Antimicrobial Effect of Monascus Strains Isolated from Ang-Khak (홍국으로부터 불리한 Monascus 균주의 향균효과)

  • Total 29 Monascus strains were isolated from Ang-Khak and 4 of them were selecte based on the relative intensity of soluble red pigment and growth rate. The optimum growth temperature of the selected isolates was 32.5$^{\circ}C$ on malt extract agar(MEZ) plate. The optimum growth pH was 5.0 on czapek yeast extract agar plate, while it was pH 6.2 or 6.5 on MEA plate. Isolate No. 116, especially, showed the strongest animicrobial activity aganist Bacillus subtilis and Staphylococcus aureus but much less aganist Escherichia coli and Enterobacter aerogenes. The maximun antimicrobial activity of isolate No. 116 against St. aureus was achieved at initial pH 5.3 on rice extract broth. The acitivity was increased with increasing amount of culture broth concentrate of isolate No. 116.

Studies on the Storage of Functional Red Soybean Curd (기능성 홍두부의 저장성)

  • We prepared a functional red soybean curd (RS) from Ang-Khak according to various concentration (RS1 : red rice powder 0.1 g/10 mL, RS2: 0.2 g/10 mL, RS3: 0.3 g/10 mL, RS4: 0.4 g/10 mL, RS5: 0.5 g/10 mL, CS: control soybean curd). During the storage period of red soybean curd, pH, acidity, and microbial counts showed a minimum change in RS5. And in the case of color, all groups have slightly increased in yellowness except RS1. In the case of texture, hardness, gumminess, and springiness appeared to be increased for a few days and then fell down. As water drained out of soybean curd during storage at 1$0^{\circ}C$, it became so stiff and then rotten that the hardness of soybean curd increased in the early stage and then decreased after all. After a week, we could find a better preservative effect of RS than CS. Resultly, we need more efforts to prolong the shelf-life of soybean curd with applying the functionality of Ang-Khak.

Screening of Monascus Strains for Antimicrobial Activity and Effect of Change of Nutrients and Incubation Conditions on Antimicrobial Activity (항균성 Monascus 균주의 Screening 및 영양원과 배양조건이 항균활성에 미치는 영향)

  • Monascus strains were isolated from Ang-Khak for the screening of antimicrobial activity. Two Monascus isolates, No.116 and No.481, were selected because they showed strong antimicrobial activity. Effect of various nutrients and incubation conditions on antimicrobial activity were different between two isolates. Strong antimicrobial activity of isolate No.116 was observed in the medium with 8% sucrose and $0.8%(NH_4)_2SO_4,$ 0.5% $KH_2PO_4and$ 0.5% $MgSO_4,$ while isolate No.481 required 8% sucrose, 1.6~2% $(NH_4)_2SO_4,$ 0.5% $MgSO_4and$ 0.5% $FeSO_4for$ the highest activity. The strong antimicroial activity was observed when both isolates were incubated on rice extract broth with initial pH of 5.3. The optiimum incubation temperature for the highest antimicrobial activity was $32.5^{\circ}C.$ With optimal conditions for the highest antimicrobial activity, isolate No.116 and No. 481 were both active for 51 hours or longer against test organisms Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, and Enterococcus faecium.

Optimization of Red Pigmentation and Effect of the Metabolites Produced by Monascus Strains on Microbial Inhibition and Colorization in Processed Ham (Monascus 균주의 적색색소 생산 특성과 육제품에서의 항균 및 착색 효과)

  • In this study, we tested possibility of replacing nitrite salts, which were always added during the meat product processing, with the metabolites produced by antimicrobial and red pigment producing Monascus strains. We have already shown that Monascus No. 116 strain has the highest antimicrobial activity among the strains isolated from Ang-Khak. Monascus isolate No. 229 was chosen due to its outstanding red pigment producing ability. The red pigment production by No. 229 was highest in the medium containing 8% sucrose, 2% yeast extract, 0.1% K2HPO4, 0.5% MgSO4. Optimum pH and temperature for the red pigment production were pH 6.2 and 3$0^{\circ}C$, was found in spot or Rf value 0.54 on TLC plate using ethyl acetate-acetone-water (4:4:1, v/v/v) as development solvent system. Isolate No. 116 and No. 229 were cultured in a optimal condition for the antimicrobial activity and red pigmentation. The culture concentrates were applied in situ to the production of instantly processed ham. Mixed application of 89 ppm Na-nitrite and 300 ppm of culture broth concentrate of Monascus isolate No. 116 and 500 ppm of red color produced by Monascus isolate No. 229 showed similar results with the single application of 94 ppm Na-nitrite. These results confirmed that the antimicrobial activity and red pigment of Monascus strains might be valuable to replace Na-nitrite salt in meat processing.
