  • Objectives : Angelica Gigantis Radix is prescribed as the root of different Angelica species on the pharmacopoeia in Korea, Japan and China. Chemical components and their biological activities were also different according to their species. A study for the development of simple method to compare Angelica roots was needed. In order to classify them, the methods such as DNA sequencing analysis and taste sensor were applied to three Angelica species like Angelica gigas, Angelica acutiloba and Angelica sinensis. Methods : PCR amplification of intergenic transcribed spacer (ITS) region was performed using ITS1 and ITS4 primer from nine Angelica roots, and then nucleotide sequence was determined. Taste pattern of samples were measured using the taste-sensing system SA402B equipped with a sensing unit, which consists of artificial lipid membrane sensor probes of anionic bitterness, astringency, saltiness, umami, and cationic bitterness (C00, AE1, CT0, AAE, and AN0, respectively). Results : As a result of comparing the similarity of the ITS region sequences, A. sinensis was discriminated from the others (A. gigas and A. acutiloba). Equally this genetic result, A. gigas and A. acutiloba showed similar taste pattern as compared to A. sinensis. Sourness, bitterness, aftertaste of bitterness, astringency, and aftertaste of astringency of A. sinensis were significantly high as compared with A. gigas and A. acutiloba. In contrast, richness was significantly low. Conclusions : These taste pattern can be used as a way of comparison of Angelica species and this technic could be applied to establish a taste pattern marker for standardization of herbs in various purposes.

  • Objectives : We compared with the effects of different parts (root head, root body and hairy root) of Angelica gigas Nakai (Angelicae Gigantis Radix, AG) with on middle cerebral artery occlusion(MCAO)-induced ischemic rats, and on LPS-induced inflammatory response in BV2 microglia. Methods : The 30% ethanol and water extracts of different parts of AG were prepared. Each extract (50 and 100 mg/kg) was administrated intraperitoneally once in MCAO-induced ischemic rats. We measured infarction volumes by TTC staining, and investigated the expression of iNOS, Bax, Bcl-2 and caspase-3 by Western blot. BV2 cells were treated with each extract for 30 min, and then stimulated with LPS. The levels of NO was measured by Griess assay. The expression of iNOS, Cox-2 and proinflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$, and IL-6) were determined RT-PCR and Western blot. The phosphorylation of ERK1/2 and JNK MAPK was determined by Western blot. Results : Among different parts of AG, the 30% ethanol and water extracts of hairy root significantly decreased infarction volume in ischemic brains and inhibited the expression of iNOS, bax and caspase-3. The extracts of hairy root significantly inhibited LPS-induced production of NO, $TNF-{\alpha}$ and IL-6 in BV2 cells, and suppressed the expression of iNOS and COX-2. The hairy root extracts attenuated LPS-induced phosphorylation of ERK1/2 and JNK MAPK in BV2 cells. Conclusions : Our results indicate that the root hairy of AG has a good neuroprotective and anti-inflammatory effects in ischemic stroke compared to other parts.

  • The purpose of the present study is a comparison of the antioxidant effects of Angelica gigas Korea (AG), Angelica sinensis of China (AS), and Angelica acutiloba of Japan (AA), and comparison of the effects of AG, AS and AA on tight-junction related genes in human keratinocyte HaCaT cells. All species showed a strong antioxidant effect, and AA was higher than AG and AS in antioxidant effects. The cytotoxicity was confirmed to be higher in AS than AG and AA at a concentration of $1,600{\mu}g/ml$ using the MTS assay in HaCaT cells. We analyzed the effects of AG, AS, and AA on mRNA expression levels of various tight-junction related genes in HaCaT cells. We found that no obvious changes in expression of Claudin 1, 3, 4, 6, 7, 8, Occludin, JAM-A, ZO-1, ZO-2, and tricellulin by treatment of all species, suggesting that there is less possibility of side effects and skin moisturizing effects due to changes in tight-junction gene expression. Our results suggest that AG, AS, and AA are thought to be effective in reducing the oxidative stress of the skin and preventing the aging of the skin.

  • This study was performed to elucidate the influence on the plasma decursinol concentration after administration of combined decursin and Cnidii Rhizoma or decurisin and Bupleuri Radix extracts in comparison with single decursin administration. The identification of decursinol isolated from Angelica gigas was carried out by GC/MS and the concentration of decursinol in the plasma after oral administration was determined by HPLC. The value of area under the plasma concentration (AUC) and the maximal concentration (Cmax) of decursinol after administration of methanol and ether Angelicae Gigantis Radix extract was higher than those of single decursin-treated group. It was also found that the AUC and the Cmax of decursinol after combined administration with decursin and Cnidii Rhizoma were higher than those of decursin administration, whereas those of combined with decursin and Bupleuri Radix was lower. Moreover, the studies were performed with decursinol only or with combined decursinol and Cnidii Rhizoma/Bupleuri Radix, both of combined treatment increased the plasma decursinol concentrations compared with decursinol-treated group in rats. On the basis of these results, it is concluded that co-existing substances or co-administration with other drugs may influence the plasma levels of decurisinol after oral administration.

  • Objectives : Araliae Continentalis Radix and Angelicae Pubescentis Radix have been used as the same medicinal name Korean and Chinese traditional medicines, respectively. The authentic Araliae Continentalis Radix is described only the root of Aralia continentalis in the Korean Pharmarcopoeia. However, the dried root of Angelica biserrata, Levisticum officinale, or Heracleum moellendorffii also has been distributed adulterants of Araliae Continentalis Radix. To develop a reliable method for identifying Araliae Continentalis Radix from adulterants, we carried out the analyses of universal DNA barcode sequences. Methods : Four plants species were collected from different habitate and nucleotide sequences of matK and rbcL were analyzed. The species-specific sequences and phylogenetic relationship were estimated using entire sequences of two DNA barcodes, respectively. Results : In comparative analysis of matK sequences, we were identified 104 positions of marker nucleotide for Ar. continentalis, 3 for An. biserrata, 4 for L. officinale and 8 for H. moellendorffii enough to distinguish individual species, respectively. Furthermore, we obtained marker nucleotides in rbcL at 42 positions for Ar. continentalis, 5 for An. biserrata and 2 for H. moellendorffii, but not for L. officinale. The phylogenetic tree of matK and rbcL were showed that all samples were clustered into four groups constituting homogeneous clades within the species. Conclusions : We confirmed that species-specific marker nucleotides of matK sequence provides distinct genetic information enough to identify four species. Therefore, we suggest that matK gene is useful DNA barcode for discriminating authentic Araliae Continentalis Radix from inauthentic adulterants.

  • Objective : The purpose of this study was to investigate the synergistic effect of Angelicae gigantis Radix (AG) and Lycii fructus (LF) ethanol extracts on skin-whitening effects. Method : LFAG extracts were prepared by extracting with 80% ethanol. The efficacy of LFAG was judged by measurement of cell viability, tyrosinase activity, melanin production, tyrosinase and microphthalmia-associated transcription factor (MITF) expression in B16F10 murine melanoma cells by lipopolysaccharides (LPS) treatment. Results : Each extract (LF or AG) inhibited the tyrosinase activity in a dose-dependent manner. The co-treatment of LFAG extracts ($25{\mu}g/mL$ LF plus $25{\mu}g/mL$ AG) markedly suppressed the LPS-induced cellular tyrosine activity, melanin production, tyrosinase and MMP-1 expression in B16F10 murine melanoma cells. These suppressive effects were synergistically increased by their combination. Conclusions : With these observations, we suggest that the extracts from Lycii fructus and Angelica gigantis Radix could be potent natural materials for whitening skin.
