Inhibitory Activity on Angiotensin Converting Enzyme and Antioxidant Activity of Hovenia dulcis Thunb. Cortex Extract (헛개나무의 Angiotensin 전환 효소 저해 및 항산화 활성)

  • To develop a new functional materials, angiotensin converting enzyme (ACE) inhibitory activity, antioxidant effect and total phenolic content of Hovenia dulcis Thunb. cortex were evaluated. Methanol and water extract of H. dulcis inhibited ACE by 81% and 76%, respectively, at the concentration of $4,000\;{\mu}g\;m{\ell}^{-1}$ which were similar level with that (85%) of commercial peptide-type ACE inhibitor. Superoxide radical scavenging activity of two extracts $(99.5%{\sim}99.9%)$ were stronger than that (69%) of ascorbic acid at the final concentration of $200\;{\mu}g\;m{\ell}^{-1}$. Among the solvent fractions, ether and ethylacetate fraction showed also potent scavenging activities (91% and 85%) for superoxide radical. Inhibitory activities of two extracts on oxidation of human low density lipoprotein (LDL) which were similar with that of ${\alpha}-tocopherol$, were higher than 80% at the concentration of $50\;{\mu}g\;m{\ell}^{-1}$. Total phenol contents of methanol and water extracts were 7.2% and 3.6%, respectively, and that of ethylacetate showed the highest value as 60.8% among the solvent fractions. Therefore, it has been suggested that H. dulcis cortex could be a effective anti-hypertention and antioxidant resource to develope a new functional material.

Inhibitory Effects of Ginseng Saponins on c-fos mRNA Expression and the Proliferation of Rat Aortic Vascular Smooth Muscle Cells Stimulated by Angiotensin II

  • To evaluate the possibility that the ginseng saponins could be developed as an anti-arteriosclerotic agent, we examined the inhibitory effects of ginseng saponins (total saponin[TS], panaxatriol[PT], panaxadiol[PD]) on the expression of c-fos mRNA and the proliferation of cultured rat aortic vascular smooth muscle cells (VSMCs) stimulated by angiotensin II (Ang II). TS and PT (1.0 mg/ml) suppressed c-fos mRNA induction in VSMCs stimulated by $10^{-5}$ M Ang II. The order of inhibitory potency was PT>TS. Ginseng saponins ($0.01{\sim}1.0$ mg/ml) inhibited the proliferation of VSMCs stimulated by Ang II in a concentration dependent manner, the inhibitory potency was TS>PT>PD at $0.1{\sim}1.0$ mg/ml. These results suggest that ginseng saponins may suppress Ang II-stimulated proliferation of aortic VSMCs which can be seen in atherosclerosis, hypertension and restenosis.

Angiotensin I-converting Enzyme Inhibitory Activities of Porcine Skeletal Muscle Proteins Following Enzyme Digestion

  • Inhibitory activities against angiotensin I-converting enzyme (ACE) of enzymatic hydrolysates of porcine skeletal muscle proteins were investigated. Myosin B, myosin, actin, tropomyosin, troponin and water-soluble proteins extracted from pork loin were digested by eight kinds of proteases, including pepsin, $\alpha$-chymotrypsin, and trypsin. After digestion, hydrolysates produced from all proteins showed ACE inhibitory activities, and the peptic hydrolysate showed the strongest activity. In the case of myosin B, the molar concentration of peptic hydrolysate required to inhibit 50% of the activity increased gradually as digestion proceeded. The hydrolysates produced by sequential digestion with pepsin and $\alpha$-chymotrypsin, pepsin and trypsin or pepsin and pancreatin showed weaker activities than those by pepsin alone, suggesting that ACE inhibitory peptides from peptic digestion might lose their active sequences after digestion by the second protease. However, the hydrolysates produced by sequential digestion showed stronger activities than those by $\alpha$-chymotrypsin, trypsin or pancreatin alone. These results suggested that the hydrolysates of porcine meat were able to show ACE inhibitory activity, even if they were digested in vivo, and that pork might be a useful source of physiologically functional factors.

Effects of age on angiotensin II response and antagonistic activity of losartan in rat aorta and liver

  • The present study was undertaken to investigate the effects of age on angiotensin II (AII) response and antagonistic activity of losartan using aortic rings and liver homogenates from rats ranging in age from 0.7 to 20 months. Whether the endothelium was present or not, the maximum contractile response to AII decreased with age. Removal of the endothelium enhanced AII-induced maximum contraction and these endothelial effects seemed to be due to endothelium-derived relaxing factor (EDRF) in all ages. Equilibrium binding studies demonstrated an age-related decrease in maximum binding $(B_{max})$ with little change in binding affinity $(K_d)$. In rat aorta, the extent of losartan-induced parallel shifts $(K_B)$ in AII concentration-response curves was not significantly different between ages. In addition, $IC_{50}$ value of losartan in competition binding was not changed with age in rat liver homogenates. These results suggest that the potency of losartan is not altered with age in rat aorta and liver, although AII-induced contractile response and the maximum AII binding decreased significantly with age.

