Characteristics and breeding of a new cultivar Pleurotus eryngii, Song-A (큰느타리버섯 신품종 '송아'의 육성 및 그 특성)

  • To develop a new cultivar of King oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strains derived from ASI 2824(Keunneutari No.2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-51($G09-21-10{\times}2844-11$) was shown the best cultural characteristics, selected to be a new cultivar and named as 'Song-A'. The 'Song-A' was formed incompatibility line distinctly in the confrontation growth of parental strains Keunneutari No.2, Aeryni 3 and ASI 2844. The optimum temperature for mycelial growth, fruiting body development and pH arrange were $25{\sim}30^{\circ}C$, $14{\sim}16^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about $94.7{\pm}29.5$ g which is almost 106% quantity compared to that of other cultuvar Keunneutari No.2. And also the stip is thick and long but the number of available stipe is few. Analysis of the genetic characteristics of the new cultivar 'Song-A' showed a different DNA profile as that of the control strains, Keunneutari No.2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primers URP4 and 7 were used. This new cultivar 'Song-A' of Pleurotus eryngii is characterized by a small number of primordia formation and the stip is thick and long. Therefore, we expect that this new strain will save of labor and cost by without culling work.

Independent Prognostic Value of the Fascin Expression in Patients with Esophageal Cancer (식도암에 있어서 Fascin의 발현과 예후와의 상관관계에 대한 연구)

  • Background: Fascin is an actin-bundling protein that induces membrane protrusions and it increases cell motility in various transformed cells. Esophageal cancer is one of the most lethal malignancies, and it exhibits extensive local invasion or frequent regional lymph node metastasis even after curative surgery. We investigate the expression of fascin by performing immunohistochemistry to evaluate the clinical characteristics and prognostic significance of its expression in esophageal cancer patients. Material and Method: Immunochemistry for fascin was performed on 76 tumor samples from 76 patients who underwent esophageal cancer operations. The expression levels of fascin in the 76 esophageal cancer tissues were compared with those in the corresponding normal esophageal epithelium. The fascin-positive samples were defined as those showing more than 75% of fascin-positive cells. Result: Overall, a fascin positive expression was detected in 39 (51.3%) out of the total 76 cases. The tumors with positive fascin expression tended to more frequently show a higher stage (p=0.030), and a higher T-factor (p=0.031). The prognosis of the fascin negative group was significantly better than that of the fascin positive group (p=0.004). Multivariate analysis revealed that lymphovascular invasion and the fascin expression were independent prognostic factors. Conclusion: Fascin was expressed in 513% of the esophageal cancer tissues and a positive expression of fascin was associated with more advanced tumor progression and recurrence. Our study suggests that the fascin expression may be an independent prognostic factor for an unfavorable clinical course few those patients suffering with esophageal cancer.

Clinical Outcome of Emergency Coronary Artery Bypass Grafting after Failed Percutaneous Transluminal Coronary Angioplasty (경피적 관상동맥 성형술의 실패에 의해 발생한 응급환자의 임상경과)

  • Background: Failed percutaneous transluminal coronary angioplasty (PTCA) is occasionally required for emergency coronary artery bypass grafting (CABG). The aim of this study was to assess the outcome of patients receiving emergency CABG after failed PTCA. Material and Method: Between May 1988 and May 2005, 5712 patients underwent PTCA, where 84 (1.4%) failed. 27 patients underwent emergency CABG after failed PTCA. The mean age was $63.7{\pm}8.9\;(46{\sim}80)$ years, with 14 male patients (51.9%). Result: All patients underwent emergent surgical revascularization within 6 hours. 22 patients underwent conventional CABG and 5 underwent off-pump CABG. The causes of PTCA failure were coronary obstruction due to new thrombi formation during the procedure (n=4), coronary dissection (n=17), coronary artery rupture (n=3) and 3 due to other causes. The rate of in-hospital operative mortality after emergent operation was 18.5% (5/27). A univariate analysis revealed that patients who died more often had left anterior descending artery disease, a preprocedural shock status, postoperative use of multiple isotropics and postoperative use of intra-aortic balloon pump. The mean follow up duration was $53.6{\pm}63.4$ months. Conclusion: Although PTCA is known to be life saving, there is still a high risk for morbidity and mortality following emergency CABG after failed PTCA, despite the advancement in PTCA techniques. This result will help identify and more effectively treat patients selected for PTCA when emergency CABG is required.

