Antioxidant and anti-inflammatory activities of water extracts and ethanol extracts from Portulaca oleracea L. (쇠비름 물, 에탄올 추출물의 항산화 및 항염증 활성)

  • Portulaca oleracea L., a species of Portulacaceae, is ubiquitous. It is a well-known traditional Chinese medicine for removing heat, counteracting toxicity, cooling blood, and maintaining hemostasia; it is also used as antidysentery agent. This study investigated the anti-oxidative and anti-inflammatory activities of water and ethanol extracts from P. oleracea. The total polyphenol content ($21.08{\pm}0.03mg\;GAE/g$) and total flavonoid content ($5.45{\pm}0.76mg\;QE/g$) of the ethanolic extracts were higher than those of the water extracts. The antioxidative activities were determined by evaluating the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activity and by the ferric reducing antioxidant potential (FRAP) assay. The ABTS radical scavenging activity of the water extract (75.53%) was higher in those of the water extract (67.03%) at concentration of $1,000{\mu}g/mL$. The DPPH radical scavenging activity and FRAP of the ethanol extract were higher than those of the water extract. We also investigated the anti-inflammatory activity of the P. oleracea extracts in LPS-stimulated Raw 264.7 cells. The production levels of nitric oxide (NO) and reactive oxygen species (ROS) significantly decreased with an increasing concentration of the extract. The expression levels of pro-inflammatory cytokines (tumor necrosis faction (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, and IL-6) were significantly lower in the ethanol extract than in the LPS alone treatment group. Based on these results, ethanolic extract from P. oleracea could be an effective antioxidant and anti-inflammatory agent.

Physiological activities of leaf extract of Lonicera morrowii A.Gray, a plant native to Ulleungdo (울릉도 자생식물인 섬괴불나무(Lonicera morrowii A.Gray) 잎 추출물의 생리활성)

  • This study was performed to investigate total phenolic contents, anti-oxidant, biological disease-related enzyme inhibitory, and anti-microbial effects of extracts of Lonicera morrowii leaves prepared with water and 40% ethanol. Anti-oxidative activities of the extracts increased in a dose-dependent manner and were very high even at low phenolic concentration. At phenolic concentrations ranging between 50 and 200 ㎍/mL, the water and ethanol extracts inhibited 39.34-76.35 and 47.53-99.83% of xanthine oxidase activity, 30.21-79.06, and 59.40-87.14% of angiotensin converting enzyme activity, and 59.81-80.35 and 36.06-63.58% of α-glucosidase activity, respectively. All inhibitory effect were increased in a dose-dependent manner. In the paper disc agar diffusion assay against seven microbes, L. morrowii extracts had anti-bacteria activities against Helicobacter pylori, Propionibacterium acne but no effect against other bacteria. These results indicate that the extracts from L. morrowii leaves have the possibility to be developed as a physiologically functional source for prevention of adult diseases.

Antioxidant potential of buffalo and cow milk Cheddar cheeses to tackle human colon adenocarcinoma (Caco-2) cells

  • Objective: The aim of present study was to assess the anti-oxidant potential of water-soluble peptides (WSPs) extract derived from buffalo and cow milk Cheddar cheeses at different stages of ripening. Methods: The antioxidant potential of WSPs extract was assessed through 2,2'-azinobis-3-ethylbenzothiazoline-6sulfonic acid (ABTS)-radical scavenging activity. In addition, impact of WSPs extract on cell viability and production of reactive oxygen species (ROS) in human colon adenocarcinoma Caco-2 (tert-butylhydroperoxide-induced) cell lines was also evaluated. Results: The ABTS-radical scavenging activity increased progressively with ripening period and dose-dependently in both cheeses. However, peptide extract from buffalo milk Cheddar cheese demonstrated relatively higher activity due to higher contents of water-soluble nitrogen. Intracellular ROS production in Caco-2 cells decreased significantly (p<0.05) till 150th day of cheese ripening and remained constant thereafter. Additionally, dose-dependent response of WSPs extract on antioxidant activity was noticed in the Caco-2 cell line. Conclusion: On the basis of current in vitro study, the Cheddar cheese WSPs extract can protect intestinal epithelium against oxidative stress due to their antioxidant activity.

