H2O2에 의해 유도된 HDF 세포 손상에 대한 그라비올라 추출물의 항산화 및 세포 보호 효과 (Antioxidative and Cytoprotective Effects of Annona muricata (Graviola) Extract for HDF Cell Damage Induced by Hydrogen Peroxide)

  • 최근 기능성과 친환경 화장품에 대한 관심이 증가하고 있으며, 이에 따라 안전하면서 효능이 우수한 식물 추출물을 활용한 소재 개발이 이루어지고 있는 실정이다. 따라서 본 연구에서도 주로 건강 기능성 소재로써 다양한 효능이 있는 것으로 알려진 그라비올라 추출물이 기능성 화장품 소재로써의 가능성을 확인하고자 하였다. 그라비올라 추출물의 항산화 활성을 확인하고자 총 폴리페놀과 총 플라보노이드 함량, DPPH radical 소거 활성을 측정하였고, HDF 세포에서의 세포 독성을 확인한 후 적정 농도에서 HDF 세포에 과산화수소($H_2O_2$)를 처리하여 산화적 스트레스에 대한 ROS 활성 억제 효과와 세포 보호 효과를 측정하였다. 본 실험 결과, 그라비올라 추출물은 항산화 지표가 되는 총 폴리페놀과 플라보노이드의 100g당 26.6 mg(CA)/100g, 14.3 mg(CA)/100g의 높은 함량을 확인하였으며, 높은 radical 소거 활성을 확인하였다. HDF 세포에 대한 세포 생존율을 측정한 결과, 모든 농도에서 유의한 세포 독성이 나타나지 않았으며, 추후 $100{\mu}g/mL$ 농도에서 실험하였다. $H_2O_2$로 유도된 HDF 세포에 ROS 활성 억제를 측정한 결과, 농도 의존적인 ROS 활성 억제 효과를 확인하였고, $H_2O_2$를 4 시간, 24 시간, 48 시간 동안 처리 후 그라비올라 추출물의 세포 보호 효과를 측정한 결과, $25{\mu}g/mL$ 농도에서 24시간까지 89.92%의 높은 세포 보호 효과를 확인하였다. 이와 같은 결과를 통하여 그라비올라 추출물은 항산화 활성이 우수하고, HDF 세포에 대한 독성이 거의 없으며, $H_2O_2$에 의해 발생하는 활성산소에 대한 효과적인 활성 억제 효과와 세포 보호 효과가 우수한 것으로 확인됨에 따라 항산화 및 세포 보호 효과를 가진 다양한 기능성 소재로서의 가능성을 확인하였다.

로스팅 온도에 따른 녹두의 항산화 활성 변화 및 일반 성분 분석 (Changes in Antioxidative Activities and General Composition of Mung Beans according to Roasting Temperature)

  • 본 연구에서는 녹두 껍질에 주로 존재하는 유용성분의 활용도를 증대시키기 위한 방안의 하나로 녹두를 로스팅 처리하였고, 이때 로스팅 온도에 따른 추출물의 항산화 활성 변화를 측정하여, 녹두의 활용도를 높이기 위한 기초 자료를 제공하고자 하였다. 녹두는 로스터기를 사용하여 볶았으며, 이때 로스팅 온도는 $90^{\circ}C$, $100^{\circ}C$, $110^{\circ}C$, $120^{\circ}C$, 로스팅 시간은 20분간으로 하였다. 로스팅한 녹두는 분말화 하여 일반 성분을 분석하였고, 70% 에탄올로 추출한 시료는 항산화 생리활성 분석 시료로 사용하였다. 대조군으로 로스팅하지 않은 생녹두를 사용하였다. 생녹두를 포함한 온도별 로스팅 녹두 시료들의 pH는 6.1에서 6.3사이였다. 환원당 함량의 경우, 로스팅 온도가 높아질수록 환원당 함량이 낮아지는 경향을 보여, 생녹두(3.14 mg/mL)의 환원당 함량이 가장 높았다. 일반성분은 수분을 제외한 나머지 성분들이 로스팅 온도가 증가할수록 증가하는 경향을 보여주었다. 추출물의 추출 수율은 로스팅 온도가 올라갈수록 증가되었다. 총 페놀 함량, 총 플라보노이드 함량, DPPH 법, ABTS 법에 의한 free radical 소거능 및 xanthine oxidase 저해 활성은 로스팅 온도가 높아질수록 증가하다가 $120^{\circ}C$ 조건에서는 오히려 감소하는 경향을 보여주었다. 결론적으로 녹두를 로스팅할 경우, 일정 수준까지의 열처리($110^{\circ}C$)는 항산화 생리활성이 증진되었으나, 그 보다 과한 열처리($120^{\circ}C$) 조건에서는 오히려 활성이 감소되었으므로, 녹두의 최적 로스팅 조건은 $110^{\circ}C$에서 20분으로 생각된다.