Clinical Analysis of Pulmonary Hamartoma: 37 Cases (폐과오종 37예에 대한 임상 고찰)

  • Background: Pulmonary hamartomas are the most common form of pulmonary benign tumors, and they occur in approximately $2{\sim}5%$ of all pulmonary neoplasm. However, only a few reports have been published on the clinical characteristics of pulmonary hamartoma in Korea. Material and Method: The charts, X-rays and pathological specimens of 37 pulmonary hamartoma patients who were diagnosed by a pathological examination from January of 2000 to May of 2005 at the Catholic Medical Center were retrospectively reviewed. Result: The peak incidence of the tumor occurred in the seventh decade of life (32.4%), There were 23 men (62.6%) and 14 women (37.8%), with mean age of 55.6 years. Twenty-six patients (70.3%) were asymptomatic and 11 patients (29.7%) had symptoms. A total of 29 tumors (78.4%) were parenchymal, and 8 (21.6%) were endobronchial. Twenty cases (54.1%) were in the right lung and 17 cases (45.9%) were in the left lung. The right lower lobe was most commonly involved. Thirty-two (86.5%) hamartomas were diagnosed by surgical resection, 4 cases (10.8%) were diagnosed by bronchoscopic biopsy and 1 case (2.7%) was diagnosed by a fine needle aspiration biopsy. Thirty-four hamartomas (91.9%) were managed by surgical resection without complication. No recurrence or malignant changes were seen during the follow up period. Conclusion: Pulmonary hamartomas are most common in males during their fifth to seventh decade and they more commonly involve the right lung. No recurrence or malignant changes were seen during the follow up period.

Construction of a Fluorescently Labeled Infectious R Peptide-Less Moloney MLV Molecular Clone for Analysis of Syncytium (합포체 분석을 위해 R 펩타이드가 결여된 형광 표지 Moloney 마우스레트로바이러스 Molecular Clone 제조)

  • Retroviruses enter host cells by membrane fusion between the viral Env proteins on the virus membrane and a virus receptor on the cellular membrane. The envelope protein of the ecotropic Moloney murine leukemia virus is synthesized as a gp85 precursor and is proteolytically cleaved into an extracellular surface unit (SU) and the transmembrane protein (TM). The cytoplasmic tail (16 amino acid; R peptide) of the TM protein is further cleaved by the viral protease during virion maturation. Unlike the wild type Env protrin bearing the R peptide, R peptide-truncated Envelope induces syncytia in susceptible cells. To understand the mechanism of R peptidetruncated Env in syncytium formation, R peptide-truncated Env expressing full-length molecular clone containing EGFP in PRR (proline rich region) of Env was constructed. This molecular clone induced syncytia in transfected NIH3T3 cells, fluorescence was detected in the cytoplasm and at the plasma membrane, while the nuclei did not stain and appeared black by fluorescence microscopy. Interestingly, virions with truncated envelope produced from transfected NIH3T3 cells induced syncytia in NIH3T3 cells, but fluorescence was not detected in the same infected cells. It is believed that cell-free viruses direct the fusion of neighboring cells without infection. Our data suggests that use of EGFP-tagged envelope for monitoring syncytium is a sensitive and convenient method. We also found that virion incorporated the R peptide-truncated Env is able to induce the formation of syncytia by fusion from without.

