Analgesic and anti-inflammatory effect of the aqueous extract of root of Angelica Dahurica

  • Angelica dahurica (Umbelliferae) grows in China, Japan, Russia, and Korea. The root of Angelica dahurica has been used as a traditional folk medicine to treat headache and toothache. In this study, the effects of the aqueous extract of Angelica dahurica on acetic acid-induced abdominal pain, carrageenan-induced edema, and thermal hyperalgesia were investigated using mice and rats. The present results showed that the aqueous extract of Angelica dahurica inhibited acetic acidinduced abdominal pain in mice and reduced carrageen-induced edema in rats. The present study showed that the aqueous extract of Angelica dahurica possesses anti-inflammatory and analgesic effects.

백지 BuOH 가용분획의 항혈전 활성에 관한 연구 (Antithrombotic Effect of the BuOH Soluble Fraction of Angelica dahurica Root)

  • Several coumarins isolated from Angelica sp. were described to show inhibitory effects against human platelet aggregation. The anti-thrombotic and anti-platelet potential was evaluated, in this paper, with the BuOH soluble fraction of Angelica dahurica root. The BuOH fraction was divided into five subfractions fr. A - E and tested in the mouse model of thrombosis. Survival was enhanced to 35% with fr. A or fr. E treated (500 mg/Kg, p.o.) group of mice compared with 5% survival of the control group. However, none of the 8 coumarin glycosides obtained from fr. A, at the conc. of 0.5 mg/ml, showed inhibitory effects against rat platelet aggregation induced by ADP or collagen.

Cytotoxic Coumarins from the Root of Angelica dahurica

  • Ten coumarins were isolated from the root of Angelica dahurica by repeated silica gel column chromatography. Their chemical structures were elucidated on the basic of physicochemical and spectroscopic data. Among them, oxypeucedanin hydrate acetonide (7) was isolated for the first time from this plant. Cytotoxicity of coumarins isolated were determined in vitro against L1210, HL-60, K562, and B16F10 tumor cell lines by MTT method. Pangelin (5) and oxypeucedanin hydrate acetonide (7) showed a potent cytotoxic activity with the $IC_{50}$ values of 8.6 to 14.6 ${\mu}g$ /mL against four kinds of tumor cell lines. Other compounds showed the moderate cytotoxic activity or no activity against the tumor cell lines.

  • Five furanocoumarins including a new one were isolated from the root of Angelica dahurica by repeated silica gel column chromatography. Their chemical structures were determined to be isoimperatorin (1), oxypeucedanin hydrate-3"-butyl ether (2), imperatorin (3), knidilin (4), and oxypeucedanin hydrate (5). This represents the first study in which the compound 2 has been isolated and identified, The long-range coupling ($^{5}J$) in the $^1H$-NMR spectrum observed in the linear furanocoumarin skeleton was also investigated in detail.

한국산(韓國産) 재배백지(栽培白芷)와 야생백지(野生白芷)에 대한 비교(比較) 연구(硏究) (A Comparative Study of between Cultivar and Wild type of Angelica dahurica in korea)

  • Objectives : This research was observed by examining the external and internal parts and the quantity of the surface components of the wild type and cultivar of Angelica dahurica in korea. Methods : The slice of the tested material mede by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by the microscope and examined the quantity of the surface components. Results : In the shape of the roots, the wild type had a very thin main root and 2 long roots, while the cultivar had a long root branched out from the short and thick main root making a square pyramid shape. Isoimperatorin was detected at Rt $17{\sim}18$ minutes. The wild type contained 0.56% while the cultivar contained it of 0.355%, thus the component of the wild type was about 1.5 times of that of the cultivar. Conclusions : It was thought that there might be also some differences in their genetic characteristics, etc.

  • Three coumarin derivatives, which stimulate collagen biosynthesis in human fibroblasts were isolated from Angelica dahurica root which has been used in traditional medicine for cold, headache, discharging blood and women's diseases. Three coumarin derivatives were identified as 5-benzofuranacrylic acid, $6,7-dihydroxy-4-(3-methyl-2-butenyl)-{\delta}-lactone\;(prangenidin)$, 5-benzofuranacrylic acid, $6,7-dihydroxy-4-methoxy-{\delta}-lactone\;(8-hydroxybergapten)$ and 5-benzofuranacrylic acid, 8-hydroxypsoralen(xanthotoxol) from their spectral data.