금은화 수용성 추출물의 LPS 유도 염증매개물 억제 효과 (Inhibitory Effect of Aqueous Extract from Lonicera japonica Flower on LPS-induced Inflammatory Mediators in RAW 264.7 Macrophages.)

  • Objective : Lonicera japonica (Caprifoliaceae) has long been used for treatment of infectious diseases in oriental countries. The aim of this study was to investigative the effect by which the aqueous extract from flower of L. japonica (LJFAE) inhibited the lipopolysaccharide (LPS)-induced inflammatory mediators in murine macrophages, RAW 264.7 cells Methods : The dried flowers of L. japonica were extracted with distilled water at $100^{\circ}C$ for 7 h. The extract was filtered through 0.45 ${\mu}m$ filter, freeze-dried. The dried extract was dissolved in Hank's balanced salt solution (HBSS) and filtered through 0.22 ${\mu}m$ filter before use. Accumulated nitrite, an oxidative product of nitric oxide (NO), was measured in the culture medium by the Griess reaction. The levels of prostaglandin E2 (PGE2), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-1$\beta$ (IL-1$\beta$), and IL-6 production, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were measured by enzyme-linked immunosorbent assay and Western blot analysis. Results: LJFAE (10-400 ${\mu}g$/ml) per se had no cytotoxic effect in unstimulated macrophages, but LJFAE concentration-dependently reduced NO, PGE2, TNF-, IL-l, and IL-6 production and COX-2 activity caused by stimulation of LPS. The levels of iNOS and COX-2 protein expressions were markedly suppressed by the treatment with LJFAE in a concentration dependent manner. Conclusions : These results suggest that LJFAE suppress the NO and PGE2production in macrophages by inhibiting iNOS and COX-2 expression and these properties may contribute to the anti-inflammatory activity of Lonicera japonica.

캐모마일, 세이지, 녹차 건분의 섭취가 노령흰쥐의 항산화능에 미치는 영향 (Effect of Dried Powders of Chamomile, Sage, and Green Tea on Antioxidative Capacity in 15-Month-Old Rats)

  • This study was performed to investigate the effect of chamomile, sage, and green tea intakes on anti oxidative capacity in 15-month-old rats. Dried powders of three plants were analyzed to determine the amount of total flavonoids, $\beta$-carotene, vitamin C, vitamin E, and dietary fibers. In order to examine the change of antioxidative capacity in old rats, forty-eight Sprague-Dawley male rats weighing 621.2$\pm$9.5 g were divided into four groups according to body weight and fed for four weeks with each experimental diet of three dried powders and control diet. Plasma and liver thiobarbituric acid reactive substances (TBARS) levels and xanthine oxidase (XO) activities, erythrocyte and liver superoxide dismutase (SOD) activities and plasma vitamin A, C, E, and total carotenoids levels were measured. The total flavonoids and vitamin C contents were the highest in green tea powder. Beta-carotene and vitamin E contents were not significantly different among all three dried powders. Total dietary fiber contents also were not different among all three dried powders, but soluble dietary fiber contents of chamomile was higher than other two dried powders. Plasma TBARS level was found to be significantly lower in all the experimental groups as compared to control group. Chamomile powder group, especially, showed the lowest level among all experimental groups. Liver TBARS levels of experimental groups were also lower than that of control group and significant differences were observed in chamomile and green tea groups compared to control. Plasma XO activity of green tea group was significantly lower than control group. Liver XO activities of green tea and chamomile groups were significantly lower than control group. Erythrocyte SOD activity was not significantly different among all the groups. However, liver SOD activities of sage and green tea groups showed a significant increase as compared to control group. Plasma vitamin A level was not significantly different among all the groups. Plasma total carotenoids levels were found to be significantly higher in experimental groups as compared to control group. Plasma vitamin E level of chamomile group and vitamin C level of green tea group showed the highest level among all the groups. In conclusion, chamomile, sage, and green tea intakes had an effect on improving antioxidative capacity in 15-month-old rats. Effects of green tea and chamomile powders, especially, were higher than sage powder.

