In the present study, an anaerobic microbial consortium for the degradation of hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) was selectively enriched with the co-addition of RDX and starch under nitrogen-deficient conditions. Microbial growth and anaerobic RDX biodegradation were effectively enhanced by the co-addition of RDX and starch, which resulted in increased RDX biotransformation to nitroso derivatives at a greater specific degradation rate than those for previously reported anaerobic RDX-degrading bacteria (isolates). The accumulation of the most toxic RDX degradation intermediate (MNX [hexahydro-1-nitroso-3,5-dinitro-1,3,5-triazine]) was significantly reduced by starch addition, suggesting improved RDX detoxification by the co-addition of RDX and starch. The subsequent MiSeq sequencing that targeted the bacterial 16S rRNA gene revealed that the Sporolactobacillus, Clostridium, and Paenibacillus populations were involved in the enhanced anaerobic RDX degradation. These results suggest that these three bacterial populations are important for anaerobic RDX degradation and detoxification. The findings from this work imply that the Sporolactobacillus, Clostridium, and Paenibacillus dominant microbial consortium may be valuable for the development of bioremediation resources for RDX-contaminated environments.
Microbial communities were analyzed in an anaerobic/aerobic sequencing batch reactor (SBR) fed with glucose as a sole carbon source. Scanning electron microscopy (SEM) showed that tetrad or cuboidal packet bacteria dominated the microbial sludge. Quinone, slot hybridization, and 165 rRNA gene sequencing analyses showed that the Proteobacteria beta subclass and the Actinobacteria group were the main microbial species in the SBR sludge. However, according to transmission electron microscopy (TEM), the packet bacteria did not contain polyphosphate granules or glycogen inclusions, but only separate coccus-shaped bacteria contained these, suggesting that coccus-shaped bacteria accumulated polyphosphate directly and the packet bacteria played other role in the enhanced biological phosphorus removal (EBPR). Based on previous reports, the Actinobacteria group and the Proteobacteria beta subclass were very likely responsible for acid formation and polyphosphate accumulation, respectively, and their cooperation achieved the EBPR in the SBR operation which was supplied with glucose.
Recently, an innovative method for wastewater treatment and nutrient removal was developed by combining the sequence batch reactor and membrane bioreactor to overcome pollution caused by shipboard sewage. This system is a modified form of the activated sludge process and involves repeated cycles of mixing and aeration. In the present study, the bacterial diversity and dominant microbial community in this wastewater treatment system were studied using the MACROGEN next generation sequencing technique. A high diversity of bacteria was observed in anaerobic and aerobic bioreactors, with approximately 486 species. Microbial diversity and the presence of beneficial species are crucial for an effective biological shipboard wastewater treatment system. The Arcobacter genus was dominant in the anaerobic tank, which mainly contained Arcobacter lanthieri (8.24%), followed by Acinetobacter jahnsonii (5.81%). However, the dominant bacterial species in the aerobic bioreactor were Terrimonas lutea (7.24%) and Rubrivivax gelatinosus (4.95%).
The sk10 isolated from kimchi was identified as W. kimchii on the basis of l6s-rDNA sequencing. Studies were made to analyze the metabolic flux shift of the sk10 on glucose under aerobic growth conditions. The sk10 produced 38.2 mM acetate, 16.3 mM ethanol, and 33.2 mM lactate under aerobic conditions, but 2.4 mM acetate, 48.0 mM ethanol, and 44.1 mM lactate under anaerobic conditions. The NADH peroxidase (NADH-dependent hydrogen peroxidase) activity of sk10 grown under aerobic conditions was 11 times higher than that under anaerobic conditions. Under the low ratio of $NADH/NAD^+$, the metabolic flux toward lactate and ethanol was shifted to the flux through acetate kinase without NADH oxidation. The kinds of enzymes and metabolites of sk10 were close to those in the pathway of Leuconostoc sp., but the metabolites produced under aerobic growth conditions were different from those of Leuconostoc sp. The stoichiometric balance calculated using the concentrations of metabolites and substrate was about 97%, coincident with the theoretical values under both aerobic and anaerobic conditions. From these results, it was concluded that the metabolic flux of W. kimchii sk10 was partially shifted from lactate and ethanol to acetate under aerobic conditions only.
The morphological characteristics of granules, which were generated in lab-scale sequencing batch reactor (SBR) for simultaneous nitrogen and phosphorus removal with denitrifying phosphorus accumulating organism (dPAO) were identified. Granular sludge was fully developed in the anaerobic-anoxic (An-Ax) SBR after 180 days of SBR operation. The average diameter of granular sludge was 2.2 mm and rod-type organisms dominated in the granules. In addition, about 1.0 mm of white precipitate was observed in the core of the granule, and the material was confirmed that it is very similar to hydroxyapatite $(HAP;\;Ca_5(PO_4)_3(OH))$ by X-ray diffraction) analysis.
