NDMA(N-nitrosodimethylamine) Removal Uising Membrane at Aerobic and Anaerobic Conditions (호기/혐기 조건에서 Membrane을 이용한 NDMA(N-nitrosodimethylamine)제거)

  • Recently, the interest in NDMA(N-nitrosodimethylamine) has increased due to its recognition as a pollutant by Ontario Ministry of Environment and Energy and California Department of Health Sciences. It is, in fact, one of the DBPs(Disinfection By-products) which appears due to chlorination and is reported to be fatal if exposed continuously to human body. Due to uncertainty in mechanism to remove it, its treatment is not yet carried out. In this experiment, treatment of biological NDMA is carried out by letting it adsorbed on Granular Sludge and then filtering the medium through MF(Microfiltration) and UF(Ultrafiltration) membranes. Granular Sludge is adapted to aerobic and anaerobic conditions for 7 days and the experimental conditions are MLSS of 8000mg/L, COD of 250mg/L, TN of 12.5mg/L, and TP of 2.5mg/L. Several batch tests were carried out and samples were collected with the interval of 1 hour. Samples were measured by LSC(Liquid scintillation counter) after filtering by MF and UF. In batch test with granular sludge the permeate concentrations(removal efficiencies) of NDMA by MF and UF were 71.7ng/L(32.0%) and 62.0ng/L(43.7%) at aerobic state, and 52.0ng/L(49.2%) and 47.6ng/L(58.9%) at anaerobic state, respectively. Hence, UF membrane showed about 10% more removal efficiency than MF and removal efficiency at anaerobic condition was 15% more than that at aerobic condition.

A comparative study on efficiency in the sulfate -added anaerobic landfill site and the semi-aerobic landfill site for the inhibition of methane genration from a landfill site (매립지의 메탄 발생억제를 위한 황산염 첨가형매립지 및 준호기성 매립지의 효율 비교에 대한 연구)

  • This study aims to observe the inhibition of methane generation, the decomposition of organic matter, and the trend of outflowing leachate, using the simulated column of the anaerobic sanitary landfill structure of sulfate addition type which is made by adding sulfate to a current anaerobic landfill structure, and the simulated column of semi-aerobic landfill structure in the laboratory which is used in the country like Japan in order to inhibit methane from a landfill site among the gases caused by a global warming these days, and at the same time to promote the decomposition of organic matter, the index of stabilization of landfill site. As a result of this study, it is thought that the ORP(Oxidation Reduction Potential) of the column of semi-aerobic landfill structure gradually represents a weak aerobic condition as time goes by, and that the inside of landfill site is likely to by in progress into anaerobic condition, unless air effectively comes into a semi-aerobic landfill structure in reality as time goes by. In addition, it can be seen that the decomposition of organic matter is promoted according to sulfate reduction in case of $R_1$, a sulfate-added anaerobic sanitary landfill structure, and that the stable decomposition of organic matter in $R_1$ makes a faster progess than $R_2$. Moreover it can be estimated that $R_1$, a sulfate-added anaerobic sanitary landfill structure has an inhibition efficiency of 55% or so, compared with $R_2$, a semi-aerobic landfill structure, in the efficiency of inhibiting methane.

  • Simultaneous removal of phosphorus and nitrogen from wastewater was studied by the anaerobic-aerobic system of activated sludge. In the anaerobic stage, most of the influent glucose was removed and orthophosphate was released, when the nitrate and/or nitrite concentration in the wastewater was almost zero. The amount of the released phosphorus was found to be directly proportional to the amount of the removed glucose. When the ratio of phosphorus to glucose in the influent was less than 0.04, the phosphorus in the wastewater was almost completely removed during the aerobic state. Under the anaerobic condition, activated sludge released phosphate and excess removal of phosphate occurred during the aerobic condition. Namely, the stress received in anaerobic period stimulated the uptake of phosphorus in aerobic period. The amounts of phosphorus release in the anaerobic and uptake in the aerobic stage were less in proportional to the concentration of $NO_x-N$. Further, if the initial ratio of $NO_2-N$/glucose was less than 0.37, the inorganic nitrogen in the influent could be completely removed.

