Resistance to anoikis, a cell-detachment induced apoptosis, is one of the malignant phenotypes which support tumor metastasis. Molecular mechanisms underlying the establishment of this phenotype require further investigation. This study aimed at exploring protein expression profiles associated with anoikis resistance of a metastatic breast cancer cell. Cell survival of suspension cultures of non-metastatic MCF-7 and metastatic MDA-MB-231 cells were compared with their adherent cultures. Trypan blue exclusion assays demonstrated a significantly higher percentage of viable cells in MDA-MB-231 than MCF-7 cell cultures, consistent with analysis of annexin V-7-AAD stained cells indicating that MDA-MB-231 possess anti-apoptotic ability 1.7 fold higher than MCF-7 cells. GeLC-MS/MS analysis of protein lysates of MDA-MB-231 and MCF-7 cells grown under both culture conditions identified 925 proteins which are differentially expressed, 54 of which were expressed only in suspended and adherent MDA-MB-231 but not in MCF-7 cells. These proteins have been implicated in various cellular processes, including DNA replication and repair, transcription, translation, protein modification, cytoskeleton, transport and cell signaling. Analysis based on the STITCH database predicted the interaction of phospholipases, PLC and PLD, and 14-3-3 beta/alpha, YWHAB, with the intrinsic and extrinsic apoptotic signaling network, suggesting putative roles in controlling anti-anoikis ability. MDA-MB-231 cells grown in the presence of inhibitors of phospholipase C, U73122, and phospholipase D, FIPI, demonstrated reduced ability to survive in suspension culture, indicating functional roles of PLC and PLD in the process of anti-anoikis. Our study identified intracellular mediators potentially associated with establishment of anoikis resistance of metastatic cells. These proteins require further clarification as prognostic and therapeutic targets for advanced breast cancer.

Gefitinib exerts anticancer effects on various types of cancer, such as lung, ovarian, breast, and colon cancers. However, the therapeutic effects of gefitinib on cervical cancer and the underlying mechanisms remain unclear. Thus, this study aimed to explore whether gefitinib can be used to treat cervical cancer and elucidate the underlying mechanisms. Results showed that gefitinib induced a caspase-dependent apoptosis of HeLa cells, which consequently became round and detached from the surface of the culture plate. Gefitinib induced the reorganization of actin cytoskeleton and downregulated the expression of p-FAK, integrin β1 and E-cadherin, which are important in cell-extracellular matrix adhesion and cell-cell interaction, respectively. Moreover, gefitinib hindered cell reattachment and spreading and suppressed interactions between detached cells in suspension, leading to poly (ADP-ribose) polymerase cleavage, a hallmark of apoptosis. It also induced detachment-induced apoptosis (anoikis) in C33A cells, another cervical cancer cell line. Taken together, these results suggest that gefitinib triggers anoikis in cervical cancer cells. Our findings may serve as a basis for broadening the range of anticancer drugs used to treat cervical cancer.
Purpose: The aim of this study was to investigate the relationship between cell adhesion and anoikis evasion among human osteosarcoma cells (MG-63), and to further study the molecular mechanisms. Materials and Methods: Human osteosarcoma cells (MG-63) were assessed for apoptosis, and caspase-3, E-cadherin and ${\beta}$-catenin expression in EDTA and control non-EDTA groups. Results: MG-63 cells were predominantly aggregated when in suspension, and the suspended cells were more dispersed in the EDTA group. Following culture in suspension for 24 h, 48 h, or 72 h, the rates of apoptosis were $34.88%{\pm}3.64%$, $59.3%{\pm}7.22%$ and $78.5%{\pm}5.21%$ in the experimental group and $7.34%{\pm}2.13%$, $14.7%{\pm}3.69%$, and $21.4%{\pm}3.60%$ in the control group, respectively. Caspase-3 expression progressively increased and E-cadherin and ${\beta}$-catenin were decreased in the experimental group, whereas there was no change in the control group. Conclusions: MG-63 cells could avoid anoikis through cell adhesion, and E-cadherin might play a role in this process.
