We studied to know the anti-inflammation effect on water extracts of Lophatheri Herba which was growing in every places in our country. We objected free radical scanvenger effect and nitrite eliminate effect of the Lophatheri Herba water extracts, and the cell viabillity, the effects of Lophatheri Herba water extracts on NO production, iNOS synthesis induced by LPS. Free radical scavenger effects were $27.91{\pm}0.12%$, $38.96{\pm}0.10%$, $46.22{\pm}0.15%$ depend on 0.5, 1.0, 2.0 mg/ml each dose of Lophatheri Herba water extracts. Nitrite eliminate effects were $9.86{\pm}0.3%$, $80.61{\pm}0.23%$, $97.62{\pm}0.56%$ in 0.1, 1.0, 2.0 mg/ml Lophatheri Herba water extracts on pH 1.2. NO production and iNOS synthesis induced by LPS were reduced in RAW 264.7 cell by Lophatheri Herba water extracts. As the above results, Lophatheri Herba water extracts have anti-inflammation effects via NO production decrease, iNOS synthesis decrease mechanism. So Lophatheri Herba water extracts will be used as the protection or treatment in chronic inflammation desease like a asthma, stomatitis etc.
Sophorae Flos (SF), a composite of flowers and flower-buds of Sophora japonica, has long been used in traditional Korean and Chinese medicines for the treatment of hemostasis and inflammation. In this study, we investigated anti-inflammatory effect of four EtOH extracts at the difference in blooming stages of flowers on LPS-induced inflammation in RAW264.7 cells. We classified the flowers of Sophora japonica with SF-1 (length of flower is shorter than calyx), SF-2 (length of calyx is shorter than flower), SF-3 (full bloom), and SF-4 (not blooming at all). We examined HPLC analysis, whether quercetin and rutin are major component of these Sophorae Flos extracts or not. As a result, SF-1 contained quercetin, but the others did not. In addition, quercetin, SF-1, and SF-4 act on the suppression of pro-inflammatory mediators including inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, nitric oxide (NO), prostaglandin E2 ($PGE_2$) against lipopolysaccharide (LPS)-induced activation in RAW264.7 cells. Of these, SF-1 showed the best anti-inflammatory effect. These results suggest that Sophorae Flos with the highest content of quercetin would be used for the treatment of various inflammation diseases.

Fructan, a polysaccharide existing in plants or produced by microorganisms, is a sugar polymer of fructose with $\beta$-2,6 linkages. In this study, we investigated some cosmeceutical properties of Fructan such as moisturizing effect, cell proliferation effect, anti-inflammation effect and cell cytotoxicity. Zymomonas mobilis, a microorganism producing Fructan, was cultured in a medium containing 10% sucrose and 2% yeast extract as main components for 24 hours at 37$^{\circ}C$ and pH 7. Fructan was obtained by precipitation from the cultured medium by adding alcohol (alcohol ratio of 1:3) after removing the enzyme by centrifuging. Fructan exhibited almost same moisturizing effect as hyaluronic acid and cell proliferation effect on human fibroblast and keratinocyte as well. Moreover, on cell proliferation test on bio-artificial skin constructed by 3-dimensional(3-D) culture after inducing primary skin inflammation with 0.5% sodium lauryl sulfate (SLS), the 3-D artificial skin treated with 0.01 mg/ml, 0.05mg/ml of Fructan exhibited higher cell proliferation than the 3-D artificial skin treated with SLS only. On anti-inflammation test on 3-D artificial skin evaluated by measuring secreted quantity of interleukin-1$\alpha$ (IL-1$\alpha$) which is a pre-inflammatory mediator induced by SLS, the quantity of IL-1$\alpha$on the 3-D artificial skin treated with 0.01 mg/ml, 0.05mg/ml of Fructan was less than the one on the 3-D artificial skin treated with SLS only. As a result of these studies, Fructan has anti-inflammation effect against inflammatory reaction by a skin irritant as well as cell proliferation effect in bio-artificial skin. Fructan was also evaluated as a safe material without any toxicity in safety tests using fibroblasts and animals.
