방 제거에 미치는 영향을 조사하였다. 침지수가 포함된 일반 사료를 섭이한 실험군의 체중 및 간과 부고환 지방 조직 내 지방 축적은 현저히 적은 것으로 나타났으며, 복부 내 내장 지방과 피하지방도 크게 발달하지 않은 것으로 나타나 침지수는 체내 축적된 지방량을 효과적으로 감소시키는 것으로 조사되었다. 침지수 식이군의 혈중 AST와 ALT의 활성은 대조군 수준으로 유지되었으며, 콜레스테롤 함량과 중성 지질 농도가 감소되는 것으로 나타나 침지수가 고지방 식이로 인한 혈중 효소 활성과 지질농도를 개선하는 것으로 조사되었다. 특히 침지수는 metabolic sensor 단백질인 AMPK와 ACC의 인산화를 촉진시켜 체내 지방산 산화에 영향을 주는 것으로 확인되었다. 또한 침지수는 복부 내 피하지방과 내장지방의 축적을 억제하는 것으로 확인되었다. 따라서 대두 부산물 중 침지수는 비만이 유도된 마우스의 혈중 지질 함량을 개선할 뿐만 아니라 체내 조직의 지방 축적을 완화시키거나 제거하는 데 효과적일 것으로 사료된다.
The world-wide rate of obesity is increasing continuously, representing a serious medical threat since it is associated with a variety of diseases including type 2 diabetes, cardiovascular disease, and numerous cancers. Sophora japonicais used as a traditional herb for medicinal purposes in eastern Asia. However, the anti-obesity effects of S. japonicafruit have not been explored. The aim of this study is to investigate the inhibition of adipocyte differentiation and adipogenesis by an ethanol extract of S. japonicafruit (EESF) in 3T3-L1 pre-adipocytes. Our results demonstrate that EESF suppressed the terminal differentiation of 3T3-L1 pre-adipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet number and lipid content through Oil Red O staining. EESF significantly reduced the accumulation of cellular triglyceride, which was associated with a significant inhibition of the levels of pro-adipogenic transcription factors, including PPARγ, C/EBPα and C/EBPβ. In addition, EESF potentially down regulated the expression levels of adipocyte-specific proteins, including aP2 and leptin. In particular, EESF treatment effectively enhanced the activation of the AMPK signaling pathway; however, the co-treatment with compound C, an inhibitor of AMPK, significantly restored the EESF-induced inhibition of pro-adipogenic transcription factors and adipocyte-specific genes. These results indicate that EESF may exert an anti-obesity effect by controlling the AMPK signaling pathway, suggesting that the fruit extract of S. japonica may be a potential anti-obesity agent.
BACKGROUND/OBJECTIVES: Different fatty acids exert different health benefits. This study investigated the potential protective effects of perilla, olive, and safflower oils on high-fat diet-induced obesity and colon inflammation. MATERIALS/METHODS: Five-week old, C57BL/6J mice were assigned to 5 groups: low-fat diet (LFD), high-fat diet (HFD) and high-fat diet supplemented with-perilla oil (HPO), olive oil (HOO), and safflower oil (HSO). After 16 weeks of the experimental period, the mice were sacrificed, and blood and tissues were collected. The serum was analyzed for obesity- and inflammation-related biomarkers. Gene expression of the biomarkers in the liver, adipose tissue, and colon tissue was analyzed. Micro-computed tomography (CT) analysis was performed one week before sacrifice. RESULTS: Treatment with all the three oils significantly improved obesity-induced increases in body weight, liver weight, and epididymal fat weight as well as serum triglyceride and leptin levels. Treatment with perilla oil (PO) and safflower oil (SO) increased adiponectin levels. The micro-CT analysis revealed that PO and SO reduced abdominal fat volume considerably. The mRNA expression of lipogenic genes was reduced in all the three oilsupplemented groups and PO upregulated lipid oxidation in the liver. Supplementation of oils improved macroscopic score, increased colon length, and decreased serum endotoxin and proinflammatory cytokine levels in the colon. The abundance of Bifidobacteria was increased and that of Enterobacteriaceae was reduced in the PO-supplemented group. All three oils reduced proinflammatory cytokine levels, as indicated by the mRNA expression. In addition, PO increased the expression of tight junction proteins. CONCLUSIONS: Taken together, our data indicate that the three oils exert similar anti-obesity effects. Interestingly, compared with olive oil and SO, PO provides better protection against high-fat diet-induced colon inflammation, suggesting that PO consumption helps manage inflammation-related diseases and provides omega-3 fatty acids needed by the body.