Development of Prevotella nigrescens ATCC $33563^T$-Specific PCR Primers (Prevotella nigrescens ATCC $33563^T$ 균주-특이 중합효소연쇄반응 프라이머 개발)

  • A Pn10 DNA probe was introduced as a Prevotella nigrescens ATCC $33563^T$-specific DNA probe. In that study, the specificity of the Pn10 was tested with only type or reference strains of 5 oral bacterial species. The purpose of this study is to evaluate the specificity of the Pn10 using the wild type strains of P. nigrescens and is to develop the P. nigrescens ATCC $33563^T$-specific PCR primers based on the nucleotide sequence of the Pn10. The specificity of the Pn10 DNA probe was determined by Southern blot analysis. The nucleotide sequence of Pn10 DNA probes was determined by chain termination method. The PCR primers were designed based on the nucleotide sequence of cloned DNA fragment. The data showed that Pn10 DNA probe were hybridized with the genomic DNAs from P. nigrescens ATCC $33563^T$ and KB6. The Pn10 homologous region, KB6-Pn10, of P. nigrescens KB6 was cloned by PCR and sequenced. The Pn10 and KB6-Pn10 DNA fragments were consisted of 1,875 bp and 1,873 bp, respectively. The percent identity of the two was 98.8% and the divergence of them was 0.6%. The two primer sets (Pn10-F-AC/ Pn10-R-AC and Pn10-F-A/ Pn10-R-A), designed base on the nucleotide sequences of Pn10 DNA probe, were specific to the P. nigrescens ATCC $33563^T$. The two PCR primer sets could detect as little as 4 pg of genomic DNA of P. nigrescens ATCC $33563^T$. These results indicate that the two PCR primer sets have proven useful for the identification of P. nigrescens ATCC $33563^T$, especially with regard to the maintenance of the strain.

A Study on the Development of a Simulation Model for Predicting Soil Moisture Content and Scheduling Irrigation (토양수분함량 예측 및 계획관개 모의 모형 개발에 관한 연구(I))

  • Two types of model were established in order to product the soil moisture content by which information on irrigation could be obtained. Model-I was to represent the soil moisture depletion and was established based on the concept of water balance in a given soil profile. Model-II was a mathematical model derived from the analysis of soil moisture variation curves which were drawn from the observed data. In establishing the Model-I, the method and procedure to estimate parameters for the determination of the variables such as evapotranspirations, effective rainfalls, and drainage amounts were discussed. Empirical equations representing soil moisture variation curves were derived from the observed data as the Model-II. The procedure for forecasting timing and amounts of irrigation under the given soil moisture content was discussed. The established models were checked by comparing the observed data with those predicted by the model. Obtained results are summarized as follows: 1. As a water balance model of a given soil profile, the soil moisture depletion D, could be represented as the equation(2). 2. Among the various empirical formulae for potential evapotranspiration (Etp), Penman's formula was best fit to the data observed with the evaporation pans and tanks in Suweon area. High degree of positive correlation between Penman's predicted data and observed data with a large evaporation pan was confirmed. and the regression enquation was Y=0.7436X+17.2918, where Y represents evaporation rate from large evaporation pan, in mm/10days, and X represents potential evapotranspiration rate estimated by use of Penman's formula. 3. Evapotranspiration, Et, could be estimated from the potential evapotranspiration, Etp, by introducing the consumptive use coefficient, Kc, which was repre sensed by the following relationship: Kc=Kco$.$Ka+Ks‥‥‥(Eq. 6) where Kco : crop coefficient Ka : coefficient depending on the soil moisture content Ks : correction coefficient a. Crop coefficient. Kco. Crop coefficients of barley, bean, and wheat for each growth stage were found to be dependent on the crop. b. Coefficient depending on the soil moisture content, Ka. The values of Ka for clay loam, sandy loam, and loamy sand revealed a similar tendency to those of Pierce type. c. Correction coefficent, Ks. Following relationships were established to estimate Ks values: Ks=Kc-Kco$.$Ka, where Ks=0 if Kc,=Kco$.$K0$\geq$1.0, otherwise Ks=1-Kco$.$Ka 4. Effective rainfall, Re, was estimated by using following relationships : Re=D, if R-D$\geq$0, otherwise, Re=R 5. The difference between rainfall, R, and the soil moisture depletion D, was taken as drainage amount, Wd. {{{{D= SUM from { {i }=1} to n (Et-Re-I+Wd)}}}} if Wd=0, otherwise, {{{{D= SUM from { {i }=tf} to n (Et-Re-I+Wd)}}}} where tf=2∼3 days. 6. The curves and their corresponding empirical equations for the variation of soil moisture depending on the soil types, soil depths are shown on Fig. 8 (a,b.c,d). The general mathematical model on soil moisture variation depending on seasons, weather, and soil types were as follow: {{{{SMC= SUM ( { C}_{i }Exp( { - lambda }_{i } { t}_{i } )+ { Re}_{i } - { Excess}_{i } )}}}} where SMC : soil moisture content C : constant depending on an initial soil moisture content $\lambda$ : constant depending on season t : time Re : effective rainfall Excess : drainage and excess soil moisture other than drainage. The values of $\lambda$ are shown on Table 1. 7. The timing and amount of irrigation could be predicted by the equation (9-a) and (9-b,c), respectively. 8. Under the given conditions, the model for scheduling irrigation was completed. Fig. 9 show computer flow charts of the model. a. To estimate a potential evapotranspiration, Penman's equation was used if a complete observed meteorological data were available, and Jensen-Haise's equation was used if a forecasted meteorological data were available, However none of the observed or forecasted data were available, the equation (15) was used. b. As an input time data, a crop carlender was used, which was made based on the time when the growth stage of the crop shows it's maximum effective leaf coverage. 9. For the purpose of validation of the models, observed data of soil moiture content under various conditions from May, 1975 to July, 1975 were compared to the data predicted by Model-I and Model-II. Model-I shows the relative error of 4.6 to 14.3 percent which is an acceptable range of error in view of engineering purpose. Model-II shows 3 to 16.7 percent of relative error which is a little larger than the one from the Model-I. 10. Comparing two models, the followings are concluded: Model-I established on the theoretical background can predict with a satisfiable reliability far practical use provided that forecasted meteorological data are available. On the other hand, Model-II was superior to Model-I in it's simplicity, but it needs long period and wide scope of observed data to predict acceptable soil moisture content. Further studies are needed on the Model-II to make it acceptable in practical use.

