In Vivo Screening for Biocontrol Agents (BCAs) against Streptomyces scabiei Causing Potato Common Scab

  • Through in vitro screening for biocontrol agents (BCAs) against Streptomyces scabiei causing potato (Solanum tuberosum) common scab, 19 streptomycete and 17 fungal isolates with antagonistic activity were selected as BCA candidates. For the selection of BCA candidates which are highly resistant to 10 kinds of antibiotics or pesticides, chemical susceptibility testing was initially performed in vitro. A remarkable degree of variation in susceptibility to antibiotics or pesticides was observed among the isolates tested. Streptomycete A020645 isolate was highly resistant to all the tested chemicals except neomycin up to 5,000 ppm. On the other hand, out of 36 antagonistic microbes subjected to in vivo pot tests using cultivar Daejima, four streptomycete isolates namely, A020645, A010321, A010564, and A020973, showed high antagonistic activity with >60% and 55% control value, respectively, and high chemical resistance to 10 kinds of chemicals. Therefore, these isolates were selected as potential BCAs for the control of potato common scab.

사과 주요 병해 방제를 위한 길항미생물 분리 및 동정 (Isolation and Identification of Antagonistic Microorganisms for Biological Control to Major Diseases of Apple Tree(Malus domestica Borkh))

  • For the purpose of acquiring microbial agents that can be utilized to biologically control the major airborne diseases to apple trees, such as canker(Botryosphaeria dothidea), bitter rot(Glomerella cingulata), alternaria leaf spot(Alternaria mali), root rot(rosellinia necatrix), canker(Valsa ceratosperma) and gray mold rot(Botrytis cinerea), the effective microorgaisms were isolated, tested for antagonistic activity to the pathogens causing major diseases to apple trees and identifed. Screening of more than 5,000 species of microorganisms collected in nature for them antagonistic action to the pathogens causing 5 major diseases to apple trees resulted in selection of effective species. Out of the 11 species, one species designated as CAP134 demonstrated outstanding activity. The bacterial strain, CAP134 exerted antagonistic efficiency of 57% on an isolated strain and 40% on a donated strain of Botryosphaeria dothidea., 52% on an isolated strain and 46% on a purchased strain of Alternaria mali, 60% on Valsa ceratosperma 25% on Glomerella cingulata, and 64% Rosellinia necatrix. The CAP134 was identified as a bacterial strain to Bacillus subtilis ATCC 6633 based on morephology, culture conditions, and physio-biochemical characteristics.

Development of W/O/W Multiple Emulsion Formulation Containing Burkholderia gladioli

  • W/O/W (water-in-oil-in-water) type multiple emulsion was applied to improve the storage stability of an antagonistic microorganism, Burkholderia gladioli. Encapsulation of microorganism into a W/O/W emulsion was conducted by using a two-step emulsification method. W/O/W emulsion was prepared by the incorporation of B. gladioli into rapeseed oil and the addition of polyglycerin polyriconolate (PGPR) and castor oil polyoxyethylene (COG 25) as the primary and secondary emulsifier, respectively. Microcrystalline cellulose was used as an emulsion stabilizer. To evaluate the usefulness of W/O/W emulsion formulation as a microbial pesticide for controlling the bacterial wilt pathogen (Ralstonia solanacearum), the storage stability and antagonistic activity of emulsion formulation were tested in vitro. The storage stability test revealed that the viability of formulated cells in emulsion was higher than that of unformulated cells in culture broth. At $4^{\circ}C$, the viabilities of formulated cells and unformulated cells at the end of 20 weeks decreased to about 2 and 5 log cycles, respectively. At $37^{\circ}C$, the viability of formulated cells decreased to only 2 log cycles at the end of storage. On the other hand, the viable cells in culture broth were not detected after 13 weeks. In activity test, formulated cells in emulsion were more effective in inhibiting the growth of pathogen than unformulated cells in culture broth. Unformulated cells completely lost their antagonistic activity during storage under similar conditions. The W/O/W multiple emulsion formulation was shown to be useful as the novel liquid formulation for biological control.