통곡물 시리얼(Whole grain cereal, WGC)이 함유된 식이는 에너지 대사 조절에 중요한 다량영양소(macronutrients)를 제공한다. 본 연구는 고지방식이(high-fat diet, HFD)로 유발된 비만 마우스를 이용하여 WGC의 근감소성 비만 예방 효과에 대해 평가하였다. C57BL/6N 마우스에 정상식이(normal diet, ND), ND+WGC, HFD, HFD+WGC를 12주 동안 제공하였다. WGC는 체중, 식이효율, 체지방 및 지방세포의 크기를 감소시켰다. 또한, WGC는 간 무게 및 간에 축적된 지방을 감소시킴으로써 HFD에 의한 비알코올성 지방간을 개선시켰다. 더욱이, WGC는 비만 마우스 및 정상 마우스의 근육 무게 및 근력을 증가시켰다. 따라서, WGC는 지방 축적을 억제하고 근육량을 증가시키므로 근감소성 비만 예방을 위한 기능성 식품으로 사용될 수 있을 것으로 기대된다.
Euphorbia humifusa Willd (Euphorbiaceae) is a functional raw material with various pharmacological activities. This study aimed to validate the inhibitory effect of Euphorbia humifusa extract (EHE) on adipocyte differentiation in vitro and in a high-fat-diet (HFD)-induced mouse model to evaluate the E.a humifusa as a novel anti-obesity and lipid metabolism enhancer agent. EHE effects on obesity and lipid metabolism were assessed in HFD-induced obese mice after 4-week treatments. Results were compared among four treatment groups (n = 7/group): low fat diet (LFD), high fat diet (HFD), and HFD-induced obese mice treated with either 100 or 200 mg/kg/day EHE (EHE100 and EHE200, respectively). EHE (50 to 200 ㎍/ml) and quercetin (50 ㎍/ml) significantly reduced 3T3-L1 preadipocyte differentiation (p < 0.001), in a concentration-dependent manner. EHE affected lipid metabolism, as evidenced by changes in serum lipid components. The HFD-EHE100 and HFD-EHE200 groups exhibited significantly (p < 0.05) reduced triglycerides (TG, 97.50 ± 6.56 and 82.50 ± 13.20 mg/dL, respectively) and low-density lipoprotein-cholesterol (LDL-c: 40.25 ± 4.99 and 41.25 ± 6.36 mg/dL, respectively) compared to the HFD group (TG: 129.25 ± 19.81 mg/dL; LDL-c: 51.75 ± 11.59 mg/dL). Haematoxylin and Eosin (H&E) and Oil red O staining showed that EHE markedly reduced lipid accumulation and inhibited lipogenesis in the liver. Interestingly, EHE significantly (p < 0.01) reduced the expression of adipogenic transcription factors in liver tissue. Our results indicated that EHE has the potential to be a therapeutic agent for addressing obesity and lipid metabolism.
선별한 신생혈관형성억제제는 EGCG보다도 제어 효과가 나은 천연산물들 중에서 전호, 파고지, 희첨 및 중심으로 그 기전을 밝히고자 하였다. 세포독성은 전호는 0.3 ppm에서 약간의 독성을 나타내었고, 파고지는 10 ppm까지는 독성을 나타내지 않았으며, 희첨 및 산수유는 25 ppm까지 독성을 나타내지 않았다. 세포부착억제작용은 신생혈관의 형성에 중요한 역할을 하기 때문에 cell adhesion 분자인 VCAM-1, ICAM-1 및 E-selectin들에 대하여 ELISA 법으로 살펴보았다. VCAM-1에 대한 천연산물의 작용은 전호 (0.2 ppm, 125%)>파고지 (0.5 ppm, 100%)>희첨 (5.0 ppm, 114%)> 산수유 (5.0 ppm, 111.8%)의 순으로 저해의 강도가 높으며, ICAM-1은 전호 (0.25 ppm, 130%)>파고지 (0.5 ppm, 100%)>희첨 (5.0 ppm, 138%)>산수유 (5.0 ppm, 66.7%)의 순으로 저해되는 것으로 나타났고, 그리고 E-Selectin은 전호 (0.25 ppm, 100%)> 파고지 (1.0 ppm, 128%)>희첨 (5.0 ppm, 120%)>산수유 (5.0 ppm, 100.7%)의 순으로 저해되는 것으로 나타났다. Western blot으로부터, 전호 추출물은 VE-cadherin과 ${\beta}$-catenin의 신호전달을 억제하였으며, 그 하위 신호 전달분자인 Akt도 억제하는 것으로 나타났고, 파고지는 ${\beta}$-catenin의 신호는 억제하지 않는 것으로 보이나, 그 하위그룹의 Akt의 신호전달을 억제하는 것으로 나타났다. 그리고, 희첨은 VE-Cadherin과 하위 그룹의 Akt를 농도의 증가에 따라 확실한 제어를 보이고 있음을 알 수 있었고, 산수유는 하위그룹의 Akt의 신호전달을 억제하는 것으로 나타났다. 4종류의 선택된 천연산물은 세포표면의 신호전달분자 그리고 그 하위 그룹의 Akt를 억제함으로써 NF-kB의 활성을 차단함으로써 신생혈관의 형성을 억제하는 것으로 판단된다. 따라서 4종류의 천연산물 전호, 파고지, 희첨 및 산수유는 전반적으로 낮은 농도에서 세포부착인자의 발현을 억제하였고, 신호전달 분자들을 억제함으로써 신생혈관 형성 억제에 따른 항비만제제로서 충분한 가능성을 보였다.
