고추품종을 달리한 전통식 고추장의 숙성중 미생물, 효소활성 및 맛성분의 변화 (Effect of Red Pepper Varieties on the Microflora, Enzyme Activities and Taste Components of Traditional Kochujang during Fermentation)

  • Microbial counts, enzyme activities and taste components of traditional kochujangs prepared with the powders from 4 different varieties of red pepper, were investigated during 90 days fermentation for the industrial production of traditional kochujang. The viable cell counts of anaerobic bacteria in kochujangs did not change remarkably during fermentation, however, aerobic bacterial counts showed a rapid increase up to 90 days of aging. The yeasts in all kochujang samples increased until 60 days of aging and than decreased. After 90 days of aging, the count of aerobic bacteria in Kumtop kochujang was higher than those of others. The activities of liquefying amylase decreased during the aging, but those of saccharogenic amylase increased at 60 days of aging. The activities of neutral protease were higher than those of acidic protease, and increased during the middle and last period of aging. The major free sugars of kochujang were maltose and glucose, and their contents were higher in Hongkwang kochujang. The major organic acids of kochujang were succinic, formic and citric acid, followed by lactic acid. Succinic acid content in kochujang decreased during fermentation, whereas formic and citirc acids were increased. The major free amino acids were serine, proline, glutamic acid, aspartic acid and alanine. Kumtop kochujang contained the highest amount of total free amino acids. Among the nucleotides and related components in kochujang, cytidine-5-monophosphate was the most abundant component at the begining of aging period, while hypoxanthine increased remarkably during fermentation. Hanwang kochujang was higher in the content of nucleotides than others. Capsaicin contents of kochujang decreased during aging and those of Jangter kochujang was higher than that of others. Sensory evaluation showed that Jangter kochujang was significantly better than Kumtop kochujang in overall acceptability, but there were no appreciable differences in color and flavor.

한국인 분변으로부터 분리된 Bacteroides fragilis Roid 8의 Glycosidase 패턴 (Glycosidase Pattern of Bacteroides fragilis Roid 8 Isolated from a Korean Adult Feces)

  • 인체의 대장은 여러 종류의 균들에 의하여 상재균총이 이루어져 있는데 이들중 혐기성 균들이 주종을 이루고 있다. 이들 혐기성 균들 중 가장 많은 수가 Bacteroides이다. 본 연구에서 한국인으로부터 분리된 Bacteroides fragilis Roid 8은 장내의 다른 혐기성 균주들에 비하여 $N-acetyl-{\beta}-glucosaminidase$, ${\alpha}-fucosidase$, ${\beta}glucuronidase$ chitobiase, PNPCase 등의 활성이 높았다. ${\beta}-galactosidase$, ${\beta}-xylosidase$, ${\alpha}-arabinofuranosidae$활성은 없었고 ${\alpha}-glucosidase$, ${\beta}-glucosidase$, ${\alpha}-galactosidae$ 등의 생산은 Bifidobacteria 에 비하여 낮았다. BHI 기본배지에 여러 종류의 탄수화물을 첨가하여 배양한 뒤 생산된 $N-acetyl-{\beta}-glucosaminidae$, ${\alpha}-fucosidase$, ${\beta}-glucuronidase$ chitobiase, PNPCase, ${\beta}-glucosidase$, ${\beta}-glucosidase$ ${\alpha}-galactosidase$ 활성을 조사한 결과 모두 glucose와 lactose 첨가배지에서 이들 효소들의 활성이 낮았다. 조사된 모든 효소들에 대하여 특이적으로 현저히 생산을 증가시키는 당은 없었다. 이들 8개의 효소에 대하여 최적 pH와 최적온도가 조사되었다.

약선식품소재의 유산균 증식 효과 (Bifidogenic Effects of Yaksun (functional herbal) Food Materials)