  • 경주지역의 토양으로부터 Fusarium 속 식물병원균에 길항력을 갖는 chitinase 생산성 길항미생물을 분리할 수 있었으며, 이를 분류학적으로 동정하여 본 결과 Serratia proteamaculans 3095로 동정할 수 있었다. 이 균주가 생성하는 chitinase의 생성조건을 조사한 결과 탄소원으로 colloidal chitin이 가장 좋았으며 그 최적 농도는 0.15%이었고, glucose에 의해 chitinase 생산 유도를 억제받는 효소임을 알 수 있었다. 질소원에 의한 영향은 $(NH_4)_2SO_4,\;(NH_4)Cl$, peptone 등에 의해 chitinase 생산성이 증가되었고, $(NH_4)_2SO_4$와 peptone을 각각 0.1%씩 첨가하였을 때 chitinase 생산이 가장 좋았다. 또한 시드름병균 Fusarium oxysporum을 대상으로 in vitro, in vivo pot 실험을 통해 Serratia sp. 3095의 강한 방제력을 검증할 수 있었다.

  • This study was carried out to research microorganisms having the antifungal activity against ginseng Alternaria blight pathogen Alternaria panax and ginseng anthracnose pathogen Colletotrichum gloeosporioides. Eleven Bacillus strains. were isolated from Korean traditional soybean paste and Kimchi. Among the 11 isolates, DJ5, KC1, KC2 and KC4 showing antagonistic activity on the mycelial growth of A. panax and C. gloeosporioides in pairing culture were finally selected as the antagonistic microorganisms. Based on 16s rRNA sequence and phylogenetic tree analysis, they were identified as Bacillus spp.. The selected microorganisms were investigated antagonistic activity by measured leaf-segment colonization in pot test. When Bacillus sp. were injected after A. panax treatment, KC1, KC2 and KC4 showed similar effect to chemical pesticides treated control. To measure preventive effect of Bacillus sp, antagonistic microorganisms were injected and C. gloeosporioides was treated in pot. When measuring the effectiveness for the prevention of Anthracnose, All Bacillus spp. showed approximately 83~90 % degree of superior preventive effect. In general, The four Bacillus spp. isolated from Korean traditional fermented foods showed therapeutic effect of Alternaria blight and preventive effect of Anthracnose.

Diversity and Bioactive Potential of Culturable Fungal Endophytes of Medicinal Shrub Berberis aristata DC.: A First Report

  • Bioactive natural compounds, isolated from fungal endophytes, play a promising role in the search for novel drugs. They are an inspiring source for researchers due to their enormous structural diversity and complexity. During the present study fungal endophytes were isolated from a well-known medicinal shrub, Berberis aristata DC. and were explored for their antagonistic and antioxidant potential. B. aristata, an important medicinal shrub with remarkable pharmacological properties, is native to Northern Himalayan region. A total of 131 endophytic fungal isolates belonging to eighteen species and nine genera were obtained from three hundred and thirty surface sterilized segments of different tissues of B. aristata. The isolated fungi were classified on the basis of morphological and molecular analysis. Diversity and species richness was found to be higher in leaf tissues as compared to root and stem. Antibacterial activity demonstrated that the crude ethyl acetate extract of 80% isolates exhibited significant results against one or more bacterial pathogens. Ethyl acetate extract of Alternaria macrospora was found to have potential antibacterial activity. Significant antioxidant activity was also found in crude ethyl acetate extracts of Alternaria alternata and Aspergillus flavus. Similarly, antagonistic activity of the fungal endophytes revealed that all antagonists possessed inhibition potential against more than one fungal pathogen. This study is an important step towards tapping endophytic fungal diversity for bioactive metabolites which could be a step forward towards development of novel therapeutic agents.

Diversity and Characterization of Endophytic Bacteria Associated with Tidal Flat Plants and their Antagonistic Effects on Oomycetous Plant Pathogens

  • Endophytic bacterial communities of tidal flat plants antagonistic to oomycete plant pathogens were studied by the isolation of 256 root colonizing endophytic bacteria from surface-disinfected root tissues of six plants ($Rosa$ $rugosa$, $Suaeda$ $maritima$, $Vitex$ $rotundifolia$, $Carex$ $scabrifolia$, $Glehnia$ $littoralis$ and $Elymus$ $mollis$) growing in a tidal flat area of Namhae Island, Korea. To understand the antagonistic potential, an $in$ $vitro$ antagonistic assay was performed to characterize and identify strains that were antagonistic to the oomycete plant pathogens $Phytophthora$ $capsici$ and $Pythium$ $ultimum$ from the total population. Nine percent of the total number of isolated bacteria exhibited in vitro inhibitory activity against target plant pathogenic oomycetes. Taxonomic and phylogenetic placement of the antagonistic bacteria was investigated by analysis of the 16S rRNA gene sequences. The sequence analysis classified the antagonistic strains into four major classes of the domain bacteria ($Firmicutes$, ${\alpha}-Proteobacteria$, ${\gamma}-Proteobacteria$ and $Actinomycetes$) and 10 different genera. Further production of secondary metabolites, hydrolytic enzymes and plant growth promoting traits were determined for the putative new species of antagonistic endophytic bacteria. These new strains could not be identified as known species of ${\alpha}-Proteobacteria$, and so may represent novel bacterial taxa. The unexpected high antagonistic bacterial diversity associated with the tidal flat plants may be indicative of their importance in tidal flat plants as a promising source of novel antimicrobial compounds and biocontrol agents.