Antioxidant Activities of Methanol Extracts from Root Parts of Korean Salad Plants

  • Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts ($319.2\;mg\;kg^{-1}$) as the greatest component, and followed by A. lappa ($96.3\;mg\;kg^{-1}$) and Y. sonchifolia ($22.9\;mg\;kg^{-1}$). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg $100\;g^{-1}$) and followed by Y. sonchifolia (11.2 mg $100\;g^{-1}$) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg $100\;g^{-1}$ exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity ($r^2$ = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.

Antioxidant Activity of Ethyl acetate Fraction of Oat in Caenorhabditis elegans (귀리 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과)

  • Oat, the seeds of Avena sativa L. (Gramineae), is an important dietary staple for people in many countries. Previous studies reported that A. sativa had various pharmacological effects such as anti-inflammatory, antitumor, neurotonic, and antispasmodic activities. In this study, Caenorhabditis elegans model system was used to investigate the antioxidant activity of methanol extract of oat. The ethyl acetate soluble fraction of the oat methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using GFP-expressing transgenic worm. As a result, the ethyl acetate fraction increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity compared to the control.

Changes in Antioxidant Activity and Total Phenolic Content of Water Spinach (Ipomoea aquatic Forsk.) under In Vitro Biomimicking System

  • The purpose of current study was to examine bioaccessibility of antioxidant activity and total phenolic content in each part of water spinach (Ipomoea aquatic Forsk.). In vitro biomimicking system simulated human digestive fluid was employed in order to measure bioavailable anti-oxidative effect and phenolic content. Antioxidant activity and total phenolic content was measured by using the DPPH method and the Folin-Ciocalteu assay, respectively. Stem of water spinach had a higher DPPH free radical scavenging effect (5.43 mg/mL for $IC_{50}$) than leaf (5.95 mg/mL for $IC_{50}$), while leaf had a greater level of total phenolic content (287.45 ${\mu}g$ GAE/mL) than stem (216.45 ${\mu}g$ GAE/mL). Bioaccessible antioxidant capacity and digestive stability of total phenolic content showed a similar pattern to what found in raw materials. Our result also indicated that total phenolic content was not found to be a major marker for prediction of antioxidant activity. It is plausible that other constituents such as vitamin E and C in water spinach could be contributors for antioxidant activities.

Antioxidant Activity of Isolated Compounds from the Shoot of Aralia elata Seem (두릅 순에서 분리된 화합물의 항산화 활성)

  • Objectives : This study was performed to investigate the antioxidant activities of isolated compounds from the shoot of Aralia elata. Methods : The methanol extract from the shoot of Aralia elata was fractionated into ethyl acetate, n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel, ODS column chromatography of n-BuOH layer afforded four flavonol glycosides. Their antioxidant activity was determined by measuring free radical scavenging activity by DPPH, ABTS and superoxide dismutase (SOD) like activity assay. Results : They were identified as quercetin 3,7-di-O-$\alpha$-rhamnopyranoside (1), quercetin 3-O-$\beta$-D-galactoside-7-O-$\alpha$-L-rhamnoside (2), kaempferol 3-O-$\beta$-glucosyl($1{\rightarrow}2$)-$\alpha$-rhamnoside-7-O-$\alpha$-rhamnoside (3) and quercetin 3-O-$\beta$-glucosyl($1{\rightarrow}2$)-$\alpha$-rhamnoside-7-O-$\beta$-rhamnoside (4) on the basis of spectroscopic data. The result showed that 1 is the most active compound in the DPPH and ABTS radical scavenging test. Conclusions : Isolated Compounds from the shoot of Aralia elata showed anti-oxidative effect.