  • 1. 약선식품소재의 물추출물 중 pH 저하효과가 가장 큰 것은 B. breve를 배양하였을 경우 Citrus aurantium (진피), Liriipe platyphylla (맥문동), Platycodon grandiflorum (길경), Schizandra chinensis (오미자)였고, 사람의 장내균총을 배양했을 경우는 Angelica gigas (당귀), Hordeum vulgare (맥아), Liriipe platyphylla (맥문동), Platycodon grandiflorum (길경), schizandra chinensis (오미자) 등이었다. 2. 약선식품소재의 $\beta$-glucosidase 효소활성억제효과는 B. breve를 배양했을 경우 Citrus aurantium(진피), Eugenia caryophyllata (정향), Liriipe platyphylla (맥문동), Platycodon grandiflorum (길경), Schizandra chinensis (오미자)가 90%. 이상의 억제효과를 나타내었고 사람의 장내균총에 대해서는 Eugenia caryophyllata (정향), Liriipe platyphylla (맥문동)가 40% 이상의 억제효과를 나타내었다. 3. $\beta$-Glucuronidase 활성을 억제하는 것으로 나타난 약선식품소재는 사람의 장내균총에 대해서는 Cinnamomum cassia(계지), Gardenia jasminoides(치자), Platycodon grandiflorum (길경) 등이였다. 4. 길경으로부터 in vitro에서 유산균증식효과를 나타내는 compound B를 분리하였으며 이 화합물은 기기분석을 한 결과 sucrose임을 밝혀졌으며, 사람의 장내균총에 대한 효과를 측정한 결과 $\beta$-glucosidase활성은 크게 억제하지 못했지만 $\beta$-glucuronidase 및 tryptophnase활성은 각각 49%, 67%의 억제효과를 나타내었다. 또한 사람의 장내우세균인 B. breve를 증식시키는 효과가 있었다.

Effects of Xylanase on Growth and Gut Development of Broiler Chickens Given a Wheat-based Diet

  • To study the working mechanisms for non-starch polysaccharidases to improve the growth performance of broiler chickens, a 21-day feeding trial was conducted. Two dietary treatments were included: 1) wheat diet (the control); 2) wheat+xylanase diet (xylanase, Allzyme PT, Alltech, Kentucky, USA). There were 8 replicates with 8 birds each for each treatment and the experimental diets were given to birds from hatch. Feed intake and body weight were measured on days 7 and 21. At the same ages, samples were taken for the determination of selected groups of luminal and mucosa-associated bacteria, mucosal morphology, brush-border membrane (BBM) bound enzyme activity and ileal nutrient digestibility. The xylanase supplement increased (p<0.05) body weight gain (BWG) and improved feed conversion ratio (FCR) at the end of the experiment but protein and starch digestibilities were not affected (p>0.05) by xylanase. Up to day 7, xylanase increased the counts of C. perfringens in the ileum and total anaerobic bacteria (TAB) in the caeca (p<0.05, p=0.07, respectively). By day 21, the counts of ileal lactobacilli (p<0.05) and TAB (p=0.07) were lower in birds given the xylanase-supplemented diet than in those on the control diet. No significant differences were observed in the counts of mucosa-associated lactobacilli and coliforms between xylanase treatment and the control at both ages. Villus height at the jejunum was not affected (p>0.05) by the supplement but crypt depth at the same site was reduced at day 7. Also, xylanase tended to increase the concentration of BBM protein (p = 0.09) and the specific activity of sucrase (p = 0.07) at day 21.

  • Bifidobacterium spp. is nonpathogenic, gram-positive and anaerobic bacteria, which inhabit the intestinal tract of humans and animals. In breast-fed infants, bifidobacteria comprise morethan 90% of the gut bacterial population. Bifidobacteria spp. are used in commericial fermented dairy products and have been suggested to exert health promoting effects on the host by maintaining intestinal microflora balances, improving lactose tolerance, reducing serum cholesterol levels, increasing synthesis of vitamins, and aiding the immune enchancement and anticarcinogenic activity for the host. These beneficial effects of Bifidobacterium are strain-related. Therefore continued efforts to improve strain characteristics are warranted. in these respect, development of vector system for Bifidobacterium is very important not only for the strain improvement but also because Bifidobacterium is most promising in serving as a delivery system for the useful gene products, such as vaccine or anticarcinogenic polypeptides, into human intestinal tract. For developing vector system, we have characterized several bifidobacterial plasmids at genetic level and developed several shuttle vectors between E. coli and Bifidobacterium using them. Also, we have cloned and sequenced several metabolic genes and food grade selection marker. Also we have obtained bifidobacterial surface protein, which will be used as the mediator for surface display of foreign genes. Recently we have succeeded in expressing amylase and GFP in Bifidobacterium using our own expression vector system. Now we are in a very exciting stage for the molecular breeding and safe delivery system using probiotic Bifidobacterium strains.