본 연구에서는 다양한 사람의 암세포주(A-549, MCF-7, MDA-MB-231, U87-MG, AGS, MKN-74 및 SNU-601 세포)와 정상세포주(MRC-5 섬유아세포 및 사랑니 유래 중간엽성 줄기세포에 토복령 추출물(Smilax china L. extract, SCLE)을 처리하여 세포 사멸 효과를 조사하였다. SCLE 처리 후, MTT 분석에서 여러 암세포주는 정상세포주보다 유의적으로 휠씬 낮은 반억제농도값을 나타내었고, 세포는 세포부착력의 소실로 인한 세포사멸(anoikis)이 관찰되었다. 또한, SCLE를 처리한 A-549, AGS 및 MCF-7 암세포주에서 세포의 생존성과 말단소립 복원효소의 활성도를 조사하였을 때, SCLE 처리 후 4일째에 세포의 생존성과 말단소립 복원효소의 활성도가 현저히 줄어드는 것을 관찰하였다. 또한, SCLE를 처리한 A-549, AGS 및 MCF-7 암세포주에서 세포 주기의 G1기에서 세포 성장이 정지되었고,세포 사멸이 유의적으로 증가하는 것을 알 수 있었다. 그러나, SCLE 처리는 rho 단백질의 활성과 관련 없는 세포부착력의 소실과 세포 사멸이 유도되는 것을 관찰하였다. 이 연구의 결과를 바탕으로 토복령 추출물은 정상 세포보다는 암세포에 특이적으로 세포부착력의 소실과 세포 사멸을 유도하여, 이 추출물에 포함된 물질을 이용한 항암 연구에 응용될 수 있을 것으로 판단된다.
Resveratrol은 포도, 오디, 땅콩과 같은 많은 과일과 채소에 존재하는 폴리페놀 화합물로써 다양한 생물학적 효과를 가진다고 보고되어있다. 그러나, resveratrol이 A549 폐암세포에서 유도하는 apoptosis에 관여하는 분자적 기전 및 anoikis에 관해서는 명백하게 밝혀지지 않았다. 본 연구에서, 우리는 정상적인 p53 유전자를 갖는 A549 세포에서 resveratrol의 효과를 조사하고, p53 유전자가 결실된 SKOV3 난소암 세포와 그 효과를 비교했다. Resveratrol은 확실하게 SKOV3 세포에 비해 농도, 시간의존적으로 A549 세포의 생존과 증식을 억제했다. 또한 resveratrol은 A549 세포의 apoptosis를 유도했지만 세포의 anoikis 저항성에는 영향을 미치지 않았다. 더불어, p53 유전자 기능이 불완전 소실된(knock-down) A549 세포의 생존과 증식은 resveratrol에 의해 변하지 않았다. 그러므로, 본 연구의 결과는 resveratrol의 항암 효과가 기능을 하는 p53 유전자의 존재에 의존한다는 것을 보여준다. 결론적으로, 우리는 resveratrol이 p53 유전자에 의존적으로 A549 세포에 대해 항암효과를 가진다는 것을 입증했다.
Background: Lung cancer has a high incidence worldwide, and most lung cancer-associated deaths are attributable to cancer metastasis. Although several medicinal properties of Panax ginseng Meyer have been reported, the effect of ginsenosides Rk1 and Rg5 on epithelial-mesenchymal transition (EMT) stimulated by transforming growth factor beta 1 (TGF-β1) and self-renewal in A549 cells is relatively unknown. Methods: We treated TGF-β1 or alternatively Rk1 and Rg5 in A549 cells. We used western blot analysis, real-time polymerase chain reaction (qPCR), wound healing assay, Matrigel invasion assay, and anoikis assays to determine the effect of Rk1 and Rg5 on TGF-mediated EMT in lung cancer cell. In addition, we performed tumorsphere formation assays and real-time PCR to evaluate the stem-like properties. Results: EMT is induced by TGF-β1 in A549 cells causing the development of cancer stem-like features. Expression of E-cadherin, an epithelial marker, decreased and an increase in vimentin expression was noted. Cell mobility, invasiveness, and anoikis resistance were enhanced with TGF-β1 treatment. In addition, the expression of stem cell markers, CD44, and CD133, was also increased. Treatment with Rk1 and Rg5 suppressed EMT by TGF-β1 and the development of stemness in a dose-dependent manner. Additionally, Rk1 and Rg5 markedly suppressed TGF-β1-induced metalloproteinase-2/9 (MMP2/9) activity, and activation of Smad2/3 and nuclear factor kappa B/extra-cellular signal regulated kinases (NF-kB/ERK) pathways in lung cancer cells. Conclusions: Rk1 and Rg5 regulate the EMT inducing TGF-β1 by suppressing the Smad and NF-κB/ERK pathways (non-Smad pathway).