Objective : To provide information about the accessory product of Broussonetia kazinoki Siebold or Broussonetia papyrifera (L.) Vent by analyzing old books of oriental medicine, domestic/international papers and related patents Methods : Old books related to the accessory product in the field of oriental medicine were reviewed. Research papers regarding the pharmacological activity of the by-products were reviewed and analyzed. Patents about the residual products were examined and classified by year and subject Results : Seven kinds of by-products from Broussonetia kazinoki Siebold or Broussonetia papyrifera (L.) Vent has been used as medicines in oriental medicine. Recently, anti-oxidating, anti-cancer, anti-mutagenic and anti-inflammation activity of the residual product of these plants has been investigated through scientific research. There were 19 patents related with the accessory products of these plants, which were in the subjects of functional cosmetics, anti-inflammation, cleansing goods, hair restorers or improvement of learning ability. Further investigations about the activity of these plants are needed in bone metabolism, water balance and hemostasis in the future. Conclusion : Residual products from these plants is being used in various ways. However, more studies on the efficacy and mechanism, as well as safety, of these plants should be conducted precisely in the future.

Objectives : Allergic diseases have a various symptoms of hyperresponsiveness and recently hyperresponsive reaction in the chronic phase is reported as the important mechanisms. Cheonggisan(CGS) is used in oriental clinics for curing various skin diseases due to effect of controlling of pruritus. There have been studies on the anti-allergic effect and anti-inflammatory effect of CGS, but there had no study of anti-allergic effects in allergic late inflammation of CGS, so we aimed to find out the effects of CGS in allergic late inflammation in our study.Methods : To investigate the anti-allergy effect and anti-inflammatory effect of CGS, RAW 264.7 macrophage cells and CSG water-extracts were used. Cytotoxic effect of CSG was examined by MTT assay, an oxidative product of NO was measured in the culture medium by the Griess reagent assay. The level of prostaglandin E2(PGE2) was measured by competitive enzyme-linked immunoassay. Cytokine(PGE2, IL-1β, IL-6, TNF-α) was measured by Bio-Plex suspension assay system and quantitative multiplexed cytokine/chemokine assay.Results : We investigated that there was no cytotoxic effect of CGS water-extract at any levels of concentration on RAW 264.7 macrophage cells by MTT assay. CGS water-extracts significantly suppressed the levels of the inflammatory mediators such as NO and PGE2, cytokine of IL-1β, TNF-α at the level of 400 ㎍/㎖ CGS concentration. But there was no significant effect on IL-6 production suppression.Conclusions : These results suggest that CSG water-extract has and anti-inflammatory effects in allergic reaction. These properties may contribute to the allergic diseases and inflammatory related disease care.
Objective : This study was performed to compare anti-inflammation, anti-oxidation and anti-bacterial effects of Ligustici Rhizoma (LR) extracted with two kinds of solvents, ethanol and distilled water. Methods : It is prepared two kinds of LR extracts 20, 50, 100 ${\mu}l/mg$ by first. MTT assay way to measure cytotoxicity is formed in Raw 264.7 cell. The anti-inflammation effect is measured by ability to inhibit production of NO in Raw 264.7 cell. The anti-oxidation effect was measured by DPPH Radical scavenging ability in HaCaT cell. The anti-bacterial effect was measured by inhibition zone diameter on Propionibacterium acnes. Results : 1. LR (20 ${\mu}l/mg$) extracted with ethanol was showed 80% cytotoxicity, LR (50 ${\mu}l/mg$) extracted with ethanol and LR (20, 50 ${\mu}l/mg$) extracted with water were showed 70% cytotoxicity, LR (100 ${\mu}l/mg$) extracted with ethanol and LR (100 ${\mu}l/mg$) extracted with water were showed 60% cytotoxicity in Raw 264.7 cell. 2. LR (100 ${\mu}l/mg$) extracted with ethanol was showed more significantly inhibitory effect on NO production than the water extraction. 3. Two kinds of LR extraction groups did not show significantly scavenging effect of DPPH radicals. 4. Two kinds of LR extractions did not have a inhibitory effect on Propionibactrium acnes. Conclusion : Two kinds of LR extracts have not cytotoxicity, statistically significant ability to scavenge DPPH radicals and effect to inhibit Propionibactrium acnes. LR extracted with ethanol only have a little effect to inhibit NO production. This study proposes that LR extracted with ethanol is more effective in anti-inflammation.

Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.
The effects of Picrorrhiza Rhizoma (PR) aqueous extracts were observed on xylene-induced acute inflammation. The xylene was topically applied 60 min after administration of 500, 250 and 125 mg/kg of PR extracts, and all animals were sacrificed 2 hrs after xylene application. The changes on ear weights, histolopathological analyses of ear were evaluated as compared with indomethacin and dexamethasone 15 mg/kg treated groups - well known anti-inflammatory agents. Xylene application resulted in marked increases in induced ear weights as compared with intact control ear. Severe vasodilation, edematous changes of ear skin and increase in the thickness of the ear tissues as acute inflammation were detected in xylene-treated control ears at histopathological observation. However, these xylene-induced acute inflammatory changes were dosedependently decreased by oral treatment of PR extracts. Therefore, it is concluded that PR extracts has favorable anti-inflammatory effects on xylene-applicated acute ear inflamed mice.

Objective : In this study, the effect of Atractylodes japonica against LPS induced inflammation in mouse microglia BV2 cells was investigated. Method : Microglia BV2 Cells viability was determined using the MTT assay. We used water, ethanol extract from Atractylodes japonica and studied on the anti-inflammatory effect of lipopolysaccharide-induced inflammation using reverse transcription polymerase chain reaction (RT-PCR), western blot, and nitric oxide detection on mouse microglia BV2 cells. Result : The MTT assay revealed that it's extract has no significant cytotoxicity in the microglia BV2 cell. Various solvent extract from Atractylodes japonica inhibited nitrite production, iNOS protein and mRNA expression levels. And also it's extracts significantly reduced lipopolysaccharide-induced COX-2 activation in RT-PCR and western blot in lipopolysaccharide-induced microglia BV2 cells Conclusion : In this study, it's extracts was shown to suppress NO production by inhibiting iNOS expression and COX-2 activity. With this effects of anti-inflammation, we suggests that, it's extracts may be a useful candidate for the development of a drug on the related inflammatory diseases in brain.
To investigate the anti-inflammatory activity of natural products, we determined the anti-inflammatory activity of purified epigallocatechin (EGC) from Camellia sinensis leaves. In the present study, we found that EGC inhibited the production of proinflammatory mediators (IL-6, TNF-${\alpha}$, NO, and $PGE_2$) in lipopolysaccharide (LPS)-stimulated Raw 264.7 cells. Suppression of IL-6 seems to be at least partly attributable to the inhibitory effect of EGC. TNF-${\alpha}$ is a major cytokine produced by LPS-induced macrophages, and they have a wide variety of biological functions including regulation of inflammation. The inhibition of IL-6 and TNF-${\alpha}$ production by EGC may downregulate the acute-phase response to LPS, thereby reducing LPS-induced inflammation. In addition to IL-6 and TNF-${\alpha}$, EGC effectively reduced the production of other key inflammatory mediators, including NO and $PGE_2$. The inhibitory effect of EGC on NO and $PGE_2$ production was supported by the suppression of inducible nitric oxide synthase and COX-2 at protein levels. These results support the traditional use of EGC in the alleviation of various inflammation-associated diseases and suggest that EGC might be useful in the development of new functional foods for inflammatory diseases.