Chito-oligosaccharides as an Alternative to Antimicrobials in Improving Performance, Digestibility and Microbial Ecology of the Gut in Weanling Pigs

  • A total of 126 crossbred weanling pigs (average body weight of $6.3{\pm}0.3$ kg) were used to investigate the effect of chito-oligosaccharide (COS) on growth performance, nutrient digestibility, pH of gastro-intestinal tract (GI), intestinal and fecal microflora of young piglets. Pigs were allocated to three dietary treatments based on body weight and gender in a single factorial arrangement. Treatments were control (No COS), T1 (0.2% COS during starter (6-13 kg) and 0.1% COS during grower (13-30 kg) phases, and T2 (0.4% COS during starter (6-13 kg) and 0.3% COS during grower (13-30 kg) phases, respectively. Each treatment had 3 replicates and 14 pigs were raised in each pen. COS is a low molecular weight water-soluble chitosan that can be obtained from chitin of the crab shell after deacetylation with concentrated sodium hydroxide at high temperature and then further decomposition by chitosanase enzyme in the presence of ascorbic acid. For the starter and grower periods, there were no significant differences (p>0.05) in average daily gain (ADG) and feed to gain ratio among treatments. However, during the overall period (6-30 kg), T2 showed better (p<0.05) feed to gain ratio than other treatments. A digestibility study was conducted at the end of grower phase which showed improvement (p<0.05) in DM and crude fat digestibility in T2 over the control. At 25 kg body weight, 6 pigs per treatment (2 per replicate) were sacrificed to determine the effect of diets on pH and microbial count at different sections of the GI tract. The pH of the cecal contents in pigs fed 0.1% COS was higher (p<0.05) than in the other treatments. Total anaerobic bacterial number increased from cecum to rectum in all treatments. The weekly total bacterial counts showed higher (p<0.05) in feces of pigs fed COS than that of untreated pigs at the $8^{th}$ week. The number of fecal E. coli in untreated pigs at $4^{th}$ wk was 7.35 log CFU/g compared to 6.71 and 6.54 log CFU/g in 0.1 and 0.3% COS-treated pigs, respectively. Similarly, at $8^{th}$ wk, fecal clostridium spp. were lower in pigs fed 0.3% COS (5.43 log CFU/g) than in untreated pigs (6.26 log CFU/g). In conclusion, these results indicated that chito-oligosaccharide could improve feed efficiency in young pigs and inhibited the growth of harmful bacteria.

  • CNP 제초제시용(除草劑施用)이 논토양(土壤)의 미생물상(微生物相)에 미치는 영향(影響)을 구명(究明)코져 유기물(有機物) 함량(含量)이 극(極)히 낮은 사양토(砂壤土)를 공시(供試)하여 유기물(有機物)의 종류(種類)를 달리하고 제초제무시용(除草劑無施用) 및 시용조건(施用條件)(1,080ppm)에서 수도무재배(水稻無栽培) 담수상태(湛水狀態)로 초자온실(硝子溫室)에 정치(靜置)하면서 미생물상(微生物相)을 조사(調査)한 결과(結果)는 다음과 같다. 1. 호기성(好氣性) 전세균(田細菌)의 균수(菌數)는 담수후(湛水後) 50일(日)째까지는 제초제시용(除草劑施用)으로 증가(增加)되었으나, 이후(以後)는 현저(顯著)한 차이(差異)를 볼 수 없었다. 유기물(有機物) 종류별(種類別)로는 제초제시용(除草劑施用) 유무(有無)에 관계(關係)없이 볏짚시용시(施用時) 호기성(好氣性) 세균수(細菌數)가 제일 많았고, glucose 시용구(施用區)는 다소(多少) 감소(減少)하는 경향(傾向)이었다. 2. 사상균수(絲狀菌數)는 제초제(除草劑) 및 유기물(有機物) 시용(施用) 유무(有無)에 관계(關係)없이 전담수기간(全湛水期間) $8{\sim}20{\times}10^3$의 수준(水準)으로 유지(維持)되었다. 3. 방선균수(放線菌數)는 제초제시용(除草劑施用)으로 감소(減少)하는 경향(傾向)이 있고 유기물시용(有機物無施用) 및 볏짚시용시(施用時)는 균수(菌數)를 현저(顯著)하게 감소(減少)시켰다. 한편 유기물시용(有機物施用)에 의한 방선균수(放線菌數)의 증가(增加)는 볼 수 없었다. 4. 혐기성(嫌氣性) 전세균(全細菌) 및 gram-음성세균수(陰性細菌數)는 제초제(除草劑) 및 유기물(有機物) 시용간(施用間)에 큰 차이(差異)를 볼 수 없었다. 5. 사상균(絲狀菌)에 대(對)한 세균(細菌)의 비(比)(B/F) 및 방선균(放線菌)에 대(對)한 세균(細菌)의 비(比)(B/A)는 제초제(除草劑) 및 유기물(有機物) 시용시(施用時) 높은 치(値)를 보였다. 6. 방선균(放線菌)의 생리적(生理的) 성질(性質)을 기준(基準)으로 분류(分類)한 결과(結果) 1) 전방선균(全放線菌)에 대(對)한 농갈색(濃褐色) 색소생산장성균(色素生産陽性菌)의 비율(比率)은 제초제시용(除草劑施用)으로 다소(多少) 높았다. 특(特)히 제초제시용(除草劑施用) 유무(有無)에 관계(關係)없이 볏짚시용시(施用時)가 현저(顯著)히 높았다. 2) Cellulase 양성균(陽性菌)의 비율(比率) 역시 제초제무시용(除草劑無施用)에 비(比)해 제초제시용(除草劑施用)으로 높았으나, 유기물(有機物) 종류간(種類間)에는 현저(顯著)한 차이(差異)가 없었다.

