Characteristics of microalgal growth was investigated using anaerobic effluent from two-phase animal waste digestor as substrate. Batch experiments were carried out to investigate the effect of the initial nitrogen and phosphorus concentrations on growth of Microcystis aeruginosa, Chlorella sp. and Euglena gracilis. In 400 times diluted anaerobic effluent (TN 3 mg/L), single cell growth of the Euglena gracilis population increased twice without delay, although Chlorella sp. and Microcystis aerugenos take over 144 hours. Similar appearance with single cell growth was observed in mixed cultures. However, microalgae population did not increase under condition of 10 times diluted influent (TP 3 mg/L) in both pure and mixed cultures, which was affected by high organic and nitrogen concentration. Logistic growth model successfully fitted to determine biokinetic parameters such as ${\lambda}$: lag time, ${\mu}m$: maximal specific growth rate, A: asymptote of growth.

The influence of mixing on biogas production and organic material removal performance of solid state anaerobic digestion of dairy manure and sawdust bedding mixtures was evaluated using 22 L volume lab-scale digesters. After 45 days of anaerobic digestion at $37^{\circ}C$, cumulative methane yield of unmixed test unit ($73.1N{\cdot}mL/g-VS$) was almost 1.3 times of that of mixed one ($56.3N{\cdot}mL/g-VS$). The biodegradable volatile solids removal rate of unmixed test unit was 67%, which was almost 28% greater than mixed one. Our results reveal that unmixed condition is better than mixed one in terms of biogas production and organic material reduction.
Characteristics of the upflow anaerobic filter process have been studied with six other conditions. When nitrate was mixed with influent in the bottom of the reactor, removal efficiencies of TBOD and TCOD were lower than those of TBOD and TCOD when nitrate was injected to the side of the reactor. In addition, when nitrate was injected to the side of the reactor the concentration of volatile acids of effluent was not high and ORP of effluent was lower than the mixture when nitrate was mingled with influent. It means that the bottom of the anaerobic filter played an important role in making volatile acids, methane production, and denitrification. Moreover, percentage of methane in the gas increased in accordance with increasing nitrate injection. It was because there were a lot of methane producing microorganisms which would rather use hydrogen than acetate. This reactor condition gets unstable due to provide nitrate. Therefore, higher hydrogen Pressure, shorter generation time, and lower standard Gibb's free energy gave great portion of methane of gas.
In order to isolate a Fe(III)-reducer from the natural environment, soil samples were collected from various patty fields and enriched with ferric citrate as a source of Fe(III) under anaerobic condition. Since the enrichment culture was serially performed, the Fe(III)-reduction activity was serially diluted and cultivated on an agar plate containing lactate and ferric citrate in an anaerobic glove box. A Gram negative, motile, rod-shaped and facultative anaerobic Fe(III)-reducer was isolated based on its highest Fe(III)-reduction activity, Bacterial growth was coupled with oxidation of lactate to Fe(III)-reduction, but the isolate fermented pyruvate without Fe(III), The isolate reduced an insoluble ferric iron (FeOOH) as well as a soluble ferric iron (ferric citrate). Using the BBL crystal enteric/non-fermentor identification kit and 16S rDNA sequence analysis, the isolate was identified as Shewanella putrefaciens IR-1.
The production of microbiologically enriched cultures that degrade cis- 1,2-dichloroethylene(DCE) under anaerobic conditions was investigated. Among 80 environmental samples, 19 displayed significant degradation of $10{\mu}M$ cis-DCE during 1 month of anaerobic incubation, and one sediment sample collected at a landfill area (Nanji-do, Seoul, Korea) showed the greatest degradation ($94\%$). When this sediment culture was subcultured repeatedly, the ability to degrade cis-DCE gradually decreased. However, under Fe(III)-reducing conditions, cis-DCE degradation by the subculture was found to be maintained effectively. In the Fe(III)-reducing subculture, vinyl chloride (VC) was also degraded at the same extent as cis-DCE No accumulation of VC during the cis-DCE degradation was observed. Thus, Fe(III)-reducing microbes might be involved in the anaerobic degradation of the chlorinated ethenes. However, the subcultures established with Fe(III) could function even in the absence of Fe(III), showing that the degradation of cis-DCE and VC was not directly coupled with the Fe(III) reduction. Consequently, the two series of enrichment cultures could not be obtained that degrade both cis-DCE and VC in the presence or absence of Fe(III). Considering the lack of VC accumulation, both cultures reported herein may involve interesting mechanism(s) for the microbial remediation of environments contaminated with chlorinated ethenes. A number of fermentative reducers (microbes) which are known to reduce Fe(III) during their anaerobic growth are potential candidates involved in cir-DCE degradation in the presence and absence of Fe(III).