It has been reported that CLA decreases fat deposition in vivo and in vitro experiments. Among CLA isomers, c9t11 and t10c12 have been shown to exert active biological activities. For example, t10c12 reduces body weight and increases lean body mass, whereas, c9t11 has little effect on body fattness. However, the underlying molecular mechanism for the anti-obesity action of CLA isomers are not well understood. The purpose of this study was to examine the effects of t10c12 and c9t11 on lipid accumulation, cell proliferation, cell death and the expression levels of Ucp genes which are proposed as targets for anti-obesity in 3T3-L1 preadipocytes. Isomers of CLA at 50$\mu$M were added into preadipocyte differentiation medium for 3, 6 and 9days. Control cells received only the vehicle in the differentiation medium. Cytochemical analyses for lipid accumulation, cell proliferation and apotosis were carried out to compare lipidogenesis and cellular activity. RT-PCR analysis of GAPDH, Ucp 2,3 and 4 were also performed to find any modulatory effects of CLA isomers on the metabolic genes. Lipid accumulation indicated by Oil Red-O staining was inhibited in CLA isomers as compared to the control. T10c12 isomer showed less lipidogenesis than c9t11 did. A decrease occurred in CLA isomers as shown by BrdU incorporation. Apotosis has occured at higher level in t10c12 when compared to that of t9c11. Ucp 2, 3 and 4 genes were also upregulated in CLA isomers. T10c12 showed higher level of Ucp gene expressions than the c9t11 did. The biological activities of CLA isomers were also found to be different during differentiation of 3T3-L1 preadipocytes, suggesting that different isomers may be active in certain stage of lipidogenesis. The results indicate that both c9t11 and t10c12 CLA isomers decrease lipidogenesis, inhibit cell proliferation, increase cell death and upregulate in Ucp gene expressions during 3T3-L1 preadipocyte differentiation. T10c12 isomer was more effective than c9t11 in overall anti-obesity activity.
Objectives : Daesiho-tang (DSHT) has been widely used in the treatment of cerebral infarct in traditional medicine. However, there was not report on the anti-obesity-related diseases efficacy of DSHT. In this study, we investigated the effects for the new formulation of DSHT, on the adipocyte differentiation cycle in 3T3-L1 cells. Methods : 3T3-L1 cells were treated with DSHT (50, 100, $200{\mu}g/m{\ell}$) during differentiation for 6 days. Also, the inhibitory effect of DSHT against 3T3-L1 adipogenesis was evaluated in various stage of adipogenesis such as early (0-2day), intermediate (2-4day), and terminal stage (4-6day). The accumulation of lipid droplets was determined by Oil Red O staining. and, the expressions of genes related to adipogenesis were measured by RT-PCR and Western blot analyses. Results : DSHT showed inhibitory activity on adipocyte differentiation at 3T3-L1 preadipocytes without affect cell toxicity as assessed by measuring fat accumulation and adipogenesis. In addition, DSHT significantly reduced the expression levels of several adipocyte marker genes including proliferator activated $receptor-{\gamma}$ ($PPAR-{\gamma}$) and CCAAT/ enhancer-binding $protein-{\alpha}$ ($C/EBP-{\alpha}$). Also, the anti-adipogenic effect of DSHT was strongly limited in the intermediate (2-4 day), terminal stage (4-6 day) of 3T3-L1 adipogenesis. In addition, the DSHT treatment down- regulated mRNA expression levels of $PPAR-{\gamma}$,, $C/EBP-{\alpha}$ in mature 3T3-L1 adipocytes. Conclusions : These results suggest that, the ability of DSHT has inhibited overall adipogenesis and lipid accumulation in the 3T3-L1 cells. The new formulation of DSHT may be a promising medicine for the treatment of obesity and related metabolic disorders.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
