Lab-on-a-chip is a miniaturized analytical device in which all of the procedures for the analysis of molecules are carried out, such as pretreatment, reaction, separation, detection, etc. Lab-on-a-chip has increasing concern as a device not only for rapid detection of molecules but also for high throughput screening and point of care, because conventional laborious and time consuming analytical procedures can be substituted. Thus, a lot of microfabrication and analytical techniques for lab-on-a-chip have been developed with microstructures smaller than a few hundreds of micrometers. Separation of the molecules is one of the most important components of lab-on-a-chip, because effective separation method can simplify the design and can provide better sensitivity. The electrokinetic separation based on capillary electrophoresis is most widely employed technique in lab-on-a-chip for the control of fluids and the separation of molecules. In this article, bioseparation techniques and its applications realized in lab-on-a-chip are reviewed.
Recently there have been increasing consumers' interests in the geographical origin of foods, due to the FTA (Free Trade Agreement) in the global market. Especially, in Korea, in relation to BSE (bovine spongiform encephalopathy), it is considered to be urgent to develop analytical techniques for distinguishing the geographical origin of beef. Korea is facing conclusion of FTA with many countries, and there is a deep national concern about the distinction of the geographical origin of food. Diverse analytical techniques have been used in many of recent researches to obtain data for distinguishing the geographical origin of foods produced in various countries. In this paper, we reviewed national and international researches about tracing of geographical origin and food authentication using stable isotopes. Improvement of the isotopic techniques and their numerous application have been provided useful information of their geographical origin in food products. Furthermore, we expect that this study could be detecting of many frauds and illegal transaction of food products. We look forward to active progressing research of detecting food origin using isotope analysis and numerous application about imported food products.

A comprehensive mapping project for agroclimatic zoning in South Korea will end by April 2010, which has required 4 years, a billion won (ca. 0.9 million US dollars) and 22 experts from 7 institutions to complete it. The map database from this project may be categorized into primary, secondary and analytical products. The primary products are called "high definition" digital climate maps (HD-DCMs) and available through the state of the art techniques in geospatial climatology. For example, daily minimum temperature surfaces were prepared by combining the climatic normals (1971-2000 and 1981-2008) of synoptic observations with the simulated thermodynamic nature of cold air by using the raster GIS and microwave temperature profiling which can quantify effects of cold air drainage on local temperature. The spatial resolution of the gridded climate data is 30m for temperature and solar irradiance, and 270m for precipitation. The secondary products are climatic indices produced by statistical analysis of the primary products and includes extremes, sums, and probabilities of climatic events relevant to farming activities at a given grid cell. The analytical products were prepared by driving agronomic models with the HD-DCMs and dates of full bloom, the risk of freezing damage, and the fruit quality are among the examples. Because the spatial resolution of local climate information for agronomic practices exceeds the current weather service scale, HD-DCMs and the value-added products are expected to supplement the insufficient spatial resolution of official climatology. In this lecture, state of the art techniques embedded in the products, how to combine the techniques with the existing geospatial information, and agroclimatic zoning for major crops and fruits in South Korea will be provided.