Purpose: In the process of anaerobic digestion, stirring of the digester and feeding of organic waste into the digester have been considered important factors for digestive efficiency. The objective of this study was to determine the most appropriate conditions for both stirring interval of the digester and organic feeding frequency in order to improve anaerobic digestion performance. Methods: A 5-L anaerobic digester was used to conduct continuous batch tests to process swine manure and food waste. Four different stirring intervals of the digester were used: 5 min/h, 10 min/2 h, 15 min/3 h, and 20 min/4 h. Results: The application of swine manure to the digester every 5 min/h resulted in the highest production of biogas as well as the highest removal rates of volatile solids (VS) and total chemical oxygen demand. Stirring the digester with a mixture of swine manure and food waste at intervals of 5min/h and 10min/2 h produced the highest biogas yields of 515.3 mL/gVS and 521.1 mL/gVS, respectively. To test different supply frequencies, organic waste was added to the digester in either a 12-hor 24-h cycle. The 24-h cycle produced 1.5-fold greater biogas production than that during the 12-h cycle. Conclusions: Thus, from the above results, to optimize anaerobic digestion performance, the ideal stirring condition must be 5min/h for swine manure feeding and 10min/2h for co-digestion of food waste and swine manure in a 24-h cycle.
This study was aimed to behaviour of organics and nitrogen on the upflow anaerobic reactor when a acid fermenter is added. Up flow anaerobic reactor (UAR) reaction will result which operates, COD removal efficiencies of reactor with nitrate loading rate 0.11, 0.66g/L/d were over 77%, but one with 1.0g/L/d was 73.5%. Especially, on NLR 0.11g/L/d, COD removal was 77% and nitrate removal efficiency was 93% simultaneously. The other side upflow anaerobic reactor and acid fermenter (UAR+AF) reaction will result witch operates, COD removal efficiencies of reactor with nitrate loading rate 0.11, 0.66g/L/d were over 85%, but one with 1.0g/L/d was 80%. Especially, on NLR 0.11g/L/d, COD removal was 85% and nitrate removal efficiency was 98% simultaneously. Also, without in reaction condition increase of influent nitrate concentration resulted in the linear decrease of nitrate removal efficiency and nitrate removal efficiency at influent nitrate-nitrogen 800mg/L was 50%. Alkalinity was increased theoretically by denitrification at low nitrate-N concentration, however, it was not increased theoretically at high nitrate-No 40% nitrate-N of UAR was denitrified until 70% height of reactor and 90% nitrate-N of UAR+AF was denitrified until 30% height of reactor Upflow anaerobic reactor was to occur accumulate acid, which TVA/Alkalinity is 0.3$\sim$0.47. Direct-seeding cultivation of rice is increasing in Asia instead of transplanting system, because of its lower cost and operational simplicity. Low-temperature germinability (LTG) and anaerobic germinability (AG) are important characters for breeding of varieties for wide-spread adoption of direct-seeding cultivation in rice. This study was performed to characterize LTG and AG of seven rice varieties and identify varieties with strong germinability on both low-temperature and anaerobic conditions. The mean germination rate and germination vigor of seven varieties were 51.7% and 6.0 under low-temperature condition, respectively. Among these varieties, Cheongcheongjinmi and Hwanggeumnodeul had the highest germination rate of 80%, indicating that Cheongcheongjinmi and Hwanggeumnodeul have a good LTG. In anaerobic conditions, the germination rate and coleoptile length for all varieties were 47.6% and 3.2 cm, respectively. Of them, the highest germination rate and coleoptile length were observed in Subo and Hopum, respectively, suggesting that these two varieties are tolerant to anaerobic during germination stage. Molecular characterization by SDS-PAGE revealed that the protein patterns differed at 50 kDa, 40 kDa, and 22 kDa between low-temperature and anaerobic conditions. Varieties identified as good LTG or AG in this study may be used for developing new direct-seeding rice cultivars through pyramiding these traits in the breeding program.
Anaerobic digester supernatant including high concentrations of nitrogen is recycled to water treatment line and make pollutant load increase in municipal wastewater treatment plant(MWTP). To treat nitrogen in anaerobic digester supernatant is suggested the method of MWTP retrofit. In this study, the lab scale reactor was operated about 200 days using supernatant of anaerobic digester. The results could draw operation condition that ammonium nitrogen removal efficiency more than 90% and nitrite conversion efficiency over 70%. Correlation between operation efficiency and operation factors was analyzed based on the operation results. Ammonium nitrogen remove efficiency and nitrite conversion efficiency were related to solid retention time (SRT), ammonium nitrogen load and ammonium nitrogen loading per unit mixed liquer suspended solid (MLSS). Results of this study can be used effective data on nitritation of supernatant of anaerobic digester, and be expected to increase availability of nitritation.
Leachate quality and methane emission from pilot-scale lysimeters operated under semi-aerobic and anaerobic conditions were monitored for 650 days. Two semi-aerobic lysimeters were filled with un-compacted and compacted municipal solid wastes whereas two anaerobic lysimeters containing compacted wastes were operated with leachate storage at 50% and 100% of waste height, respectively. Despite having high moisture in wastes and operating under tropical rainfall events, leachate stabilization in semi-aerobic lysimeters took place much faster resulting in BOD reduction by 90% within 60 days, significantly shorter than 180-210 days observed in anaerobic lysimeters. Nitrogen concentration in leachate from semi-aerobic lysimeter could be reduced by 90%. In term of gas emission, semi-aerobic lysimeter with un-compacted wastes had much lower methane emission rate of $2.8g/m^2/day$ compare to anaerobic lysimeters ($62.6g/m^2/day$) through seasonal fluctuation was observed. Nevertheless, semi-aerobic lysimeter with waste compaction has similar performance to anaerobic lysimeter.