Evaluation of Reproductive Growth in a Mature Stand of Korean Pine under Simulated Climatic Condition (국지기후가 잣나무 성숙임분의 생식생장에 미치는 영향분석)

  • This study was conducted to reveal the effects of local climatic conditions on reproductive growth in a mature stand of Korean white pine based on climatic estimates. For this, the reproductive growth such as production and characteristics of cone and seed were first measured and summarized for seven years from 1974 to 1980. The local climatic conditions in the study site were also estimated by both a topoclimatological method and a spatial statistical technique. The local climatic conditions were then correlated with and regressed on the growth factors to reveal the relationships between the climatic estimates and the reproductive growth. Average number of conelet formation per tree showed highly negative correlation with some climatic variables related to minimum temperature in the year of flower bud differentiation. Especially, the most significant negative correlation were found between average of the minimum temperature for June and July of flower bud differentiation year and the number of conelet formation. There was no significant correlation between the number of cone production and climatic variables. However, total precipitation from December of the flowering year to February of the cone production year showed the most high correlation (r=0.6036) with the number of cone production. It was found that significant climatic variables affecting the amount of cone drop and cone drop percentage were the sum of cloudy days from June of the flowering year to August of the cone production year. Positive correlation was significantly recognized between the average weight of empty seed per cone and total precipitation from December of the flowering year to February of the cone production year. For the percentage of empty seed, five climatic variables among 19 variables were significantly correlated at 10% level. The average weight of a cone showed negative correlation with total precipitation from June of the flowering year to August of the cone production year. It was also found that average weight of a seed had highly negative correlation with total precipitation from December of the flowering year to February of the cone production year. The average weight of cone coat was negatively correlated with two climatic variables derived from clear days, which are sum of clear days from November of the flowering year to March of the cone production year and sum of clear days from December of the flowering year to February of the cone production year. On the other hand, it showed positive correlation with mean temperature of May in the flowering year. The exactly same results were obtained in correlation analysis for the percentage of cone coat.