Alcohol-induced Hyperlipidemia Is Ameliorated by Orally Administered DWP208, a Sodium Succinate Form of ZYM201

  • DWP208 is a sodium succinate form of ZYM-201 which is a triterpenoid glycoside isolated from Sanguisorba officinalis, a medicinal plant prescribed for various diseases, such as duodenal ulcers and bleeding in East Asian counties. We demonstrated that this compound is able to normalize the altered lipid metabolism induced by hyperglycemia and a high fat diet. In this study, we determined whether hyperlipidemic conditions induced with chronically treated alcohol can also be restored by DWP208. Similar to our previous results, orally administered DWP208 (1 to 10 mg/kg) also ameliorated the hyperlipidemia that was induced by alcohol. This compound reversed the alcohol-induced hyperlipidemia including (i) up-regulated hyperlipidemic parameters such as low-density lipoprotein (LDL), very low-density lipoprotein (VLDL), atherosclerotic index (AI), triglyceride, and total cholesterol, and (ii) down-regulated hyperlipidemic parameters such as absolute body weight, superoxide dismutase (SOD) activity, and high-density lipoprotein (HDL) in serum and liver. According to our data, the ameliorative activity of DWP208 is due to its indirect anti-oxidative activity as a result of which lipid peroxide and hydroxyl radical levels were reduced and the activity of SOD was enhanced. Therefore, our data strongly suggest that DWP208 can be used as a remedy against alcohol-induced hyperlipidemia.

Study on the Anti-oxidative Activity and Anti-inflammatory Effects of Processed Sulfur with Cordyceps Militaris Mycelium (동충하초 균사체를 이용한 법제 유황의 항산화활성 및 항염증 효과)

  • The purpose of this study was evaluated physiological activity of processed sulfur with Cordyceps militaris mycelium that antioxidative and antiinflammatory effects. Proliferation of processed sulfur (PS) with Cordyceps militaris mycelium was increased in dose-dependent manner. In organic sulfur contents of Cordyceps militalis mycelium fortified processed sulfur, CM+PSH (CM+3000 ppm of PS) was significantly higher than other groups. However, CM+PSL (CM+1500 ppm of PS) was almost changed organic sulfur. Content of total polyphenol compounds was similarity to CM, CM+PSL and CM+PSH. The EDA (electron donating ability) and SOD-like activity was increased in dose-dependent manner and the activity of CM were significantly higher than CM+PSL and CM+PSH. We examined cytotoxicity, nitric oxide production of Raw 264.7 cell and inhibition of HT 1080 cell by MTT assay. CM, CM+PSL and CM+PSH do not have any toxic effects in macrophages (Raw 264.7). And CM+PSL and CM+PSH inhibited the production of nitrite in Raw 264.7 cells activated with LPS. The antitumor effects of processed sulfur with Cordyceps militaris mycelium on HT 1080 cell was indicated a significantly inhibition activity. These results suggested that processed sulfur with Cordyceps militaris mycelium have activities of antioxidant, antiinflammatory effects.

Synergistic Effect of Methanol Extract from Kalopanax pictus and Ascorbic Acid on Antioxidant, Anticancer and Immunomodulatory Activities (음나무(Kalopanax pictus) 추출물과 비타민 C의 항산화, 항암 및 면역활성 상승효과)

  • The 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS), nitric oxide (NO) scavenging activities and ferric- reducing/antioxidant power (FRAP) assay against extracts of Kalopanax pictus (KP) were measured. Radical scavenging and antioxidant activities were increased depend on the concentration and the effects were enhanced by ascorbic acid (AA). KP extracts and AA had a good anti-proliferating activity against HepG2 cells by MTT assay and induces cells apoptosis, which was demonstrated by flow cytometric analysis. KP extracts and AA caused the arrest of cell-cycle progression at either G0-G1-phase or G2/M-phase, which might be depending upon the KP extracts concentration. In addition, KP extracts and AA are effective in enhancing immunity and nitric oxide production by RAW 264.7 macrophages cells. KP extracts and AA inhibited tumor cell growth and exerted antioxidant effects as compared to controls. These results demonstrate that simultaneous AA and KP extracts treatment could be useful in preventing the oxidative damage and anti-proliferating HepG2 cells, and are effective in enhancing immunomodulatory and antioxidant activity.