  • The studies for antithrombotic substances from medicinal plants in my laboratory were started from the studies on PAF-antagonistic substances from Korean medicinal plants. The screening studies of PAF-receptor binding antagonistic activity were conducted on the extracts of 300 Korean medicinal plants, 37 tropical medicinal plants, 20 mushrooms, and 30 vegetables. From the results of screening studies, it was possible to select two Korean medicinal plants, i.e. 1) the leaf of Biota orientalis and 2) the seed of Arctium lappa, and two tropical medicinal plants, i.e. 3) the rhizome of Alpinia officinarum and 4) the leaf of Ardisia crispa as the candidates for the activity guided isolation of PAF-antagonistic substances.

Biological Control of Apple Anthracnose by Paenibacillus polymyxa APEC128, an Antagonistic Rhizobacterium

  • The present study investigated the suppression of the disease development of anthracnose caused by Colletotrichum gloeosporioides and C. acutatum in harvested apples using an antagonistic rhizobacterium Paenibacillus polymyxa APEC128 (APEC128). Out of 30 bacterial isolates from apple rhizosphere screened for antagonistic activity, the most effective strain was APEC128 as inferred from the size of the inhibition zone. This strain showed a greater growth in brain-heart infusion (BHI) broth compared to other growth media. There was a reduction in anthracnose symptoms caused by the two fungal pathogens in harvested apples after their treatment with APEC128 in comparison with non-treated control. This effect is explained by the increased production of protease and amylase by APEC128, which might have inhibited mycelial growth. In apples treated with different APEC128 suspensions, the disease caused by C. gloeosporioides and C. acutatum was greatly suppressed (by 83.6% and 79%, respectively) in treatments with the concentration of $1{\times}10^8$ colony forming units (cfu)/ml compared to other lower dosages, suggesting that the suppression of anthracnose development on harvested apples is dose-dependent. These results indicated that APEC128 is one of the promising agents in the biocontrol of apple anthracnose, which might help to increase the shelf-life of apple fruit during the post-harvest period.

토양병원균(土壤病原菌) 길항성(拮抗性) Streptomyces sp. A-2 활성물질(活性物質)의 특성(特性)에 관한 연구(硏究) (Studies on Characterization of Active Substances from Antagonistic Streptomyces sp. A-2 Strain against Soil-borne Phytopathogen)

  • 우리나라 28개 지역(地域) 논밭 토양에서 수집한 시료에서 방선균(放線菌) 108주를 분리하고, 고추역병균, 참깨역병균 및 채소입고병균에 종합적으로 항균력을 갖는 Streptomyces sp. A-2를 선발하여 그 활성물질(活性物質)의 특성을 조사한 결과는 다음과 같다. 1. G.Y.B., $M\ddot{u}eller$, B.H.I., Czapek 등의 배지중에서 길항성(拮抗性)의 효과가 가장 잘 이전되는 배지는 G.Y.B. 및 B.H.I. 배지이었고, 주(主) 활성물질(活性物質)은 에칠아세테이트로 이전(移轉)되는 군과 수층(水層)에 잔존하는 군으로 구분되었으며 이들 물질은 열(熱)과 압력(壓力)에 매우 강한 결태형태(結台形態)를 보였다. 2. 길항성(拮抗性) Streptomyces sp. A-2에 의해 배양된 여액의 pH를 3이나 12로 조정하여 길항물질을 추출하여 3개 식물병원균에 대한 길항력 검정을 실시한 결과 pH를 달리한 추출법에서 별다른 차이점이 없었다. 이들 분획의 UV 스펙트럼은 폴리엔 길항활성의 마크로라이드의 그것과 유사하였으므로 폴리엔계 항생제가 이 균주의 길항성에 관련하는 주(主) 물질(物質)이라고 추정되었다.