질경이의 영양성분 분석과 항산화 활성 (Component Analysis and Antioxidant Activity of Plantago asiatica L.)

  • 본 연구는 질경이의 기능성식품 및 화장품 소재로서의 이용가능성을 조사하기 위해 질경이의 영양성분 분석을 통한 식품영양학적 접근, 생리활성 기능을 기대할 수 있는 관련 물질 함량의 분석하였다. 식품영양학적 접근에서의 질경이의 일반성분은 건량기준으로 탄수화물 63.71%, 조단백질 18.75%, 조지방 1.67% 및 조회분 6.48%이었고, 질경이 100 g의 함유 열량은 466.71 Kcal로 분석되었으며, 총 식이섬유소 함량은 건량기준으로 22.68%로 나타났다. 또한, 필수아미노산과 비필수아미노산 함량은 각각 4.815.22 mg, 6.591.04 mg이었고, 무기질 중 캄륨의 함유량이 가장 높았고 그 다음이 칼슘, 마그네슘, 인으로 나타나 알칼리성 재료임을 알 수 있었으며, 지방산 함량의 경우 불포화지방산의 함량이 높은 것으로 나타났다. 질경이의 총 페놀 함량은 $79.65{\mu}g/g$, 플라보노이드 함량은 $4.43{\mu}g/g$으로 분석되었으며, DPPH 소거 활성을 농도별로 측정하여 비교한 결과 $500{\mu}g/mL$ 농도에서 70.30%로 높은 활성을 보였다. 또한, 환원력의 경우에는 질경이 추출물이 25 mg/mL 농도에서 3.5로 높은 환원력을 보였다. 이상의 결과로부터 질경이 물추출물은 항산화능이 있음을 확인할 수 있었다. 따라서 질경이를 식품 내 첨가물로서, 혹은 다른 약용으로의 활용 가능성을 제시하고 있다.

수오연수단(首烏延壽丹)이 노화유발 흰쥐의 항산화능에 미치는 영향 (Effect of Suoyounsoodan(首烏延壽丹) on Antioxidant Capacity in G-galactose induced Aging Rats)

  • Objectives : Suoyounsoodan(首烏延壽丹) composed of Polygonum multiflorum THUNB. and some medical herbs is known as formula of senescence delay effect The purpose of this study is to investigate the effect of Suoyounsoodan(首烏延壽丹) on antioxidant enzyme activity such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase(GSH-px) in rat plasma and liver. Methods: Sprague-Dawley rats divided into 4 groups, Young group(8 weeks old, N-8), Aging group(18 weeks old, N-18), pathologically induced aging group(injected D-galactose 50mg/kg, 1time/day for 6 weeks, CON) and Suoyounsoodan(首烏延壽丹) administered group(D-galactose 50mg/kg and Suoyounsoodan extracts 840.0mg/kg 1time/day for 6 weeks, SOY). Rats were sacrificed and TBARS, SOD, CAT, and GSH-px were mesured in rat plasma and liver. Results: Plasma and liver TBARS concentrations of SOY group was sinificantly lower than that of control. Red blood cell(RBC) SOD activities of SOY group was increased(F=3.405, p=0.034, ANOVA test), and RBC catalase activities of all experimental groups were not significantly different. RBC GSH-px activities of SOY group was increased(F=9.261, p=0.0001, ANOVA test). Liver SOD activities of SOY group was higher than that of control(F=3.806, p=0.023, ANOVA test). Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of SOY group was significantly higher than that of control(F=3.572, p=0.029, ANOVA test). Conclusions: According to the above results, It is considered Suoyounsoodan is effective in inhibiting lipid peroxidation and increasing anti oxidative enzyme activities in D-galactose induced aging rat.