Effect of Nitrogen-Load Condition on Hydrogen Production and Bacterial Community in Continuous Anaerobic Hydrogen Fermentation Process

  • Effect of nitrogen-load condition on hydrogen ($H_2$) production and bacterial community in a continuous anaerobic hydrogen fermentation were investigated. The slight $H_2$ production on extremely low nitrogen-load condition (C/N ratio: 180) at the start-up period. The highest $H_2$ production was obtained when the C/N ratio was 36, the $H_2$ production yield ($mol-H_2/mol-glucose$) reached to 1.7, and it was indicated that Clostridium pasteurianum mainly contributed to the $H_2$ production. The $H_2$ production was decreased on both the lower (C/N: 72) and higher (C/N: 18) nitrogen-load conditions. The excess nitrogen-load was not always suitable for the hydrogen production. The fluctuation of $H_2$ production seemed to be caused by a change in the bacterial community according to the nitrogen-load condition, while a recovery of $H_2$ productivity was possible by a control of nitrogen-load condition through the bacterial community change. When the nitrogen-load condition was not suitable for hydrogen production, the lactic acid concentration was increased and also lactic acid bacteria were definitely detected, which suggested that the competition between hydrogen fermentator and lactic acid producer was occurred. These results demonstrated that the nitrogen-load condition affect on the $H_2$ productivity through the change of bacterial community in anaerobic hydrogen fermentation.

Effects of Temperature in Anaerobic Nitrogen Removal Process from Piggery Waste : Activities in Ranges of Low Field-temperature (돈사폐수의 혐기성 질소제거에 있어서 온도의 영향 : 낮은 현장 온도범위에서의 활성)

  • ANAMMOX (Anaerobic ammonium oxidation) reactor, which was cultivated ANAMMOX bacteria in mesophilic condition ($35^{\circ}C$), was operated to investigate the effects of temperature. In $20{\sim}30^{\circ}C$ of operation condition, which was assumed as field-temperature, total N removal and $NH_4-N$ removal rate were declined from about 2.50 and $1.27kg\;N/{m^3}_{reactor}-day$ (0.06 and 0.03 kg N/kgVSS/day) to 1.62 and $0.41kg\;N/{m^3}_{reactor}-day$ (0.04 and 0.01 kg N/kgVSS/day), In this range of temperature, ANAMMOX had very low activities but acid fermentation bacteria and denitrifiers, which were competitors of substrates, had high activities relatively. Though operation temperature was higher than inhibition condition for two months, ANAMMOX activities could not been recovered once they were inhibited by low temperature. This fact was resulted from very slow doubling time of ANAMMOX bacteria. This study shows that maintenance device of optimal temperature is necessary required in field application of ANAMMOX.

Survivals of Lactic Acid Bacteria and its Characteristics under the Acidic and Anaerobic Condition (혐기적 산성조건하에서 젖산균의 생존과 그 특성)

  • We investigated the survival, $\beta$-galactosidase activity and cellular permeability of lactic acid bacteria such as Lactobacillus acidophilus ATCC 4356, Lactobacillus casei subsp. casei IFO 3533, Streptococcus thermophilus KCTC 2185, Lactobacillus delbrueckii subsp. lactis ATCC 4797, and Lactobacillus delbrueckii subsp. bulgaricus ATCC 11842 in anaerobic condition of pH 1.5-3.5 range. Numbers of all tested viable cells did not decrease at pH 3.5, but decreased rapidly at pH 1.5 and pH 2.5 during 2 hour incubation at modified EG medium. Immediately after 2 hour incubation, the decrease in population at pH 1.5 and pH 2.5 was about 6-8 and 5-7 log cycles/ml, respectively. Lactobacillus acidophilus ATCC 4356 showed the higest survival of all tested bacteria. The $\beta$-galactosidase activity from Lactobacillus delbrueckii subsp. bulgaricus ATCC 11842 and Streptococcus thermophilus KCTC 2185 decreased rapidly at pH 1.5 and 2.5, but there was a little decrease at pH 3.5. The cellular permeability that was measured by the leakage of intracellular materials increased with decrease of pH. These results suggest that the ingested lactic acid bacteria may be destroyed in contact with low pH of gastric acid.

  • There are many of anaerobic culture methods and equipments for isolation and cultivation of anaerabic bacteria, but most of these methods are used without pre-reduced media. Gas-jet method is a recommend. able method for the culture of anaerobes, resently developed. Bacteriological studies were experimented of liver abscess of cattle by the use of gas. jet method. The results were summarised as follows; 1. Gas-jet method for anaerobic culture are expedient for the making of pre-reduced media, maintaining of oxygen free condition in the culture tube, picking of bacteria from colony and colony counting etc. 2. A 121 strains of facultative anaerobic and anaerobic bacteria were isolated from liver abscess of 27 head of cattle, and the isolated anaerobic bacteria were as follows. Peptostreptococcus spp. 7 strains Acid aminococcus fermentans 1 Veillonella spp. 1 Bacterioides spp. 6 Bifidobacterium spp. 4 Arachinia propionica 2 Lactobacillus spp. 4 Propionibacterium acnes 1 3. Liver abscess were infected with many of bacteria, about $10^3-10^9$ numbers per gram of abcessed tissue. Almost of abscess were mixed infection of various bacterial species rather than simple species.

