Production of Polyhydroxyalkanoates (PHAs) from Sequencing Batch Reactor Using Synthetic Wastewater (연속회분식 처리시스템에서 인공하수를 이용한 Polyhydroxyalkanoates (PHAs)의 생산)

  • The production of polyhydroxyalkanoates (PHAs) using Sequencing Batch Reactor (SBR) was investigated. The experiments were performed in two fabricated SBRs (4 L) of different oxidation state. Synthetic wastewater was used as substrate, using C/N/P ratio of 42:10:1. SBR 1 and SBR 2 were operated in aerobic dynamic feeding (ADF) and anaerobic/oxic dynamic feeding (AODF) condition, respectively. ADF provide feast and famine in aerobic condition, while AODF in anaerobic/oxic condition. PHAs production was found high in AODF than AOF. Maximum PHAs content of 40.0% (w/w)of biomass were produced in AODF mode. Produced PHAs structural and thermal property were good.

  • The total production of food waste was about 11,398ton/day('03) in Korea. Also, food waste was treated by landfill, incineration, reuse and anaerobic digestion. The method of food waste treatment depended primarily on landfill. However, the method of landfill causing social problems was prevented to treat food waste in the first of January 2005.12) Thus, anaerobic digestion is an important method to treat food waste because of possibility of energy recovery as methane gas. In this study, the possibility of food waste treatment containing high organic material and low pH in the one stage anaerobic reactor to save cost and time and energy recovery using $CH_{4}$ gas by the hybrid anaerobic reactor (HAR) was measured. The HAR was designed by combing the merits of the anaerobic filter (AF) to minimize the microorganism shock when food waste of very low pH was injected and up-flow anaerobic sludge blanket (UASB) to prevent from plugging and channeling phenomena by large suspended solids when semi solids were injected. Granule was packed in the section of HAR. The purpose of the BMP experiment was to measure the amount of methane generated when organic material was resolved under anaerobic conditions, to grasp bio resolution of organic material. Total accumulated methane production per VS amount was $0.471(m^{3}/\cal{kg}\;VS)$. So, the value was about $81.2\%$ of theoretical methane production which was $0.58(m^{3}/\cal{kg}\;VS)$ by elementary analysis and organic matter removal velocity (K) was $0.18(d^{-1})$. From these results, food waste was treated by anaerobic treatment. From this study, $CH_{4}$ generation from food waste (11,398 ton/day) could be estimated. By using an energy conversion factor of Braun's study, $5.97KWh/m^{3}\;CH4,\;60\%\;of\;CH_{4}$ gas generation, the amount of total energy producing food waste is to 6,727MWh/day. It could be confirmed that energy recovery using $CH_{4}$ gas was possible. Above these results, food waste containing organic matters of high concentration could be treated in HRT 30 days under an anaerobic condition, using the hybrid anaerobic reactor and reuse of $CH_{4}$ gas was possible.

  • Salmonella typhimurium can encounter a wide variety of environments during its life cycle. In nature, S. typhimurium can experience and survive dramatic acid stresses that occur in diverse ecological niches ranging from pond water to phagolysosomes. These survival mechanism is aquired by the Acid Tolerance Response(ATR) in Salmonella. The ATR of S. typhimurium is a complex inducible phenomenon in which exposures to slight or moderate low pH will produce a stress response capable of protecting the organism against more severe acid challenges. ATR in Salmonella has two different systems that are called RpoS dependent and independent. We found that ATR in anaerobic was showed RpoS independent because rpoS$\Omega$AP had ATR as S. typhimurium UK1. Using the P22 MudJ(Km, lacZ) operon fusion technique and a lethal selection procedure combining low pH(pH4.5) and sodium acetate(10mM, pH4.5), we isolated LF487 aatA::MudJ which showed acid sensitive in anaerobic condition. aatA locus was determined at 12 min on Salmonella Genetic Map. The survival rate of aatA mutant was showed significantly diminished at pH4.3 than virulent wild type Salmonella in anaerobic condition(5% $CO_2$, 5% H$_2$, 90% $N_2$). Therefore isolated gene was confirmed important gene for anaerobic ATR system.

The Influence of Ammonium-Nitrogen on Anaerobic Microorganisms in Swine Wastewater by Batch-Fermentation. (혐기성 회분식 배양에서 양돈폐수의 NH$_4$-Nitrogen이 혐기성 미생물에 미치는 영향)

  • This study presents the influence of ammonium-nitrogen on microorganisms in swine wastewater. For the anaerobic batch fermentation, two different methods were used. One is the dilution of wastewater with water. The other method is the elimination of ammonium-nitrogen from the wastewater. By addition of MgO into wastewater, non-soluble crystall was formed under alkaline condition as MgNH$_4$PO$_4$6$H_2O$ (MAP). The master culture was adapted in swine wastewater for more than 3 months, in water-dilution method, the dilution of wastewater with 25% water gave us the best result in efficiency of COD removal. Two hundred hours later MAP-treated wastewater showed the efficiency of the COD removal more than 80%. Under same condition obtained none MAP-treated wastewater about 50%. MAP treatment carried out the very effective anaerobic digestion with swine wastewater. The important result in this study is that the low ratio of C:N influenced on anaerobic microorganisms more than high concentration of ammonium nitrogen in swine wastewater. The struvite for the crystallforming has no toxic effect on methanogenic bacteria.