Effects of Tributyrin on Intestinal Energy Status, Antioxidative Capacity and Immune Response to Lipopolysaccharide Challenge in Broilers

  • This study was carried out to investigate the effects of tributyrin (TB) on the growth performance, pro-inflammatory cytokines, intestinal morphology, energy status, disaccharidase activity, and antioxidative capacity of broilers challenged with lipopolysaccharide (LPS). A total of 160 one-day-old Cobb broilers were allocated to 1 of 4 treatments, with 4 replicated pens per treatment and 10 birds per pen. The experiment consisted of a $2{\times}2$ factorial arrangements of treatments with TB supplementation (0 or 500 mg/kg) and LPS challenge (0 or $500{\mu}g/kg$ body weight [BW]). On days 22, 24, and 26 of the trial, broilers received an intraperitoneal administration of $500{\mu}g/kg$ BW LPS or saline. Dietary TB showed no effect on growth performance. However, LPS challenge decreased the average daily gain of broilers from day 22 to day 26 of the trial. Dietary TB supplementation inhibited the increase of interleukin-$1{\beta}$ (in the jejunum and ileum), interleukin-6 (in the duodenum and jejunum), and prostaglandin $E_2$ (in the duodenum) of LPS-challenged broilers. Similar inhibitory effects of TB in the activities of total nitric oxide synthase (in the ileum) and inducible nitric oxide synthase (in the jejunum) were also observed in birds challenged with LPS. Additionally, TB supplementation mitigated the decrease of ileal adenosine triphosphate, adenosine diphosphate and total adenine nucleotide and the reduction of jejunal catalase activity induced by LPS. Taken together, these results suggest that the TB supplementation was able to reduce the release of pro-inflammatory cytokines and improve the energy status and anti-oxidative capacity in the small intestine of LPS-challenged broilers.

발효 스쿠알렌을 함유한 마이크로좀의 제조 및 효능효과 (Effectiveness and Preparation of Microsome containing Fermented Squalene)

  • 본 연구에서는 미생물을 이용하여 개발한 발효 스쿠알렌의 안정성 향상을 위하여 마이크로좀-SQ20 (Microsome-SQ20)을 제조하고, 이에 대한 물리적 거동 및 특성과 효능효과에 대하여 연구하였다. 마이크로좀-SQ20의 외관은 투명한 액체로 고유의 냄새를 가지고 있었다. 색상은 무색이고, 비중은 0.928, pH는 5.82 (20%용액)로 화장품에 사용하는데 적합한 나노에멀젼이 형성되었다. 스쿠알렌의 주성분 함량은 20.05%로 안정하게 봉입이 된 것을 확인할 수 있었다. 마이크로좀-SQ20을 0.1% 수용액화하여 측정한 입자크기는 134.8 nm로 bluish한 유화상을 얻었다. F-SQ(Fermented squalene)과 MF-SQ(Microsome squalene)의 DPPH라디칼에 의한 항산화 효과는 각각 80.72%, 81.5%로, L-ascorbic acid와 비교하여 동등한 효과를 보였다. F-SQ 및 MF-SQ은 10 ppm에서 각각 150.3%, 129.9%의 세포생존율을 나타냈다. SQ, F-SQ, MF-SQ의 elastase 저해능은 10 ppm에서 각각 8.7%, 10.33%, 8.7%의 저해능력을 가지고 있음을 알 수 있었다. MMP-1 저해능력은 SQ, F-SQ, MF-SQ 모두 10 ppm에서 각각 1.55%, 41.44%, 31.79%를 나타내 F-SQ의 collagenase 저해능이 우수한 것을 알 수 있었다.