  • The physiological activities of cultivated Korean blue mussel (Mytilus edulis) and New Zealand green-lipped mussel (Perna canaliculus) were analyzed and compared. Both hot water extracts of blue mussel flesh (BMF) and green-lipped mussel flesh (GMF) showed increased activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). BMF showed increased ADH and slightly decreased ALDH activities compared to GMF. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of BMF was higher than that of GMF at the same concentration. BMF and GMF showed similar inhibitory activity against angiotensin converting enzyme at a concentration of 30 mg/mL. These results suggest that cultivated Korean blue mussel has similar physiological activity with New Zealand green-lipped mussel.

  • The extracellular polysaccharide...(EPS) has been used in a wide variety of industrial applications because of gel formation, emulsion stability, control of the surface tension, water absorption and film formation etc. In this study, inhibitory activity on tyrosinase and inhibitory activity on angiotensin converting enzyme...(ACE) are determined. UV adsorption wavelength, beads formation of EPS were investigated. In the result, in 1%(w/v) EPS concentration, EPS had inhibitory activity of 71.8% on tyrosinase and inhibitory activity of 61.5% on angiotensin converting enzyme in 1.5% EPS concentration. Adsorption wavelength of EPS was UV-B,C . Beads based on EPS were prepared by w/o emulsion method and the shape of EPS beads observed by SEM was spherical and uniform.

  • We experienced a case of severe intraoperative hyperkalemia during laparoscopic radical nephrectomy in a 60-year-old male patient with renal insufficiency, whose hypertension had been managed by preoperative angiotensin II receptor blocker (ARB) and adrenergic beta-antagonist. After renal vessel ligation, his intraoperative potassium concentration suddenly increased to 7.0 mEq/L, but his electrocardiography (ECG) did not show any significant change. While preoperative ARB therapy has been regarded as a contributing factor for further aggravation of underlying renal insufficiency, we assumed that nephrectomy itself and rhabdomyolysis caused by surgical trauma also aggravated the underlying renal dysfunction and resulted in sudden hyperkalemia. Hyperkalemia was managed successfully with calcium gluconate, insulin, furosemide and crystalloid loading during the intraoperative and immediate postoperative periods, and potassium concentration decreased to 5.0 mEq/L at 8 hours after the operation. The patient's hospital course was uncomplicated, but his renal function deteriorated further.

  • The effect of nicotine on the secretion of $[Met^{5}]-enkephalin$ (ME) in addition to proenkephalin A (proENK) mRNA levels and effects of indomethacin, nordihydroguaiaretic acid (NDGA), and captopril on nicotine-induced responses were studied in bovine adrenal medullary chromaffrin (BAMC) cells. Long-term exposure of BAMC cells to nicotine at a concentration of $10{\mu}M$ significantly increased proENK mRNA level and the secretion of ME into the medium. Treatment of BAMC cells with NDGA (a lipoxygenase inhibitor, $10{\mu}M$), indomethacin (a cycloooxygenase inhibitor) or captopril (an angiotensin converting enzyme inhibitor) alone did not affect ME secretion and proENK mRNA levels. The pretreatment of BAMC cells with NDGA inhibited the increased ME secretion and proENK mRNA level induced by nicotine. However, indomethacin and captopril did not affect nicotine-induced responses. Our results indicate that neuronal regulations of ME secretion and proENK mRNA level induced by nicotine in BAMC cells are in part mediated by a lipoxygenase-but not cyclooxygenase-and endogenous renin-angiotensin pathway.

Angiotensin-I Converting Enzyme Inhibitory Activity of Enzymatic Hydrolysates of Food Proteins (식품단백질 효소가수분해물의 Angiotensin-I 전환효소 저해작용)

  • Enzymatic hydrolysates of food proteins (defatted soybean cake, egg albumin and casein) were tested for inhibitory activity against angiotensin-I converting enzyme (ACE). Food proteins were hydrolysed with complex enzyme, bromelain, alcalase, $\alpha$-chymotrypsin, trypsin, papain and pepsin by heating method. The hydrolysates obtained from the treatment of complex enzyme and bromelain showed the higher ACE inhibitory activity. ACE inhibitory activity of hydrolysates exhibited a tendency to be increased until 8hrs and increased with increment of concentration. The activity was also stable by heat treatment at 10$0^{\circ}C$ for 20min. Molecular weight of active fraction was about 1, 400 and defatted soybean cake hydrolysate below 1, 400 in case of defatted soybean cake hydrolysate treated with alcalase. Amino acid of the active fractions was abundant in Asp, Glu, Lys, lle, Leu, Ala and Val.

  • An angiotensin converting enzyme (ACE) inhibitor was isolated and partially purified from bovine blood plasma. Bovine blood plasma was obtained after removing blood cells by centrifugation, followed by the addition of anticoagulant to whole bovine blood. To precipitate plasma proteins, bovine blood plasma was treated with 4% trichloroacetic acid (TCA) as a final concentration .An ACE inhibitor was isolated from TCA supernatnat, using ultrafiltration, gel permeation chormatography, and reverse-phase high pressure liquid chromatogrpahy. The ACE inhibitor purified from TCA supernatant had IC50 values of 9.4$\mu$M.