정상 신생아의 대변에서 생후 1주일 동안 장내세균총의 변화 (The Changes of Intestinal Normal Flora in Neonates for Seven Days Postnatally)

  • 목 적: 자궁 내에서 태아의 소화기는 무균상태에 있다가 출생 후 신생아기에 점차 장내세균의 집락화가 이루어진다. 출생 초기에 처음으로 나타나는 세균은 Staphylococcus, Enterobacteriaceae, Streptococcus가 있으며, 이후 Bifidobacterium spp.와 같은 혐기성 세균이 나타난다. 이에 저자들은 정상 신생아의 대변에서 생후 1주일 동안 장내세균총의 분포와 변화를 규명하고자 하였다. 방 법: 한림의대 강동성심병원에서 출생한 정상 신생아 15명(자연분만 3명, 제왕절개 12명)(모유수유 1명, 분유수유 8명, 혼합수유 6명)을 대상으로 하였다. 출생 첫 대변과 1일, 3일, 7일째의 대변을 채취하였다. 대변 배양 배지와 조건를 달리하여 호기성균(blood agar plate), E. coli (trypton bile X-glucuronide 배지), 그람 양성 혐기성균(phenylethyl alcohol agar), lactobacillus (MRS 배지), Bifidobacterium spp. (bifidobacterium selective agar), Clostridium difficile (cefoxitincycloserine-fructose agar)을 검출하여 배양된 균 집락의 수를 측정하여 기록하였다. 결 과: 출생 첫 대변에서는 균이 거의 검출되지 않았다. 호기성균, E. coli, 그람 양성 혐기성균은 출생 시부터 생후 7일까지 유의하게 증가하였다(p<0.05). Lactobacilli는 한명의 신생아에서 생후 7일에 검출되었다. Bifidobacterium spp.은 2명에서 검출되었다. Clostridium difficile은 생후 7일까지 검출되지 않았다. 분만 방법과 수유 방법에 따른 균 집락의 차이는 없었다. 결 론: 정상 신생아에서 생후 1주일 동안 장내세균총의 다양한 변화를 관찰할 수 있었다. 앞으로 생후 1주일 이후의 장내세균총의 변화와 더 많은 환아에서의 연구를 진행한다면 생균제 사용에 도움이 되리라 생각된다.