슈퍼자미의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidant Activity of Superjami)

  • This study attempted to investigate the characteristics of Superjami, which had a high C3G (cyandin-3-glucoside) content by comparing it with common rice (Ilpum) and black rice (Heougjinju, Suwon 415) for their components and physio-chemical characteristics. There were no significant differences in the water content, however there were significant differences in crude protein and crude fat in the order of Ilpum < Superjami < Heougjinju. As for the morphological characteristics of grains, it had been learned that Superjami was heavier and longer than the common rice, moreover Superjami had bigger seeds. As for the hardness of grains, the longer the time of steeping, the more decreased their hardness was at large. In the case of color, they showed significant differences in all the L, a, and b values of grains and flours. As a result of the experiment of pigment elution after steeping of the rice flours at $20^{\circ}C$ and $40^{\circ}C$, the longer the time steeping, the more increased the pigment elution became. The values are bigger with Heougjinju and Superjami than with Ilpum. As for the elution pH after rice steeping, the longer the time of steeping, the lower pH all the assays tended to have. As a result of the analysis of the total polyphenol contents of Ilpum, Heougjinju, and Superjami, it had turned out that the total polyphenol contents of Heougjinju and Superjami are 1.2 times as high as Ilpum, a common rice, and that Superjami is significantly higher than Heougjinju. As a result of the experiment of DPPH radical scavenging ability, there are significant differences among the assays in the order of Superjami > Heougjinju > Ilpum. Futher, it has turned out that Superjami has a higher DPPH radical scavenging ability than Heougjinju. Consequently, it can be stated that Superjami has a strong anti-oxidative ability. Thus, we should more precisely grasp the cooking characteristics of Superjami, which is in the state of brown rice, via comparing it with a common brown rice, and also provide opportunities to apply Superjamii to more foods by studying its starch characteristics in addition to its grain and flour properties.

In-vitro와 In-vivo에서 산수유의 남성갱년기 개선효과 (Effect of Corni Fructus on Testosterone Deficiency Syndrome in In vitro and In vivo)

  • This study was carried out to evaluate the preventive effect of the Corni Fructus (SSU) 50 % EtOH extract (SSU-E50) against bisphenol A (BPA) toxicity in Leydig cells and improving testosterone deficiency syndrome in orchidectomized Sprague-Dawly (SD) rats. Antioxidant properties were measured by radical scavenging activity of SSU-E50 in ABTS assay and DPPH assay. Also, real-time polymerase chain reaction(real-time PCR) was performed to quantify the mRNA expression levels of antioxidant enzyme. SD rats were divided into eight group: normal, sham operation (Sham), orchidectomized (ORX), ORX treated with testosterone 1 mg/kg (Tes. 1), ORX treated with SSU water extract 100 mg/kg (SSU-A 100) and 300 mg/kg (SSU-A 300), ORX treated with SSU 50 % EtOH extract 100 mg/kg (SSU-E 100) and 300 mg/kg (SSU-E 300). On a comparative basis, the SSU showed better activity quenching ABTS with an IC50 value of 0.29 mg/ml and DPPH with an IC50 value of 0.33 mg/ml. Cell viability was evaluated by MTS assay as described not cytotoxic at the highest concentration of $500{\mu}g/ml$. Cytotoxicity of BPA showed in $200{\mu}M$, but definitely survived by treatment with SSU in Leydig cells. In addition, SSU increased the mRNA expression levels of antioxidant enzyme in BPA induced Leydig cells. Superoxide dismutase (SOD) level was slightly increased and malondialdehyde (MDA) level was decreased with SSU-A 100 in in-vivo. These results suggest that Corni Fructus extracts have the greatest property as a natural anti-oxidative and improves testosterone deficiency syndrome source.