  • In this study, we analyzed and quantified the amounts of bioactive phenolic constituents, xanthotoxin (1), oxypeucedanin (2), and imperatorin (3) in each part of the root of Angelica dahurica by HPLC, which validated by ICH guide lines comparing the linearity, intra day precision, inter-day precision. As a result, the amount of imperatorin 2.96% in the rootlet was two fold higher than that of the main root 1.32%. On the other hand, the amounts of xanthotoxin and oxypeucedanin in the rootlet showed similar to those of main root. In addition, the cortex of root is more plentiful of three consitituents (0.66%, 0.53%, and 1.85%) than those of xylem (0.29%, 0.05%, and 0.07%). These results show that the rootlet and cortex contain a large amount of bioactive phenolic constituent including xanthotoxin, oxypeucedanin, and imperatorin than other parts of the root.

  • 피부노화는 피부의 이상현상이나 질환은 아니며 나이가 들어감에 따라 시간의 진행에 따라 일어나는 퇴행성 변화로 외적형태의 변화로는 피부 건조 및 주름 등이 있다. 주름을 생성하는 원인에는 자외선, 건조 및 물리적, 화학적 자극 등 환경요인에 기인하는 피부 스트레스와 각질층의 수분량 저하, 비후화, 진피의 교원섬유, 탄력섬유의 양적, 질적 변화 등이 유발하는 피부의 탄력성이나 신축성의 저하를 들 수 있다. 본 연구에서는 Angelim dahurica root에서 얻어진 3종의 주름개선 유효성분의 구조분석 및 피부 노화에 대한 효과를 살펴보았다.

  • Essential oil of the root of Angelica dahurica Benth et Hook(Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain eleven monoterpenes such as ${\alpha}-pinene(4.74%),\;campben, {\beta}-pinene,\;myrcene,\;{\alpha}-phellandrene,\;{\delta}-3-carene(39.4%),\;{\alpha}-terpinene,\;{\rho}-cymene,\;{\beta}-phellandrene,\;{\alpha}-terpinene,\;terpinolene\;and\;also\;found\;to\;contain\;4-vinylguaiacol,\;iso-elemicin,\;{\beta}-elemene$, caryophyllene, ligustilide, osthol and seven tentatively identified sesquiterpenes.

구릿대(Angelica dahurica) 뿌리의 성분 분석 (Analysis on the Components of the Angelica dahurica Root)

  • 구릿대 뿌리의 생리적 기능과 이용 가능성에 대한 연구의 일환으로 일반성분과 환원당, 유리당, 무기질 성분 및 아미노산의 함량과 아미노산 유도체 등에 대하여 분석하였다. 일반성분 중에는 수분 함유량이 71.7%로 가장 높았으며, 탄수화물 10.6%, 조단백질 9.3%, 조회분 6.6%, 그리고 조지방이 1.9%로 나타났다. 환원당의 함량은 1,850 mg/100 g이었으며, 유리당의 총 함량은 80 mg/100 g으로 sucrose의 함량이 28.4 mg/100 g으로 가장 높았다. 무기질은 K가 2,145.03 mg/100 g으로 가장 높았으며, Mg(286.35 mg/ 100 g), Ca(145.23 mg/ 100 g), Zn(4.78 mg/100 g), Fe(4.20 mg/100g), Mn(3.76 mg/100 g) 등이 검출되었다. 구성아미노산의 함량은 71.68 mg/ 100 g으로 필수아미노산이 20.98 mg/100 g, 비필수아미노산이 50.70 mg/100 g으로 분석되었으며 proline(11.74 mg/100 g)의 함량이 가장 높았다. 유리아미노산은 필수아미노산 6.67 mg/100 g, 비필수아미노산 10.37 mg/100 g으로 총 17.04 mg/ 100 g이 함유된 것으로 나타났으며, alanine(5.96 mg/100 g)의 함량이 가장 높았다. 아미노산 유도체의 함량은 3.37 mg/100 g으로 나타났다.