The behavior of total organic carbon (TOC) and phosphate were observed for 15 days with immobilized activated sludge using polyacrylamide (PAA) by sequencing batch reactor (SBR). In the preparation of immobilized sludge by PAA, it was found that suitable acrylamide concentration for actual wastewater treatment was to be 15% through the batch test. When SBR system was operated in the repeated aerobic and anaerobic conditions, TOC removal efficiency was 92%. The uptake rate of phosphate was increased from 1.78 mg-P/g cell/hr on the 5th day of acclimation to 2.5 mg-P/g cell/hr on the 15th day of acclimation. And the total phosphorus content in PAA bead was increased from 40 mg-P/g cell on the 1st day of operation to 55 mg-P/g cell on the 15th day of operation. From this study, lowering the volume of aeration tank was possible when PAA bead was used in wastewater treatment and long operation was also possible without the settler.
Various analytical methods such as electron microscopy, quinone analysis, and 16S rDNA sequencing studies were used to investigate the microbial communities and to identify the microorganisms responsible for enhanced biological phosphorus removal (EBPR) in an anaerobic/aerobic sequencing batch reactor (SBR) fed with acetate. Electron photomicrographs showed that oval-shaped microorganisms of about $0.7\;{\sim}\;1\;{\mu}m$ in diameter dominated the microbial sludge. These microorganisms contained polyphosphate granules and glycogen inclusions, which suggests that they are a kind of phosphorus accumulating organism. Quinone and 16S rRNA sequence analyses showed that the members of Proteobacteria beta subclass were the most abundant species, which were affiliated with the Rhodocyclus-likes group. Phylogenetic analysis revealed that the two dominating clones of the beta subclass were most distantly related to Propionivibrio dicarboxylicus DSM 5885 and Rhodocyclus tenuis DSM 109 with about 95% and 96% sequence similarity, respectively. Therefore, it was concluded that the oval-shaped organisms related to the Rhodocyclus-likes group are likely to be responsible for biological phosphorus removal in SBR operation supplied with acetate.
In the study, we Investigated the behavior and removal efficiency of organics, nitrogen. phosphorus with operating conditions in SBRs. Substrate used was synthetic wastewater in which the ratio of $COD_{cr}$. : N : P was 100 : 12 : 2. The cycling the in SBRs was adjusted at 6 hours and 8 hours, and then certainly Included anaerobic and aerobic conditions. Also, for each cycling time. we performed 2 series of experiment simultaneously which was set up 10 days and 20 days as SRT. The removal efficiency of $COD_{cr}$. was over 97% in all operating conditions. In the 6 hours cycling time, the removal efficiency of $PO_4^{3-}-P$ reached almost 100% in steady state. And then we could observe a typical phonemena of phosphorus release and uptake, and the removal efficiency of N was 67%, Residual N source was almost TKN and most of the rest remained as $NO_2-N$. Also the difference in both SRTs was not observed practically. In the 8 hours cycling time, dissolution of sludge appeared. and, $PO_4^{3-}-P was not nearly removed but nitrogen was removed up to 75%, And the residual nitrogen was accumulated as $NO_2^--N$.
To better understand the ecology of tetrade forming organisms (TFOs) floating in a large amount of dairy wastewater treatment plant (WWTP) effluent (sequencing batch reactor [SBR]) during the inefficient phosphorus (P) removal process of an enhanced biological P removal system, the TFOs from the effluent of a full scale WWTP were separated and attempts made to culture the TFOs in presence/absence of oxygen. The intact TFOs only grew aerobically in the form of unicellular short-rods. Furthermore, to identify the intact TFOs and unicellular short-rods the DNAs of both were extracted, analyzed using their denaturing gradient gel electrophoresis (DGGE)-profiles and then sequenced. The TFOs and unicellular short-rods exhibited the same banding pattern in their DGGE-profiles, and those sequencing data resulted in their identification as Acinetobacter sp. The intact TFOs appeared in clumps and packages of tetrade cells, and were identified as Acinetobacter sp., which are known as strict aerobes and efficient P-removers. The thick layer of extracellular polymeric substance surrounding Acinetobacter sp. may inhibit phosphate uptake, and the cell morphology of TFOs might subsequently be connected with their survival strategy under the anaerobic regime of the SBR system.
The rational utilization of crop straw as a raw material for natural gas production is of economic significance. In order to increase the efficiency of biogas production from agricultural straw, seasonal restrictions must be overcome. Therefore, the potential for biogas production via anaerobic straw digestion was assessed by exposing fresh, silage, and dry yellow corn straw to cow dung liquid extract as a nitrogen source. The characteristics of anaerobic corn straw digestion were comprehensively evaluated by measuring the pH, gas production, chemical oxygen demand, methane production, and volatile fatty acid content, as well as applying a modified Gompertz model and high-throughput sequencing technology to the resident microbial community. The efficiency of biogas production from fresh straw (433.8 ml/g) was higher than that of production from straw silage and dry yellow straw (46.55 ml/g and 68.75 ml/g, respectively). The cumulative biogas production from fresh straw, silage straw, and dry yellow straw was 365 l-1 g-1 VS, 322 l-1 g-1 VS, and 304 l-1 g-1 VS, respectively, whereas cumulative methane production was 1,426.33%, 1,351.35%, and 1,286.14%, respectively, and potential biogas production was 470.06 ml-1 g-1 VS, 461.73 ml-1 g-1 VS, and 451.76 ml-1 g-1 VS, respectively. Microbial community analysis showed that the corn straw was mainly metabolized by acetate-utilizing methanogens, with Methanosaeta as the dominant archaeal community. These findings provide important guidance to the biogas industry and farmers with respect to rational and efficient utilization of crop straw resources as material for biogas production.