Untargeted metabolomics is a useful tool for drug development focusing on novel chemotherapeutic and chemopreventative agents against cancer cells. In recent years, quadrupole time of flight liquid chromatography-mass spectrometry (Q-TOF LC/MS)-based untargeted metabolomic approaches have gained importance to evaluate the effect of these agents at the molecular level. The researchers working on cell culture studies still do not apply standardized methodologies on sample preparation for untargeted metabolomics approaches. In this study, the rough and wet lysis techniques performed on MCF-7 breast cancer cells were compared with each other via the Q-TOF LC/MS-based metabolomic approach. The C18 and hydrophilic interaction liquid chromatography (HILIC) columns were used for the separation of the metabolites in MCF-7 cell lysates. 505 peaks were detected through the HILIC column and 551 peaks were found through the C18 column for the wet lysis technique. This situation supported by the base peak chromatograms showed that the wet lysis technique allowed us to extract higher number of non-polar metabolites. Almost equal number of metabolites was found for the C18 and HILIC columns (697 peaks for the HILIC column and 695 peaks for the C18 column) when the rough lysis technique was used. However, the intensities of polar metabolites were higher for the rough lysis technique on base peak chromatograms for both the HILIC and C18 columns. Although cell lysis technique, which is the first step in the sample preparation for cell culture studies, does not cause dramatic differences in the number of the detected metabolite peaks, it affects the polar and non-polar metabolite ratio significantly. Therefore, it must be considered carefully especially for in vitro drug development studies.
The techniques for manipulation of various micro-tools were essential for particle isolation and chemical analysis of micro-size particles. This report described the detailed techniques for the preparation and handling of several micro-tools. Presence of uranium particles in smeared filter paper were identified by using the solid track detector. The uranium particles were isolated using the micro-tools under the stereomicroscope and then transferred to the filament of TIMS for the determination of isotope ratios.
Dissolved organic matter (DOM), which changes according to various factors, is ubiquitously present from natural environments to engineered treatment systems. Only limited information is available regarding the environmental functions of DOM after bulk analyses are only applied for characterization. In this paper, latest DOM analytical techniques are briefly introduced, which include fluorescence excitation-emission matrix with parallel factor analysis (EEM-PARAFAC), size-exclusion chromatography with an organic carbon detector (SEC-OCD), carbon/nitrogen stable-isotope ratio, and Fourier transform-ion cyclotron resonance-mass spectroscopy (FT-ICR-MS). Recent examples of using advanced analyses to interpret the phenomena associated with DOM occurring in natural and engineered systems are presented here. Through EEM-PARAFAC, different components like protein-like, fulvic-like, and humic-like can be identified and tracked individually through the investigated systems. SEC-OCD allows researchers to quantify different size fractions. FT-ICR-MS provides thousands of molecular formulas present in bulk DOM samples. Lastly, carbon/nitrogen stable-isotope ratio offers reasonable tools for tracking the sources in environments. We also discuss the advantages and weakness of the above-mentioned characterizing tools. Specifically, they focus on single environmental factors (different sourced-DOM and interaction of sediment-pore water) or simple changes after individual treatment processes. Through collaboration with the advanced techniques later, they help the researchers to better understand environmental behaviors in aquatic systems and serve as essential tools for addressing various pending problems associated with DOM.
Current status of soil contamination with fluorine and its source were investigated. The basic principles and procedures of various techniques for the analysis of fluorine contents in soil and solid phase samples were summarized in this review. Analysis of fluorine in solid matrices can be achieved by two types of techniques: (i) UV/Vis spectrophotometer or ion selective electrode (ISE) analysis after performing appropriate extraction steps and (ii) direct solid analysis. As the former cases, the standard method of Korean ministry of environment, alkali fusion-ISE method, pyrohydrolysis, oxygen bomb combustion, aqua regia digestion-automatic analysis, and sequential extraction-ISE method were introduced. In addition, direct analysis methods (i.e., X-ray fluorescence spectrometry and proton induced gamma-ray emission spectrometry) and atomic spectrometry combining with the equipment for introducing solid phase sample were also reviewed. Fluorine analysis techniques can be reasonably selected through site-specific information such as matrix condition, contamination level, the amount of samples and the principles of various methods for the analysis of fluorine presented in this review.

Safety-related equipments of nuclear power plant must be seismically qualified to demonstrate their ability to function as required during and/or after the earthquake, The seismic qualification is usually achieved through analysis and testing. Analysis method is preferably adopted for structurally simple equipments which are easy to be mathematically modeled. However even for relatively complex equipments analysis method is occasionally used for computing the input motion or supporting information for the component test followed. Electrical cabinet is a typical example for which analysis method is combinedly used with test to get modal properties of the enclosing cabinet structure. Usually the structural elements and doors of the cabinet are loosely interconnected with small-size bolts or spot welding. Therefore cabinet-type equipment usually has high and complex nonlinear properties which are not easily idealized by simple practical modeling techniques. in this paper with respect to a typical cabinet-type structure(instrumentation cabinet of nuclear power plant) a comparative study has been performed between three different state-of-the -art modeling techniques: lumped mass model frame model and FEM modal. Form the study results it has been found that modal properties of the cabinet-type structure in the elastic behavior range can be reasonably computed through any type of modeling techniques in the practice with slight modification of model properties to get better accuracy. However it needs additional modeling techniques to get reasonable results up to nonlinear range.