  • Tourism is multi-faceted phenomenon which involves movement to stay in destinations outside the normal place of residence. Thus, tourism is a composite phenomenon which incorporates the diversity of variables and relationships to be found in the tourist travel process. Tourism-behaviors are of major economic and social significance. With the growth of mass-tourism, the government departments of tourism is to provide many tourist facilities, to develop travel destination and tourism resources. At same time, the growth of mass-tourism has prompted perceptive travellers to raise many questions concerning the many travel destination and the tourism resources of encouraging further tourism demand. Most of tourism have a mental in their minds about various objects. This allowed him to choose tourism destination as a important rules. In tourism geography, behavioral approach to wildness, tourism resources and places has risen since 1960's. The tourist profile can be viewed under two major categories: the tourist social-economic and behavioral characteristic. Particulary, the motivations, attitude, need, values and preference of travel destinations are of crucial importance in contributing to their desion-making process. In this view, this paper is emprical study investigated travel patterns and behavioral characteristic of tourists and potential tourist in East Chonnam. The purpose of this paper is to show the spatial preference and tourism behavior of travel destinations. For this study, we are undertaken the questionary method employed by recreation geographers in order to collect research data for the East Chonnam-citizens functioned as a major tourist demand. The East Chonnam Province have many tourist-places including national park, provincial park, and local tourist-places. Thus, citizen of the East Chonnam evaluate many tourist-places very high, but evaluate the facilities of tourist place very low. The high ranks of preferences to travel destinations among tourist-places are Hyangilam, Geomoon island, Odongis land, Songkwang temple. The major travel destinations occurred the temporary travel trip are Odong island, Sunam temple, Songkwang temple. Heungkook temple, Hyangilam which are the type of mountainous in located inland. The relationship between the degree of preference and tourism behavior of travel trip for the travel destinations does not correlate each other. The result of analysis about the degree of seasonal preferences presented spatial differences according to characteristics of tourist-places. The typical travel destination are divided into seasons : Odong island. Mt. Backwoon, Goemoon island of spring, Goemoon island, Sungbul valley, Banggukpo beach of summer, Songkwang temple, Mt. Pal young of fall. The future of tourism will be rapidly grow with increase of personal mobility and leisure time, chang of tourism behavior. Thus, it is imperative that planning and development for tour-root, facilities of tourist-places should be implemented to increase tourism demand.

The Cross-Sectional Characteristic and Spring-Neap Variation of Residual Current and Net Volume Transport at the Yeomha Channel (경기만 염하수로에서의 잔차류 및 수송량의 대조-소조 변동과 단면 특성)

  • The object of this study is to estimate the net volume transport and the residual flow that changed by space and time at southern part of Yeomha channel, Gyeonggi Bay. The cross-section observation was conducted at the mid-part (Line2) and the southern end (Line1) of Yeomha channel for 13 hours during neap and spring-tides, respectively. The Lagrange flux is calculated as the sum of Eulerian flux and Stokes drift, and the residual flow is calculated by using least square method. It is necessary to unify the spatial area of the observed cross-section and average time during the tidal cycle. In order to unify the cross-sectional area containing such a large vertical tidal variation, it was necessary to convert into sigma coordinate system by horizontally and vertically for every hour. The converted sigma coordinate system is estimated to be 3~5% error when compared with the z-level coordinate system which shows that there is no problem for analyzing the data. As a result, the cross-sectional residual flow shows a southward flow pattern in both spring and neap tides at Line2, and also have characteristic of the spatial residual flow fluctuation: it northwards in the main line direction and southwards at the end of both side of the waterway. It was confirmed that the residual flow characteristics at Line2 were changed by the net pressure due to the sea level difference. The analysis of the net volume transport showed that it tends to southwards at $576m^3s^{-1}$, $67m^3s^{-1}$ in each spring tide and neap tide at Line2. On the other hand, in the control Line1, it has tendency to northwards at $359m^3s^{-1}$ and $248m^3s^{-1}$. Based on the difference between the two observation lines, it is estimated that net volume transport will be out flow about $935m^3s^{-1}$ at spring tide stage and about $315m^3s^{-1}$ at neap tide stage as the intertidal zone between Yeongjong Island and Ganghwa Island. In other words, the difference of pressure gradient and Stokes drift during spring and neap tide is main causes of variation for residual current and net volume transport.