Regulatory Mutations for Anaerobic Inducible Gene Expression in Salmonella typhimurium

  • New regulatory, loci which participate in the regulation of anaerobic inducible gene expression in Salmonella typhimurium were identified. We observed the regulatory network of new regulator mutations to various anaerobic inducible gene (1). Some anaerobic inducible lac fusions were also induced at low pH condition which was severe environment to withstand for its virulence at the place like phagolysosome. Sic oxygen-regulated regulatory mutants (oxr) isolated by Tn10 mutagenesis were divided into two groups. Five of them were found to show negative effect on the regulation of anaerobic gene expression, while on e showed positive effect on the regulation. Genetic loci of four oxr were identified with 54 Mud-P22 lysogens covering the whole chromosome of S. typhimurium, in the nearby region of map unit 87 min (oxr101), 63 min (oxr104), 97 min (oxr 105), and 57 min (oxr 106), respectively. Two oxr mutants were subjected to two-dimensional polyacrylamide electrophoretic analysis of anaerobic inducible proteins for searching the control circuitry of our oxr mutants.

Anaerobic Degradation of Petroleum Hydrocarbons in Soil by Application of a Digestion Sludge (소화슬러지를 이용한 토양 내 석유계 탄화수소의 혐기성 분해)

  • Anaerobic degradation of petroleum hydrocarbons in a soil artificially contaminated with 10,000 mg/kg soil of diesel fuel was tested by adding an anaerobic sludge taken from a sludge digestion tank. Treatments of soil(50 g) with 15 mL/kg soil and 30 mL/kg soil of the digestion sludge(2,000 mg/L of vss(volatile suspended solids)) showed 37.2% and 58.0% of total petroleum hydrocarbons(TPH) removal during 90 days incubation, respectively. In evaluation of several anaerobic conditions including nitrate reducing, sulfate reducing, methanogenic, and mixed electron accepters condition, treatments with the digested sludge showed significant degradation of diesel fuel under all anaerobic conditions compare to a control treatment of soil without the sludge and a treatment of autoclaved soil treatment with autoclaved digestion sludge. The rate of diesel fuel degradation was the highest in the treatment with the sludge and mixed electron accepters (75% removal of TPH) for 120 days incubation followed in order by sulfate reducing, nitrate reducing, methanogenic condition as 67%, 53%, 43%, respectively. However, the removal rate of non-biodegradable isoprenoid was the highest in the sulfate reducing condition. These results suggest that anaerobic degradation of diesel fuel in soil with digested sludge is effective for practical remediation of soil contaminated with petroleum hydrocarbons.

  • The behaviors of inorganic nitrogen derived from solid animal waste in soil has been received too much concern partly because nitrate which occurred from nitrification can act as a pollutant to soil and groundwater and partly because the loss of nitrogen from surface soil by downward movement of water is disadvantageous in the view of plant nutrient. This present study was conducted to get fundamental imformations on nitrogen behavior and to provide improved basical concepts on the management of animal waste. Fresh or fermented pig manure was mixed with a sandy loam soil in the ratio of 2:1(soil:pig manure), packed into test tube and incubated at $30^+/-1^{\circ}C$ for 8 weeks under aerobic- or anaerobic condition. Sample tubes were taken at the one week interval and analyzed on pH, the amount of $CH_4$ produced under anaerobic condition and inorganic nitrogen. The pH of soil treated with fresh pig manure under anaerobic condition was lowered by 1.87 unit compared to that of under aerobic condition, but at the treatment with fermented pig manure, pH change was very little between aerobic and anaerobic condition. The coefficients of regressional equations which were obtained from pH and incubation time were -0.114 in fresh pig manure and -0.089 in fermented pig manure, and the extent of pH decrease due to incubation was greater in fresh pig manure than that of fermented pig waste. No differences in the amounts of $CH_4$ produced under anaerobic condition between fresh and fermented pig manure was observed until 3 weeks of incubation, however, after that the amount of $CH_4$ produced in fresh pig manure was abruptly increased and cumulative amont of $CH_4$ was reached 8.6 mole/g. K values on $CH_4$ production in fresh and fermented pig manure was 0.211 mole/g/day and 0.046 mole/g/day, respectively, for 5 weeks from the 3rd to the 8th week. $NH_4-N$ concentration at aerobic condition with fresh pig manure treatment was lowered by passing time of incubation, but $NO_3-N$ concentration was elevated from 11.2 ppm at initial state to 67.3 ppm after incubation and this trend on $NH_4-N$, $NO_3-N$ concentration was very similar to the treatment of fermented pig manure. While $NH_4-N$ concentration under anaerobic condition was greatly increased. $NO_3-N$ concentartion was not only very low but also no great changes, that was ranged from 4 to 8 ppm.