본 연구에서는 증폭시킨 홍삼으로부터 분리한 ginsenoside Rh2(Rh2)와 compound K(CK)의 융복합적 항염증 및 항암효과를 연구하여 홍삼 내 유용한 기능성분에 대한 기초 자료를 제공하고자 한다. 이에 Hep3B에서의 세포독성과 IL-6 유도 STAT3 루시퍼라아제 활성, B16F10과 Hacat 세포의 생존 농도를 측정하였고 Apoptosis와 관련된 분자의 발현양상을 확인하기 위해 FAC (fluorescence activated cell sorting) 분석을 수행하였다. 실험결과 Rh2, CK mixture가 10 ug/ml일 때 Hep3B 세포에서 세포독성이 없고 IL-6 감소율이 102%로 항염증 효과가 있는 것으로 나타났다. 또한 Rh2, CK mixture 50 uM에서 melanoma 세포인 B16F10과 human keratinocyte인 Hacat에서 독성을 보여 사멸하는 것을 관찰하였다. FACS 분석 결과 annexin V가 발현되지 않고 흑색종 세포와 keratinocyte가 탈착되면서 사멸되는 것을 확인하였다. 이러한 현상을 통하여 사멸되는 메카니즘이 anoikis 방식의 세포사멸로 인한 것으로 추정할 수 있으며 그것에 대한 명확한 세포사 신호 체계 규명을 위하여 향후 세포부착 단백질의 변화에 대한 연구가 필요할 것으로 판단된다.
Epithelial-mesenchymal transition (EMT) is widely-considered to be a modulating factor of anoikis and cancer metastasis. We found that, in MDA-MB-231 cells, TP53I11 (tumor protein P53 inducible protein 11) suppressed EMT and migration in vitro, and inhibited metastasis in vivo. Our findings showed that hypoxic treatment upregulated the expression of $HIF1{\alpha}$, but reduced TP53I11 protein levels and TP53I11 overexpression reduced $HIF1{\alpha}$ expression under normal culture and hypoxicconditions, and in xenografts of MDA-MB-231 cells. Considering $HIF1{\alpha}$ is a master regulator of the hypoxic response and that hypoxia is a crucial trigger of cancer metastasis, our study suggests that TP53I11 may suppress EMT and metastasis by reducing $HIF1{\alpha}$ protein levels in breast cancer cells.
이 연구는 단일 세포로 분리된 유도만능줄기세포(induced pluripotent stem cells, iPSCs)에 anoikis 세포사멸을 억제할 수 있는 Rho-associated protein kinase (ROCK)의 억제제를 처리하여 iPSCs의 세포 생존성을 향상하고자 하였다. Episomal plasmid 방법으로 확립된 iPSCs를 단일세포로 분리한 후, ROCK 억제제 Y-27632 dihydrochloride (Y-27632)를 0 uM, 0.5 uM, 1 uM, 2.5 uM, 5 uM, 7.5 uM 및 10 uM 농도별로 5주일 동안 각각 처리하였을 때, 5 uM 이상의 농도에서 세포의 생존율이 유의적으로 향상되었고, 10 uM의 Y-27632을 0일, 1일, 2일, 3일, 4일 및 5일 동안 처리하였을 때, Y-27632의 노출 기간이 길어질수록 세포의 생존율이 유의적으로 향상되는 것을 관찰하였다. 그러나, Y-27632의 노출 후, iPSCs의 형태학적 분화가 관찰되어 10 uM의 Y-27632에서 5일 동안 iPSCs에 처리 한 후, 줄기세포학적인 특성을 비교 조사하였다. 우선, octamer-binding transcription factor 4 (OCT-4), homeobox protein NANOG (NONOG) 및 SRY-box 2 (SOX-2) 줄기세포 특이 유전자의 발현은 Y-27632를 처리한 실험군은 Y-27632를 처리하지 않은 대조군에서 서로 유의적인 차이를 나타내지 않았다. 또한, Y-27632를 처리한 실험군은 Y-27632를 처리하지 않은 대조군과 비교하여 telomerase 활성과 이것의 활성과 관련된 telomerase reverse transcriptase (TERT) 및 telomerase RNA component (TERC)의 유전자 발현에는 유의적인 차이가 없었다. 이상의 결과로 보아, iPSCs에 Y-27632를 처리하였을 때, iPSCs의 줄기세포의 특정을 유지하면서 anoikis에 의한 세포사멸을 감소시켜 세포 생존율이 증가한다는 것을 알 수 있었다.