  • 농약(農藥)이 토양(土壤) 미생물(微生物)의 상변화(相變化)에 끼치는 영향(影響)을 구명(究明)하기 위하여 담수토양(湛水土壤) 상태하(狀態下) 수도(水稻)를 이앙(移秧)한 구(區)와 이앙(移秧)하지 않은 구(區)에 살충제(殺蟲劑)인 carbofuran, 제초제(除草劑) pyrazolate 및 pyrazolate+pretilachlor 합제(合劑)를 처리(處理)한 후(後) pH 변화(變化)와 총세균(總細菌), 그람 음성균(陰性菌), 사상균(絲狀菌)등의 수(數)를 경시적으로 조사분석(調査分析)한 결과(結果)는 다음과 같다. 1. pH 변화(變化)는 수도재배(水稻栽培) 약제(藥劑) 처리후(處理後) 시일(時日)이 경과(經過)함에 따라 약간(若干) 낮아지다 6주(週)째에는 다시 상승(上昇)하였고, 수도(水稻)를 재배(栽培)한지 않은 경우(境遇) 약제처리후(藥劑處理後)의 pH는 거의 변화(變化)가 없었다. 2. 공시약제(供試藥劑) 모두 수도재배(水稻栽培) 유무(有無)에 관계(關係)없이 약제처리(藥劑處理) 후(後)의 총세균수(總細菌數)는 대조구(對照區)에 비하여 적었고 처리약량(處理藥量)이 증가(增加)함에 따라 감소(減少)되었으나 시일(時日)이 경과(經過)함에 따라 증가(增加)되었다. 3. 농약처리후(農藥處理後) 6주(週)째까지 그람 음성균(陰性菌)의 수(數)는 대조구(對照區)보다 적었고 시일(時日)이 경과(經過)함에 따라 carbofuran처리구(處理區)의 그람 음성균(陰性菌)의 수(數)는 감소(減少)되었으나 pyrazolate 및 합제처리(合劑處理)의 그람 음성균(陰性菌) 수(數)는 증가(增加)되는 경향(傾向)이었다. 4. 약제처리구(藥劑處理區)의 혐기성(嫌氣性) 세균(細菌)의 수(數)도 대조구(對照區)보다도 적었으나 시일(時日)이 경과(經過)하면서 증가(增加)되었다. 5. 사상균(絲狀菌)의 수(數)는 carbofuran 처리(處理)의 경우(境遇) 대조구(對照區)와 비슷하였으나 pyrazolate와 그 합제처리구(合劑處理區)의 경우(境遇)는 처리농도(處理濃度)가 증가(增加)하면서 4-5주(週)째 까지는 더욱 감소(減少)되다가 6주(週)째 까지는 더욱 감소(減少)되다가 6주(週)째에는 증가(增加)한는 경향(傾向)이었다.

Evaluation of Multi-microbial Probiotics Produced by Submerged Liquid and Solid Substrate Fermentation Methods in Broilers

  • Two experiments were conducted to evaluate multi-microbe submerged liquid (SLF) and solid substrate (SSF) fermented probiotic products in broilers. The SLF and SSF probiotics were comprised of Lactobacillus acidophilus ($1.1{\times}10^9$ and $4{\times}10^8$ cfu/g), Bacillus subtilis ($1.1{\times}10^9$ and $4{\times}10^9$ cfu/g), Saccharomyces cerevisiae ($1.5{\times}10^7$ and $1.0{\times}10^4$ cfu/g) and Aspergillus oryzae ($2.6{\times}10^7$ and $4.3{\times}10^7$ cfu/g), respectively. In Exp. 1, 640 day-old Ross chicks were allotted to 4 treatments, each comprising 4 replicates (40 chicks/replicate). The basal diet was prepared without any antimicrobials (negative control, NC), and 20 mg/kg avilamycin (positive control, PC), 0.3% SLF and 0.3% SSF probiotics were added to the basal diets as treatments. Birds fed PC and SSF diets showed improved (p<0.001) overall weight gain and F/G than birds fed NC and SLF diets; whereas, birds fed SLF diet had better weight gain and F/G than birds fed NC diet. Retention of CP was higher (p<0.05) in birds fed the SSF diet than birds fed PC, SLF and NC diets. Birds fed the SLF diet tended to have higher (p<0.10) cecal total anaerobic bacteria than birds fed PC and NC diets; whereas, lesser cecal coliforms were noticed in birds fed PC, SLF and SSF diets than birds fed the NC diet. In Exp. 2, 640 day-old Ross chicks were randomly allotted to 4 treatments in a $2{\times}2$ factorial arrangement. Each treatment had 4 replicates (40 chicks/replicate). Two different multi-microbe probiotic products (0.3% SLF or SSF) each with two different antibiotics (10 mg/kg colistin, or 20 mg/kg avilamycin) were used as dietary treatments. Birds fed the SSF diet had greater weight gain (p<0.001), better F/G (p<0.05), greater retention of energy (p<0.001) and protein (p<0.05), and lesser cecal Clostridium (d 35) than birds fed SLF diet. Birds fed the colistin-supplemented diet had less (p<0.01) cecal coliforms when compared with birds fed the avilamycin diet. Additionally, birds fed the avilamycin diet had greater energy retention (p<0.05) than birds fed the colistin diet. Thus, the results of this study suggest the multi-microbe probiotic product prepared by a solid substrate fermentation method to be superior to the probiotic product prepared by submerged liquid fermentation; moreover, feeding of probiotics with different antibiotics did not elicit any interaction effect between probiotic and antibiotic.