Probabilistic Models to Predict Listeria monocytogenes Growth at Low Concentrations of NaNO2 and NaCl in Frankfurters

  • This study developed probabilistic models to describe Listeria monocytogenes growth responses in meat products with low concentrations of NaNO2 and NaCl. A five-strain mixture of L. monocytogenes was inoculated in NBYE (nutrient broth plus 0.6% yeast extract) supplemented with NaNO2 (0-141 ppm) and NaCl (0-1.75%). The inoculated samples were then stored under aerobic and anaerobic conditions at 4, 7, 10, 12, and 15℃ for up to 60 d. Growth response data [growth (1) or no growth (0)] for each combination were determined by turbidity. The growth response data were analyzed using logistic regression to predict the growth probability of L. monocytogenes as a function of NaNO2 and NaCl. The model performance was validated with the observed growth responses. The effect of an obvious NaNO2 and NaCl combination was not observed under aerobic storage condition, but the antimicrobial effect of NaNO2 on the inhibition of L. monocytogenes growth generally increased as NaCl concentration increased under anaerobic condition, especially at 7-10℃. A single application of NaNO2 or NaCl significantly (p<0.05) inhibited L. monocytogenes growth at 4-15℃, but the combination of NaNO2 or NaCl more effectively (p<0.05) inhibited L. monocytogenes growth than single application of either compound under anaerobic condition. Validation results showed 92% agreement between predicted and observed growth response data. These results indicate that the developed model is useful in predicting L. monocytogenes growth response at low concentrations of NaNO2 and NaCl, and the antilisterial effect of NaNO2 increased by NaCl under anaerobic condition.

  • Yoghurt was prepared with Bifidobacterium longum TK-100 and Lactobacillus acidophilus TK-2070. The prepared yoghurt showed the increase of the titratable acidity under cold storage condition. This was derived from the active L. acidophilus TK-2070 on the logarithmic phase rather than from the B. longumn TK-2070. B. longum TK-100 grew well in the facultative anaerobic condition as well as in the strict anaerobic condition. Reinforced clostridial agar medium with 0.1% aniline blue was tried for the differential viable cell counts in the mixed culture and in the yoghurt. B. longurn TK-2070 had the light gray, blue-dotted colonies of about 2 mm diameter. L. acidophilus TK-2070 had the light gray colonies of about 1 mm diameter.

  • Korean green tea leaves which were harvested three times(May, June, August) were treated with anaerobic conditions and were measured changes of ${\gamma}-aminobutyric$ acid(GABA) and other constituents. In anaerobically treated green tea leaves, the content of ${\gamma}-aminobutyric$ acid(GABA) and alanine increased while glutamic acid decreased. Whereas theanine, arginine, caffeine and tannin showed little change and the content of vitamine C slightly decreased with the passing of the anaerobic treatment time. Formation of GABA, a hypotensive constituents, was proportioned to the content of glutamic acid and the optimum time of the anaerobic treatment was about 12 hours. In the anaerobic treatment of green tea leaves, effect of nitrogen gas and vacuum condition was no difference between two.

Hypoglycemic Effect of Mulberry Leaves with Anaerobic Treatment in Alloxan-induced Diabetic Mice (혐기처리한 뽕잎의 혈당강하 효과)

  • The hypoglycemic effect of mulberry leaves using alloxan-induced diabetic mice was investigated. The hypoglycemic effect of mulberry leaves was significantly affected with anaerobic incubation up to for 6 hrs. The body weight of mice treated with 20% anaerobic conditioned mulberry leaves was higher than that in the control group. Mulberry leaves which were treated with anaerobic condition significantly decreased blood glucose level in alloxan-induced type I diabetes mice. Furthermore, glucose tolerance was significantly recovered by feeding mulberry leaves anaerobically treated. Pathological analysis revealed that the disappearance of insulin-secreting beta-cells in the islets of Langerhans from the alloxan induced diabetic mice was strongly inhibited in the mulberry leaves feeding group. The content of 1-deoxynojirimycin increased up to 5% in detached mulberry leaves incubated under nitrogen gas for 6 hrs. Nevertheless, it is not clear whether the synthesis of 1-deoxynojirimycin is increased or the degradation of it is decreased. Overall, results obtained from this study indicate that the hypoglycemic effect was increased by the mulberry leaves with anaerobic treatment.

Molecular Analysis of the Salmonella Typhimurium tdc Operon Regulation

  • Efficient expression of the Salmonella Typhimurium tdc ABCDEG operon involved in the degradation of L-serine and L-threonine requires TdcA, the transcriptional activator of the tdc operon. We found that the tdcA gene was transiently activated when the bacterial growth condition was changed from aerobic to anaerobic, but this was not observed if Salmonella was grown anaerobically from the beginning of the culture. Expression kinetics of six tdc genes after anaerobic shock demonstrated by a real-time PCR assay showed that the tdc CDEG genes were not induced in the tdcA mutant but tdcB maintained its inducibility by anaerobic shock even in the absence of tdcA, suggesting that an additional unknown transcriptional regulation may be working for the tdcB expression. We also investigated the effects of nucleoid-associated proteins by primer extension analysis and found that H-NS repressed tdcA under anaerobic shock conditions, and fis mutation delayed the peak expression time of the tdc operon. DNA microarray analysis of genes regulated by TdcA revealed that the genes involved in N-acetylmannosamine, maltose, and propanediol utilization were significantly induced in a tdcA mutant. These findings suggest that Tdc enzymes may playa pivotal role in energy metabolism under a sudden change of oxygen tension.
